Abstract:Brown adipose tissue (BAT) is a heater organ that expresses thermogenic uncoupling protein 1 (UCP1) to maintain high body temperatures during cold stress. BAT thermogenesis is considered an overarching mammalian trait, but its evolutionary origin is unknown. We show that adipose tissue of marsupials, which diverged from eutherian mammals ~ 150 million years ago, expresses a nonthermogenic UCP1 variant governed by a partial transcriptomic BAT signature similar to that found in eutherian beige adipose tissue. We found that the reconstructed UCP1 sequence of the common eutherian ancestor displayed typical thermogenic activity, whereas therian ancestor UCP1 is nonthermogenic. Thus, mammalian adipose tissue thermogenesis may have evolved in two distinct stages, with a prethermogenic stage in the common therian ancestor linking UCP1 expression to adipose tissue and thermal stress. We propose that in a second stage, UCP1 acquired its thermogenic function specifically in eutherians, such that the onset of mammalian BAT thermogenesis occurred only after the divergence from marsupials.

Abstract:The epidemic of obesity and type 2 diabetes has increased interest in pathways that affect energy balance in mammalian systems. Brown fat, in all of its dimensions, can increase energy expenditure through the dissipation of chemical energy in the form of heat, using mitochondrial uncoupling and perhaps other pathways. We discuss here some of the thermodynamic and cellular aspects of recent progress in brown fat research. This includes studies of developmental lineages of UCP1+ adipocytes, including the discovery of beige fat cells, a new thermogenic cell type. We also discuss the physiology and transcriptional control of brown and beige cells in rodents and the state of current knowledge about human brown fat.

Abstract:Mammalian cellular machinery has evolved to function best at ~37 °C, making maintenance of core body temperature (at ~37 °C) critical for optimal organ function. The physiological importance of thermoregulation has been appreciated for many years, but our mechanistic understanding of the diversity and functioning of thermogenic mechanisms has advanced considerably in the past century. A brief analysis by Prusiner and Poe (published in 1968) marked a pivotal shift in thermoregulation research, transitioning from a coarse physiological perspective to biochemical interpretations of thermogenic pathways. In the thermoneutral zone, core body temperature is sustained with minimal metabolic effort, facilitated by anatomical features and baseline metabolism. The thermoneutral zone is a subjective temperature range that might be around 20–22 °C for a lean, lightly clothed man. When environmental temperatures drop below the thermoneutral zone, the body activates a series of thermoregulatory mechanisms to preserve core temperature and re-establish thermal homeostasis. Physiologically, thermogenic mechanisms are classified as shivering thermogenesis, which occurs in muscle, and non-shivering thermogenesis (NST). NST comprises a group of processes that use different energy sources and mechanisms that ultimately converge in the release of heat. Prusiner and Poe distinguished the molecular mechanisms of thermogenic pathways and proposed a classification scheme based on the biochemical origins of metabolic heat rather than a one based on physiology. In addition, several other suggestions presented in their paper have had lasting implications for ongoing research on futile cycles and thermogenesis. Specifically, the authors highlighted two points that remain relevant for contemporary studies: first, the relative thermogenic contribution of ATP synthesis versus ATP hydrolysis, and second, the thermogenic role of the ATP-consuming futile cycle.

Thermal Adaptations in Animals: Genes, Development, and Evolution

Abstract:Thermal adaptation to environmental temperature is a driving force in animal evolution. This chapter presents thermal adaptation in ectotherms and endotherms from the perspective of developmental biology. In ectotherms, there are known examples of temperature influencing morphological characteristics, such as seasonal color change, melanization, and sex determination. Furthermore, the timing of embryonic development also varies with environmental temperature. This review will introduce the cellular and molecular mechanisms underlying temperature-dependent embryogenesis. The evolution of thermal adaptation in endotherms is also important for survival in cold climates. Recent genome-wide studies have revealed adaptive mutations in the genomes of extant humans as well as extinct species such as woolly mammoths and Neanderthals. These studies have shown that single-nucleotide polymorphisms in physiologically related genes (e.g., CPT1A, LRP5, THATA, PRKG1, and FADS1-3) allow humans to live in cold climates. At the end of this chapter, we present the remaining questions in terms of genetic assimilation, heat shock protein Hsp90, and embryonic development.

Abstract:This is a comparative survey embracing basal metabolism measurements of all adult animal species studied at the Nutrition Laboratory. Considerable space is devoted to a painstaking consideration of the conditions necessary for comparative measurements of heat production, both intraspecific and interspecific, with analyses of the difficulties involved and the criteria employed. Data are presented for the various warm-blooded species on the basis of twenty-four hour heat production with reference to body weight. Then interspecific comparisons are made, expressed in terms of total heat production per kilogram of body weight and per square meter of surface area. Abstract:Brown adipose tissue (BAT) is responsible for regulating body temperature through adaptive thermogenesis. The ability of thermogenic adipocytes to dissipate chemical energy as heat counteracts weight gain and has gained considerable attention as a strategy against obesity. BAT undergoes major remodeling in a cold environment. This remodeling results from changes in the number and function of brown adipocytes, expanding the network of blood vessels and sympathetic nerves, and changes in the composition and function of immune cells. Such synergistic adaptation requires extensive crosstalk between individual cells in the tissue to coordinate their responses. To understand the mechanisms of intercellular communication in BAT, we apply the CellChat algorithm to single-cell transcriptomic data of mouse BAT. We construct an integrative network of the ligand-receptor interactome in BAT and identify the major signaling inputs and outputs of each cell type. By comparing the ligand-receptor interactions in BAT of mice housed at different environmental temperatures, we show that cold exposure enhances the intercellular interactions among the major cell types in BAT, including adipocytes, adipocyte progenitors, lymphatic and vascular endothelial cells, myelinated and non-myelinated Schwann cells, and immune cells. These interactions are predicted to regulate the remodeling of the extracellular matrix, the inflammatory response, angiogenesis, and neurite growth. Together, our integrative analysis of intercellular communications in BAT and their dynamic regulation in response to housing temperatures provides a new understanding of the mechanisms underlying BAT thermogenesis. The resources presented in this study offer a valuable platform for future investigations of BAT development and thermogenesis.

Abstract:Adaptive thermogenesis by brown adipose tissue (BAT) dissipates calories as heat, making it an attractive anti-obesity target. Yet how BAT contributes to circulating metabolite exchange remains unclear. Here, we quantified metabolite exchange in BAT and skeletal muscle by arteriovenous metabolomics during cold exposure in fed male mice. This identified unexpected metabolites consumed, released and shared between organs. Quantitative analysis of tissue fluxes showed that glucose and lactate provide ~85% of carbon for adaptive thermogenesis and that cold and CL316,243 trigger markedly divergent fuel utilization profiles. In cold adaptation, BAT also dramatically increases nitrogen uptake by net consuming amino acids, except glutamine. Isotope tracing and functional studies suggest glutamine catabolism concurrent with synthesis via glutamine synthetase, which avoids ammonia buildup and boosts fuel oxidation. These data underscore the ability of BAT to function as a glucose and amino acid sink and provide a quantitative and comprehensive landscape of BAT fuel utilization to guide translational studies.

Abstract:Mammals are at the end of a gradual metabolic evolution in the course of which the step from anaerobic to aerobic cellular metabolism and the transition from water to land life formed the basis for an increase in metabolic rate (from brady- to tachymetabolism). The increased metabolic rate and the resulting endogenous heat production were the preconditions for enhanced long-term performance as well as for homeothermy which allowed mammals and birds to invade temperate zones. However, the underlying increase in membrane permeability also results in an increased energy demand (for membrane pump activity) which leads to the reduced hypoxia tolerance of mammals and requires a permanent substrate supply. As an adaptation to a seasonal discrepancy between increased thermoregulatory energy demand and decreased food supply, some small mammals apparently extended the newly evolved non-REM-sleep into hibernation. Mammalian hibernation is characterized by a profound metabolic reduction which is influenced by acidosis and limited to a tolerable degree by maintained thermoregulation. The lower limit of cooling seems to be determined by a critical minimal metabolic rate which is common to all mammals. The higher the normothermic metabolic rate, the lower is the temperature at which this minimal metabolic rate is reached. Since specific (i. e., weight-corrected) basal metabolic rate increases with decreasing body mass, small mammals exhibit a higher hypothermia tolerance than larger ones. On the other hand, the metabolic decrease to a uniform minimal level reflects an inactivation of the overall metabolic size relationship and, thus, forms a counterpart to the metabolic increase from a lower fetomaternal to the higher size-related level, occurring after birth. The postnatal metabolic increase which favours the onset of thermoregulation, parallels the increase in oxygen tension at the transition from fetal to adult circulation and, thus, probably enables mammalian neonates to readjust their metabolic needs in response to hypoxia. There is increasing evidence that, similar to the step from anaerobiosis to aerobiosis, the increase in metabolic rate resulting from any increase in oxygen supply is a general principle of evolution that, apart from its further adaptive benefits, protects tissues from oxygen excess and subsequent oxidative stress.

Abstract:The body of mammals harbors two distinct types of adipose tissue: while cells within the white adipose tissue (WAT) store surplus energy as lipids, brown adipose tissue (BAT) is nowadays recognized as the main tissue for transforming chemical energy into heat. This process, referred to as 'non-shivering thermogenesis', is facilitated by the uncoupling of the electron transport across mitochondrial membranes from ATP production. BAT-dependent thermogenesis acts as a safeguarding mechanism under reduced ambient temperature but also plays a critical role in metabolic and energy homeostasis in health and disease. In this review, we summarize the evolutionary structure, function and regulation of the BAT organ under neuronal and hormonal control and discuss its mutual interaction with the central nervous system. We conclude by conceptualizing how better understanding the multifaceted communicative links between the brain and BAT opens avenues for novel therapeutic approaches to treat obesity and related metabolic disorders.

Abstract:Thermogenic fat, consisting of brown and beige adipocytes, dissipates energy in the form of heat, in contrast to the characteristics of white adipocytes that store energy. Increasing energy expenditure by activating brown adipocytes or inducing beige adipocytes is a potential therapeutic strategy for treating obesity and type 2 diabetes. Thus, a better understanding of the underlying mechanisms of thermogenesis provides novel therapeutic interventions for metabolic diseases. In this review, we summarize the recent advances in the molecular regulation of thermogenesis, focusing on transcription factors, epigenetic regulators, metabolites, and non-coding RNAs. We further discuss the intercellular and inter-organ crosstalk that regulate thermogenesis, considering the heterogeneity and complex tissue microenvironment of thermogenic fat.

Abstract:In rodents, brown adipose tissue (BAT) is a metabolic organ that produces heat in response to cold and dietary intake through mitochondrial uncoupling. For long time, BAT was considered to be solely important in small mammals and infants, however recent studies have shown that BAT is also functional in adult humans. Interestingly, the presence and/or functionality of this thermogenic tissue is diminished in obese people, suggesting a link between human BAT and body weight regulation. In the last years, evidence has also emerged for the existence of adipocytes that may have an intermediate thermogenic phenotype between white and brown adipocytes, so called brite or beige adipocytes. Together, these findings have resulted in a renewed interested in (human) brown adipose tissue and pathways to increase the activity and recruitment of these thermogenic cells. Stimulating BAT hypertrophy and hyperplasia in humans could be a potential strategy to target obesity. Here we will review suggested pathways leading to BAT activation in humans, and discuss novel putative BAT activators in rodents into human perspective.

Abstract:Adipose tissue serves as the body’s primary energy storage site; however, findings in recent decades have transformed our understanding of the multifaceted roles of this adaptable organ. The ability of adipose tissue to undergo energy expenditure through heat generation is termed adaptive thermogenesis, a process carried out by thermogenic adipocytes. Adipocytes are the primary parenchymal cell type in adipose tissue, yet these cells are sustained within a rich stromal vascular microenvironment comprised of adipose stem cells and progenitors, immune cells, neuronal cells, fibroblasts, and endothelial cells. Intricate cross talk between these diverse cell types is essential in regulating the activation of thermogenic fat, and the past decade has shed significant light on how this intercellular communication functions. This review will draw upon recent findings and current perspectives on the sophisticated repertoire of cellular and molecular features that comprise the adipose thermogenic milieu.

Abstract:The advent of placentation and permanent homeothermy depended on major increases in metabolic rates and coincided with expansions in the number of microRNA families ( ). Since models of the evolution of brain size in the human lineage have not accounted for thermoregulatory constraints, we reparametrized rates of energy expenditure from log-based specific s to -based cellular s ( s). We demonstrate that mammalian brain and liver sizes vary inversely relative to a thermoregulatory Core (kidney+heart+liver+brain). Liver s also vary inversely with respect to body mass and the s of kidney, heart and brain, recapitulating the downregulation of gluconeogenesis during developmental growth of whole body and liver. With as a proxy for energy reliability, a logistic feedback model of relative brain size detected adaptation to a low-energy regime among the smallest primates and, among the largest primates, adaptation to the physiological limit on the rate of heat dissipation.

Abstract:Adipose tissue is a complex organ consisting of a mixture of mature adipocytes and stromal vascular cells. It displays a remarkable ability to adapt to environmental and dietary cues by changing its morphology and metabolic capacity. This plasticity is demonstrated by the emergence of interspersed thermogenic beige adipocytes within white depots in response to catecholamines secretion. Coordinated cellular interaction between different cell types within the tissue and a fine-tuned transcriptional program synergistically take place to promote beige remodeling. However, both cell-cell interactions and molecular mechanisms governing beige adipocyte appearance and maintenance are poorly understood. In this and the previous issue of Genes & Development, Shao and colleagues (pp. 1461-1474) and Shan and colleagues (pp. 1333-1338) advance our understanding of these issues and, in doing so, highlight potential therapeutic strategies to combat obesity-associated diseases.

Abstract:Hibernation has evolved several times in mammals to overcome harsh winter climates and food scarcity. During hibernation, animals undergo extreme shifts in metabolic rate, heart rate, respiration, and body temperature. These changes reduce energy consumption and allow animals to survive solely on their fat reserves. Understanding the mechanisms for these extreme shifts has long been recognized as a model for translational medicine as hibernators do not exhibit the same adverse effects of extended immobility that non-hibernating mammals suffer. Though work on individual species has illuminated important mechanisms of these functional changes, the genomic basis of this phenotype remains largely unknown, and few studies have drawn on comparative work to elucidate commonalities across diverse hibernating mammals. Synthesizing both single species and comparative approaches, we use metabolomic data from active and denning black bears (Ursus americanus) to guide bioinformatic analyses of genes using tests of selection and evolutionary rate convergence across independent lineages of hibernating mammals. We identify several genes with significant signatures of selection and evolutionary rate convergence in hibernators that represent both previously known and novel genetic mechanisms of the hibernation phenotype. These data provide novel insights into the genetic basis of this adaptation and serve to direct clinical research in hibernation-based therapies.

Abstract:Endothermy requires a source of endogenous heat production. In birds, this is derived primarily from shivering, but in mammals it is mostly non-shivering thermogenesis (NST). Brown adipose tissue (BAT) is a specialized tissue found in Eutherian mammals that is the source of most NST. Heat production in BAT depends primarily on the activity of uncoupling protein 1 (UCP1), which decouples transport of protons across the inner mitochondrial membrane from synthesis of ATP. UCP1 and hence heat production of BAT is regulated by many factors. In this paper we discuss the main factors activating UCP1 and increasing heat production. Probably the most well-known activator is the catecholamine norepinephrine (NE) which is released from sympathetic nerve endings and binds to adrenergic receptors that are abundantly expressed on BAT. NE stimulates release of free-fatty acids. It was previously thought that such FFAs were essential for activation of UCP1. However recent work has suggested intracellular lipolysis is not essential and FFAs can be derived from extracellular sources. Thyroid hormones also exert impacts on metabolic rate via effects on brown adipocytes which express type 2 deiodinase. Knocking out DIO2 makes mice cold intolerant. Parathyroid hormone appears to also be a potent regulator of BAT activity and may be an important mediator of elevated expenditure during cancer cachexia, although this is disputed by observations that cachexia wasting is not blunted in UCP1 KO mice. Cardiac natriuretic peptides have also been implicated in regulating BAT thermogenesis and the interconversion of beige adipocytes from their white to brown form. Activation of BAT thermogenesis may be an important component of the post-ingestion rise in heat production. Recent work suggests the gut derived hormone secretin may play a key role in this effect, directly linking BAT activation to the alimentary tract. Not only gut hormones but also metabolites derived from gut microbiota such as butyrate may be an important activator of BAT during cold exposure. Additional regulatory factors include bone morphogenic proteins, fibroblast growth factor 21, Vascular endothelial growth factors and transient receptor potential vanilloid receptors which are important components of thermal sensing and hence how brown adipose tissue responds to the cold. In the future the main challenge is to understand how these regulatory factors combine with each other and with inhibitory factors to control heat production from BAT, and what their relative importance is in differing circumstances. Knocking out UCP1 has revealed other sources of heat production in BAT including creatine-dependent cycles and a futile cycle of Ca2+ shuttling into and out of the endoplasmic reticulum via the SERCA and ryanodine receptors.

It is the balance between these two conflicting goals that determines the mammal's size, heart rate and amount of hair.
