Abstract:The innate immune system serves as the first line of host defense. Transforming growth factor-β–activated kinase 1 (TAK1) is a key regulator of innate immunity, cell survival, and cellular homeostasis. Because of its importance in immunity, several pathogens have evolved to carry TAK1 inhibitors. In response, hosts have evolved to sense TAK1 inhibition and induce robust lytic cell death, PANoptosis, mediated by the RIPK1-PANoptosome. PANoptosis is a unique innate immune inflammatory lytic cell death pathway initiated by an innate immune sensor and driven by caspases and RIPKs. While PANoptosis can be beneficial to clear pathogens, excess activation is linked to pathology. Therefore, understanding the molecular mechanisms regulating TAK1 inhibitor (TAK1i)-induced PANoptosis is central to our understanding of RIPK1 in health and disease.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to understand the mechanism of action of protein phosphatase PP6 in TAK1 inhibitor (TAK1i) - induced PANoptosis. Specifically, through genome - wide CRISPR screening, the researchers found that PP6 and its regulatory subunits play a crucial role in TAK1i - induced cell death. The paper further explored how PP6 affects the occurrence of PANoptosis by regulating the phosphorylation state of RIPK1. In particular, PP6 promotes the autophosphorylation of the death - signal phosphorylation site S166 of RIPK1 by reducing the phosphorylation of the survival - signal phosphorylation site S321 of RIPK1, thereby driving PANoptosis. ### Research Background - **Innate Immune System**: As the first line of defense in host defense, the innate immune system senses pathogen - associated molecular patterns (PAMPs), damage - associated molecular patterns (DAMPs), and homeostatic - alteration molecular processes (HAMPs) through pattern - recognition receptors (PRRs) to initiate inflammatory responses and cell death. - **The Role of TAK1**: TAK1 is a key regulator of innate immunity, cell survival, and cell homeostasis. Many pathogens have evolved the ability to carry TAK1 inhibitors to evade or disrupt host defenses. The host has evolved the ability to sense TAK1 inhibition and induce strong lytic cell death (PANoptosis). - **PANoptosis**: This is a unique innate - immune - inflammatory - lytic cell death pathway, initiated by innate - immune sensors and driven by caspases and RIPKs. Although PANoptosis helps to clear pathogens, excessive activation is associated with pathology. ### Research Methods - **Genome - wide CRISPR Screening**: The researchers used genome - wide CRISPR screening to identify the role of PP6 and its regulatory subunits in TAK1i - induced cell death. - **siRNA Knock - down Experiments**: By knocking down different subunits of PP6 with siRNA, their functions in TAK1i - induced cell death were verified. - **Immunoblotting Analysis**: The expression and phosphorylation states of related proteins in cell lysates were detected, especially the S166 and S321 phosphorylation sites of RIPK1. ### Main Findings - **The Role of PP6 in TAK1i - induced Cell Death**: The catalytic subunit PPP6C of PP6 and its regulatory subunits PPP6R1, PPP6R2, and PPP6R3 play a crucial role in TAK1i - induced cell death. Knocking down PPP6C significantly reduced TAK1i - induced cell death, and knocking down all three regulatory subunits in combination was required to completely block TAK1i - induced cell death. - **PP6 Regulates RIPK1 Phosphorylation**: PP6 promotes the autophosphorylation of the death - signal phosphorylation site S166 of RIPK1 by reducing the phosphorylation of the survival - signal phosphorylation site S321 of RIPK1, thereby driving PANoptosis. ### Conclusions - **The Importance of PP6 in TAK1i - induced PANoptosis**: The research results indicate that the PP6 complex plays a crucial role in TAK1i - induced, RIPK1 - dependent PANoptosis, which may provide new targets for the treatment of inflammatory diseases. ### Future Research Directions - **Further Exploration of the Roles of Other Phosphatases**: Investigate whether other phosphatases are also involved in regulating the phosphorylation state of RIPK1. - **The Cooperative Mechanism between PP6 and PP1γ**: Explore whether PP6 and its subunits cooperate with PP1γ to regulate TAK1i - induced PANoptosis. - **The Role of mtROS in TAK1i - induced Cell Death**: Study the role of mitochondrial ROS (mtROS) in TAK1i - induced cell death. Through these studies, the authors hope to provide new insights into the molecular mechanisms of TAK1i - induced PANoptosis and potential targets for the development of new strategies for the treatment of inflammatory diseases.
