Abstract:Cyclodextrins (CDs), which are a class of cyclic oligosaccharides extracted from the enzymatic degradation of starch, are often utilized in molecular recognition and assembly constructs, primarily via host-guest interactions in water. In this review, recent progress in CD-based supramolecular nanoassemblies that are sensitive to chemical, biological, and physical stimuli is updated and reviewed, and intriguing examples of the biological functions of these nanoassemblies are presented, including pH- and redox-responsive drug and gene delivery, enzyme-activated specific cargo release, photoswitchable morphological interconversion, microtubular aggregation, and cell-cell communication, as well as a geomagnetism-controlled nanosystem for the suppression of tumor invasion and metastasis. Moreover, future perspectives and challenges in the fabrication of intelligent CD-based biofunctional materials are also discussed at the end of this review, which is expected to promote the translational development of these nanomaterials in the biomedical field.

Abstract:As a class of macrocyclic host with excellent water solubility, low biotoxicity and high charge density, multi-charged cyclodextrins have been extensively studied because they can interact with organic/inorganic/biological molecules through multiple synergistic forces such as the hydrophobic cavities and electrostatic interactions to construct the smart supramolecular assemblies with stable topology, functional diversity and stimulus responsiveness. The latest research progress of pH-, photo-, enzyme-, redox-, magnetic- and multi-stimulus responsive smart supramolecular assemblies, which are constructed by typical positively/negatively charged and amphiphilic multi-charged cyclodextrins including their application in the fields of drug delivery, controlled release and sensory detection is introduced, and the challenges and future developments of multi-charged cyclodextrin smart supramolecular assemblies are discussed.

Abstract:The excellent biocompatibility and unique inclusion capability as well as powerful functionalization capacity of cyclodextrins and their derivatives make them especially attractive for engineering novel functional materials for biomedical applications. There has been increasing interest recently to fabricate supramolecular systems for drug and gene delivery based on cyclodextrin materials. This review focuses on state of the art and recent advances in the construction of cyclodextrin-based assemblies and their applications for controlled drug delivery. First, we introduce cyclodextrin materials utilized for self-assembly. The fabrication technologies of supramolecular systems including nanoplatforms and hydrogels as well as their applications in nanomedicine and pharmaceutical sciences are then highlighted. At the end, the future directions of this field are discussed.

Abstract:Construction of biocompatible nanoparticles is a significant topic of research at the chemistry/biology interface. Presently, more and more researchers have focused on supramolecular amphiphiles constructed by macrocycle-induced aggregation, a feasible and universal strategy in the fabrication of functional nanomaterials. In this work, we report a pH-responsive nanoparticle through a noncovalent complexation between per-6-thiolated beta-cyclodextrin sodium salt (SACD) and amphiphilic ferrocene derivative (FC12+Br-) in water. The formation and composition of the obtained supramolecular nanoparticles were comprehensively characterized by UV/Vis absorption, dynamic laser scattering, and microscopic observations. Furthermore, the drug loading and release studies demonstrated that the supramolecular binary nanoparticles could efficiently encapsulate doxorubicin hydrochloride (DOX) in their nanoparticulate cores, and the entrapped DOX could be rapidly released in a low pH environment. The present study suggests that the obtained binary supramolecular amphiphiles with good stability, tunable assembling/disassembling behaviors, and controlled drug loading/release properties have great potential in the construction of on-demand drug-delivery systems.

Abstract:Stimuli responsive polymers have been extensively studied as nanocarriers for drug delivery systems (DDSs), especially those based on supramolecular interactions. Cycludextrin (CD) is one kind of widely applied host molecule, and the host guest interactions between CD and different counterparts can respond to different stimuli and thus can be applied as responsive linkers for polymeric DDSs. In this review, the polymeric nanocarriers based on the host guest interactions between CD and ferrocene, azobenzene, and benzimidazole as DDSs are summarized, with redox, light, and pH sensitivity, respectively. The mechanisms for the stimuli responsive ability of the linkers, the application of them for construction of DDSs with different polymer structures, and the controlled release behaviors have been focused. In addition, the outlook and challenge of these systems are discussed.

Abstract:In the biomedical and pharmaceutical fields, cyclodextrin (CD) is undoubtedly one of the most frequently used macrocyclic compounds as the host molecule because it has good biocompatibility and can increase the solubility, bioavailability, and stability of hydrophobic drug guests. In this review, we generalized the unique properties of CDs, CD-related supramolecular nanocarriers, supramolecular controlled release systems, and targeting systems based on CDs, and introduced the paradigms of these nanomedicines. In addition, we also discussed the prospects and challenges of CD-based supramolecular nanomedicines to facilitate the development and clinical translation of these nanomedicines.
A Smart Core-Crosslinked Supramolecular Drug Delivery System (SDDS) Enabled by Pendant Cyclodextrins Encapsulation of Drug Dimers Via Host-Guest Interaction

Abstract:Owing to poor aqueous solubility and low delivery efficiency, most of anti-cancer chemodrugs depend on various smart drug delivery platforms to enhance the treatment efficacy. Herein, a stimuli-responsive supramolecular drug delivery system (SDDS) is developed based on polymeric cyclodextrins (PCD) which crosslinked by stimuli-cleavable drug dimers via host-guest interaction. PEGylated PCD was precisely controlled synthesized by ring-opening polymerization and azide-alkyne click chemistry, and two doxorubicins (DOX) were linked with a disulfide bond to form a drug dimer (ss-DOX). They then co-assembled into supramolecular micelles. Drug dimers were utilized as cross-linkers to stabilize the micelles. The drug loading efficiency was very high that could be up to 98%. The size and morphology were measured by DLS and TEM. Owing to the disulfide bonds of drug dimers, these supramolecular micelles were dissociated by treating with dithiothreitol (DTT). In the meanwhile, the free DOXs were recovered and released from cavities of cyclodextrins because of dynamic equilibrium and hydrophilicity changes. The release profile was studied under mimic physiological conditions. Furthermore, in vitro cytotoxicity study showed excellent anti-cancer efficacy of reduced-responsive supramolecular polymeric micelles. Therefore, it can be served as a safe and stimuli-responsive SDDS for cancer therapy.

Abstract:From motor proteins tomolecularmachines, supramolecular chemistry has been revealed as a remarkable link to bridge the gap between biology, chemistry, and materials. Taking inspiration from the dynamic, reversible, and directional manner of non-covalent interactions that can respond to diversiform external stimuli, supramolecular nanovalves installed on the surface of inorganic or hybrid nanocarriers have received extensive attention in stimuli-responsive delivery of small drug molecules, implying their outspread bioapplications in cancer/gene therapy, biomedical application, and antimicrobial regulations. The state of supramolecular nanovalves could be well regulated by switching the conformation or the assembled/disassembled state of the assemblies. This tutorial review lays the foundation for a better understanding of the significant and typical intelligent drug-delivery systems immobilized with supramolecular polymers or supramolecular pseudorotaxanes as the gating entities of nanovalve-based molecular machines, by showing their chemical structures, operation modes, and release modalities triggered by various actuations at molecular scale. In particular, relevant perspectives of supramolecular therapeutics will also be elaborated upon.

Abstract:Novel vesicles assembled by 'supramolecular amphiphiles' based on the inclusion complexes between 1 and β-CDs (β-CD and HP-β-CD) were found to carry drugs and be successively-responsive to external stimuli for the first time. These vesicles were observed by TEM and SEM and confirmed by DLS. The formation patterns of 'supramolecular amphiphiles' characterized by UV and NMR can be controlled by choosing different hosts. Unlike traditional drug delivery and releasing systems, the 'useless' inclusion complexes 1·β-CDs in thinking formula can assemble into vesicles to carry both cytotoxic 1 and drugs (piroxicam and ampicillin) at one time, confirmed by TEM, UV, and NMR data. Also the vesicles assembled by 1·β-CDs can be successively-responsive to acidifying and oxidizing, and release drugs and cytotoxic compounds in order.

Abstract:Cyclodextrins (CDs) are widely used macrocycles in supramolecular assembly due to their easy availability, versatile functionality and excellent biocompatibility. Although they are well-known for forming host-guest complexes with a wide range of guests and this host-guest chemistry has long been utilized in industry and academia, new opportunities have arisen in recent years, particularly in supramolecular assembly. In the present review, we will first provide a basic introduction to CDs and then summarize their emerging roles in the fields of supramolecular chemistry and materials. This includes their involvement in hybrid frameworks with inorganic components such as metal ions and polyoxometalates, crystalline self-assembly with amphiphilic molecules, and their new possibility of "catassembly" and induced chiral supramolecular structures that have previously been overlooked. Finally, we will comment on the future perspectives of CDs to inspire more ideas and efforts, with the aim of promoting diverse applications of CDs in supramolecular materials.
A fluorescent, chiratity-responsive, and water-soluble cage as a multifunctional molecular container for drug delivery

Abstract:In recent years, the rational design and construction of drug delivery systems (DDSs) via a supramolecular approach for improving chemical therapeutics have gained significant attention. Here, we report a host-guest DDS formed from a fluorescent, chirality-responsive, and water-soluble tetraphenylethene-based octacationic cage as a fluorescent/chiral probe, solubilizer, and molecular cargo, which can recognize chiral nucleoside drugs, enhance the solubility of insoluble drugs, and protect drugs from the outside environment by forming host guest complexes in aqueous solution. Given the fluorescence properties and dynamically rotational conformation of tetraphenylethene (TPE) units, this fluorescent and chirality-responsive cage exhibits different responses including turn-on/turn-off fluorescence and negative/positive circular dichroism (CD) when binding with different chiral nucleoside drugs in water, resulting in multiple-responsive photophysical behaviors for these chiral drugs. Furthermore, this water-soluble cationic cage with a hydrophobic cavity can improve the water solubility of insoluble drugs (e.g., CPT) by forming host-guest complexes in water. More importantly, this multifunctional cage exhibits a low toxicity to both human colon and breast cancer cell lines in vitro, and drugs encapsulated by the cage are more effective in killing cancer cells than drugs alone. Finally, the on off -on fluorescence responses in the formation and dissociation processes of the cage superset of drug complexes have been successfully used to monitor drug release and track drug delivery by fluorescence microscopy in vitro. Therefore, this fluorescent, chirality-responsive, and water-soluble cage as a multifunctional molecular container can be used to construct a smart drug delivery system with several functions of fluorescence and CD detection, water solubilization, real-time monitoring, and chemotherapy.

Abstract:Developing smart controlled-release systems for cancer therapy is highly desired in biomedicine. In order to improve therapeutic efficacy and lower undesired side effects, the construction of stimulus-responsive nanocarriers is a favorable solution. Emerging supramolecular self-assemblies possessing intrinsic dynamic and adaptive features present promising capabilities for the fabrication of stimulus-responsive drug-release systems. Dynamic supramolecular vesicles have attracted considerable attention as therapeutic carriers ascribed to their advantages of high-cargo-loading capacity/feasibility, excellent biocompatibility, and facile functionalization. Here, a summary and discussion of recent significant development of vesicles constructed by supramolecular self-assembly for stimulus-responsive drug delivery and therapeutics is given. Through presenting some representative studies, strategies regarding the design, synthesis, characterization, and biomedical applications of supramolecular vesicle carriers are highlighted according to various stimulus triggers. The aim is to provide a quick research update in this rapidly developing field.
Tunable supramolecular self-assemblies based on cyclodextrin polymer as a loading platform for water-soluble drugs

Abstract:Drug loading capacity is a crucial character of nano-scaled drug carriers to achieve high quality pharmaceutical preparations. However, efficient encapsulation of water-soluble small molecular drugs still faces large obstacles in many cases. Herein, we designed a novel supramolecular delivery system constructed by poly(β-cyclodextrin) containing benzoic acid groups (PCD-PA) and adamantyl terminated poly(ethylene glycol) (PEG-AD) to provide multiple intermolecular interactions for competent loading of water-soluble small-molecular drugs. PCD-PA had multiple host molecules, and PEG-AD could be inserted via host-guest interaction in different proportion to adjust the composition of supramolecular carrier. Meanwhile, π-π stacking and electrostatic interaction furnished by benzoic acid groups served as binding force for drug entrapment, which led to considerable loading capacity for several water-soluble drugs. Among the drugs with different chemical structures, mitoxantrone hydrochloride and doxorubicin hydrochloride bearing anthraquinone rings and several protonable amino groups acquired the highest loading content as about 14 % in PCD-PA3/PEG-AD supramolecular self-assemblies. Further computational simulations investigated the mechanism of drug loading based on the interactions between the carrier materials and the payloads. In addition, the weakly acidic environment obviously accelerated the release of certain drugs. All in all, this self-assembled supramolecular nano-system displayed great potentials as a delivery platform for diverse water-soluble drugs.

Abstract:Health has always been a hot topic of concern, whereas cancer is one of the largest security risks to human health. Although the existing drug delivery systems (DDSs) have been extensively reported and commercially applied, there are still some issues that have yet to be well-resolved, including the toxicity, side-effects, and targeted therapy efficiency of drugs. Consequently, it is still necessary to develop a novel, highly efficient, controlled and targeted DDS for cancer therapy. For this, a supramolecular polymer, beta-CD-g-PDMAEMA@Azo-PCL, was designed and developed through the host-guest inclusion complexation interactions between a host polymer, beta-cyclodextrin-graf t-poly(2-(dimethylamino)ethyl methacrylate) (beta-CD-g-PDMAEMA), and a guest polymer, azobenzene modified poly(e-caprolactone) (Azo-PCL), and was characterized by various analysis techniques. The supramolecular assembly was examined in various pH environments and/or under UV-vis irradiation, showing the formation of supramolecular assemblies from regular spherical shapes to irregular aggregates with various hydrodynamic diameters. The 2D NOESY NMR studies showed the formation of inclusion complexation between Azo-PCL and beta-CD-g-PDMAEMA and between beta-CD and the side groups of PDMAEMA. The supramolecular assemblies could encapsulate doxorubicin to form spherical core-shell drug-carrying micelles with an entrapment efficiency of 66.1%. The effects of external environment stimuli on the in vitro drug release were investigated, showing light- and pH-modulated drug release properties. The cytotoxicity assessment indicated that the blank supramolecular micelles were nontoxic, whereas the drug-loaded micelles exhibited comparable or even superior anticancer activity to the anticancer activity of free DOX and inhibition of cancer cell proliferation. Therefore, the developed supramolecular assemblies can potentially be used as drug-controlled release carriers.

Abstract:Supramolecular self-assembly provides a new strategy for the development of drug delivery systems from molecular components. With non-covalent interactions as driving forces, supramolecular drug delivery systems (SDDSs) can realize precise component control at the molecular level, increased predictability of the self-assembled structures, tunable control of morphologies and sizes, controlled release of delivered drugs. In this review, we first concisely introduce the background for the design of SDDSs and then describe important advances in SDDS development that involves the applications of cyclodextrin, calixarene, pillararene and cucurbituril based on host-guest interactions. Following these popular design principles, we further present the applications of water-soluble supramolecular organic frameworks as SDDSs. Finally , the challenges that need to be addressed for the practical translation of SDDSs are discussed.

Abstract:This paper reports the synthesis and drug delivery properties of a novel supramolecular nanoparticle. α-Cyclodextrins (α-CD) were threaded on cinnamic acid modified poly(ethylene glycol) to form inclusion complex nanoparticles by supramolecular self-assemble. The anti-tumor drug doxorubicin was loaded in the nanoparticles and released in vitro to study the drug release behavior and the anti-tumor effects. The structure and morphology of the nanoparticles were characterized by nuclear magnetic resonance, X-ray diffraction, ultraviolet absorbance, dynamic laser scattering, scanning electronic microscopy, transmission electron microscopy and atom force microscopy. The distribution of the drug loaded nanoparticles in cells and the anti-tumor effects were studied by confocal laser microscopy. The results demonstrate that the supramolecular nanoparticle is biocompatible and it is a promising carrier for drug delivery systems.

Abstract:Modification of natural polysaccharides such as chitosan (CS), β‑cyclodextrin (β-CD), and alginic acid offers a promising strategy for the preparation of smart drug carriers, and latest innovations on such carriers are focused on stimuli-responsive biomaterials. In this study, highly hydrophilic three-demensional (3D) porous CS-grafted β-CD (CS-g-β-CD) was prepared through the Williamson ether synthesis reaction with epichlorohydrin (ECH) as the crosslinker and the consequent nucleophilic reaction between the epoxide ring of ECH and the primary amine of CS, which was then characterized by 1H nuclear (1H NMR), Fourier transform infrared (FT-IR) spectra, X-ray diffraction (XRD) analysis, scanning electron microscope (SEM), thermogravimetry (TG), and N2 adsorption/desorption isotherms. When etoposide (VP16), an anti-cancer drug, was encapsulated in the CS-g-β-CD, the encapsulation ratio was up to 73.6%. Finally, the resultant CS-g-β-CD was successfully used as the responsive drug carrier for pH- and thermo-sensitive release of VP16. This work opens a new avenue for the preparation of stimuli-responsive drug carriers with modified natural polysaccharides.
