Abstract:The development of porous composite materials is of great significance for their potentially improved performance over those of individual components and extensive applications in separation, energy storage, and heterogeneous catalysis. Now mesoporous metal-organic frameworks (MOFs) with macroporous melamine foam (MF) have been integrated using a one-pot process, generating a series of MOF/MF composite materials with preserved crystallinity, hierarchical porosity, and increased stability over that of melamine foam. The MOF nanocrystals were threaded by the melamine foam networks, resembling a ball-and-stick model overall. The resulting MOF/MF composite materials were employed as an effective heterogeneous catalyst for the epoxidation of cholesteryl esters. Combining the advantages of interpenetrative mesoporous and macroporous structures, the MOF/melamine foam composite has higher dispersibility and more accessibility of catalytic sites, exhibiting excellent catalytic performance.

Abstract:Metal‐organic frameworks (MOFs), with attractive properties such as high surface area, tunable porosity, designable functionality and excellent stability, have aroused tremendous interests of researchers as the matrices for enzyme immobilization. Recently, several efficient strategies including surface immobilization, post‐synthetic infiltration and in situ encapsulation have been explored. The MOFs‐immobilized enzymes, named enzymes@MOFs, show remarkably enhanced stability and recyclability, accelerating the cell‐free biocatalysis in diverse applications. This concept will impart the typical strategies for enzyme immobilization with MOFs, and their potential applications.

Abstract:The creation of a new metal-organic framework (MOF) with a hollow hierarchical porous structure has gained significant attention in the realm of enzyme immobilization. The present work employed a novel, facile, and effective combinatorial technique to synthesize modified MOF (N-PVP/HZIF-8) with a hierarchically porous core-shell structure, allowing for the preservation of the structural integrity of the encapsulated enzyme molecules. Scanning electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy, confocal laser scanning microscopy, and other characterization tools were used to fully explore the changes of morphological structure and surface properties in different stages of the preparation of immobilization enzyme CRL-N-PVP/HZIF-8, thus showing the superiority of N-PVP/HZIF-8 as an enzyme immobilization platform and the logic of the immobilization process on the carrier. Additionally, the maximum enzyme loading was 216.3 mg mL-1, the relative activity of CRL-N-PVP/HZIF-8 increased by 15 times compared with the CRL@ZIF-8 immobilized in situ, and exhibited quite good thermal, chemical, and operational stability. With a maximal conversion of 88.8%, CRL-N-PVP/HZIF-8 demonstrated good catalytic performance in the biosynthesis of phytosterol esters as a proof of concept. It is anticipated that this work will offer fresh concepts from several perspectives for the creation of MOF-based immobilized enzymes for biotechnological uses.

Abstract:In situ immobilization of enzyme into metal–organic frameworks (MOFs) is performed through a one-step and facile method. Candida antarctica lipase B (CalB) is directly embedded in zeolitic imidazolate framework (ZIF)-8 by simply mixing an aqueous solution of 2-methylimidazole and zinc nitrate hexahydrate [Zn(NO3)2⋅6H2O] containing CalB at room temperature. Due to the intrinsic micropores of ZIF-8, the obtained CalB@ZIF composite is successfully applied in size-selective transesterification reaction in organic solvent. CalB@ZIF not only shows much higher catalytic activity but also exhibits higher thermal stability than free CalB. Besides, the robust ZIF-8 shell also offers the hybrid composites excellent reusability.

Abstract:Enzymes, as natural catalysts with remarkable catalytic activity and high region-selectivities, hold great promise in industrial catalysis. However, applications of enzymatic transformation are hampered by the fragility of enzymes in harsh conditions. Recently, metal-organic frameworks (MOFs), due to their high stability and available structural properties, have emerged as a promising platform for enzyme immobilization. Synthetic strategies of enzyme-MOF composites mainly including surface immobilization, covalent linkage, pore entrapment and in situ synthesis. Compared with free enzymes, most immobilized enzymes exhibit enhanced resistance against solvents and high temperatures. Besides, MOFs serving as matrixes for enzyme immobilization show extraordinary superiority in many aspects compared with other supporting materials. The advantages of using MOFs to support enzymes are discussed. To obtain a high enzyme loading capacity and to reduce the diffusion resistance of reactants and products during the reaction, the mesoporous MOFs have been designed and constructed. This review also covers the applications of enzyme-MOF composites in bio-sensing and detection, bio-catalysis, and cancer therapy, which is concerned with interdisciplinary nano-chemistry, material science and medical chemistry. Finally, some perspectives on reservation or enhancement of bio-catalytic activity of enzyme-MOF composites and the future of enzyme immobilization strategies are discussed.

Abstract:Enzymes are natural catalysts with high catalytic activity, substrate specificity and selectivity. Their widespread utilization in industrial applications is limited by their sensitivity to harsh reaction conditions and difficulties relating to their removal and re-use after the reaction is complete. These limitations can be addressed by immobilizing the enzymes in solid porous supports. Covalent organic frameworks (COFs) are ideal candidate carriers because of their good biocompatibility, long-term water stability and large surface area. In post-synthetic immobilization, the enzyme is added to an existing COF; this has had limited success because of enzyme leaching and pore blockage by enzymes that are too large. Direct-immobilization methods—building the COF around the enzyme—allow tailored incorporation of proteins of any size and result in materials with lower levels of leaching and better mass transport of reactants and products. This protocol describes direct-immobilization methods that can be used to fabricate enzyme@COF (@ = engulfing) biocomposites with rationally programmed structures and functions. If COF construction requires harsh reaction conditions, the enzyme can be protected by using a removable metal-organic framework. Alternatively, a direct in situ approach, in which the enzyme and the COF monomers assemble under very mild conditions, can be used. Examples of both approaches are described: enzyme@COF-42-B/43-B capsules (enzymes including catalase, glucose oxidase, etc.) with ZIF-90 or ZPF-2 as protectors, and lipase@NKCOF-98/99 via in situ direct-immobilization methods (synthesis timing: 30–100 min). Example assays for physical and functional characterization of the COF and enzyme@COF materials are also described.

Abstract:Because of their efficiency, selectivity, and environmental sustainability, there are significant opportunities for enzymes in chemical synthesis and biotechnology. However, as the three-dimensional active structure of enzymes is predominantly maintained by weaker noncovalent interactions, thermal, pH, and chemical stressors can modify or eliminate activity. Metal-organic frameworks (MOFs), which are extended porous network materials assembled by a bottom-up building block approach from metal-based nodes and organic linkers, can be used to afford protection to enzymes. The self-assembled structures of MOFs can be used to encase an enzyme in a process called encapsulation when the MOF is synthesized in the presence of the biomolecule. Alternatively, enzymes can be infiltrated into mesoporous MOF structures or surface bound via covalent or noncovalent processes. Integration of MOF materials and enzymes in this way affords protection and allows the enzyme to maintain activity in challenge conditions (e.g., denaturing agents, elevated temperature, non-native pH, and organic solvents). In addition to forming simple enzyme/MOF biocomposites, other materials can be introduced to the composites to improve recovery or facilitate advanced applications in sensing and fuel cell technology. This review canvasses enzyme protection via encapsulation, pore infiltration, and surface adsorption and summarizes strategies to form multicomponent composites. Also, given that enzyme/MOF biocomposites straddle materials chemistry and enzymology, this review provides an assessment of the characterization methodologies used for MOF-immobilized enzymes and identifies some key parameters to facilitate development of the field.

Abstract:As a class of widely used biocatalysts, enzymes possess advantages including high catalytic efficiency, strong specificity and mild reaction condition. However, most free enzymes have high requirements on the reaction environment and are easy to deactivate. Immobilization of enzymes on nanomaterial‐based substrates is a good way to solve this problem. Metal‐organic framework (MOFs), with ultra‐high specific surface area and adjustable porosity, can provide a large space to carry enzymes. And the tightly surrounded protective layer of MOFs can stabilize the enzyme structure to a great extent. In addition, the unique porous network structure enables selective mass transfer of substrates and facilitates catalytic processes. Therefore, these enzyme‐immobilized MOFs have been widely used in various research fields, such as molecule/biomolecule sensing and imaging, disease treatment, energy and environment protection. In this review, the preparation strategies and applications of enzymes‐immobilized MOFs are illustrated and the prospects and current challenges are discussed.

Abstract:Metal-organic frameworks (MOFs) represent an emerging class of crystalline materials with well-defined pore structures and hold great potentials in a wide range of important applications. The functionality of MOFs can be further extended by integration with other functional materials, e.g., encapsulating metal nanoparticles, to form hybrid materials with novel properties. In spite of various synthetic approaches that have been developed recently, a facile method to prepare hierarchical hollow MOF nanostructures still remains a challenge. Here we describe a facile emulsion-based interfacial reaction method for the large-scale synthesis of hollow zeolitic imidazolate framework 8 (ZIF-8) nanospheres with controllable shell thickness. We further demonstrate that functional metal nanoparticles such as Pd nanocubes can be encapsulated during the emulsification process and used for heterogeneous catalysis. The inherently porous structure of ZIF-8 shells enables encapsulated catalysts to show size-selective hydrogenation reactions.

Abstract:Unlike inorganic crystals, metal-organic frameworks do not have a well-developed nanostructure library, and establishing their appropriately diverse and complex architectures remains a major challenge. Here, we demonstrate a general route to control metal-organic framework structure by a solvent-assisted ligand exchange approach. Thirteen different types of metal-organic framework structures have been prepared successfully. To demonstrate a proof of concept application, we used the obtained metal-organic framework materials as precursors for synthesizing nanoporous carbons and investigated their electrochemical Na + storage properties. Due to the unique architecture, the one-dimensional nanoporous carbon derived from double-shelled ZnCo bimetallic zeolitic imidazolate framework nanotubes exhibits high specific capacity as well as superior rate capability and cycling stability. Our study offers an avenue for the controllable preparation of well-designed meta-organic framework structures and their derivatives, which would further broaden the application opportunities of metal-organic framework materials.

Abstract:The immobilization of enzymes on or in solid supports is crucial for the industrialization of enzymes as chemical catalysts because immobilization provides stabilization, easy separation, and recyclability. Here, we show that a water-stable metal-organic framework, NU-1000, with hierarchical pore structure has the right combination of properties to be particularly well-suited as a scaffold for immobilizing enzymes such that they maintain full enzymatic catalytic activity. The immobilized enzyme shows greater resistance to organic solvent and denaturant than does the free enzyme and is characterized by greater reactant accessibility and higher activity than the same enzyme encapsulated in topologically simpler metal-organic frameworks. These findings suggest design rules for hierarchical pore structuring of host frameworks for enzyme-encapsulation applications by demonstrating enzyme immobilization in a solid support whereby the enzyme is highly accessible and retains catalytic activity under chemically challenging conditions.

Abstract:Enzymatic processes have great potential for applications in industrial biocatalysis, biosensing, and biomedical engineering. However, poor stability and difficulty in the reuse of enzymes still create challenges for their use in catalysis and other applications. In recent years, enzyme-incorporated metal-organic frameworks (enzyme@MOF) have emerged as a new type of armored enzymes and are thought to be an appealing solution to the above challenges. To date, the preparation of enzyme-MOF composites can be divided into three categories, including physical adsorption, covalent conjugation, and coprecipitation. The coprecipitation method integrates the synthesis of MOFs and encapsulation of enzymes into one step, allowing common MOFs with regular pore sizes to be used as carriers for enzyme binding. In this chapter, we present a protocol for the synthesis of enzyme@MOF composites using the coprecipitation approach with particular details.

Abstract:As a typical green methodology, enzymatic catalysis has been extensively employed in multitudinous chemical and biological transformation procedures. However, intrinsic fragile nature of enzymes makes them prone to denaturation or destabilization in harsh practical conditions, leading to unavoidably shortened lifespan and extremely high cost. It was proven that enzyme immobilization is an efficient strategy for enhancing their catalytic performance in continuous industrial practices. Metal-Organic Frameworks (MOFs) with extremely high specific surface area, abundant porosity, extraordinary multifunctionality, and relatively high stability, in recent years, have attracted remarkable research interests as novel supporting matrices for efficient enzyme immobilization and protection. Many reported MOFs-enzyme composites exhibit unprecedented catalytic performances than those of free enzymes, including improved enzyme efficiency, stability, selectivity, and recyclability, due to the protection of enzymes by highly ordered frameworks. To present a systematic overview of this emerging and developing field, herein, we summarize an update review about the most recent advances in MOFs immobilizing enzymes from the aspects of general synthetic approaches, critical impact factors, enhanced catalytic performances, and the practical applications. Subsequently, the emerging theories, methodologies and technologies in this thriving area are briefly introduced. Finally, barriers and future perspectives about MOFs for enzyme immobilization are also discussed.

Abstract:Encapsulating enzymes within metal-organic frameworks has enhanced their structural stability and interface tunability for catalysis. However, the small apertures of the frameworks restrict their effectiveness to small organic molecules. Herein, we present a green strategy directed by visible linker micelles for the aqueous synthesis of MAF-6 that enables enzymes for the catalytic asymmetric synthesis of chiral molecules. Due to the large pore aperture (7.6 Å), double the aperture size of benchmark ZIF-8 (3.4 Å), MAF-6 allows encapsulated enzyme BCL to access larger substrates and do so faster. Through the optimization of surfactants' effect during synthesis, BCL@MAF-6-SDS (SDS = sodium dodecyl sulfate) displayed a catalytic efficiency (Kcat/Km) that was 420 times greater than that of BCL@ZIF-8. This biocomposite efficiently catalyzed the synthesis of drug precursor molecules with 94-99% enantioselectivity and nearly quantitative yields. These findings represent a deeper understanding of de novo synthetic encapsulation of enzyme in MOFs, thereby unfolding the great potential of enzyme@MAF catalysts for asymmetric synthesis of organics and pharmaceuticals.

Abstract:Enzyme immobilization is a widely reported method to favor the applicability of enzymes by enhancing their stability and re-usability. Among the various existing solid supports and immobilization strategies, the in situ encapsulation of enzymes within crystalline porous matrices is a powerful tool to design biohybrids with a stable and protected catalytic activity. However, to date, only a few metal-organic frameworks (MOFs) and hydrogen-bonded organic frameworks (HOFs) have been reported. Excitingly, for the first time, Y. Chen and co-workers expanded the in situ bio-encapsulation to a new class of crystalline porous materials, namely covalent organic frameworks (COFs). The enzyme@COF materials not only exhibited high enzyme loading with minimal leaching, high catalytic activity and selectivity, chemical and long-term stability and recyclability but could also be scaled up to a few grams. Undoubtedly, this work opens new striking opportunities for enzymatic immobilization and will stimulate new research on COF-based matrices.
