Abstract:Aerobic granular sludge (AGS) was rapidly cultivated in an integrated oxidation ditch with two-zone clarifiers by using a novel external sludge treatment and return mode to treat low concentrations of actual domestic sewage. The selective pressure created by the two-zone clarifiers can retain the well-settling granules and discharge light flocs with poor settleability. The granules stayed in the reactor, which induced bacterial attachment to the granules that acted as nuclei, while the discharged flocs can stimulate microorganisms to secrete large amounts of extracellular polymeric substances (EPS) under the external conditioning of CaCl2 and natural air drying. Then, this surplus sludge was returned to the reactor to create more small granules that combined with each other through the action of hydraulic shear forces to achieve rapid granulation. The results showed that AGS was formed successfully in the reactor on day 18, and after 51 days of continuous operation, the biomass concentration and settling ability were further improved (the mixed liquor suspended solids (MLSS) and sludge volume index at 5 min (SVI5) were stable at approximately 3500 mg/L and 40.0 mL/g, respectively). During the whole experimental period, the biological sludge activity was greatly improved, and the EPS and microbial community changed significantly, including an enrichment of microbes with EPS secretion and granule stabilization functions. The study results reveal that the pollutant removal efficiency improved after granulation. Furthermore, this approach required less energy and is eco-friendly for potential full-scale implementation.

Abstract:A pilot-scale sequencing batch reactor (SBR) treating 120 m3/d of a town's wastewater was set up in 2009 and aerobic granules with a mean diameter of 0.28 mm, mixed liquor suspended solids (MLSS) of 7,500 mg/L and sludge volume index (SVI)30 of 43 mL/g were achieved. A full-scale SBR with 50,000 m3/d for treating a town's wastewater was operated in 2010 and aerobic granules with a mean MLSS of 2,285 mg/L and SVI30 of 52.5 mL/g were obtained. Aerobic granules had excellent performances of chemical oxygen demand (COD) and NH4+-N removal and remained stable for a long time. Raw wastewater and SBR operating mode had a positive effect on aerobic granule formation. Therefore, aerobic granular technology could be successfully applied in the full-scale bioreactor under specific conditions. Future development of aerobic granular technology is the application in full-scale continuous-flow reactors.

Abstract:Aerobic granular sludge (AGS) has been recently studied and developed as a way to circumvent the poor settling and biomass retention capacity of conventional activated sludge. While it is known that AGS allows the simultaneous presence of oxic and anoxic zones on the surface or within granules, respectively, the actual composition dynamics of those granules in relation to their underlying wastewater influent have receive little attention. The main goal of this study is thus to assess the relationship between wastewater composition, microbial community structure and epistylis abundance. Two SBRs (sequencing batch reactors) comprising the same inoculum and fed with either domestic or synthetic wastewater were used in this regard. The more complex composition of domestic wastewater promoted a higher bacterial richness than their synthetic counterparts. Indeed, the vast majority of the bacterial community in the SBR fed with synthetic wastewater was dominated by the genus Thauera. The SBR fed with domestic wastewater also showed a more thorough granulation and greater treatment efficiency. Surprisingly, the abundance of epistylis was positively correlated with remaining suspended solids, hinting that ciliates might be responsible for SS (suspended solids) removal and would be a desirable trait to include in wastewater treatment plants. In sum, our study gives insight into the differing population dynamics shaped by domestic and synthetic wastewaters inoculated with the same initial consortium, along with their overall pollutant removal efficiency.
Aerobic granular sludge process: a fast growing biological treatment for sustainable wastewater treatment

Abstract:Aerobic granular sludge (AGS) was developed as a better replacement for activated sludge (AS) and overcoming sludge–water separation issues in the biological treatment of municipal and industrial wastewaters. AGS consists of dense self-immobilized microbial granules that have compact microbial structure, high biopolymer content and higher settling velocities. Granular microbial growth allows effective biomass-water separation, higher biomass concentrations in the bioreactor and simultaneous nitrification, denitrification and phosphate removal from wastewater in a single treatment tank. In this way, AGS process minimizes the land footprint (50%–75% lower) and energy (30%–48% lower) of wastewater treatment plants (WWTPs) as compared to conventional AS process. In the last 20 years, AGS process has grown into a mature biotechnological solution from laboratory-scale to full-scale WWTPs. The other benefits of AGS process are lower sludge production and resource utility of excess sludge. Prevailing microbial groups and their unique metabolism contributes to lower sludge production in AGS process. Extraction of alginate-like exopolymer increases the resource recovery and aid in the sludge management. However, the long start-up periods for AGS formation and disintegration of AGS are seen as constraints for widespread implementation of AGS technology. Extensive research in this area already identified key operational parameters and strategies for improved start-up of granulation. The bioreactor operating conditions such as anaerobic feeding, feast-famine regime and short settling periods impose selection of slow-growing bacteria with unique metabolic traits and favour granulation. Anaerobic feeding and sludge removal strategies are important in maintaining the stability of granules. Further studies on molecular aspects of granulation would be crucial for widespread implementation of this emerging technology in WWTPs.

Abstract:Treatment of municipal wastewaters with aerobic granular sludge (AGS) has been extensively researched in the past decade and has now become a mature option for implementation. Aerobic granules are distinguished from activated sludge flocs through their larger size and more compact and spherical structure. Due to these properties, granules settle rapidly and can therefore contribute to compact treatment processes through high sludge concentrations and short settling times. In this review, the factors that promote granulation in treatment processes are identified and discussed and the experience of municipal wastewater treatment with AGS at laboratory-, pilot-, and full-scale are critically evaluated. The most important factors to promote granulation include exposing the biomass to relatively high concentrations of contaminants in sequencing batch reactors, promoting slow-growing microorganisms and applying a relatively short settling time. Enhanced biological phosphorus removal is preferably integrated with AGS and the large size of the granules makes simultaneous nitrification (at the surface of the granules) and denitrification (at the inner, anoxic parts) feasible. We propose directions for future research including further optimization of AGS to obtain stable and low effluent nutrient concentrations in line with increasingly stringent upcoming effluent demands.

Abstract:Aerobic granular sludge (AGS) is a novel microbial community which allows simultaneous removal of carbon, nitrogen, phosphorus and other pollutants in a single sludge system. AGS is distinct from activated sludge in physical, chemical and microbiological properties and offers compact and cost-effective treatment for removing oxidized and reduced contaminants from wastewater. AGS sequencing batch reactors have shown their utility in the treatment of abattoir, live-stock, rubber, landfill leachate, dairy, brewery, textile and other effluents. AGS is extensively researched for wide-spread implementation in sewage treatment plants. However, formation of AGS takes relatively much longer time while treating low-strength wastewaters like sewage. Strategies like increased volumetric flow by means of short cycles and mixing of sewage with industrial wastewaters can promote AGS formation while treating low-strength sewage. This article reviewed the state of research on AGS formation mechanisms, bioremediation capabilities and biotechnological applications of AGS technology in domestic and industrial wastewater treatment.

Abstract:It was an important discovery in wastewater treatment that the microorganisms in the traditional activated sludge can form aerobic granular sludge (AGS) by self-aggregation under appropriate water quality and operation conditions. With a typical three-dimensional spherical structure, AGS has high sludge-water separation efficiency, great treatment capacity, and strong tolerance to toxic and harmful substances, so it has been considered to be one of the most promising wastewater treatment technologies. This paper comprehensively reviewed AGS from multiple perspectives over the past two decades, including the culture conditions, granulation mechanisms, metabolic and structural stability, storage, and its diverse applications. Some important issues, such as the reproducibility of culture conditions and the structural and functional stability during application and storage, were also summarized, and the research prospects were put forward. The aggregation behavior of microorganisms in AGS was explained from the perspectives of physiology and ecology of complex populations. The storage of AGS is considered to have large commercial potential value with the increase of large-scale applications. The purpose of this paper is to provide a reference for the systematic and in-depth study on the sludge aerobic granulation process.

Abstract:The granulation of sludge is a process of microbial self-immobilization during the wastewater treatment process under favorable conditions.Granular sludge is a denser,larger and better microbial aggregate.According to the electron acceptors of microbial metabolism,granular sludge can be divided into two general groups,that is,anaerobic and aerobic granular sludge.Anaerobic granular sludge has been studied extensively for decades,whereas the interest in aerobic granular sludge was started just several years ago.To date,the researches on the aerobic granulation have been focused on two aspects: one is on the formation of aerobic granular sludge on macro-scale,in order to optimize the operational parameters;the other is on the granulation mechanisms and the microbial diversity.This paper reviewed the up-to-date research and application of aerobic granular sludge not only in China,but also worldwide.The main contents include: the performance of aerobic granular sludge(morphology and size,settling ability,density and strength,biological activity,cell surface hydrophobicity,extracellular polysaccharides etc.),the cultivating conditions,granulation mechanisms and the influencing factors(organic loading rates,substrates constituents,hydrodynamic shear force,settling time,operational period,feeding mode,periodic starvation,reactor configuration,dissolved oxygen and temperature etc.),as well as the application of aerobic granular sludge(industrial wastewater treatment,municipal sewage treatment,toxic organics degradation,removal of nitrogen and phosphorus,and biosorption of heavy metal ions and radioactive wastes etc.).The completely autotrophic granular sludge,such as nitrifying granular sludge,was also mentioned.The future concern will focus on the up-scaling,manipulating,and optimizing the formation of aerobic granules,the changes in microbial activity and community in mature aerobic granules,as well as their long-term stability in the reactors.The intensive and successful researches of this technology will promote it from laboratory study to the pilot-and full-scale applications,which will foresee its promising application in practical wastewater treatment.

Abstract:Aerobic granular sludge (AGS) technology is gaining increasing interest globally. However, several aspects are still challenging its installation, as the integration into existing municipal wastewater treatment plants. The operation of AGS units in continuous flow mode could facilitate that integration; yet has only been experienced at lab and pilot scales. Further, handling of excess granular sludge constitutes a prominent challenge. Even that, adopting effective strategies for its management holds promising contributions to the circular economy by fostering the reuse and recovery of valuable resources. The recovery of phosphorus and alginate‐like polymers holds significant commercial potential worldwide, with broad applications in agriculture, food, pharmaceuticals, and biotechnology.

Abstract:Aerobic granular sludge can be classified as a type of self-immobilized microbial consortium, consisting mainly of aerobic and facultative bacteria and is distinct from anaerobic granular methanogenic sludge. Aerobic granular technology has been proposed as a promising technology for wastewater treatment, but is not yet established as a large-scale application. Aerobic granules have been cultured mainly in sequenced batch reactors (SBR) under hydraulic selection pressure. The factors influencing aerobic granulation, granulation mechanisms, microbial communities and the potential applications for the treatment of various wastewaters have been studied comprehensively on the laboratory-scale. Aerobic granular sludge has shown a potential for nitrogen removal, but is less competitive for the high strength organic wastewater treatments. This technology has been developed from the laboratory-scale to pilot scale applications, but with limited and unpublished full-scale applications for municipal wastewater treatment. The future needs and limitations for aerobic granular technology are discussed.

Abstract:Aerobic granular sludge (AGS) in continuous-flow reactors (CFRs) has attracted significant interest, with notable progress in research and application over the past two decades. Cumulative studies have shown that AGS-CFRs exhibit comparable morphology, settleability, and pollutant removal efficiency to AGS cultivated in sequencing batch reactors, despite their smaller particle sizes. Shear force and selection pressure are the primary drivers of granulation. While not mandatory for granulation, feast/famine conditions play a crucial role in ensuring long-term stability and nutrient removal. Additionally, bioaugmentation can facilitate the granulation process. Furthermore, this paper comprehensively assesses the application of AGS-CFRs in full-scale wastewater treatment plants (WWTPs). Currently, AGS-CFRs have been implemented in nine WWTPs, encompassing two distinct processes. Hydrocyclone-based densified activated sludge significantly enhances sludge density, settleability, and biological phosphorus removal efficiency, thus increasing treatment capacity. The microaerobic–aerobic configuration with internal separators can induce granulation, ensuring long-term stability, eliminating the need for external clarifiers, and reducing land and energy requirements. This review demonstrates the high potential of AGS-CFRs for intensifying existing WWTPs with minimal retrofitting needs. However, further research is required in granulation mechanisms, long-term stability, and nutrient removal to promote the widespread adoption of AGS.

Abstract:Aerobic granular sludge (AGS), a self-immobilized microbial consortium containing different functional microorganisms, is receiving growing attention, since it has shown great technological and economical potentials in the field of wastewater treatment. Microbial community is crucial for the formation, stability, and pollutant removal efficiency of aerobic granules. This mini-review systematically summarizes the recent findings of the microbial community structure and function of AGS and discusses the new research progress in the microbial community dynamics during the granulation process and spatial distribution patterns of the microbiota in AGS. The presented information may be helpful for the in-depth theoretical study and practical application of AGS technology in the future.

Abstract:Aerobic granular sludge (AGS) is a recent innovative technology and is considered a forthcoming biological process for sustainable wastewater treatment. AGS is composed of the dense microbial consortium of aerobic, anaerobic, and facultative types of bacteria. The mechanism of AGS formation and its stability for long-term operation is still a subject of current research. On the other hand, AGS makes the treatment process sustainable in a cost-effective way. However, in order for AGS to be applied in a broader range of applications, there are several challenges to overcome, such as slow-speed granulation and the disintegration of AGS after granulation. Many factors play a role in the stability of granules. The storage of granules and the later use of them for granulation startup is a feasible method for reducing the time for granulation and maintaining stability. This review focuses on the granulation process and characteristics of AGS, granulation time and the stability of AGS under different conditions, the comparison of different storage methods of granules, and their recovery and rejuvenation. From this review, it is evident that additional research is required to assess the effectiveness of regenerated AGS after prolonged storage to promote AGS technology for commercial applications.

Abstract:Aerobic granular sludge (AGS), as an aggregate of numerous self-immobilized functional microorganisms, has been recognized as an increasingly viable option for wastewater treatment and reuse water production. This study aimed at evaluating the application of AGS reactor as primary, ultrafiltration (UF) as secondary and nanofiltration (NF) as tertiary treatment for the treatment of municipal wastewater, focusing on determining the membrane fouling mechanism and reuse water production. AGS reactor, UF and NF exhibit high removal efficiency for COD (51.33%, 90.48% and 99.26%) and nutrients (53.63%, 94.84% and 98.06% total nitrogen, and 49.8%, 97.07% and 98.73% phosphorus), indicating that the integrated aerobic granular sludge and UF/NF process could provide high pollution removal and quality reuse water. As for filtration behavior of UF and NF, shear stress produced by agitation speed could significantly improve flux, due to the dispersion of pollutants. Besides, for AGS, improved simultaneous nitrification and denitrification by the internal anaerobic, anoxic, and aerobic structure and the richer biological community increased foulant removal, while the larger pore size and mature structure also reduced foulant deposition. Moreover, shear stress also diminished the total fouling resistance, reversible fouling and irreversible fouling, as well membrane cleaning efficiency was promoted, by controlling AGS deposition on membrane. In addition, the fouling mechanism of conventional floc sludge and that of AGS were deeply analyzed and respectively compared, from three aspects, namely, Scanning Electron Microscope (SEM), Attenuated Total Reflection Fourier Transform Infrared Spectroscopy (ATR-FTIR) and Atomic Force Microscope (AFM). Overall, the results demonstrated an alternative option for water treatment and reuse water production in an integrated AGS and membrane process.

Abstract:Activated sludge is one of the most widely implemented technologies for municipal wastewater treatment. Yet, more restrictive environmental standards demand for more efficient technologies. Aerobic granular sludge (AGS) is a promising alternative in this context since this technology has shown potential for simultaneous organic matter and nutrient removal using smaller bioreactors and consuming less energy. However, despite such engaging claims, only ca. 40 full-scale AGS systems have been installed worldwide after 30 years of development. This reduced implementation suggests the existence of significant bottlenecks for this technology, which currently only have partially been overcome. This overview aims to analyze the recent progress in R&D concerning aerobic sludge granulation for municipal wastewater treatment via the analysis of research articles and invention patents as well as to elucidate exiting technological gaps and development opportunities. Culturing methods aiming at fast granulation, long-term stability and excellent process performance are of utmost interest for promoting massive implementation of full-scale AGS systems. Moreover, the recovery of biomaterials from waste sludge could contribute to the implementation of the biorefinery paradigm in wastewater treatment plants.

Abstract:Aerobic granular sludge (AGS) comprises an aggregation of microbial cells in a tridimensional matrix, which is able to remove carbon, nitrogen and phosphorous as well as other pollutants in a single bioreactor under the same operational conditions. During the past decades, the feasibility of implementing AGS in wastewater treatment plants (WWTPs) for treating sewage using fundamentally sequential batch reactors (SBRs) has been studied. However, granular sludge technology using SBRs has several disadvantages. For instance, it can present certain drawbacks for the treatment of high flow rates; furthermore, the quantity of retained biomass is limited by volume exchange. Therefore, the development of continuous flow reactors (CFRs) has come to be regarded as a more competitive option. This is why numerous investigations have been undertaken in recent years in search of different designs of CFR systems that would enable the effective treatment of urban and industrial wastewater, keeping the stability of granular biomass. However, despite these efforts, satisfactory results have yet to be achieved. Consequently, it remains necessary to carry out new technical approaches that would provide more effective and efficient AGS-CFR systems. In particular, it is imperative to develop continuous flow granular systems that can both retain granular biomass and efficiently treat wastewater, obviously with low construction, maintenance and exploitation cost. In this review, we collect the most recent information on different technological approaches aimed at establishing AGS-CFR systems, making possible their upscaling to real plant conditions. We discuss the advantages and disadvantages of these proposals and suggest future trends in the application of aerobic granular systems. Accordingly, we analyze the most significant technical and biological implications of this innovative technology.

Abstract:As a biological promising wastewater treatment technology, aerobic granular sludge (AGS) technology had been widely studied in sequencing batch reactors (SBRs) for the decades. Presently, the whole processes of its granulation, long-term operation, storage, and reactivation have not been thoroughly evaluated, and also the relationships among microbial diversity, granular size, and characteristics were still not that clear. Hence, they were systematically evaluated in an AGS-SBR in this work. The results demonstrated that Proteobacteria and Bacteroidetes were the dominant phyla, Flavobacterium, Acinetobacter, Azoarcus, and Chryseobacterium were the core genera with discrepant abundances in diverse stages or granular size. Microbial immigration was significant in various stages due to microbial diversity had a line relationship with COD/MLVSS ratio (R2 = 0.367). However, microbial diversity had no line relationship with granular size (R2 = 0.001), indicating the microbial diversity in different-sized AGS was similar, although granular size had a line relationship with settleability (R2 = 0.978). Overall, compared to sludge traits (e.g., sludge size, settleability), COD/MLVSS played a key role on microbial evolution. This study revealed the relationships between granule characteristics and microbial community, and contributed to the future AGS-related studies.

Abstract:Aerobic granular sludge (AGS) capable of nitrogen and phosphorus removal is mainly limited to the applications in sequencing batch reactors. This study introduced an innovative continuous self-circulating up-flow fluidized bed process (Zier process) using separate aeration. The process was composed of an anoxic column (Zier-A), aeration column (Zier-OO) and aerobic column (Zier-O), and was used to treat actual municipal sewage continuously for 170 days. The process achieved self-circulation of 20-32 times and an up-flow velocity within the reactor of 7-16 m/h which were accurately controlled with only separate aeration. The larger proportion of self-circulating multiple times contributed to particle formation and stability, providing hydraulic shear conditions, and screened the precipitation performance of the granular sludge (GS). Meanwhile, the dissolved oxygen (DO) of Zier-O was controlled at 0.1-0.3 mg/L, and the DO of Zier-A input water was zero. The accurate oxygen supply enhanced simultaneous nitrification and denitrification (SND) as well as short-cut nitrification and denitrification in Zier-O and improved the COD utilization rate and the nitrogen removal rate in Zier-A. The COD treatment capacity reached 2.46 kg-COD/(m³·d). With a hydraulic retention time of 10 h, the process consistently ensured that the average concentrations of ammonia nitrogen and total nitrogen in the effluent were maintained below 5 and 15 mg/L, respectively. Moreover, the process maintained the shape and stability of GS, the median diameter of GS ranged between 300-1210 μm, the percentage of mass with particle size distribution < 200 μm at a height of 150 cm within Zier-A and Zier-O accounted for as low as 0.04%-0.05%, and showed good settling performance. The suspended solids in effluent can be maintained at 50-80 mg/L. Overall, the unique structural setting and control method of the Zier process provide another approach for the application of continuous AGS treatment for municipal sewage.
Full-scale upgrade activated sludge to continuous-flow aerobic granular sludge: Implementing microaerobic-aerobic configuration with internal separators

Abstract:Aerobic granular sludge (AGS) has been successfully used in sequencing batch reactors. However, their application to existing continuous-flow systems remains challenging. In this study, a novel microaerobic-aerobic configuration with internal separators was implemented in a full-scale municipal wastewater treatment facility with a nominal capacity of 2.5 × 104 m3 d-1. Sludge characteristics, pollutant removal and associated pathways, shifts in the microbial community, and underlying granulation mechanisms were investigated. Following a two-month operation period, the transition from flocculent-activated sludge to well-defined AGS with distinct boundaries and compact structures was successfully achieved. The average size of sludge increased from 31.9 to 138.5 μm, with granules larger than 200 μm constituting 28.9 % of the total sludge and SVI30 averaging 51.4 ± 8.2 mL g-1. The 95th percentile effluent COD, NH4+-N, and TN concentrations were 35.0, 1.2, and 13.3 mg L-1, respectively. The primary pathways for pollutant removal were identified as simultaneous nitrification, denitrification, and phosphorus removal within the microaerobic tanks. The enrichment of denitrifying phosphorus-accumulating organisms, including Hydrogenophaga, Accumulibacter, Azospira, Dechloromonas, and Pseudomonas, provides an essential microbial foundation. Furthermore, computational fluid dynamics modeling revealed that the incorporation of internal separators in aerobic tanks induced shifts in the flow pattern, transitioning from a single-circulation cell to multiple vortical cells. This alteration amplified the local velocity gradients, generating the required shear forces to drive granulation. Moreover, mass balance analysis revealed that the microaerobic and aerobic tanks operated under feast and famine conditions, respectively, creating a microbial selection pressure that favored granulation. This process eliminates the need for external clarifiers, resulting in a footprint reduction of 38.2 % and one-third energy savings for sludge reflux. This study offers valuable insights into the application of continuous-flow AGS to upgrade existing activated sludge systems with limited retrofitting requirements.

The Science of The Total Environment

Abstract:The evolution of emerging contaminants (ECs) has caused greater requirements and challenges to the current biological wastewater treatment technology. As one of the most promising biological treatment technologies, the aerobic granular sludge (AGS) process also faces the challenge of ECs. This study summarizes the recent progress and characteristics of several representative ECs (persistent organic pollutants, endocrine disrupting chemicals, antibiotics, and microplastics) in AGS systems that have garnered widespread attention. Additionally, the biodegradation and adsorption mechanisms of ECs were discussed, and the interactions between various ECs and AGS was elucidated. The importance of extracellular polymeric substances for the stabilization of AGS and the removal of ECs is also discussed. Knowledge gaps and future research directions that may enable the practical application of AGS are highlighted. Overall, AGS processes show great application potential and this review provides guidance for the future implementation of AGS technology as well as elucidating the mechanism of its interaction with ECs.
