Abstract:Intracellular bacteria in dormant states can escape the immune response and tolerate high-dose antibiotic treatment, leading to severe infections. To overcome this challenge, cascade-targeted nanoplatforms that can target macrophages and intracellular bacteria, exhibiting synergetic antibiotic/reactive oxygen species (ROS)/nitric oxide (NO)/immunotherapy, were developed. These nanoplatforms were fabricated by encapsulating trehalose (Tr) and vancomycin (Van) into phosphatidylserine (PS)-coated poly[(4-allylcarbamoylphenylboric acid)-ran-(arginine-methacrylamide)-ran-(N,N'-bisacryloylcystamine)] nanoparticles (PABS), denoted as PTVP. PS on PTVP simulates a signal of "eat me" to macrophages to promote cell uptake (the first-step targeting). After the uptake, the nanoplatform in the acidic phagolysosomes could release Tr, and the exposed phenylboronic acid on the nanoplatform could target bacteria (the second-step targeting). Nanoplatforms can release Van in response to infected intracellular overexpressed glutathione (GSH) and weak acid microenvironment. l-arginine (Arg) on the nanoplatforms could be catalyzed by upregulated inducible nitric oxide synthase (iNOS) in the infected macrophages to generate nitric oxide (NO). N,N'-Bisacryloylcystamine (BAC) on nanoplatforms could deplete GSH, allow the generation of ROS in macrophages, and then upregulate proinflammatory activity, leading to the reinforced antibacterial capacity. This nanoplatform possesses macrophage and bacteria-targeting antibiotic delivery, intracellular ROS, and NO generation, and pro-inflammatory activities (immunotherapy) provides a new strategy for eradicating intracellular bacterial infections.

Abstract:Abstract Catalytic therapy utilizing special substances (e.g., hydrogen peroxide and glucose) in tumor sites has attracted wide interest to achieve tumor‐specific therapy or improve the efficacy of other treatments. Benefiting from the selectivity and high activity of catalytic chemistry, efficient cancer therapy is available with negligible side effects. Numerous works, which are focused on further augmenting catalytic therapy with cascade strategies, have been reported with biocompatible nanomaterials composed of natural enzymes and/or nanozymes. Herein, we summarized catalytic reactions and nanomaterials in cascade strategy‐involved cancer therapy. With rapid advances in chemistry and nanomaterials, developing more selective and efficient cascade catalytic strategies will continue to be promising and challenging for cancer therapy.

Abstract:Chemical transformation in cellular environment is critical for regulating biological processes and metabolic pathways. Harnessing biocatalytic cascades to produce chemicals of interest has become a research focus to benefit industrial and pharmaceutic areas. Nanoreactors, which can act as artificial cell‐like devices to organize cascade reactions, have been recently proposed for potential therapeutic applications for life‐threatening illnesses. Among various types of nanomaterials, there is a growing interest in 2D metal–organic frameworks (MOFs). By virtue of the ultralarge specific surface area, high porosity, and structural diversity, 2D MOF nanosheets hold great promise for a broad spectrum of biomedical use. Herein, a unique planar MOF‐based hybrid architecture (GMOF‐LA) is introduced by incorporating ultrasmall gold nanoparticles (Au NPs) as nanozyme and l‐Arginine (l‐Arg) as nitric oxide (NO) donor. The prepared Au NPs enable oxidation of glucose into hydrogen peroxide, which drives biocatalytic cascades to covert l‐Arg into NO. Interestingly, the well‐designed nanosheets not only possess excellent catalytical activity for NO generation, resulting in gas therapeutic effect, but also serve as a desired photosensitizer for photodynamic therapy. This study establishes a good example of exploring bioinspired nanoreactors for cooperative anticancer effect, which may pave the path for future "bench‐to‐bedside" design of nanomedicine.

Abstract:Wound infections caused by drug-resistant bacteria pose a great threat to human health, and the development of non-drug-resistant antibacterial approaches has become a research priority. In this study, we developed Cu2O-SnO2 doped polydopamine (CSPDA) triple cubic antibacterial nanoenzymes with high photothermal conversion efficiency and good Fenton-like catalase performance. CSPDA antibacterial nanoplat-form can catalyze the generation of hydroxyl radical ( center dot OH) from H2O2 at low concentration (50 mu g .mL-1) under 808 nm near-infrared (NIR) irradiation to achieve a combined photothermal therapy (PTT) and chemodynamic therapy (CDT). And the CSPDA antibacterial nanoplatform displays broad-spectrum and long-lasting antibacterial effects against both Gram-negative Escherichia coli (100 %) and Gram-positive Staphylococcus aureus (100 %) in vitro . Moreover, in a mouse wound model with mixed bacterial infection, the nanoplatform demonstrates a significant in vivo bactericidal effect while remaining good cytocompatible. To conclude, this study successfully develops an efficient and long-lasting bacterial infection treatment system. This system provided different options for future studies on the design of synergis-tic antimicrobial therapy. Hence, the as-synthesized synergetic photothermal therapy and chemodynamic therapy nanoenzymes have rapid and long-term bactericidal ability, well-conglutinant performance and effectively preventing wound infection for clinical application.

Abstract:Cascade nanoreactors (NRs) which can be activated in tumor microenvironment and catalyze the local substance to toxic species for killing cancer cells is an appealing emerging agent for cancer treatment in recent years. Peroxidase-like nanozymes are often integrated into such kind of systems for cancer therapy. In this study, magnetic metal-organic framework (MOF: zeolitic imidazolate framework 8, ZIF-8) nanoparticles (NPs) with ferrimagnetic structures inside are fabricated via the mineralization of iron during the growth of ZIF-8. The Fe-base magnetic structures can serve as peroxidase-like nanozyme. After the loading of GOx on the magnetic MOF NPs, magnetic cascade MOF/enzyme hybrid NRs (Fe@ZIF-8@GOx NRs) are successfully constructed. In the presence of glucose, the NRs can perform high cascade reaction activity to produce hydroxyl radicals center dot OH) under mild acidic condition at physiological temperature. The NRs can be more internalized by cancer cells under a magnetic field and take advantage of acidic environment in lysosomes to produce high toxic center dot OH by cascade reaction to kill cancer cells. Magnetic targeting to kill cancer cells at the specified location can be achieved by applying a magnetic field to the targeted area and then damage to normal cells will be minimized.

Abstract:Chemodynamic therapy (CDT) based on cascade catalytic nanomedicine has emerged as a promising cancer treatment strategy. However, most of the reported cascade catalytic systems are designed based on symmetric- or co-assembly of multiple catalytic active sites, in which their functions are difficult to perform independently and may interfere with each other. Especially in cascade catalytic system that involves fragile natural-enzymes, the strong oxidation of free-radicals toward natural-enzymes should be carefully considered, and the spatial distribution of the multiple catalytic active sites should be carefully organized to avoid the degradation of the enzyme catalytic activity. Herein, a spatially-asymmetric cascade nanocatalyst is developed for enhanced CDT, which is composed by a Fe 3 O 4 head and a closely connected mesoporous silica nanorod immobilized with glucose oxidase (mSiO 2 -GOx). The mSiO 2 -GOx subunit could effectively deplete glucose in tumor cells, and meanwhile produce a considerable amount of H 2 O 2 for subsequent Fenton reaction under the catalysis of Fe 3 O 4 subunit in the tumor microenvironment. Taking the advantage of the spatial isolation of mSiO 2 -GOx and Fe 3 O 4 subunits, the catalysis of GOx and free-radicals generation occur at different domains of the asymmetric nanocomposite, minimizing the strong oxidation of free-radicals toward the activity of GOx at the other side. In addition, direct exposure of Fe 3 O 4 subunit without any shelter could further enhance the strong oxidation of free-radicals toward objectives. So, compared with traditional core@shell structure, the long-term stability and efficiency of the asymmetric cascade catalytic for CDT is greatly increased by 138%, thus realizing improved cancer cell killing and tumor restrain efficiency.

Abstract:It is of great significance to develop effective antibacterial agents and methods to combat drug resistant bacterial infections due to its increasing threaten to human health and the ineffectiveness of antibiotics. Herein, a multifunctional hybrid nano-assembly (M1-Fe NPs) based on conjugated oligomer and ferrous ion was engineered with favorable bactericidal activity for synergetic antibacterial therapy. The chelation of ferrous ion not only enhances the photothermal conversion efficiency of M1 but also endows the nano-assembly with catalytic capability of transferring H2O2 into stronger oxidant hydroxyl radicals (•OH). Meanwhile, the generated heat can further promote the Fenton reaction activity. By generating cytotoxic heat and oxidative •OH, M1-Fe NPs can effectively kill Staphylococcus aureus in vitro and in vivo with the aid of low dosage of H2O2. The work provides a new multifunctional platform for combinational drug resistant antibacterial therapy and even antitumor therapy.

Abstract:Cascade catalytic reaction exhibits simple procedure and high efficiency, such as that from the orderly assembly of different enzymes in biological systems. Mimicking of the natural cascade procedure becomes critical, but the orderly assembly of different enzymes is still challenging. Herein, we reported single Au‐Pt nanozyme with "three‐in‐one" functions to initiate cascade conversions for O2 supply as mimic catalase, H2O2 production with its glucose oxidase‐like property, and •OH generation as mimic peroxidase for chemodynamic therapy (CDT). Thus, the complex assembly and cross‐talk among the different enzymes are avoided. To this end, metastable Cu2O NPs, as scaffolds, are used to anchor ultrasmall Au‐Pt nanozyme, while metal‐organic framework (MOF) is used to encapsulate the nanozyme for tumor microenvironment (TME) response and shielding protein adsorption. Pluronic® F127 is then modified on the surface to improve hydrophilicity and biocompatibility of the composite. The endogenous acidity and glutathione in tumor degrade MOF to expose nanozyme for cascade catalytic CDT. The high photothermal conversion ability of the composite also enhances the CDT, while Cu2+ ions consume GSH to further improve CDT efficiency as augmented cascade catalytic tumor therapy. Abstract:Disulfiram (DSF), a new potential anticancer drug, has been shown to exhibit anticancer activity dependent on the formation of CuET, the chelation product of DSF with Cu2+. However, the poor stability of DSF and insufficient physiological concentration of Cu2+ hinder its practical application. To achieve the co-delivery of DSF and Cu2+ while overcoming the inefficiency of single chemotherapy, in this study, a cascade nanoplatform, DSF/Ce6@ZIF-8@CuO2, was constructed by encapsulating DSF and chlorin e6 (Ce6, a photosensitizer) in zeolite imidazole framework-8 (ZIF-8, a nanocarrier) and then loading CuO2, which self-supplied H2O2/O2, onto DSF/Ce6@ZIF-8. By triggering the response of DSF/Ce6@ZIF-8@CuO2 to the acidic tumor microenvironment, encapsulated DSF, Ce6 and CuO2 were released to achieve multimodal synergistic treatment with enhanced DSF chemotherapy and chemodynamic/photodynamic therapy (CDT/PDT). In vitro and animal studies indicated that the designed DSF/Ce6@ZIF-8@CuO2 has strong tumor-inhibitory effects and provides a promising paradigm for designing smart nanoplatforms.

Abstract:The one-pot combination of biocatalytic and chemocatalytic reactions represents an economically and ecologically attractive concept in the emerging cascade processes for manufacturing. The mutual incompatibility of biocatalysis and chemocatalysis, however, usually causes the deactivation of catalysts, the mismatching of reaction dynamic, and further challenges their integration into concurrent chemo-biocascades. Herein, we have developed a convenient strategy to construct versatile functional metal-organic framework micro-nanoreactors (MOF-MNRs), which can realize not only the encapsulation and protection of biocatalysts but also the controllable transmission of substances and the mutual communication of the incompatible chemo-biosystems. Importantly, the MOFs serving as the shell of MNRs have the capability of enriching the chemocatalysts on the surface and improving the activity of the chemocatalysts to sufficiently match the optimum aqueous reaction system of biocatalysts, which greatly increase the efficiency in the combined concurrent chemo-biocatalysis. Such strategy of constructing MOF-MNRs provides a unique platform for connecting the "two worlds" of chemocatalysis and biocatalysis.

Abstract:Nanozymes are a class of nanomaterials with enzyme-like activities that have been extensively studied for their possibilities and superiorities over natural enzymes. Intelligent design and fabrication of a nanozyme platform with highly efficient and stable tandem enzyme characteristics is necessary. Herein, a nanozyme cascade promoting bioplatform based on protein/bimetallic nanoparticles attached to ZIF-derived multilayer N-doped carbon nanosheets is proposed. Due to the carbon-based large surface area and solitary pairs of nitrogen electrons, the metal could be riveted to the carbon-based surface and reduced in situ to nanoparticles. Simultaneously, the addition of BSA has made it possible to establish a biological platform. The obtained BSA-PtAu@CNS has a linear range of 2 × 10-6 to 2 × 10-3 M for H2O2 and apparent dynamic Km of 0.005 × 10-3 M, which shows stronger affinity with H2O2 than HRP. The nanohybrid could adapt to severe reaction environment and enable prominent tandem dual-enzyme activity for glucose sensing. As a result, the cascade nanozyme bioplatform based on BSA-PtAu@CNS is accomplished and provides insights for subsequent biocompatible nanomaterials for use in medical and diagnostic applications.

Abstract:Cu‐HHTP, a readily synthesizable 2D MOF nanozyme is proposed for antibacterial therapy. In response to the substrate adsorption and decomposition process of POD reaction, non coordination saturation and highly conjugated structure are proposed. Compared to traditional commercially available 3D nanozyme, Cu‐HHTP exhibits superior visible light catalytic performance, demonstrating enormous application potential for antibacterial therapy. The misuse of antibiotics leads to emergence of drug‐resistant bacteria. Developing artificial nanozymes with high stability and excellent catalytic performance is desirable to substitute for antibiotics, but still remains huge challenge. Herein, the study reports a new strategy for cascaded enhanced peroxidase (POD) reaction driving antibacterial therapy via constructing 2D conjugated metal‐organic frameworks (MOFs) with non coordination saturation as nanozyme. It is demonstrated that the non coordination saturation Cu atom promotes the adsorption of H2O2, followed by the Cu‐HHTP MOFs with high conjugated structure can enhance the photogenerated electron transfer for ·OH generation in POD reaction. The sequential enhancement of substrate adsorption and decomposition endows the as‐made Cu‐HHTP MOFs with outstanding antibacterial property of >99.9999% elimination with a concentration only 1/133 that of traditional nanozyme. In addition, the study also certifies its excellent in vivo antimicrobial activity and biocompatibility, indicating the therapeutic potential for infections by drug‐resistant bacteria.

Abstract:Infections caused by drug-resistant bacteria pose a great threat to human health. Non-antibiotic-dependent antibacterial strategies have become the focus of research. Among them, chemical dynamic treatment-based (CDT) therapeutic systems, which catalyze the production of hydroxyl radicals by enzymes, have achieved tremendous success for antibacterial purposes. However, limited kinetics of the Fenton reaction, poor permeability, and short half-life of hydroxyl radicals compromise the antibacterial effects of CDT. In addition, difficulties in the early diagnosis of infection lead to drug abuse and delayed treatment. Herein, a polydopamine coated ferrous sulfide theranostic platform adsorbing a hypochlorite responsive probe with photothermal treatment (PTT) enhanced CDT was synthesized. The probe component was used for the early diagnosis of infection. PTT not only inactivated bacteria by hyperthermia but also accelerated the Fenton reaction to produce more ·OH. In vitro antibacterial experiments demonstrated that the multifunctional theranostic platform has a broad antibacterial spectrum, including methicillin-resistant Staphylococcus aureus (MRSA), drug-resistant Escherichia coli (DR E. coli), and Pseudomonas aeruginosa (P. aeruginosa). A Synergistic Antibacterial Study of Copper-Doped Polydopamine on Ti3C2Tx Nanosheets with Enhanced Photothermal and Fenton-like Activities

Abstract:In response to the trend of drug-resistant and super bacteria, the existing single antibacterial methods are not sufficient to kill bacteria, and the development of multifunctional antibacterial nanomaterials is urgent. Our study aims to construct copper-doped polydopamine-coated Ti3C2Tx (CuPDA@Ti3C2Tx) with an enhanced photothermal property and Fenton-like activity. The nanocomposite hydrogel consisting of CuPDA@Ti3C2Tx and alginate can improve the antioxidant activity of two-dimensional MXene nanosheets by coating them with a thin layer of PDA nanofilm. Meanwhile, Cu ions are adsorbed through the coordination of PDA-rich oxygen-containing functional groups and amino groups. Calcium ions were further used to crosslink sodium alginate to obtain antibacterial hydrogel materials with combined chemotherapy and photothermal therapy properties. The photothermal conversion efficiency of CuPDA@Ti3C2Tx is as high as 57.7% and the antibacterial rate of Escherichia coli reaches 96.12%. The photothermal effect leads to oxidative stress in bacteria, increases cell membrane permeability, and a high amount of ROS and copper ions enter the interior of the bacteria, causing protein denaturation and DNA damage, synergistically leading to bacterial death. Our study involves a multifunctional synergistic antibacterial nanodrug platform, which is conducive to the development of high-performance antibacterial agents and provides important research ideas for solving the problem of drug-resistant bacteria.

Abstract:This work reports a nanocatalytic antimicrobial agent (Si@Cu NSs) with enhanced CDT via p‐d orbital hybridization strategy and synergistic photothermal effect. This strategy enhances the Fenton‐like reaction of copper atoms and addresses the limited application of nanoparticles in pathophysiological environments. Excellent therapeutic effects demonstrate the feasibility of constructing novel CDT agents by orbital hybridization for the catalytic treatment of IAIs. Recurrent bacterial infections, impenetrable microbial biofilms, and irremediable antibiotic resistance are the most perilous threats in orthopedic implant‐associated infections (IAIs). Recently, chemodynamic therapy (CDT) has been considered a promising therapy, while the clinical practices for existing CDT agents are limited by the low therapeutic efficiency in physiological circumstances. Herein, it is demonstrated that silica‐copper nanosheets (Si@Cu NSs) exhibit a combined therapeutic photothermal‐chemodynamic strategy for IAIs treatment with superior capacity and biocompatibility. This unique design endows the nanostructure with enhanced copper‐based Fenton‐like properties via p‐d orbital hybridization by incorporating copper nanoclusters (Cu NCs) on silicene nanosheets (Si NSs) matrix, integrating inherent photothermal performance of Si NSs with specific catalytic anti‐infection of Cu NCs against planktonic bacteria and biofilms both in vitro and in vivo. This study not only reveals the promising bio‐application prospects of 2D nanocatalytic biomaterials but also demonstrates the feasibility of constructing novel CDT agents by orbital hybridization for the catalytic treatment of IAIs.
