A novel animal model of a spinal cord injury platform

Abstract:Objective:Using the novel spinal cord injury platform (Feng’s Standard 1st Generation spinal cord injury platform, FSI) and minimally invasive operation, to establish standardized, significant injury grading, low complications and highly reproducible criteria for traumatic spinal cord injury.Methods:A total of 69 C57BL/6 mice, randomly numbered, were divided into three injury groups (
n=12 each): mild, moderate and severe, and control group (
n=9). The T9 was positioned by 3 different methods. The mouse spinal cord injury model was established using a coaxial spinal cord contusion platform and the forces generated by the hits at different heights (Kdyn) were recorded before the experiment. The hind limb mobility of mice was observed using behavioral test on days 1, 14, 28, and 42 after injury, while histopathological changes were observed simultaneously. The sample means of each group were compared using ANOVA and multiple rate comparisons were analyzed using the chi-square test.
Results:The results of force testing suggested that the coaxial spinal contusion platform had high hitting accuracy, with forces of (35.7±1.0), (55.0±1.3), and (72.3±1.7) Kdyn generated after landing at 20, 35, and 50 mm altitude, respectively. Accurate variability was high among the 3 methods of spinal cord stage localization (

P<0.01). The BMS score suggested a statistical difference in the degree of injury among the experimental groups (

P<0.01), severe versus moderate injury group:
χ2=313.500,
P<0.05; severe versus mild injury group:
χ2=274.500,
P<0.01. The results of the slant plate test suggested a statistical difference between the moderately and severely injured groups and the normal and mildly injured groups (

P<0.05). The pathological results showed that the mice in each group had significant differences in different time periods (
F=682.600,
P<0.01).
Conclusion:Coaxial spinal cord contusion platform combined with the minimally invasive method to create C57BL/6 mouse traumatic spinal cord injury model can complete the experimental process with simple steps, accurate and reproducible strike intensity.
