Study of Association Between Nut Traits and SCOT Molecular Marker in Some Old Iranian Walnut Trees

Abstract:Association analysis study correlates molecular and morphological data and provides important practical genetic information to the plant breeder. To identify markers related to 14 nut quantitative traits of walnuts including morphometric (nut length, nut width, and nut thickness; kernel width, kernel length, and kernel thickness; shell thickness; packing tissue thickness; packing tissue length) and weight (kernel weight, nut weight, shell weight, packing tissue weight, and kernel percentage) traits in 45 old Iranian walnut genotype, 16 SCOT primers were used. Association analysis between SCOT molecular data (as independent variables) and quantitative data (as dependent variable) was performed using stepwise regression separately for both years of the experiment. In total, out of the 243 loci in the first year and 302 loci in the second year, 40 common loci related to quantitative traits in both years entered the regression model. The highest numbers of common significant loci in the 2 years were related to the kernel weight and kernel width, with 10 and 9 loci, respectively. Nut width, thickness, and weight, as well as the percentage of the kernel, had the least number of loci. The thickness and weight of packing tissue had one locus. The locus SCOT31-1200 was related to nut thickness, kernel width, and kernel weight, SCOT30-2000 was related to kernel length and nut weight, SCOT7-1100 was related to kernel length and packing tissue weight, SCOT34-500 was related to kernel width and kernel thickness, SCOT15-800 was related to kernel weight and packing tissue thickness, and SCOT31-1300 was related to shell thickness and shell weight. These relationships enabled the selection of several traits at the same time. The highest R 2 value was observed in relation to nut length (SCOT17-900), kernel length (SCOT7-1100), kernel width (SCOT16-1400 and SCOT32-1000), kernel thickness (SCOT13-500), shell thickness (SCOT34-1200), kernel weight (SCOT35-1200, SCOT34-600, SCOT14-1400), and shell weight (SCOT31-1300). Since these relationships were most effective in marker-assisted coding of the traits, they can be used in walnut breeding programs.
