Abstract:Deep eutectic solvent-aqueous two-phase systems (DES-ATPSs) have been greatly studied in the research field for downstream bioprocessing since 2014. This is mostly due to their merits such as ease of synthesis, excellent solubility for most solutes, low toxicity, and many more. This chapter aims to provide a comprehensive review of recent literature focusing on sugar-based DES-ATPS, where its working principle, synthesis process, and characterization method were detailed. The applications and advancement of sugar-based DES-ATPS in the separation and purification of various biomolecules were extensively discussed. Five different parameters consisting of mass fraction of DES, type of hydrogen bond acceptor (HBA) and hydrogen bond donor (HBD), HBA to HBD ratios, temperature, and type of phase forming agents were highlighted to elucidate their correlation to the performance of sugar-based DES-ATPS. Furthermore, insights into the recycling of DES for consequent extractions were provided.

Abstract:Aqueous two-phase systems (ATPSs) are formed by the phase separation of immiscible aqueous solutions with above-critical concentrations. Particularly, ATPS emulsions have been demonstrated to exhibit significant potential in a wide variety of biomedical applications because of their remarkable permeability, affinity partitioning, and biocompatibility. In this review, we provide comprehensive insights into ATPS emulsion droplets, covering their unique properties and fabrication methods. In addition, we summarize ATPS droplet-derived functional materials, with emphasis on protocells and microparticles, and present their applications. We believe that this review will highlight the unique characteristics of ATPS droplets towards more widespread application.

Abstract:The aqueous two-phase system (ATPS) is an all-aqueous system fabricated from two immiscible aqueous phases. It is spontaneously assembled through physical liquid-liquid phase separation (LLPS) and can create suitable templates like the multicompartment of the intracellular environment. Delicate structures containing multiple compartments make it possible to endow materials with advanced functions. Due to the properties of ATPSs, ATPS-based drug delivery systems exhibit excellent biocompatibility, extraordinary loading efficiency, and intelligently controlled content release, which are particularly advantageous for delivering drugs in vivo . Therefore, we will systematically review and evaluate ATPSs as an ideal drug delivery system. Based on the basic mechanisms and influencing factors in forming ATPSs, the transformation of ATPSs into valuable biomaterials is described. Afterward, we concentrate on the most recent cutting-edge research on ATPS-based delivery systems. Finally, the potential for further collaborations between ATPS-based drug-carrying biomaterials and disease diagnosis and treatment is also explored.

Affinity-Controlled Partitioning of Biomolecules at Aqueous Interfaces and Their Bioanalytic Applications

Abstract:All-aqueous phase separation systems play essential roles in bioanalytical and biochemical applications. Compared to conventional oil and organic solvent-based systems, these systems are characterized by their rich bulk and interfacial properties, offering superior biocompatibility. In particular, phase separation in all-aqueous systems facilitates the creation of compartments with specific physicochemical properties, and therefore largely enhances the accessibility of the systems. In addition, the all-aqueous compartments have diverse affinities, with an important property known as partitioning, which can concentrate (bio)molecules toward distinct immiscible phases. This partitioning affinity imparts all-aqueous interfaces with selective permeability, enabling the controlled enrichment of target (bio)molecules. This review introduces the basic principles and applications of partitioning-induced interfacial phenomena in a typical all-aqueous system, namely aqueous two-phase systems (ATPSs); these applications include interfacial chemical reactions, bioprinting, and assembly, as well as bio-sensing and detection. The primary challenges associated with designing all-aqueous phase separation systems and several future directions are also discussed, such as the stabilization of aqueous interfaces, the handling of low-volume samples, and exploration of suitable ATPSs compositions with the efficient protocol.

Abstract:Abstract Aqueous two-phase systems (ATPSs) are liquid–liquid equilibria between two aqueous phases that usually contain over 70% water content each, which results in a nontoxic organic solvent-free environment for biological compounds and biomolecules. ATPSs have attracted significant interest in applications for formulating carriers (microparticles, nanoparticles, hydrogels, and polymersomes) which can be prepared using the spontaneous phase separation of ATPSs as a driving force, and loaded with a wide range of bioactive materials, including small molecule drugs, proteins, and cells, for delivery applications. This review provides a detailed analysis of various ATPSs, including strategies employed for particle formation, polymerization of droplets in ATPSs, phase-guided block copolymer assemblies, and stimulus-responsive carriers. Processes for loading various bioactive payloads are discussed, and applications of these systems for drug delivery are summarized and discussed.

Abstract:Aqueous two-phase systems (ATPS), also known as Aqueous Biphasic Systems (ABS), have been extensively studied as platforms for the separation and purification of biomolecules and other valuable compounds. These liquid–liquid extraction (LLE) systems have been a tool for biotechnology since its origin (Albertsson, 1950's), recently expanding to exciting fields such as health, biomedicine and material sciences. Due to their biocompatibility, amenability, flexibility, and versatility, ATPS have been applied across various research areas, addressing many challenges associated with conventional methodologies. In this feature article, we first discuss the fundamentals of ATPS and the molecular mechanisms that govern their formation and are crucial for their application. We then explore the most prominent and innovative applications of these systems in downstream processing. Additionally, we provide insights into the design of in situ upstream–downstream integrated platforms, and their use as pre-treatment and analytical tools. The latest advancements in ATPS applications within disruptive bioengineering and biotechnology fields are presented, along with their pioneering use in emerging scientific areas, such as the formation of all-aqueous (water-in-water) emulsions, microfluidic systems, and membrane-free batteries. Overall, this work underscores the transformative potential of ATPS in various branches of science, pinpointing directions for future research to fully explore and maximize ATPS capabilities, overcome existing hurdles, and drive innovation forward.

Abstract:Biomolecules, specifically proteins, polysaccharides, and secondary metabolites are potential lead compounds due to their remarkable pharmacological properties. However, the complex molecular structure of the biomolecules makes their separation processes of great challenges. The conventional downstream processes require multistep protocols that are less efficient, high solvent consumption, expensive, time-consuming, and laborious. Hence, aqueous two-phase system (ATPS) is a reliable technique for the extraction and purification of biomolecules from a complex mixture. ATPS is an environmentally friendly, simple, cost effective, and easily scalable process. It requires a short processing time to separate biomolecules of industrial values simultaneously in a single process. Modifications have also been performed by introducing deep eutectic solvents, ionic liquids, carbohydrates, amino acids or copolymers to enhance the process efficiency with an increased yield, purity and bioactivity of recovered biomolecules. This review attempts to review the recent developed ATPSs and their efficiency to extract, isolate, and purify biomolecules such as proteins, polysaccharides, secondary metabolites and other biological substances. The review provides insights into the feasibility and reliability of ATPS for biomolecule recovery.

Abstract:Over the last years, aqueous two-phase systems (ATPS) regained an increasing interest due to their potential in the downstream processing of biomolecules. After many years with only a few articles published, a lot of effort and work has been put into studying these systems for the partitioning of a range of compounds including proteins, organic low-molecular weight molecules or metal ions. Although several research and review articles appeared, a background review on ATPS partitioning fundamentals is needed. In this article, partitioning theories and main effects of several important factors for partitioning, such as molecular weight of the polymer, effect of added salts, pH, electrical charges, and temperature on phase diagrams, tie-line lengths, interfacial tension and settling time of the two aqueous phases are extensively reviewed. The trend in ATPS research is given compiling the recent 2008–2013 research articles published in the field.

Abstract:Superhydrophilic–superhydrophobic patterned surfaces constitute a branch of surface chemistry involving the two extreme states of superhydrophilicity and superhydrophobicity combined on the same surface in precise patterns. Such surfaces have many advantages, including controllable wettability, enrichment ability, accessibility, and the ability to manipulate and pattern water droplets, and they offer new functionalities and possibilities for a wide variety of emerging applications, such as microarrays, biomedical assays, microfluidics, and environmental protection. This review presents the basic theory, simplified fabrication, and emerging applications of superhydrophilic–superhydrophobic patterned surfaces. First, the fundamental theories of wettability that explain the spreading of a droplet on a solid surface are described. Then, the fabrication methods for preparing superhydrophilic–superhydrophobic patterned surfaces are introduced, and the emerging applications of such surfaces that are currently being explored are highlighted. Finally, the remaining challenges of constructing such surfaces and future applications that would benefit from their use are discussed.

Abstract:Double emulsions have been extensively used in scientific researches and industrial applications due to their attractive unique feature of multiple phases. However, constructing droplets with such a complex structure is not a simple task for all time. The simultaneous existence of two contradictory interfaces makes it hard to prepare stable double emulsions in principle and in practice. Over the past century, tremendous efforts have been devoted by myriads of scientists to make progresses in both theory and preparation of double emulsions. In this review, the current understanding of double emulsions is systematically revealed. In addition to emphasizing the corresponding pioneer and landmark works as many as possible, the state-of-the-art achievements will also be discussed. By regulating the oil-water interface with smartly designed interface-active agents in combination with varying the phase volume fractions, the basic theory framework based on the phase inversion from simple emulsions to double emulsions is also summarized. Technical preparation strategies of emulsification are introduced to show the building process of the two contradictory interfaces in one system. Furthermore, some specific biomedical applications of double emulsions are also discussed, which is expected to stimulate further innovation and utilization of double emulsions.

Abstract:In recent years, the increasing need for energy conservation and environmental protection has driven industries to explore more efficient and sustainable processes. Liquid-liquid extraction (LLE) is a common method used in various sectors for separating components of liquid mixtures. However, the traditional use of toxic solvents poses significant health and environmental risks, prompting the shift toward green solvents. This review deals with the principles, applications, and advantages of aqueous two-phase systems (ATPS) as an alternative to conventional LLE. ATPS, which typically utilize water and nontoxic components, offer significant benefits such as high purity and single-step biomolecule extraction. Abstract:In this study, Janus droplets are fabricated by inducing phase separation of single‐phase droplets made up of an aqueous two‐phase system (ATPS). The resultant Janus droplets are highly monodisperse in structures and internal morphologies. Due to the affinity partitioning properties of ATPS, encapsulated ingredients can automatically partition into different compartments of the resultant Janus droplets. The use of biocompatible and cytocompatible ATPS also enables the encapsulation of enzymes and cells in the resultant droplets with their activity preserved at a relatively high level. image

Abstract:This review covers the analytical applications of protein partitioning in aqueous two-phase systems (ATPSs). We review the advancements in the analytical application of protein partitioning in ATPSs that have been achieved over the last two decades. Multiple examples of different applications, such as the quality control of recombinant proteins, analysis of protein misfolding, characterization of structural changes as small as a single-point mutation, conformational changes upon binding of different ligands, detection of protein–protein interactions, and analysis of structurally different isoforms of a protein are presented. The new approach to discovering new drugs for a known target (e.g., a receptor) is described when one or more previous drugs are already available with well-characterized biological efficacy profiles.

Abstract:Aqueous biphasic system (ABS) is widely used in the recovery, extraction, purification and separation of proteins, enzymes, nucleic acids and antibodies. The ABS with high water content and low interfacial tension offers a biocompatible environment for the recovery of labile biomolecules. Process integration can be achieved using ABS by incorporating multiple-steps of purification, concentration and purification of biomolecules in a single-step operation which often results in high product recovery yield and purity. Conventional ABS is usually formed by aqueous solutions of two polymers or a polymer and a salt above a critical concentration. The high viscosity of polymer-based ABS causes slow phase separation and hinders the mass transfer of biomolecules, whereas polymer/salt ABS is characterized by high ionic strength resulting in the loss of bioactivity of recovered biomolecules. These limitations have encouraged the development of novel ABS which is more cost-effective for various biotechnological applications. This review discusses the characteristics and mechanisms of several types of emerging unconventional ABS using phase-forming components such as hyperbranched polymers, special salts, surfactants, magnetic fields, the addition of nanoparticles and incorporation of various solvent. Moreover, several novel applications of ABS for different separation purposes such as microfluidic-based ABS, ABS bioreactors, application of ABS as an analytical tool, and ABS micropatterning are discussed in this review. In the last section of this review, a comprehensive summary of process integration using ABS for extractive fermentations, bioconversion, crystallization and precipitation is also supplemented for the comprehensive review of various types and applications of ABS in recent years.

Aqueous two-phase system based adherent droplet microfluidics for enhanced enzymatic reaction

Abstract:Utilizing the aqueous two-phase system（ATPS） based droplet microfluidics, a highly efficient enzymatic reaction platform has been developed that takes the advantage of the properties of the ATPS adherent droplet microfluidics including rapid mass transfer, efficient mixing, and reaction-separation coupling. The proposed platform utilizes droplet microfluidics, overcoming the limitation of low mass transfer rate and the high time/energy consumption of the traditional bulk ATPS. A wall-mounted droplet microreactor is established, further enhancing mass transfer due to the creation of a larger vortex flow. In addition, the ATPS droplet with the ability of molecular containing and selective enzyme and product partitioning are investigated. By comparing the enzymatic reaction effects of two types of droplet microreactors with and without microchannel walls, it is found that the conversion of droplet microreactors with microchannel walls can reach 40% in only 6 min. It is 9.4 times higher than that of the droplet microreactor not attached to the wall of the microchannel. Therefore, this study paves a new way to enhance the enzyme-catalyzed reaction processes at the microscale through the ATPS droplet microfluidic technique.
Phase Separation of Parallel Laminar Flow for Aqueous Two Phase Systems in Branched Microchannel

Abstract:Aqueous two phase systems (ATPSs) have good biocompatibility and special selectivity. Their phase equilibrium and applications in biological analysis have received much attention. Herein, parallel laminar flow (PLF) in the microchannel can provide an effective platform to enhance mass transfer and preserve separate phases simultaneously. As fundamentals in feasible and convenient sampling of PLF for ATPS, the phase separation methods and rules in branched microchannel were studied in this work, selecting PEG 4000 + Na2SO4 + H2O as a model system. The exploration of flow pattern showed that a stable PFL was easily to form in the shallow microchannel of 200 μm (depth) × 600 μm (width), as long as the velocity of lower phase was higher than 0.51 mm/s. The phase interface of PLF could be easily controlled by the flow ratio of two phases. Single-phase separation could be reliably achieved in T-junction outlets when the flow rate of outlet ascertains to be smaller or larger than that of inlet on the same side. The trifurcate outlets with an extra middle channel could help realize a simultaneous two-phase separation. The flow rate of the extra channel is the key for the phase separation performance, the range of which available for simultaneous two-phase separation is determined by the resistance balance and the flow rates deviation offsetting as well. It is favor for increasing phase separation efficiency to make the products of flow rate, viscosity, and the length of corresponding outlet channel close with each other for the upper phase and the lower phase. The adjustable lengths of three channels can provide flexible choices to enhance simultaneous two-phase separation of diversified ATPSs at various operating flow ratios. A multiport microchip with T-junction inlets and trifurcate outlets designed for adjusting the lengths of branched channels on-chip is a convenient tool for PLF contact and in situ phase separation of ATPSs in varieties of application.

Abstract:This review article provides an overview of the fundamental background and application of aqueous two-phase systems (ATPSs) in protein and enzyme extraction. The types of ATPSs, including polymer/salt, polymer/polymer, alcohol/salt, surfactant-based, and ionic liquid-based ATPSs, are discussed, along with the unconventional ATPSs. Factors affecting partitioning in ATPSs, such as molecular weight and polymer concentration, pH, temperature, hydrophobicity, and affinity, are also examined. The article then focuses on the application of ATPSs in protein and enzyme extraction, including continuous processing and scaling-up. The future prospects, challenges, and limitations of ATPSs in this field are discussed, along with the challenges associated with their use in industry. The results section highlights the potential of citrate green salts as an alternative to sulfate and phosphate salts in salt-based ATPSs and the need for more research on using ionic liquids as an additive in ATPS types for protein and enzyme extraction. Overall, this review suggests combining cheap and environmentally friendly materials in ATPSs can be a practical solution for using ATPSs in the industry.

Abstract:Liquid-liquid segmented flows in microchannels have been extensively investigated in the context of nanoparticle synthesis. The enhanced mixing in the slugs results in monodispersed particles. Earlier studies have focused on Organic-Aqueous Systems (OAS). The nanoparticles synthesized in the presence of organic solutions have limited applications. An alternative green route for the synthesis can be developed using an Aqueous Two-Phase System (ATPS). These systems are characterized by interfacial tensions, which are two orders of magnitude lower than typical organic aqueous systems. In this work, flow patterns and hydrodynamics of ATPS are investigated as a first step. Polyethylene glycol -trisodium citrate system was chosen as ATPS. The objective of this work is to see if any new physics arises in an ATPS system. The low interfacial tension results in high Capillary numbers (Ca >> 3) in a microfluidic system. Consequently, the flow observed here is parallel or core-annular. However, in a millichannel, the capillary number becomes lower (Ca << 1) for an ATPS system. In this work, experiments were carried out in a millichannel to span different flow patterns. The pattern formation was analyzed and classified into three categories, i.e., slug flow (interfacial tension dominated), transition flow, and core annular flow (inertia dominated). Flow regime maps based on the Reynolds number, Capillary number, and Weber number of each phase were found to be qualitatively similar to those of OAS. Simulations were performed for various interfacial tension values. An interfacial tension value of 1.25x10-4 N/m was found to yield slug sizes which fitted well with the experimental data. Film thickness was measured experimentally and with simulations compared favorably with the correlations available in the literature for OAS.

Among various emulsion forming methods, microfluidic technology, with good monodispersity, high controllability and operation simplicity, has been widely used in the preparation of multiphase emulsions with different systems. This review would focus on the basic principles of forming multiphase emulsions, the recent progress in controlling multiphase flow in microfluidics, and preparation of functional materials with microfluidics mainly by the authors’ research group. We believe that the review will contribute to the readers in this prospective area very well.

Abstract:Energy conversion and harvesting technologies primarily rely on interface interactions at the liquid–liquid and liquid–solid interfaces. The efficient functioning of these technologies can be achieved through interface engineering, which regulates mass transfer and charge transportation. Several strategies have been employed to control the properties of the interface and enhance energy conversion efficiency. Significant progress has been made in understanding reactions at the liquid interface, opening new opportunities for its application. This review summarizes recent advancements in liquid interface manipulation methods, such as structured liquids and ultra-stable emulsions, and various liquid interface-based applications, including hydrovoltaic, triboelectric nanogenerators, all-liquid batteries, and water desalination, offering insights into the understanding of the complicated liquid interface and the reactions happening at liquid interfaces.
