Abstract:Abstract Background While the rapidly increasing global population has led to a dramatically increased demand for the agricultural production, there have been heavy economic losses owing to various pest attacks on different food crops. The advancement of various biotechnological techniques have come as a boon in addressing the global concern and leads to the development of novel varieties that have proven to be highly economical, pesticide resistant and environmentally safe. Main body The present review was aimed to update the recent developments that have taken place in the field of crop production. Major focus was laid predominantly on such genes that have demonstrated positive effects and proved to be of commercial success at the market primarily due to the development of pest-resistant transgenic food crops with expression of Bacillus thuringiensis toxins. This technology has been effective against a wide range of pests including coleopterans, lepidopterans, hemipterans, dipterans, strongylida (nematodes) and rhabditida. In similar lines various plant derived toxic proteins were also discussed along with different genes that code for insect resistant proteins such as δ -endotoxins and secreted toxins. This article also helps in understanding the structural features of the genes that are endowed with insect resistance followed by their mechanism of action on pests. Further the role of secondary metabolites in controlling the pests was addressed. The Pros and Cons of existing tools of insect pest management were demonstrated. Conclusions Novel technologies are necessary in crop improvement to progress the pace of the breeding programs, to confer insect resistance in crop plants. Therefore, the future aim of crop biotechnology is to engineer a sustainable, multi-mechanistic resistance to insect pests considering the diversity of plant responses to insect attack.

Abstract:Genetic modification in plants was first recorded 10,000 years ago in Southwest Asia where humans first bred plants through artificial selection and selective breeding. Since then, advancements in agriculture science and technology have brought about the current GM crop revolution. GM crops are promising to mitigate current and future problems in commercial agriculture, with proven case studies in Indian cotton and Australian canola. However, controversial studies such as the Monarch Butterfly study (1999) and the Séralini affair (2012) along with current problems linked to insect resistance and potential health risks have jeopardised its standing with the public and policymakers, even leading to full and partial bans in certain countries. Nevertheless, the current growth rate of the GM seed market at 9.83-10% CAGR along with promising research avenues in biofortification, precise DNA integration and stress tolerance have forecast it to bring productivity and prosperity to commercial agriculture.

Abstract:New advances in crop genetic engineering can significantly pace up the development of genetically improved varieties with enhanced yield, nutrition and tolerance to biotic and abiotic stresses. Genetically modified (GM) crops can act as powerful complement to the crops produced by laborious and time consuming conventional breeding methods to meet the worldwide demand for quality foods. GM crops can help fight malnutrition due to enhanced yield, nutritional quality and increased resistance to various biotic and abiotic stresses. However, several biosafety issues and public concerns are associated with cultivation of GM crops developed by transgenesis, i.e., introduction of genes from distantly related organism. To meet these concerns, researchers have developed alternative concepts of cisgenesis and intragenesis which involve transformation of plants with genetic material derived from the species itself or from closely related species capable of sexual hybridization, respectively. Recombinase technology aimed at site-specific integration of transgene can help to overcome limitations of traditional genetic engineering methods based on random integration of multiple copy of transgene into plant genome leading to gene silencing and unpredictable expression pattern. Besides, recently developed technology of genome editing using engineered nucleases, permit the modification or mutation of genes of interest without involving foreign DNA, and as a result, plants developed with this technology might be considered as non-transgenic genetically altered plants. This would open the doors for the development and commercialization of transgenic plants with superior phenotypes even in countries where GM crops are poorly accepted. This review is an attempt to summarize various past achievements of GM technology in crop improvement, recent progress and new advances in the field to develop improved varieties aimed for better consumer acceptance.

This approach, integrating ecological intelligence with modern scientific advancements, is seeing increasing adoption in various regions, notably in India. Drawing inspiration from nature's own mechanisms, biocontrol employs specific organisms or their biological derivatives to manage and mitigate pests, thereby decreasing the dependency on chemical agents. Through an in-depth exploration of biocontrol applications, this study places a special emphasis on India's initiatives and contrasts them with global endeavors. Noteworthy examples include the innovative use of parasitic wasps to combat the papaya mealybug menace in Tamil Nadu, the strategic application of fungi like Trichoderma for disease control in Maharashtra, and the successful introduction of parasitoid wasps to manage olive flies in regions like California. Rooted in foundational ecological principles, these methodologies exhibit potential benefits that span improved crop yields, economic viability, and most importantly, reduced environmental adversities. Effective deployment and scaling of these techniques require an integrated approach, emphasizing collaboration among researchers, farming communities, and industry stakeholders. Abstract:Plants are challenged incessantly by several biotic and abiotic stresses during their entire growth period. As with other biotic stress factors, insect pests have also posed serious concerns related to yield losses due to which agricultural productivity is at stake. In plants, trait modification for crop improvement was initiated with breeding approaches followed by genetic engineering. However, stringent regulatory policies for risk assessment and lack of social acceptance for genetically modified crops worldwide have incited researchers toward alternate strategies. Genome engineering or genome editing has emerged as a new breeding technique with the ability to edit the genomes of plants, animals, microbes, and human beings. Several gene editing strategies are being executed with continuous emergence of variants. The scientific community has unraveled the utility of various editing tools from endonucleases to CRISPR/Cas in several aspects related to plant growth, development, and mitigation of stresses. The categorical focus on the development of tools and techniques including designing of binary vectors to facilitate ease in genome engineering are being pursued. Through this Review, we embark upon the conglomeration of various genome editing strategies that can be and are being used to design insect pest resistance in plants. Case studies and novel crop-based approaches that reiterate the successful use of these tools in insects as well as in plants are highlighted. Further, the Review also provides implications for the requirement of a specific regulatory framework and risk assessment of the edited crops. Genome editing toward insect pest management is here to stay, provided uncompromising efforts are made toward the identification of amiable target genes.This article has not yet been cited by other publications.

Abstract:Insect pests indicate serious hazards to agriculture, human health and global food security, which are compounded by climate change and changing pest behaviors. Traditional pest management systems, such as chemical controls and integrated pest management, are under threat due to resistance and environmental concerns. This review explores innovative pest control methods, focusing on Genetic Pest Management (GPM) and the Sterile Insect Technique (SIT). GPM technologies, such as CRISPR-based gene editing and gene drives, offer promising solutions by introducing targeted genetic modifications to pest populations. SIT, which involves releasing sterilized insects to reduce pest populations, is enhanced by advances in sexing and genetic techniques. Biological control agents (BCAs), including entomopathogenic bacteria and fungi, provide sustainable alternatives for pest management. The effectiveness of these methods depends on understanding the underlying mechanisms and optimizing application conditions. Regulatory frameworks play a crucial role in the adoption of these technologies, with varying requirements across countries. We believe that ongoing advancements in biological and genetic approaches, when effectively regulated and thoughtfully integrated, will be crucial in shaping the next generation of pest management techniques. Abstract:Insecticidal transgenes, when incorporated and expressed in plants, confer resistance against insects by producing several products having insecticidal properties. Protease inhibitors, lectins, amylase inhibitors, and chitinase genes are associated with the natural defenses developed by plants to counter insect attacks. Several toxin genes are also derived from spiders and scorpions for protection against insects. Bacillus thuringiensis Berliner is a microbial source of insecticidal toxins. Several methods have facilitated the large-scale production of transgenic plants. Bt-derived cry, cyt, vip, and sip genes, plant-derived genes such as lectins, protease inhibitors, and alpha-amylase inhibitors, insect cell wall-degrading enzymes like chitinase and some proteins like arcelins, plant defensins, and ribosome-inactivating proteins have been successfully utilized to impart resistance to insects. Besides, transgenic plants expressing double-stranded RNA have been developed with enhanced resistance. However, the long-term effects of transgenes on insect resistance, the environment, and human health must be thoroughly investigated before they are made available for commercial planting. In this chapter, the present status, prospects, and future scope of transgenes for insect pest management have been summarized and discussed.

Abstract:Amid concerns over chemical pesticide resistance and its associated environmental hazards, parasitoids offer an alternative long-term solution to manage insect pests in agriculture. India's use of parasitoids in pest management has developed in tandem with the rest of the world, and this review summarizes the history of parasitoid-based biocontrol from the past to the present, focusing on problems such as climate adaptability, ecological compatibility, research-based advances, and policy-making. It focuses on successful classical, conservative, and augmentative techniques that form the foundation for implementing effective and sustainable biological control strategies involving parasitoids in India. The components that influence the efficiency of biocontrol activities, such as suitable phenological stages of parasitoids, field deployment techniques, quality assurance, environmental conditions, area-wide approaches, the need for sound habitat management, policy interventions, and public-private partnership are highlighted. Recent advancements in parasitoid mass production, quality control, and understanding competitive ecological interactions have provided prospects for designing effective parasitoid-centered biocontrol programs. The review presents historical breakthroughs in explaining how parasitoids help stabilize the agroecological dynamics that support sustainable food systems, primarily in India.

Abstract:Recent developments in modern biotechnology such as the use of RNA interference (RNAi) have broadened the scope of crop genetic modification. RNAi strategies have led to significant achievements in crop protection against biotic and abiotic stresses, modification of plant traits, and yield improvement. As RNAi-derived varieties of crops become more useful in the field, it is important to examine the capacity of current regulatory systems to deal with such varieties, and to determine if changes are needed to improve the existing frameworks. We review the biosafety frameworks from the perspective of developing countries that are increasingly involved in modern biotechnology research, including RNAi applications, and make some recommendations. Malaysia and India have approved laws regulating living modified organisms and products thereof, highlighting that the use of any genetically modified step requires regulatory scrutiny. In view of production methods for exogenously applied double-stranded RNAs and potential risks from the resulting double-stranded RNA-based products, we argue that a process-based system may be inappropriate for the non-transformative RNAi technology. We here propose that the current legislation needs rewording to take account of the non-transgenic RNAi technology, and discuss the best alternative for regulatory systems in India and Malaysia in comparison with the existing frameworks in other countries.

Abstract:Annually, the cost of insect pest control in agriculture crosses billions of dollars around the world. Until recently, broad-spectrum synthetic pesticides were considered as the most effective means of pest control in agriculture. However, over the years, the overreliance on pesticides has caused adverse effects on beneficial insects, human health and the environment, and has led to the development of pesticide resistant insects. There is a critical need for the development of alternative pest management strategies aiming for minimum use of pesticides and conservation of natural enemies for maintaining the ecological balance of the environment. Host plant resistance plays a vital role in integrated pest management but the development of insect-resistant varieties through conventional ways of host plant resistance takes time, and is challenging as it involves many quantitative traits positioned at various loci. Biotechnological approaches such as gene editing, gene transformation, marker-assisted selection etc. in this direction have recently opened up a new era of insect control options. These could contribute towards about exploring a much wider array of novel insecticidal genes that would otherwise be beyond the scope of conventional breeding. Biotechnological interventions can alter the gene expression level and pattern as well as the development of transgenic varieties with insecticidal genes and can improve pest management by providing access to novel molecules. This review will discuss the emerging biotechnological tools available to develop insect-resistant engineered crop genotypes with a better ability to resist the attack of insect pests.

Abstract:With the emergence of transgenic technologies, new ways to improve the agronomic performance of crops for food, feed, and processing applications have been devised. In addition, ability to express foreign genes using transgenic technologies has opened up options for producing large quantities of commercially important industrial or pharmaceutical products in plants. Despite this high adoption rate and future promises, there is a multitude of concerns about the impact of genetically modified (GM) crops on the environment. Potential contamination of the environment and food chains has prompted detailed consideration of how such crops and the molecules that they produce can be effectively isolated and contained. One of the reasonable steps after creating a transgenic plant is to evaluate its potential benefits and risks to the environment and these should be compared to those generated by traditional agricultural practices. The precautionary approach in risk management of GM plants may make it necessary to monitor significant wild and weed populations that might be affected by transgene escape. Effective risk assessment and monitoring mechanisms are the basic prerequisites of any legal framework to adequately address the risks and watch out for new risks. Several agencies in different countries monitor the release of GM organisms or frame guidelines for the appropriate application of recombinant organisms in agro-industries so as to assure the safe use of recombinant organisms and to achieve sound overall development. We feel that it is important to establish an internationally harmonized framework for the safe handling of recombinant DNA organisms within a few years.

Abstract:Global demand for food is increasing day by day due to an increase in population and shrinkage of the arable land area. To meet this increasing demand, there is a need to develop high-yielding varieties that are nutritionally enriched and tolerant against environmental stresses. Various techniques are developed for improving crop quality such as mutagenesis, intergeneric crosses, and translocation breeding. Later, with the development of genetic engineering, genetically modified crops came up with the transgene insertion approach which helps to withstand adverse conditions. The process or product-focused approaches are used for regulating genetically modified crops with their risk analysis on the environment and public health. However, recent advances in gene-editing technologies have led to a new era of plant breeding by developing techniques including site-directed nucleases, zinc finger nucleases, and the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR associated protein 9 (Cas9) that involve precise gene editing without the transfer of foreign genes. But these techniques always remain in debate for their regulation status and public acceptance. The European countries and New Zealand, consider the gene-edited plants under the category of genetically modified organism (GMO) regulation while the USA frees the gene-edited plants from such type of regulations. Considering them under the category of GMO makes a long and complicated approval process to use them, which would decrease their immediate commercial value. There is a need to develop strong regulatory approaches for emerging technologies that expedite crop research and attract people to adopt these new varieties without hesitation.

Abstract:Genetic engineering has emerged as a powerful tool in the field of horticulture to enhance the traits of crops, leading to improved yield, quality, and resistance to biotic and abiotic stresses. This comprehensive review explores the recent advancements in genetic engineering techniques applied to horticulture crops, providing a detailed overview of the innovative strategies employed to manipulate plant genomes. The review begins by discussing the evolution of genetic engineering in horticulture, highlighting key milestones and breakthroughs that have paved the way for current advancements. It covers a range of techniques such as CRISPR-Cas9, RNA interference, and synthetic biology, shedding light on their applications in modifying specific genes responsible for desired traits. The main focus of the review is on the enhancement of key horticultural traits, including disease resistance, pest resistance, abiotic stress tolerance, and post-harvest attributes. Examples from various horticultural crops, such as fruits, vegetables, and ornamental plants, illustrate the success stories and potential applications of genetic engineering in each category. The ethical and regulatory aspects of genetic engineering in horticulture are also explored, addressing concerns related to environmental impact, biodiversity, and consumer acceptance. Host-Delivered RNA Interference for Durable Pest Resistance in Plants: Advanced Methods, Challenges, and Applications

Abstract:Insect-pests infestation greatly affects global agricultural production and is projected to become more severe in upcoming years. There is concern about pesticide application being ineffective due to insect resistance and environmental toxicity. Reduced effectiveness of Bt toxins also made the scientific community shift toward alternative strategies to control devastating agricultural pests. With the advent of host-delivered RNA interference, also known as host-induced gene silencing, targeted insect genes have been suppressed through genetic engineering tools to deliver a novel insect-pest resistance strategy for combating a number of agricultural pests. This review recapitulates the possible mechanism of host-delivered RNA interference (HD-RNAi), in particular, the silencing of target genes of insect-pests. We emphasize the development of the latest strategies against evolving insect targets including designing of artificial microRNAs, vector constructs, and the benefit of using plastid transformation to transform target RNA-interfering genes. Advantages of using HD-RNAi over other small RNA delivery modes and also the supremacy of HD-RNAi over the CRISPR-Cas system particularly for insect resistance have been described. However, the broader application of this technology is restricted due to its several limitations. Using artificial miRNA designs, the host-delivered RNAi + Bt combinatorial approach and chloroplast transformation can overcome limitations of RNAi. With careful design and delivery approaches, RNAi promises to be extremely valuable and effective plant protection strategy to attain durable insect-pest resistance in crops.

Abstract:The changing agro-climatic conditions in India largely intensify the incidence of various insect pests and their associated losses. Pests, particularly insects, diseases, and weeds, are threatening the productivity of crops farmed for human consumption. In order to minimise their infestation, a wide range of pesticides are being used to save the crops. However, traditional insect control techniques, such as chemicals, are expensive, labor-intensive, and require repeated application. Their injudicious use over the years has resulted in problems like pest resurgence, secondary pest outbreaks, pesticide resistance, and environmental pollution, which have all deteriorated human health. Because of the disadvantages of traditional chemical control approaches, biological control of insect pests is increasingly being considered and applied as a safe and cost-effective solution to the invasive plant problem. In the modern era, new tools such as genome editing, including CRISPR-CAS technology for natural enemy native DNA modification, combined with other traditional approaches such as natural enemy importation and release, can improve the efficacy and durability of bio-control research and technology. The global perspective of biological control presents an overview of many approaches and new innovative technologies being developed in different parts of the world to manage insect pests at an acceptable level.
