What problem does this paper attempt to address?
The problem that this paper attempts to solve is to explore the heterogeneity and clonal relationships of adaptive immune cells in inflammatory bowel disease (IBD), especially ulcerative colitis (UC). Specifically, the authors integrated single - cell RNA sequencing (scRNA - seq), T - cell receptor sequencing (scTCR - seq) and B - cell receptor sequencing (scBCR - seq) techniques to reveal the key components, cell states and clonal relationships of the peripheral and gastrointestinal mucosal immune systems in healthy states and under UC conditions. The main findings of the paper include: 1. **Changes in plasma cells**: The number of IgG1+ plasma cells in the colon tissues of UC patients increases. 2. **Changes in regulatory T cells**: The number of regulatory T cells (Treg) in the colon tissues of UC patients increases, and these cells express high levels of the transcription factor ZEB2. 3. **Changes in γδ T cells**: There is an enrichment of specific γδ T - cell subsets in the peripheral blood of UC patients. 4. **Changes in CD8+ tissue - resident memory T cells (TRM)**: Heterogeneity of CD8+ TRM cells is observed in the colon tissues of UC patients.
