Abstract:All eukaryotes have evolved complex sensory systems, which give them the ability to recognize information in the external environment through a variety of sense organs, including those for smelling, hearing, seeing, touching, taste, etc. This information is first perceived by the sensory organ with the help of specific receptor proteins and then transmitted to the brain, where it is processed and interpreted. The mechanism involved in this perception and interpretation of information through sensory organs is an area of research which has been difficult but fascinating. The olfactory system is perhaps the first of our sensory systems that has been deciphered in its entirety using the techniques of molecular biology, and the 2004 award of Nobel Prize in Physiology or Medicine to Richard Axel and Linda Buck is a recognition of this work. Research in the field of neurobiology (including olfactory system) has been central to the efforts in dealing with many human neurological disorders and behavioral patterns. In view of this, during 1990s, several Nobel Prizes in Physiology or Medicine were awarded for discoveries in this area of research. The Nobel Prizes in Physiology or Medicine awarded in recent years in the area of sensory systems and neurobiology include the following: (i) the 1994 Nobel Prize to Alfred Gilman and Martin Rodbell (both from the US), for discovery of G-proteins that help cells respond to outside signals; (ii) the 1998 Nobel Prize to Robert Furchgott, Louis Ignarro, and Ferid Murad (all from the US) for discovering that nitric oxide acts as a signal in the cardiovascular system, and (iv) the 2000 Nobel Prize to Arvid Carlsson (Sweden), Paul Greengard and Eric Kandel (both from the US) for discoveries concerning signal transduction in the nervous system. This year’s Nobel laureates, Axel and Buck, are both currently working as HHMI (Howard Hughes Memorial Institute) investigators (a prestigious position in USA awarded to eminent scientists on a competitive basis). Axel is working at Columbia University College of Physicians and Surgeons, New York, USA; and Buck, who earlier worked as a postdoctoral fellow with Axel, now works at Fred Hutchinson Cancer Research Center, Seattle, USA. A series of discoveries initiated jointly by these two Nobel laureates and followed by not only their two independent groups, but also by several other groups around the world completely dissected the pathway of olfaction sensing and memory. While studying the olfactory system, instead of hunting for the receptor proteins directly, Axel and Buck looked for genes encoding receptor proteins that are expressed only in the olfactory epithelium (for receptors and olfactory system, see later). They discovered a large gene family (perhaps the largest gene family known so far), which is now known to comprise some 1000 different genes in the human genome (3% of all genes) and some 1500 genes in the mouse genome (3–5% of all genes) that give rise to either an equivalent or a smaller number of olfactory receptor types (reports about odorant receptor (OR) genes and the OR types vary, partly because a large number of OR genes detected earlier are turning out to be pseudogenes). In a recent commentary, Buck summarized how did she, while working in Axel’s laboratory, discover the genes for ORs, and how did this discovery subsequently lead to the exploration of the mechanism of odour perception by several laboratories around the world. Axel and Buck both are currently continuing their work on olfactory system, although recently, Axel has largely been working on insects (Drosophila, Manducta, cockroach, etc.), while Buck continues working on mammals, including humans and mice.
