Abstract:Abstract Insertion/Deletion (InDel) polymorphisms have smaller amplicons, lower mutation rates, and facilitate the popularization and application of the existing capillary electrophoresis (CE) platform of forensic laboratories. Given its rich and diverse resource of human genetics, the multi-ethnic region of southwest China’s Guizhou is an important area for studying the genetic structure and origin of ethnic groups and human evolution. However, DNA data and the sparse sampling of present-day populations are lacking, especially in relation to the InDel markers. Here, we report data on 47 autosomal InDels from 592 individuals from four populations in Guizhou, Han, Dong, Yi, and Chuanqing. Genotyping was performed with the AGCU InDel 50 kit to evaluate their utility for forensic purposes and to explore the population genetic structure of the studied populations. First, we found no significant deviations from Hardy–Weinberg equilibrium and linkage equilibrium at the 47 autosomal InDels in the studied populations after Bonferroni correction. The combined powers of discrimination for the Han, Dong, Yi, and Chuanqing populations were 0.999999999999999999757 9, 0.999999999999999999644 9 and 0.999999999999999999778 7, 0.999999999999999999582 3, respectively, and the combined powers of exclusion were 0.999759 9, 0.999562 5, 0.999746 3 and 0.999745 9, respectively. These findings indicated that the AGCU InDel 50 kit could be applied to forensic personal identification and was an effective supplement for parentage testing in the studied populations. A variety of genetic structure analysis results for principal component analysis, multidimensional scaling, genetic distance, STRUCTURE, and phylogenetic analysis, revealed that genetic proximity of the studied populations correlates with language, geography, and culture. The differences among the studied populations were smaller than the differences between populations in different regions. Guizhou Han, Dong, and Chuanqing were better clustered with the linguistically close populations than Guizhou Yi.
