Palbociclib-letrozole loaded solid self-nano emulsifying drug delivery system for oral treatment of breast cancer: In-vitro and In-vivo characterization

Abstract:The current research aimed to formulate a self-nanoemulsifying drug delivery system (SNEDDS) co-loaded with palbociclib (PCB) and letrozole (LTZ) to enhance anticancer efficacy by improving oral solubility and bioavailability. Liquid PCB-loaded SNEDDS (L-PSNE-1 to L-PSNE-9) were optimized for high % transmittance, % cumulative drug release (%CDR), and low self-emulsification time using a 3 2 full factorial design. LTZ was incorporated into the optimized liquid PCB-loaded SNEDDS (L-PLSNE-8), solidified using Neusilin-US2 (S-PLSNE-8), and characterized for morphology by transmission electron microscopy (TEM) (L-PLSNE-8), crystallinity by X-ray diffraction, in vitro drug release, Caco-2 cell permeability, cytotoxicity, apoptotic potential, and pharmacokinetic parameters. The optimized L-PSNE-8, composed of Maisine (15%), Cremophor RH-40 (40%), and labrasol, demonstrated reduced self-emulsification time (90 ± 1.08 sec) and a high transmittance (99.26 ± 1.71%). L-PLSNE-8 showed nanoscale spherical globules in TEM analysis. The FTIR spectra of S-PLSNE-8 revealed no incompatibility between the drug and excipients. DSC and XRD analysis of the S-PLSNE-8 demonstrated the amorphization and molecular dispersion of PCB and LTZ. S-PLSNE-8 exhibited a complete drug release (99.76 ± 0.75%) compared to plain drug dispersions in 0.1N HCl after 20 min of release study. The release profiles in phosphate buffer at pH 6.8 were observed to be slightly greater than those in 0.1N HCl. S-PLSNE-8 significantly (P<0.001) decreased the growth of MCF-7 breast cancer cells and increased their apoptosis (P < 0.001). Compared to plain drugs, S-PLSNE-8 showed almost a 4- and 2-fold increase in Caco-2 cell permeability of PCB and LTZ, respectively. Oral pharmacokinetic study of S-PLSNE-8 revealed a 7.5- and 4-fold increase in bioavailability for LTZ and PCB, respectively, compared to plain drugs. These findings suggest that S-PLSNE-8 is a promising oral drug delivery system for poorly soluble and permeable drugs in breast cancer treatment.
