Enhancing extraction of shikonin and phenolic antioxidants from Echium italicum L. using ultrasound and response surface methodology: optimizing temperature, time, and liquid–solid ratio

Abstract:Shikonin, an important secondary metabolite present in the roots of Echium italicum , holds significant importance in the pharmaceutical and cosmeceutical sectors. This study aimed to optimize the extraction of shikonin, DPPH scavenging activity, total phenol content (TPC), and total flavonoid content (TFC) from E. italicum roots in ultrasound-assisted extraction (UAE) parameters such as temperature (30 to 60 °C), time (15 to 45 min), and liquid–solid ratio (10–0.2 to 1 mL/g), utilizing response surface methodology (RSM). The results indicated that the effect of a quadratic polynomial model was significant for all responses. The optimal conditions for the independent variables were determined to be a temperature of 41.84 °C, an extraction time of 45 min, and a liquid–solid ratio of 10–0.78 mL/g, resulting in the optimizing extraction of DPPH scavenging activity (79.64%), TPC (90.37 mg gallic acid equivalent (GAE) g −1 ), TFC (28.38 mg quercetin (Qu) g −1 ), and shikonin content (40.24 mg g −1 ), which aligned with the predicted value. In general, this result affirms the benefit of RSM with UAE in increasing shikonin and phenolic antioxidants, which could be used in the pharmaceutical and food industries.
