Abstract:A catalytic annulation cascade is achieved that affords an efficient access to phthalides from readily available benzoic acids and saturated ketones. The reaction is catalyzed by a bimetallic Ir/Cu system and proceeds via the combination of Cu-catalyzed dehydrogenation of ketones and Ir-catalyzed direct C-H functionalization under the assistance of the weakly coordinating carboxyl followed by the beta-H elimination and intramolecular Michael addition. This method shows a broad scope of substrates and good functional group tolerance. The protocol furnishes an alternative strategy for the construction of diverse 3-substituted phthalides.

Abstract:A Cu(I)‐catalyzed [4 + 2] oxidative annulation of α,β‐unsaturated ketoxime acetates with cyclopropanols has been developed for the synthesis of 2,4,5‐trisubstituted pyridines in 28−76% yields. This method employs cyclopropanols as C2 synthons and features various functional group compatibility and gram‐scale synthesis. In addition, a plausible reaction pathway was proposed based on the mechanism studies.

Abstract:The first copper-catalyzed three-component annulation of α,β-unsaturated ketoximes, 1,3-dicarbonyls and paraformaldehyde has been documented. This novel strategy achieved the two C−C bonds cleavage of 1,3-dicarbonyl compounds directly as a single-carbon synthon and provided a new and highly efficient method for the synthesis of 2,3-disubstituted pyrroles in moderate to good yields with broad functional group compatibility.

Abstract:A novel synthesis method has been developed for the construction of functionalized 2,4-diarylpyridines. The reaction involves several key processes, including a reduction reaction, Michael reaction, decarboxylation, intramolecular cyclization, oxidation, ring-opening and aromatization reaction. All of these processes are facilitated by heating a mixture of the starting materials in the environmentally benign solvent, propylene carbonate (PC), with the oxidant (air) and the catalyst Cu(OAc)2. This cascade reaction not only forms two new bonds but also cleaves three existing bonds in a single pot. The protocol presented in this study is a valuable tool for synthesizing functionalized unsymmetrical 2,4-diarylpyridines.

Abstract:A Cu(I)‐catalyzed ring‐opening (3+2) annulation of cyclopropenones with ketoxime acetates has been developed. In disclosed protocol, ketoxime acetates are selected as C‐N splicing unit to participate in the transformation process, furnishing a sequence of highly functional 1,2‐dihydro‐pyrrol‐3‐one derivatives in good isolated yields. Besides, scale‐up reaction is conducted to demonstrate the practicability of developed methodology and a tentative mechanism for (3+2) annulation is also proposed.

Abstract:An unprecedented copper-catalyzed oxidative dehydrogenative [3 + 2] annulation of O-acyl oximes containing α-C(sp3)-H bonds with α-amino ketones for the preparation of 3-aminopyrroles is developed. This method handles O-acyl oximes as both internal oxidants and three-atom-unit components combined with the copper catalyst to drive catalytically generated oxime nitrogen-centered radicals and oxidative dehydrogenation of the α-amino ketones to the key 2-iminoethan-1-one intermediates, enabling site-selective cleavage of the multiple α-C(sp3)-H bonds in O-acyl oximes and α-amino ketones to create two new bonds, including a C(sp2)-C(sp2) bond and a C(sp2)-N bond, between the carbon atoms and the nitrogen atom in a cross-selective fashion.

Abstract:Herein, we disclose a general and practical iminoarylation of alkenes by nickel-catalyzed reductive cross-coupling of unsaturated oxime esters with readily available aryl halides, providing an expedient approach for constructing pyrroline derivatives. The absence of organometallic reagents enables the reaction to occur under mild conditions with a broad substrate scope and good functional group tolerance. Moreover, other C-based electrophiles, including alkenyl, alkynyl and alkyl halides, or pseudohalides, were also competent substrates for this reaction.

Abstract:Herein, a novel protocol was constructed for the synthesis of highly functionalized 2,4-diarylpyridines. In this reaction, α,β-unsaturated ketoxime acetates and enaminones were used as substrates in a one-pot cascade reaction, which included a Michael reaction, intramolecular cyclization, dehydrogenation, and the removal of HOAc. This reaction resulted in the formation and cleavage of two bonds by refluxing a mixture of the starting materials in toluene and in the presence of the catalyst Fe(OTf)3. As a result, a series of functionalized pyridines were produced through a complex cascade reaction. This protocol can be used to synthesize functionalized asymmetrical 2,4-diarylpyridines. This approach is suitable for the combinatorial and parallel syntheses of pyridine derivatives in a one-pot reaction rather than traditional tedious multi-step reactions.

Abstract:A highly efficient protocol for the synthesis of pyrrolidones by the copper‐catalyzed alkynylation/annulation of aliphatic amides with alkynyl carboxylic acids is discussed in this paper. A broad range of easily accessible alkynyl carboxylic acids were introduced at the β‐methyl group of aliphatic amides with the assistance of an 8‐aminoquinolyl auxiliary group via decarboxylation to achieve the subsequent cyclic CN bond formation within one hour. High selectivity of β‐methyl groups over methylene groups was observed, and the extension of this catalytic system to the activation of methylene CH bonds failed. The substrates with two different groups at the α‐position of the aliphatic amides lead to the formation of diastereoisomers which is determined by 1H NMR spectroscopy. The initially produced products with Z‐configurations can be easily transformed to the corresponding products with E‐configurations by the treatment with dilute p‐toluenesulfonic acid after the reaction. This catalytic tandem decarboxylative cyclization provides a new opportunity for the direct functionalization of sp3 CH bonds.

Abstract:A copper catalyzed annulation-aromatization of benzyl trifluoromethyl ketimines with 3-acryloyloxazolidin-2-ones for the synthesis of 3-fluoropyridines through double C-F bond cleavages has been developed. In this approach, the annulation occurred between the in situ formed dienes from trifluoromethyl ketimines via the first C-F bond cleavage and 3-acryloyloxazolidin-2-ones. Then the aromatization afforded 3-fluoropyridines in moderate yields through the second C-F bond cleavage. The 3-fluoropyridine products could be further hydrolyzed to multi-substituted 3-pyridinecarboxylic acids.

Abstract:A novel, efficient, and practical method for the synthesis of imidazopyridine derivatives has been developed through the copper-catalyzed aerobic oxidative CH functionalization of substituted pyridines with N-(alkylidene)-4H-1,2,4-triazol-4-amines. The procedure occurs by cleavage of the NN bond in the N-(alkylidene)-4H-1,2,4-triazol-4-amines and activation of an aryl CH bond in the substituted pyridines. This is the first example of the preparation of imidazopyridine derivatives by using pyridines as the substrates by transition-metal-catalyzed CH functionalization. This method should provide a novel and efficient strategy for the synthesis of other nitrogen heterocycles.

Abstract:An efficient copper-catalyzed strategy for the synthesis of α-ketoamides via cross-coupling of methyl ketones and pyridin-2-amines is described. This transformation has provided a simple process for the formation of C−N and C=O bonds to prepare α-ketoamides, which are important substrates and intermediates for the preparation of fine chemicals. The reaction mechanism is investigated, which suggests that the reaction proceeds via a radical pathway.

Abstract:Copper‐promoted annulation reactions of terminal alkynes with 2‐aminopyridines have been developed for the synthesis of 2‐halogenated imidazo[1,2‐a]pyridines using copper halide as the halogen source. A variety of substrates survived under the reaction conditions and gave the desired products in good yields. This reaction features advantages such as easily available starting materials, broad substrate scope, and mild reaction conditions.

Abstract:An iron(III)-porphyrin-catalyzed cyclization of ketones with dimethyl sulfoxide (DMSO) and ammonium acetate for the synthesis of unsymmetrical and symmetrical pyridines by employing DMSO as C-4 or C-6 source has been developed. Various aryl ketone derivatives react readily with DMSO, producing the pyridines in yields of 30%similar to 85%. This method uses non-noble metals and proceeds under mild reaction conditions with operational simplicity, which thus allows the expedient assembly of pyridines from readily available ketones. Based on the preliminary experiments, a plausible mechanism of this transformation is disclosed.

Abstract:An efficient, practical, and scalable Cs2CO3‐catalyzed [4+2] annulation reaction between ynones and acetates is presented, wherein ynones serve as the four‐atom annulation partners. This methodology facilitates the synthesis of substituted 2‐pyrones with yields ranging from good to excellent. The ready availability of the starting materials, coupled with the simplicity of the annulation protocol, renders this approach highly amenable to the preparation of a diverse range of 2‐pyrones. Furthermore, the feasibility of the reaction on a gram scale and its application in the synthesis of a potent and selective cyclooxygenase‐2 inhibitor underscore the practical significance and utility of this method.
