Abstract:Organoboron compounds play an irreplaceable role in synthetic chemistry and the related transformations based on the unique reactivity of C-B bond are potentially the most efficient methods for the synthesis of organic molecules. The synthetic importance of multiboron compounds in C-C bond formation and function transformation reactions is growing and the related borations of activated or nonactivated alkenes have been developed recently. However, introducing directly two boron moieties into the terminal sites of alkenes giving 1,1-diborylalkanes in a catalytic fashion has not been explored yet. Here we describe a synthetic strategy of 1,1-diborylalkanes via a Ni-catalyzed 1,1-diboration of readily available terminal alkenes. This methodology shows high level of chemoselectivity and regioselectivity and can be used to convert a large variety of terminal alkenes, such as vinylarenes, aliphatic alkenes and lower alkenes, to 1,1-diborylalkanes.1,1-diborylalkanes are useful building blocks in synthetic chemistry. Here, the authors present a highly chemo- and regioselective Ni-catalyzed reaction for the synthesis of 1,1-diborylalkanes from a wide variety of readily available terminal alkenes.

Abstract:Chiral tertiary alcohols are an important structural motif, however, the general and efficient methodologies for their synthesis are less reported. Herein, we report a Ni(ІІ)-catalyzed asymmetric alkenylation and arylation of aryl ketones with organoborons under air via a 1,5-metalate shift strategy to obtain chiral tertiary allylic alcohols and diaryl alcohols. The reaction demonstrates good functional group tolerance and delivers chiral tertiary alcohols with good to excellent results. Furthermore, this method can be applied to the late-stage modification of drugs and the efficient synthesis of natural products. Notably, the reaction proceeds through an outer-sphere mechanism. The Ni(II) complex functions both as a Lewis acid to activate the ketone and create a chiral environment, and as coordination bridge linking the ketone and the organoboron-derived "ate" complex, facilitating the 1,5-metalate shift without forming a C-Ni bond. This approach contrasts with traditional transition metal-catalyzed nucleophilic addition reactions that involve carbon-metal bond formation.

Abstract:Great advances have been made by leveraging high-valent Pd(II)/Pd(IV) catalysis in the areas of C–H activation and alkene difunctionalisation, thus representing a powerful approach for the construction of carbon-carbon and carbon-heteroatom bond. However, the catalytic reactions involving high-valent Ni(II)/Ni(IV) catalysis are largely underdeveloped. Here we report a Ni(II)-catalysed dicarbofunctionalisation of unactivated alkenes via high-valent Ni(II)/Ni(IV) catalysis. This dicarbofunctionalisation protocol provides a highly efficient and direct route towards vicinal substituted alkanes using primary, secondary, and tertiary amides, as well as secondary and tertiary amines as the native directing group under redox-neutral conditions that are challenging to access through conventional methods. The key to the success of this reaction is the use of a bulky β-diketone ligand, which could enable the insertion of alkene to aryl-Ni(II) species, stabilize the alkyl-Ni(II) species and inhibit the homolytic alkyl-Ni(II) cleavage. This dicarbofunctionalisation reaction features the use of native directing group, a broad substrate scope, and excellent scalability. The resulting Weinreb amide-derived products can be readily derivatized to a variety of ketones and aldehyde, which are fundamentally useful in synthetic chemistry. In addition, this protocol has been employed for the efficient preparation of several bioactive compounds, showcasing the significant synthetic values of our current method.

Abstract:Ketones, prevalent in many biologically significant molecules, require the development of novel methods to synthesize these structures, which is a critical endeavor in organic synthesis. Transition metal catalysis has proven to be an effective method for synthesizing ketones. However, the scope of these substrates remains relatively limited, particularly due to their incompatibility with sensitive functional groups. Herein, we report a Ni-catalyzed three-component 1,2-carboacylation of alkenes, which activates secondary/tertiary alkyl bromides. This method offers significant advantages: simplicity of operation, ready availability of substrates, and broad substrate applicability. A series of experimental studies have helped clarify the key mechanistic pathways involved in this cascade reaction.

Abstract:The transition metal-catalyzed hydrofunctionalization of alkenes offers an efficient solution for the rapid construction of complex functional molecules, and significant progress has been made during last decades. However, the hydrofunctionalization of internal alkenes remains a significant challenge due to low reactivity and the difficulties of controlling the regioselectivity. Here, we report the hydroarylation and hydroalkenylation of internal alkenes lacking a directing group with aryl and alkenyl boronic acids in the presence of a nickel catalyst, featuring a broad substrate scope and wide functional group tolerance under redox-neutral conditions. The key to achieving this reaction is the identification of a bulky 1-adamantyl β -diketone ligand, which is capable of overcoming the low reactivity of internal 1,2-disubstituted alkenes. Preliminary mechanistic studies unveiled that this reaction undergoes an Ar-Ni(II)-H initiated hydroarylation process, which is generated by the oxidative addition of alcoholic solvent with Ni(0) species and sequential transmetalation. In addition, the oxidative addition of the alcoholic solvent proves to be the turnover-limiting step.

Abstract:German cystic fibrosis (CF) chromosomes were screened for molecular lesions in exon 20 of the cystic fibrosis transmembrane conductance regulator (CFTR) gene by chemical cleavage of mismatch. An 3884G‐to‐A transition was detected in two patients which leads to an exchange of a serine by an asparagine in the Walker motif A of the second nucleotide binding fold. The affected serine residue is evolutionarily strongly conserved among the pro‐ and eukaryotic members of the protein superfamily of traffic ATPases. The two S1251N alleles were linked to the benign missense mutation F508C which is located in another conserved region of CFTR, the center region of the first nucleotide binding fold. Both patients with the complex allele F508C‐S1251N are carrying ΔF508 on the other CF chromosome and are suffering from severe pulmonary and gastrointestinal CF disease. Although F508C has been classified as a neutral sequence variation because of its discovery in healthy ΔF508 gene carriers, it may nevertheless influence CFTR dysfunction caused by the S1251N mutation. © 1992 Wiley‐Liss, Inc.
Redox‐Neutral Nickel(II) Catalysis: Hydroarylation of Unactivated Alkenes with Arylboronic Acids

Abstract:Reported here is the discovery of a redox-neutral NiII /NiII catalytic cycle which is capable of the linear-selective hydroarylation of unactivated alkenes with arylboronic acids for the first time. This novel catalytic cycle, enabled by the use of an electron-rich diimine ligand, features broad substrate scope, and excellent functional-group and heterocycle compatibility under mild reaction conditions in the absence of additional oxidants and reductants. Mechanistic investigations using kinetic analysis and deuterium-labelling experiments revealed the protonation to be the rate-determining step in this redox-neutral catalysis, and the reversible chain-walking nature of the newly developed diimine-Ni catalyst.

Abstract:The Ni-catalyzed decarbonylative borylation of (hetero)aryl carboxylic acids with B(2)cat(2) has been achieved without recourse to any additives. This Ni-catalyzed method exhibits a broad substrate scope covering poorly reactive non-ortho-substituted (hetero)aryl carboxylic acids, and tolerates diverse functional groups including some of the groups active to Ni-0 catalysts. The key to achieve this decarbonylative borylation reaction is the choice of B(2)cat(2) as a coupling partner that not only acts as a borylating reagent, but also chemoselectively activates aryl carboxylic acids towards oxidative addition of their C(acyl)-O bond to Ni-0 catalyst via the formation of acyloxyboron compounds. A combination of experimental and computational studies reveals a detailed plausible mechanism for this reaction system, which involves a hitherto unknown concerted decarbonylation and reductive elimination step that generates the aryl boronic ester product. This mode of boron-promoted carboxylic acid activation is also applicable to other types of reactions.

A series of trisubstituted fluoroalkenes can be obtained in a highly stereoselective manner by mixing of simple starting materials alkynes, ethyl bromodifluoroacetate, and arylboronic acids in the presence of nickel catalyst. This method features low-cost catalyst, mild reaction conditions, and excellent functional group compatibility.

Abstract:Selective construction of two distinct alkyl–alkyl bonds across the C=C bond of simple unactivated olefins is a persistent challenge in organic synthesis. Transition metal-catalysed alkene-dicarbofunctionalization reactions proceeding via classic organometallic elementary reaction mechanism (oxidative addition/transmetalation/reductive elimination) suffered from non-bond forming side reactivities of alkyl–metal intermediates (facile β-hydride elimination) and thus strongly limited the substrate scope of olefins and coupling reagents. Here we demonstrate that the problem of alkyl–metal side reactivities can be overcome with an electron-shuttle catalysis approach, wherein the metal catalyst acts as an electron shuttle to induce and quench the radical intermediates and facilitate the alkyl–alkyl bond formation via radical-unsaturated bond addition. This strategy was applied to a modular alkylative aminomethylation of olefins with two distinct alkyl–alkyl bonds generated through nickel electron-shuttle catalysis. Mechanistic studies supported the formation of carbon-centred radical species and the involvement of radical addition processes.

Abstract:We report herein a three-component 1,2-arylboration of alkenyl amines bearing a cleavable picolinamide directing group. With aryl halides as electrophiles and B<sub>2</sub>Pin<sub>2</sub> as nucleophiles, a wide range of alkenes could be converted into valuable boryl-functionalized aliphatic amines. The reaction proceeds with high levels of chemo- and regiocontrol and exhibits high functional group tolerance. In addition, the pinacol boronic ester group could undergo various transformations, indicating that the protocol could potentially provide a platform for versatile regioselective difunctionalization of alkenyl amines.

Abstract:An unprecedented regioselective trans-hydroboration and carboboration of unbiased electronically internal alkynes were realized via nickel catalysis system with the aid of the directing group strategy. Furthermore, the excellent α- and β-regioselectivity could be accurately switched by the nitrogen ligand (terpy) and phosphine ligand (Xantphos). Mechanism studies get an insight into the rational reaction process, that underwent the cis-to-trans isomerization of alkenyl nickel species. This transformation not only expands the scope of transition-metal-catalyzed boration of internal alkynes but also, more particularly, portraits the vast perspectives of the directing group strategy in the selective functionalization of unactivated alkynes.
