Abstract:An effective synthetic method to achieve difluoromethylated oxazolidin-2-imine has been developed via visible-light promoted three-component tandem reaction of aryl allylamines, 2-BTSO2CF2H (BT = Benzothiazole) and isocyanates. This method features mild reaction conditions and good functional group tolerance, and the reaction mechanism was confirmed by experiments and interpreted by quantum chemical calculations.

Abstract:Described herein is an efficient protocol for the regioselective introduction of a vinyl trifluoromethylthioether to remote unactivated C(sp(3))-H bonds. The cascade process involves the vinyl radical-mediated 1,5-hydrogen atom transfer (HAT) and remote vinyl migration. During the transformation, inert C-H and C-C bonds are consecutively cleaved under mild conditions. The reaction features good functional group tolerance, broad substrate scope, and high regio-/stereo-selectivity.

Abstract:Copper-mediated trifluoromethylation of α-diazo esters with TMSCF(3) reagent has been developed as a new method to prepare α-trifluoromethyl esters. This trifluoromethylation reaction represents the first example of fluoroalkylation of a non-fluorinated carbene precursor. Water plays an important role in promoting the reaction by activating the "CuCF(3)" species prepared from CuI/TMSCF(3)/CsF (1.0:1.1:1.1). The scope of this trifluoromethylation reaction is broad, and its efficiency is demonstrated in the synthesis of a variety of aryl-, benzyl-, and alkyl-substituted 3,3,3-trifluoropropanoates.

Abstract:A visible-light-induced trifluoromethylsulfonylation reaction of diazo compounds is herein reported. This developed synthetic method captures the relatively rare trifluoromethyl sulfone radicals via coordination to the Mn(acac)(3) catalyst, delivering the corresponding alpha-trifluoromethyl sulfone esters in good to moderate yields (up to 82%). This protocol exhibits broad substrate scope and is easily carried out under mild reaction conditions. Furthermore, a plausible mechanism of the reaction was investigated through DFT calculations.

Abstract:We unveil a novel strategy that leverages cost-effective fluorine-containing sources for rapid synthesis of fluorinated com-pounds through multicomponent reactions. Our study introduces a visible light-driven palladium-catalyzed 1,4-difluoromethyl difunctionalization of conjugated dienes, capitalizing on the abundant industrial resource chlorodifluoro-methane (ClCF2H, Freon-22). Illuminated by blue LED light, the palladium catalyst orchestrates the efficient single electron reduction of Freon-22's C-Cl bond, yielding CF2H radicals. This versatile protocol accommodates an extensive array of amines, sulfinates, and enolates as nucleophiles, delivering regioselective difunctionalization products via SN2' substitution of the allylic palladium intermediate. Redox-neutral in nature, this transformation finds utility in late-stage functionalization of drug molecules, while the broad compatibility with diverse functional groups on conjugated dienes underscores its versa-tility. Initial insights suggest the involvement of a difluoromethyl radical intermediate, unveiling a direct and mild avenue to harness CF2H radicals from Freon-22.

Abstract:Vinylboronates and alkylboronates are key components in variegated transformations in all aspects of chemical science. This work describes a sequential radical difunctionalization strategy for the construction of fluorine-containing vinylboronates and alkylboronates with the integrated redox-active reagent N-trifluoromethylthiophthalimide. Abstract:A copper-catalyzed enantioselective 1,5-cyanotrifluoromethylation of vinylcyclopropanes has been developed using a radical relay strategy. This asymmetric reaction has demonstrated high enantioselective control, broad substrate scope, and mild conditions. Initiated by the in situ generated CF3 radical from Togni’s reagent, this method offers a new solution for remote enantioselective bifunctionalization of alkenes and thus provides a straightforward way for the synthesis of chiral CF3-containing internal alkenylnitriles.

Abstract:Herein, we develop a base-promoted regioselective synthesis of N-vinyl pyrazoles from vinyl sulfonium salts with diazo compounds. This metal-free synthetic protocol provides an efficient and practical approach to diverse N-vinyl pyrazoles in good to excellent yields under mild conditions. The reaction appears to experience a [3 + 2] annulation of vinyl sulfonium salts and diazo anions rather than diazo compounds, followed by N-vinylation.

Abstract:A highly efficient and regioselective direct C–H trifluoromethylation of pyridine based on an N-methylpyridine quaternary ammonium activation strategy has been developed. A variety of trifluoromethylpyridines can be obtained in good yield and excellent regioselectivity by treating the pyridinium iodide salts with trifluoroacetic acid in the presence of silver carbonate in N,N-dimethylformamide. The protocol features good functional group compatibility, easily available starting materials, and operational simplicity. Abstract:The fluorine-containing organic motif is becoming privileged in pharmaceuticals, agrochemicals, and functional materials, owing to its unique properties such as electron-withdrawing character, metabolic stability, and lipophilicity. Described herein is the PhI(OAc)2 -mediated radical trifluoromethylation of vinyl azides with Me3 SiCF3 to efficiently generate α-trifluoromethyl azines. The resulting α-trifluoromethyl azines were successfully transformed to valuable fluorine-containing molecules such as α-trifluoromethyl ketones, β-trifluoromethyl amines, 5-fluoropyrazoles, and trifluoroethyl isoquinolines.

Abstract:A variety of vinylic bromides and iodides undergo smooth trifluoromethylation and pentafluoroethylation with RfH-derived CuRf (Rf = CF3, C2F5) to give the corresponding fluoroalkylated olefins. These reactions employing the low-cost CuRf reagents occur in high yield with excellent chemo- and stereoselectivity under mild conditions (23–80 °C). Crystal structures of one trifluoromethyl and one pentafluoroethyl derivative have been determined.

Abstract:The effects of different substituents located at the para position of the aromatic ring and β carbon atom of the styrene on the reaction were investigated. The results showed that the reaction steps with higher energy barriers changed a little with the substituents of the reactants, which indicates that the reaction has a good adaptability to reactants containing different substituents. It was found the proton transfer in the final tautomerism step of nitroso intermediate to oxime is the rate limiting step under anhydrous conditions. Although the solvent effect did not influence the the rate limiting step significantly, the water mediated proton transfer significantly decreased the energy barrier of final tautomerism step. Compared with the direct proton transfer in vacuum, the energy barrier of the final tautomerism step decreased from 57.80kcal/mol in vacuum to 12.98kcal/mol with the water mediated proton transfer in water, which declined by 77.5%. When water participates in rate-limiting steps in organic solvents, the energy barrier also decreases significantly, which indicates that a small amount of water in the organic solvent is conducive to the reaction. This study is of great significance for the application of bifunctionalized reaction in the synthesis of organic fluoride compounds with different substituents.

Abstract:Nickel(II)-catalyzed regioselective trifluorovinylation and 1,2,2-trifluoroethylation of allyl alcohols with trifluorovinyl reagents were performed. The reaction of (E)-3-phenylprop-2-en-1-ol and trifluorovinylzinc bromide (TFVZ) afforded 1,2,2-trifluorovinyl diene in moderate to high yields. The reaction of (E)-3-phenylprop-2-en-1-ol and trifluorovinyl trimethylsilane (TFVTMS) resulted in the novel nucleophilic addition product 1,1,2-trifluoroethoxy aryl olefin.
