Abstract:Objective:To study the sterile conditions of leaves,induction of callus,induction of embryogenesis and plant regeneration of loquat.Methods:Loquat leaves were set up different sterilization time by diping in the saturation chloride of lime and 0.1% HgCl2,and were cut into small pieces and inoculated in the medium which added 6-BA,2,4-D and NAA.The leaves callus and plant regeneration were inducted.Results:Saturation chloride of lime dip in 25 min,0.1% HgCl2 dip in 4 min were better for younger leaf.Callus induced from leaves in MS+6-BA(0.5 mg/L)+2,4-D(0.5 mg/L)+NAA(2.0 mg/L) medium.The grain-like,light yellow and loose callus were embryonic callus.During the inducing of embryoid,the culture mediums with different grouth regulator concentration were used.Finally,the culture medium with the highest of reduction frequency was MS+6-BA(0.5 mg/L)+2,4-D(0.5 mg/L)+NAA(1.5 mg/L).The better culture medium for plant regeneration from embryoid of loquat was MS+6BA(1 mg/L)+NAA(0.5 mg/L).The root medium was MS+NAA(0.3 mg/L).The better matrix ratio of transplant was perlite,pert and sand as 1∶2∶1.Conclusion:Induction callus from loquat leaves and plant regeneration are the new way for large-scale production

Abstract:Loquat（Eriobotrya japonica Lindl. ）was an evergreen fruit produced in subtropical region. China had about 70% of the world loquat yield,which provided a large quantity of loquat flowers. The flower had abundant bioactive constituents,such as flavonoid,carotenoid,oleanolic acid,ursolic acid,amygdalin etc. The recent research developments of the loquat flowers were reviewed.

Abstract:Seedless fruit is a desirable trait that has been achieved in many plant cultivars,such as grape,citrus,and loquat,etc. In this paper,the reasons and mechanism of seedless fruit were studied and reviewed based on physiological and genetic research of fruit crops. Generally speaking,fruit physiology mainly include male sterility,gastrula abortion,endosperm abortion,self-incompatibility,balance and synergetic mechanism of plant hormones. While fruit genetics mostly consist of natural parthenocarpy,mutation,polyploidy seedless mechanism,triploid application,chemical induction,adoption of seedless genes and so on. Furthermore,the research prospect of seedless fruit was proposed.

Abstract:Loquat (Eriobotrya japonica Lindl.) is an economically important subtropical fruit tree, originating and commercially produced mainly in China and Japan. The aseptic seed germination of 13 loquat cultivars, including 9 white-fleshed cultivars and 4 red-fleshed ones, was studied, and the relationship between the germination rate and the content of endogenous ABA in seeds was explored. The germination rate and the seedling height at 80 days after sowing of white-fleshed cultivars were generally higher than that of red-fleshed ones. The ABA content in seeds was generally lower in white-fleshed cultivars, and the ABA content negatively correlated with the germination rate at 80 days after sowing. A moderate detrimental effect of 75% ethanol disinfection on aseptic germination of loquat seeds was observed, especially, for the cultivar Dahongpao, and the germination rate and seedling height were reduced by around half and one-third, respectively. The addition of plant preservative mixture (PPM) to the germination medium at a final concentration of 25 mg·L−1 effectively controlled contamination when the 75% ethanol disinfection procedure was omitted.

Abstract:In order to clarify the seedless formation mechanism of the loquat cultivar Dawuxing(Eriobotrya japonica Lindl.cv.Dawuxing) treated with GA3 before flowering,the effects on the embryo development,pollen fertility and seedless were investigated by paraffin wax cut and pollen germination examination etc.The results showed that:(1) the fruit ovule was anat-ropous ovule with double layers integument and nuclear type endosperm;(2) the seedless mainly was caused by embryo abortion induced from endosperm and nucellus degeneration after treated with GA3;the endosperm degeneration caused the nutrition deficiency which made failure of embryo development,so the embryo abnormally divided and eventually degenerated.On the other hand,the integument could be developed at the early stage of the fruit development and sterile seed was formed.

Abstract:So far Little information was available on breaking the Luffa cylindrical seed dormancy.The four treatments,soaking in 30% H_2O_2 for 10min,soaking in 0.1M HNO_3 for 12h,ultrasonic stimulation for 10min and breaking coat in one side of the seed by cutting,were conducted to investigate the effects of breaking seed dormancy using three lines of towel gourd.The results showed that the optimal treatment was breaking coat in one side of the seed,which increased the seed germination percentage from 7.8%-30.7% to 88.2%-100%.Moreover,the other three treatments were effective on breaking the seed dormancy,but there were no significant differences among them.

Abstract:The aim was to study the proper ratio of hormones for seed germination.Lilium speciosum var.gloriosoides widely distributed in Zhejiang Province,was used as matericals.The seeds of induction and germination medium adding different concentration of hormones were studied,and the MS basal medium without any hormones was used as control.The germination rate,shoot induction rate,number of shoots inducted were measured,and the growth of plantlets were observed during the experiment.Meanwhile,the seeds induced form plantlets were analyzed using the conventional chromosome preparation in this paper.The results showed that medium adding hormones could enhance the germination rate significantly,the optimum medium was MS media supplemented with 6-BA 1.0 mg/L and NAA 0.1 mg/L,the germination percentage,induction rate and average indefinite buds were 88.3%,88.9% and 2.5,respectively.Seeds without peeling treatment did not germinate after inoculating 30 days at all.The induced seedlings were transferred to MS medium,the growth condition sustained as before,which illustrated testa was the key factor limiting seed germination and hormone treatment or other physical measures can effectively improve the germination rate.The ploidy levels of seeds induction plantlets were diploid(2n=2x=24),and no genetic variation was found.
Seed formation in triploid loquat (Eriobotrya japonica) through cross-hybridization with pollen of diploid cultivars

Abstract:As the fruits of loquat (Eriobotrya japonica, 2n = 2x = 34) carry large seeds, the breeding of seedless loquat has long been a goal. The recent creation of triploid cultivars (2n = 3x = 51) and the application of gibberellins allow commercial production of seedless loquat, but the possibility of seed formation in triploid loquats has not been carefully investigated. Through crossing experiments and cytological observations of meiosis and pollen tube growth, we found that the triploid line 3N-N28 was essentially self-sterile, but developed seeds on pollination with pollen from diploid cultivars at rates of up to 5.5%. Almost half of the seedlings survived to 5 months, and carried diploid (2n = 34), tetraploid (2n = 68), or aneuploid chromosome numbers. Our results suggest that triploid loquat cultivars might retain the risk of seed formation. Protection from pollination by diploid cultivars or the development of new triploid cultivars will be necessary to ensure the production of seedless loquat fruits.

Abstract:After more than 10 years of development,DNA molecular markers were widely used in loquat.Types and characteristics of molecular markers utilized in the study of loquat were summarized.The progresses and achievements have been reviewed on genetic diversity and phylogenetic relationship,genetic linkage mapping of molecular markers,fingerprinting and identification of germplasm.Meanwhile,the problems presented in Loquat were discussed.Besides,development and use of more types of molecular markers of loquat,germplasm collection and evaluation based on molecular markers,construction of high-density genetic linkage mapping of molecular markers and fingerprinting,were considered as the direction of development in the future.

Abstract:In this paper,the biological and economic characteristics of loquat cultivars introduced from Sichuan and Fujian Province were studied.The cultivated techniques were also discussed.The result showed that the Zaozhong 6 was the most suitable cultivar in the northern of Guangxi Province.The Zaozhong-6 grew best with ripening date,20 days earlier than others.After planted for years,the yield of each tree and single fruit weight were 10.630 kg and 37.494 g on average,respectively,and soluble solid content was 11.303%.The flesh was delicious and good quality.

Abstract:The morphological characteristics of pistil and embryogenesis of Chinese plum cultivar Zuili were studied for clari-fying the reason of poor fruit setting with respect to reproductive biologically. Materials were dehydrated, embedded and cut in Paraffin wax. The results were as follows: The style is stuffed; two ovules are contained in the ovary; the ovule was anat-ropous,biteguminous and tenuinucellate. The embryo sac was staying at the 2 to 4 nucleis stage and on the 3rd day after an-thesis reached to the early 8 nucleis stage. The development of the embryo sacs was of polygonum type. The first division of the zygote took place after a dormancy of 2 to 3 days. The development of endosperm conforms to the nuclear type. The pri-mary endosperm nucleus began to divide on the 7th days after anthesis. The globular embryo stage began on 12th day after anthesis. Degeneration of nucellar cells and integument at early stage and abnormal embryo sac structure at mature stage was observed during the development of embryo.

Abstract:Loquats have drawn much attention due to their essential nutrients and unusual phenology, which fills a market gap in early spring. Fruit firmness (FF) is one of the most important quality attributes. Dynamic changes in FF, cell wall (CW) polysaccharides, CW hydrolase activity, and expression of CW metabolism-related genes during the fruit development and ripening stages of two contrasting loquat cultivars were compared. Although the two cultivars possessed similar FF at the initial fruitlet stage, Dawuxing was significantly firmer than Ninghaibai at all subsequent time points. FF was positively correlated with the contents of covalent-soluble pectin and hemicellulose, activity of peroxidase, and gene expressions of PME, EG, CAD6, and POD; and negatively correlated with the contents of water-soluble pectin, activities of polygalacturonase, endo-glucanase, cellobiohydrolase, and xylanase, and gene expressions of PG, EG2, PAL1, PAL3, and CAD5. Identifying molecular mechanisms underlying the differences in FF is useful for fundamental research and crop improvement in future.

A Study on Cold Resistance Function of Dehydrin Gene EjDHN5 from Loquat

Abstract:In order to examine the cold resistance function of dehydrin gene EjDHN5 from loquat (Eriobotrya japonica),it was overexpressed in tobacco plants.Two lines with relatively higher transcripts levels of the target gene,L24 and L26 were chosen for function characterization.Both of wide type (WT) and transgenic lines (L24 and L26) were treated with 0,2,4 and 8 μmol · L-1 methyl viologen (MV).After being exposed to 0,2,4 and 8 μmol · L-1 MV under light,the survival rate of WT plants decreased to 73.6％ and 56.9％,respectively,and the PSⅡ activity reduced dramatically,while the transgenic lines still remained a survival rate of 100％ as well as a much higher PSⅡ activity.MV treatment in leaf discs from 6-leaf-stage tobaccos confirmed that transgenic lines maintained higher chlorophyll content,accumulated less reactive oxygen species (ROS) and MDA.Moreover,compared to the WT plants,the EjDHN5-overexpressing plants exhibited a better growth status,had less ROS and suffered lighter membrane damage in leaves after cold stress.In summary,these results indicated that overexpressing EjDHN5 improved oxidative and cold tolerance in tobacco.It also provides evidence that dehydrins contribute to cold tolerance in loquat,which may be partly ascribed to its oxidative stress defense in plant.

Abstract:Genetic relationship of 85 loquat accessions was analyzed using 11 SSR primers of high polymorphism selected from 15 primers.A total of 85 polymorphic amplified fragments (Na) were produced from 11 primers.Na of each pair of primers ranged from 4 to 10,with an average of 7.7 alleles,and the valid Ne ranged from 1.93 to 6.40.The result indicated that the selected loquat germplasm accessions had an abundant genetic diversity.UPGMA cluster analysis showed the 85 loquat accessions were divided into two groups,A and B,respectively.The group A included 5 accessions from Zhejiang Tangqi.Group B were consisted of 80 accessions from Jiangsu,Zhejiang,Chongqing,Fujian of China and Spain.Group B could be further divided into the subgroups of B1,B2,B3 and B4.In addition,these accessions from the same origin were clustered together.Accessions of Spain No.2,Spain No.4 and Spain No.5 introduced to Tangqi town were the same variety Algerie.Baisha and Zhubao varieties of loquat in Tangqi town were one variety with different names.
Low Female Gametophyte Fertility Contributes to the Low Seed Formation of the Diploid Loquat [Eriobotrya Japonica (Thunb.) Lindl.] Line H30-6

Abstract:Loquat is a widely grown subtropic fruit because of its unique ripening season, nutrient content, and smooth texture of its fruits. However, loquat is not well-received because the fruits contain many large seeds. Therefore, the development of seedless or few-seed loquat varieties is the main objective of loquat breeding. Polyploidization is an effective approach for few-seed loquat breeding, but the resource is rare. The few-seed loquat line H30-6 was derived from a seedy variety. Additionally, H30-6 was systematically studied for its fruit characteristics, gamete fertility, pollen mother cell (PMC) meiosis, stigma receptivity, in situ pollen germination, fruit set, and karyotype. The results were as follows. (1) H30-6 produced only 1.54 seeds per fruit and the fruit edible rate was 70.77%. The fruit setting rate was 14.44% under open pollination, and the other qualities were equivalent to those of two other seedy varieties. (2) The in vitro pollen germination rate was only 4.04 and 77.46% of the H30-6 embryo sacs were abnormal. Stigma receptivity and self-compatibility in H30-6 were verified by in situ pollen germination and artificial pollination. Furthermore, the seed numbers in the fruits of H30-6 did not significantly differ among any of the pollination treatments (from 1.59 ±0.14 to 2 ± 0.17). (3) The chromosome configuration at meiotic diakinesis of H30-6 was 6.87I + 9.99II + 1.07III +0.69IV +0.24V (H30-6), and a total of 89.55% of H30-6 PMCs presented univalent chromosomes. Furthermore, chromosome lagging was the main abnormal phenomenon. Karyotype analysis showed that chromosomes of H30-6 had no recognizable karyotype abnormalities leading to unusual synapsis on the large scale above. (4) The abnormal embryo sacs of H30-6 could be divided into three main types: those remaining in the tetrad stage (13.38%), those remaining in the binucleate embryo sac stage (1.41%), and those without embryo sacs (52.82%). Therefore, we conclude that the loquat line H30-6 is a potential few-seed loquat resource. The diploid loquat line H30-6 was with low gametophyte fertility, which may be driven by abnormal meiotic synapses. The low female gamete fertility was the main reason for the few seeds. This diploid loquat line provides a new possibility for breeding a few-seed loquat at the diploid level.

Abstract:Loquat (Eriobotrya japonica Lindl.) is an evergreen fruit tree of Chinese origin, and its autumn–winter flowering and fruiting growth habit means that its fruit development is susceptible to low-temperature stress. In a previous study, the triploid loquat (B431 × GZ23) has been identified with high photosynthetic efficiency and strong resistance under low-temperature stress. Analysis of transcriptomic and lipidomic data revealed that the fatty acid desaturase gene EjFAD8 was closely associated with low temperatures. Phenotypic observations and measurements of physiological indicators in Arabidopsis showed that overexpressing-EjFAD8 transgenic plants were significantly more tolerant to low temperatures compared to the wild-type. Heterologous overexpression of EjFAD8 enhanced some lipid metabolism genes in Arabidopsis, and the unsaturation of lipids was increased, especially for SQDG (16:0/18:1; 16:0/18:3), thereby improving the cold tolerance of transgenic lines. The expression of ICE-CBF-COR genes were further analyzed so that the relationship between fatty acid desaturase and the ICE-CBF-COR pathway can be clarified. These results revealed the important role of EjFAD8 under low-temperature stress in triploid loquat, the increase expression of FAD8 in loquat under low temperatures lead to desaturation of fatty acids. On the one hand, overexpression of EjFAD8 in Arabidopsis increased the expression of ICE-CBF-COR genes in response to low temperatures. On the other hand, upregulation of EjFAD8 at low temperatures increased fatty acid desaturation of SQDG to maintain the stability of photosynthesis under low temperatures. This study not only indicates that the EjFAD8 gene plays an important role in loquat under low temperatures, but also provides a theoretical basis for future molecular breeding of loquat for cold resistance.

Abstract:Loquat (Eriobotrya japonica Lindl.) is a subtropical fruit tree with high medicinal value native to China. Different organs of loquat have been used historically as folk medicines and this has been recorded in Chinese history for thousands of years. Research shows that loquat extracts contain many antioxidants, and different extracts exhibit bioactivity capable of counteracting inflammation, diabetes, cancer, bacterial infection, aging, pain, allergy and other health issues. Bioactive compounds such as phenolics and terpenoids have been isolated and characterized to provide a better understanding of the chemical mechanisms underlying the biological activities of loquat extracts. As the identification of compounds progresses, studies investigating the in vivo metabolism, bioavailability, and structure–activity relationships, as well as potential toxicity of loquat extracts in animal or cell models are receiving more attention. In addition, genetic studies and breeding of loquat germplasms for high contents of health-benefiting compounds may provide new insight for the loquat industry and research. This review is focused on the main medicinal properties reported and the possible pharmaceutically active compounds identified in different loquat extracts.
Comparative transcriptome analysis reveals key genes potentially related to organic acid and sugar accumulation in loquat

Abstract:Organic acids and sugars are the primary components that determine the quality and flavor of loquat fruits. In the present study, major organic acids, sugar content, enzyme activities, and the expression of related genes were analyzed during fruit development in two loquat cultivars, ’JieFangZhong’ (JFZ) and ’BaiLi’ (BL). Our results showed that the sugar content increased during fruit development in the two cultivars; however, the organic acid content dramatically decreased in the later stages of fruit development. The differences in organic acid and sugar content between the two cultivars primarily occured in the late stage of fruit development and the related enzymes showed dynamic changes in activies during development. Phosphoenolpyruvate carboxylase (PEPC) and mNAD malic dehydrogenase (mNAD-MDH) showed higher activities in JFZ at 95 days after flowering (DAF) than in BL. However, NADP-dependent malic enzyme (NADP-ME) activity was the lowest at 95 DAF in both JFZ and BL with BL showing higher activity compared with JFZ. At 125 DAF, the activity of fructokinase (FRK) was significantly higher in JFZ than in BL. The activity of sucrose synthase (SUSY) in the sucrose cleavage direction (SS-C) was low at early stages of fruit development and increased at 125 DAF. SS-C activity was higher in JFZ than in BL. vAI and sucrose phosphate synthase (SPS) activities were similar in the two both cultivars and increased with fruit development. RNA-sequencing was performed to determine the candidate genes for organic acid and sugar metabolism. Our results showed that the differentially expressed genes (DEGs) with the greated fold changes in the later stages of fruit development between the two cultivars were phosphoenolpyruvate carboxylase 2 ( PEPC2 ), mNAD-malate dehydrogenase ( mNAD-MDH ), cytosolic NADP-ME ( cyNADP-ME2 ), aluminum-activated malate transporter ( ALMT9 ), subunit A of vacuolar H + -ATPase ( VHA-A ), vacuolar H + -PPase ( VHP1 ), NAD-sorbitol dehydrogenase ( NAD-SDH ), fructokinase ( FK ), sucrose synthase in sucrose cleavage ( SS-C ), sucrose-phosphate synthase 1 ( SPS1) , neutral invertase (NI), and vacuolar acid invertase ( vAI) . The expression of 12 key DEGs was validated by quantitative reverese transcription PCR (RT-qPCR). Our findings will help understand the molecular mechanism of organic acid and sugar formation in loquat, which will aid in breeding high-quality loquat cultivars.

Abstract:Loquat ( Eriobotrya japonica Lindl.) is one of the most economically important evergreen fruit crops in China, while it often suffered the injury of cold stress in winter and earlier spring, and the annual yield loss of loquat fruits caused by cold or freezing stress was immeasurable. However, knowledge about the physiological response and molecular mechanism under cold stress is still limited. To investigate the potential regulation mechanism pre- and post-cold stress in loquat and the changes in physiological indicators, a comparative transcriptome analysis was performed against a cold-resistant cv. "Huoju" and a cold-sensitive cv. "Ninghaibai". The results of physiological indicators related to cold resistance indicated that rachis was most sensitive to cold stress and was considered as the representative organ to directly evaluate cold resistance of loquat based on subordinate function analysis. Here, we compared the transcriptome profiles of rachis pre- and under cold stress in "Huoju" and "Ninghaibai". A total of 4,347 and 3,513 differentially expressed genes (DEGs) were detected in "Ninghaibai" and "Huoju", among which 223 and 166 were newly identified genes, respectively, most of them were functionally enriched in plant hormone signal transduction (Huoju: 142; Ninghaibai: 200), and there were higher plant hormone content and related DEG expression levels in "Huoju" than that of "Ninghaibai". Moreover, a total of 3,309 differentially expressed transcription factors (DETFs) were identified, and some DEGs and DETFs were screened to be subjected to co-expression network analysis based on the gene expression profile data. Some candidate DEGs, including UDP-glycosyltransferase (UGT), glycosyltransferase (GT), sugar phosphate/phosphate translocator (SPT), sugar transport protein (STP), proline-rich receptor-like protein kinase (PERK), and peroxidise (POD), were significantly affected by cold stress, and the expression level of these genes obtained from real-time quantitative RT-PCR was consistent with the pattern of transcriptome profile, which suggested that these genes might play the vital roles in cold resistance of loquat. Our results provide an invaluable resource for the identification of specific genes and TFs and help to clarify gene transcription during the cold stress response of loquat.

Abstract:The experiment was undertaken to promote an enlargement of frost-induced seedless loquat fruit through applications of gibberellic acid (GA) and GA combined with N- (2-chloropyridyl) -N'-phenylurea (KT).Spraying with an aqueous solution of 500 ppm GA or 500 ppm GA+20 ppm KT greatly stimulated frost-induced seedless fruit to attain the same size as seeded control ones. The GA-treated seedless fruits were more slender but had a thicker pulp, compared to seeded untreated fruits. Application of GA + KT was more effective for enlargement than a single application of GA. If sprayed immediately after a frost, the enlargement response of the seedless fruits was significant. Although treated seedless fruits turned yellow earlier, the total soluble solids contents in the juice at harvest were slightly lower than that of seeded fruits. No difference in titratable acidity was found between the treated and control fruits.
