Abstract:<p>Although it is widely accepted that human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) are readily available, robustly reproducible, and physiologically appropriate human cells for clinical applications and research in the cardiovascular field, hiPSC-CMs cultured in vitro retain an immature metabolic phenotype that limits their application, and little is known about the underlying molecular mechanism controlling mitochondrial metabolic maturation during hiPSCs differentiation into cardiomyocytes. In this study, we found that peroxisome proliferator-activated receptor γ coactivator-1α (PGC-1α) plays an important role in inducing mitochondrial biogenesis and establishing oxidative phosphorylation (OXPHOS) during the cardiac differentiation of hiPSCs. Knocking down PGC-1α by siRNA impaired mitochondrial respiration, while upregulating PGC-1α by ZLN005 promoted mitochondrial biosynthesis and function by regulating the expression of downstream genes involved in mitochondrial dynamics and oxidative metabolism in hiPSC-CMs. Furthermore, we found that estrogen-related receptor α (<em>ERRα</em>) was required for the induction of PGC-1α stimulatory effects in hiPSC-CMs. These findings provide key insights into the molecular control of mitochondrial metabolism during cardiac differentiation and may be used to generate more metabolically mature cardiomyocytes for application.</p>

What problem does this paper attempt to address?
### The Problem the Paper Attempts to Solve This paper aims to address the issue of mitochondrial metabolic immaturity during the differentiation of human induced pluripotent stem cells (hiPSCs) into cardiomyocytes (hiPSC-CMs). Specifically, the study focuses on the role of peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α) in this process. ### Background and Motivation Although hiPSC-CMs have broad application prospects in clinical applications and research in the cardiovascular field, these cells retain an immature metabolic phenotype when cultured in vitro, which limits their application. Currently, little is known about the molecular mechanisms controlling mitochondrial metabolic maturation during the differentiation of hiPSCs into cardiomyocytes. Therefore, studying the role of PGC-1α in this process helps generate metabolically more mature cardiomyocytes, thereby enhancing their value in clinical and research applications. ### Main Findings 1. **PGC-1α Promotes Mitochondrial Respiration and Biogenesis**: - The study found that PGC-1α significantly promotes mitochondrial biogenesis and the establishment of oxidative phosphorylation (OXPHOS) during the differentiation of hiPSCs into cardiomyocytes. - Knockdown of PGC-1α expression impairs mitochondrial respiratory function, while upregulation of PGC-1α expression through the small molecule compound ZLN005 promotes mitochondrial biogenesis and function. 2. **The Role of ERRα in PGC-1α Regulation**: - The study also found that estrogen-related receptor alpha (ERRα) is essential for the stimulatory effect of PGC-1α. ERRα forms a coactivator complex with PGC-1α, jointly regulating the expression of downstream genes involved in mitochondrial dynamics and oxidative metabolism. 3. **Relationship Between Metabolic Changes and PGC-1α Expression**: - Through Western blot analysis, the study found that the expression of PGC-1α and its downstream metabolic transcription factors NRF1 and ERRα significantly increased in differentiated cardiomyocytes. Additionally, the expression of proteins related to fatty acid transport and the tricarboxylic acid cycle (TCA) also significantly increased, indicating a positive correlation between mitochondrial activity and the expression of PGC-1α and its downstream targets. ### Conclusion These findings preliminarily reveal the important role of PGC-1α in inducing oxidative metabolism and mitochondrial biogenesis during the differentiation of hiPSCs into cardiomyocytes and uncover its possible regulatory mechanisms. These research results provide a foundation for improving the mitochondrial metabolic maturity of hiPSC-CMs and enhancing their applications.
