Abstract:The increasing demand for tea consumption calls for the development of more products with distinct characteristics. The sensory quality of tencha is significantly determined by innate differences among tea cultivars. However, the correlations between the chemical composition and sensory traits of tencha are still unclear. To enhance the understanding of the flavor formation mechanism in tencha and further to develop new cultivars resources, we investigated non-volatiles and volatile metabolites as well as sensory traits in tencha from different tea cultivars (Camellia sinensis cv. Yabukita, Longjing 43 and Baiye 1); the relationships between the flavor traits and non-volatiles/volatiles were further evaluated by partial least squares - discriminate analysis (PLS-DA), multiple factor analysis (MFA) and multidimensional alignment (MDA) analysis. A total of 64 non-volatiles and 116 volatiles were detected in all samples, among which 71 metabolites were identified as key flavor-chemical contributors involving amino acids, flavonol glycosides, flavones, catechins, ketones, alcohols, hydrocarbons, aldehydes, esters and acids. The levels of taste-related amino acids, flavonol glycosides and gallic acid varied significantly among the tencha samples made from different tea cultivars. All the samples exhibited typical quality characteristics of tencha. The tencha from Camellia sinensis cv. Longjing 43 and Camellia sinensis cv. Baiye 1 (cultivated in the open) exhibited higher levels of amino acids and gallic acid, which were associated with the umami taste and mellow taste of tea infusion. Abundant flavonol glycosides were related to the astringency, while partial tri-glycosides specifically quercetin-3-O-galactoside-rhamnoside-glucoside and total of flavonol galactoside-rhamnoside-glucoside were associated with mellow taste. The floral alcohols were identified as significant contributors to the refreshing aroma traits of tencha. The green, almond-like, acidic and fruity odorants were associated with a green and fresh aroma, while the green, cheesy and waxy odorants such as ketones, esters, acids and hydrocarbons were associated with seaweed-like aroma. This study provides insight into sensory-related chemical profiles of tencha from different tea cultivars, supplying valuable information on flavor and quality identification for tencha.

Abstract:In the present work, a novel infrared-assisted extraction coupled to headspace solid-phase microextraction (IRAE-HS-SPME) followed by gas chromatography-mass spectrometry (GC-MS) was developed for rapid determination of the volatile components in green tea. The extraction parameters such as fiber type, sample amount, infrared power, extraction time, and infrared lamp distance were optimized by orthogonal experimental design. Under optimum conditions, a total of 82 volatile compounds in 21 green tea samples from different geographical origins were identified. Compared with classical water-bath heating, the proposed technique has remarkable advantages of considerably reducing the analytical time and high efficiency. In addition, an effective classification of green teas based on their volatile profiles was achieved by partial least square-discriminant analysis (PLS-DA) and hierarchical clustering analysis (HCA). Furthermore, the application of a dual criterion based on the variable importance in the projection (VIP) values of the PLS-DA models and on the category from one-way univariate analysis (ANOVA) allowed the identification of 12 potential volatile markers, which were considered to make the most important contribution to the discrimination of the samples. The results suggest that IRAE-HS-SPME/GC-MS technique combined with multivariate analysis offers a valuable tool to assess geographical traceability of different tea varieties.

Abstract:In this study, the chemical constituents of pu-erh tea, black tea, and green tea, as well as those of pu-erh tea products of different ages, were analyzed and compared using a chemical profiling approach. Differences in tea processing resulted in differences in the chemical constituents and the color of tea infusions. Human biological responses to pu-erh tea ingestion were also studied by using ultraperformance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOFMS) in conjunction with multivariate statistical techniques. Metabolic alterations during and after pu-erh tea ingestion were characterized by increased urinary excretion of 5-hydroxytryptophan, inositol, and 4-methoxyphenylacetic acid, along with reduced excretion of 3-chlorotyrosine and creatinine. This study highlights the potential for metabonomic technology to assess nutritional interventions and is an important step toward a full understanding of pu-erh tea and its influence on human metabolism.

Abstract:<p>Aroma compounds are one of the most important components in tea, as well as an important indicator for its quality. In this paper, using HS‐SPME/GC‐MS method, we established the finger‐print for aromatic components in green tea produced in Guizhou Province, China. In this finger‐print, there are 9 volatile aromatic components which have significant correlation with the quality scores of green teas' aroma (p<0.05). They contain ethers and aldehydes (dimethyl sulfide and phenylacetaldehyde) of aromatic compounds, and alcohols and esters (1‐penten‐3‐ol, linalool, phenethyl alcohol, geraniol and Methyl salicylate) of aliphatic compounds, and pyrrole compounds of heterocyclic compounds (3‐ethyl‐1‐pyrrole and 1‐ethyl‐2‐carbaldehyde‐1‐pyrrole). The Regression Model has been established for predicting evaluation scores of green teas' aroma. A good result was obtained via verification, indicating that GC‐MS is a feasible way to analyze key volatile aromatic components in green tea, which also can be used to predict quality scores aroma and different grades.</p>

Abstract:Based on the fully automatic headspace solid-phase microextraction (HS-SPME)/gas chromatography-mass spectrometry (GC-MS) and multivariate statistical methods, a novel model of identifying and evaluating the quality of Yunnan Pu-erh green tea was constructed for the first time in this work. Twelve Pu-erh green teas from 12 typical production sites of Pu-erh district in Yunnan Province and 6 regular green teas from Zhejiang, Sichuan, Anhui, Henan, Hubei, and Jiangsu provinces of China were used to construct the model. Data from 18 green tea samples by GC-MS were processed with fingerprint technology and chemometric methods. The GC-MS fingerprints from 12 Pu-erh green teas whose correlation coefficients and congruence coefficients were over 0.850 and demonstrated Pu-erh green tea samples from different production sites in Yunnan were consistent to some extent in spite of slightly different chemical indexes. A total of 77 volatile compounds were identified in 18 green teas, mainly including linalool, linalool oxides, phytol, caffeine, geraniol, and dihydroactinidiolide, and their chemical compositions were slightly similar. Cluster analysis (CA) and principal component analysis (PCA) demonstrated that 12 Pu-erh green teas could be clearly distinguished from other six regular green teas according to their chemical characteristics. Our results thus indicate that the chromatographic fingerprint combined with multivariate statistical techniques is useful for the identity and consistency evaluation of Pu-erh green teas. Such an approach is believed to be equally applicable to other green teas.

Abstract:Tea (Camellia sinensis L), an important drink and a natural medicine for thousands of years, contains many health beneficial compounds. Growing season, geographical region, and fermentation methods create many variations in tea compositions, which contribute to each tea's uniqueness. In this study, a simple, rapid, and efficient ultra-performance liquid chromatography (UPLC) method combined with diode array detector (DAD) and mass spectroscopic (MS) detection and chemometrics analysis was used to analyse three different types of teas (green pu-erh, green tea, white tea). Using the developed method, 68 compounds were identified and 54 were quantified based on retention times. UV spectra, and MS spectra by referencing to available standards and data in the literatures. The results showed the chemical differences between the tested teas. Principal component analysis (PCA) was applied to classify and distinguish between tea samples. Published by Elsevier Ltd.

Abstract:As the basic element of tea, aroma is one of the most important factors determining the quality of tea. In this paper, a novel and objective method for rapid discrimination and classification of green tea aromas was developed by coupling of dynamic headspace sampling high-pressure photoionization/chemical ionization-time-of-flight mass spectrometer (HPPI-TOFMS) and multivariate statistical analysis techniques, such as partial least squares discrimination analysis (PLS-DA) and layer cluster analysis (HCA). The classification efficiency was evaluated by analyzing the receiver operating characteristic (ROC) curve. The observations and the robustness were assessed using 200 permutation tests. The volatile fingerprint mass spectra of 31 green tea samples of 4 sub. types (tender chestnut-like, tender-like, chestnut-like and roasted chestnut-like aroma) collected from different production areas in Sichuan, Guizhou, Zhejiang, Anhui, etc, were obtained based on this method. In the HPPI/CI mass spectra of the aroma VOCs from different sub-types of green teas, the types and relative intensities of the characteristic mass peaks were obviously different. The results of multivariate statistical analysis showed that there were little differences among the aroma VOCs of the same sub-type, but significant differences were exhibited among different sub-types, which could achieve accurate classification and identification. The prediction ability of the model for unknown samples reaches 85.1%. This method provided a new idea and approach for the effective identification of green tea aromas, indicating that it had potential application value and broad development prospects in the fields of food quality and safety evaluation.
Characterization of the aroma-active compounds in Xiaokeng green tea by three pretreatment methods combined with gas chromatography-olfactometry (GC-O)

Abstract:Chinese Xiaokeng green tea (XKGT) possesses elegant and fascinating aroma characteristics, but its key odorants are still unknown. In this study, 124 volatile compounds in the XKGT infusion were identified by headspace-solid phase microextraction (HS-SPME), stir bar sorptive extraction (SBSE), and solvent extraction-solid phase extraction (SE-SPE) combined with gas chromatography-mass spectrometry (GC-MS). Comparing these three pretreatments, we found HS-SPME was more efficient for headspace compounds while SE-SPE was more efficient for volatiles with higher boiling points. Furthermore, SBSE showed more sensitive to capture ketones then was effective to the application of pretreatment of aroma analysis in green tea. The aroma intensities (AIs) were further identified by gas chromatography-olfactometry (GC-O). According to the AI and relative odor activity value (rOAV), 27 compounds were identified as aroma-active compounds. Quantitative descriptive analysis (QDA) showed that the characteristic aroma attributes of XKGT were chestnut-like, corn-like, fresh, and so on. The results of network analysis showed that (E, Z)-2,6-nonadienal, nonanal, octanal and nerolidol were responsible for the fresh aroma. Similarly, dimethyl sulfide, (E, E)-2,4-heptadienal, (E)-2-octenal and β-cyclocitral contributed to the corn-like aroma. Furthermore, indole was responsible for the chestnut-like and soybean-like aroma. This study contributes to a better understanding of the molecular mechanism of the aroma characteristics of XKGT.

Abstract:The quality of tea is presently evaluated by the sensory assessment of professional tea tasters, however, this approach is both inconsistent and inaccurate. A more standardized and efficient method is urgently needed to objectively evaluate tea quality. In this study, the chemical fingerprint of 7 different Dayi Pu-erh tea brands and 3 different Ya'an tea brands on the market were analyzed using fully automatic headspace solid-phase microextraction (HS-SPME) combined with gas chromatography-mass spectrometry (GC-MS). A total of 78 volatiles were separated, among 75 volatiles were identified by GC-MS in seven Dayi Pu-erh teas, and the major chemical components included methoxyphenolic compounds, hydrocarbons, and alcohol compounds, such as 1,2,3-trimethoxybenzene, 1,2,4-trimethoxybenzene, 2,6,10,14-tetramethyl-pentadecane, linalool and its oxides, α-terpineol, and phytol. The overlapping ratio of peaks (ORP) of the chromatogram in the seven Dayi Pu-erh tea samples was greater than 89.55%, whereas the ORP of Ya'an tea samples was less than 79.10%. The similarity and differences of the Dayi Pu-erh tea samples were also characterized using correlation coefficient similarity and principal component analysis (PCA). The results showed that the correlation coefficient of similarity of the seven Dayi Pu-erh tea samples was greater than 0.820 and was gathered in a specific area, which showed that samples from different brands were basically the same, despite have some slightly differences of chemical indexes was found. These results showed that the GC-MS fingerprint combined with the PCA approach can be used as an effective tool for the quality assessment and control of Pu-erh tea.

Abstract:The volatile components of Meitan Cuiya tea in Guizhou were determined, which provides theoretical basis to improve and enhance the aroma quality of Meitan Cuiya tea. Headspace solid-phase microextraction method (HS-SPME) combined with gas chromatography-mass spectrometry (GC-MS) was used for the analysis of aroma components from Meitan Cuiya tea. The relative contents of volatile substances in samples were determined and compared. Meitan Cuiya tea had 43 kinds of volatile compounds, among them, the predominant compounts were alcohols, esters, aldehydes, ketones, hydrocarbons and others, of which the major ones were ethanal (2.444%), methyl sulfide (5.264%), linalool (5.106%), trans-Geraniol (2.587%), dihydroactinidiolide (2.364%), hexadecane (3.605%) and caffeine (26.348%). The GC-MS method had the advantages of rapid, simple and accurate and it was suitable for the study of aroma components in Meitan Cuiya tea. It provides advanced technical support for further studies on the chemical composition of tea aroma components and formation mechanism of Meitan Cuiya tea aroma quality.
GC-MS Combined with Proteomic Analysis of Volatile Compounds and Formation Mechanisms in Green Teas with Different Aroma Types

Abstract:Aroma is one of the key factors for evaluating the quality of green tea. A tender aroma (NX) and floral-like aroma (HX) are two types of high-quality aroma of green tea. In this work, the different aroma types of baked green tea were classified by sensory evaluation. Then, seven tea samples with a typical tender or floral-like aroma were selected for further volatile component analysis by GC-MS. A total of 43 aroma compounds were identified in two different aroma types of baked green tea samples. The PCA showed that linalool, geraniol, 3-hexenyl butyrate, and 3-hexenyl hexanoate were the major volatiles contributing to the HX. On the other hand, most of the alcohol volatiles, such as 1-octanol, 1-octen-3-ol, 1-dodecanol, 1-hexadecanol, phenylethyl alcohol, benzyl alcohol, aldehydes and some hydrocarbons contributed more to the NX. In addition, the chemical composition analysis showed that the content of free amino acids was higher in NX green tea samples, while the content of catechins was relatively higher in HX tea samples. A proteomic analysis revealed that most of the enzymes involved in VPBs pathways, such as phenylalanine ammonialyase, peroxidase, and shikimate-O-hydroxycinnamoyl transferase, were more abundant in NX than in HX tea samples. These results laid a foundation for the aroma formation mechanism of different aroma types of baked green tea and provided some theoretical guidance for the breeding of specific aroma varieties.
Evaluation of Aroma Components in Chinese Southwest Tobacco by Headspace Gas Chromatography-Mass Spectrometry

Abstract:In this paper, a simple method of headspace analysis (HSA) was applied to determine aroma components in Chinese southwest tobacco. Ninety-seven aroma compounds were extracted from tobacco under optimized experimental conditions. Meanwhile, it took 4 h for traditional method namely simultaneous distillation and extraction (SDE) for analysis, which is time-consuming and a waste of solvent. Compared with simultaneous distillation and extraction, headspace analysis is faster and more convenient in extracting tobacco aroma compounds. Moreover, six aroma compounds were first proposed in the research. The main aromatic components in tobacco detected by headspace can evaluate the tobacco quality and distinguish a specific variety from others. Headspace analysis method was proposed as an easy, rapid and environment-friendly method for the determination of aroma components in tobacco. The proposed method can not only enhance the extracting rate of aromas but meet the demand of qualitative analysis.

Abstract:Aroma substances are the most crucial criteria for the sensory evaluation of tea quality, and also key attractors influencing consumers to make the decision for purchasing tea. Understanding the aromatic properties of tea infusion during different brewing time is crucial to control the tea aromatic quality. Here, headspace and direct immersion solid-phase microextraction (HS-SPME and DI-SPME), coupled with GC-MS, were employed to investigate the impact of brewing time on the changes of the volatile features of green tea infusion. Esters, aldehydes, alcohols, fatty acids, and alkaloids were the predominant volatile groups from tea infusions. Two to three minutes was identified as the best duration for the tea brewing that can maximize the abundance of aromatic chemicals in the headspace emitted from the tea infusions. The variation of the key aromatic contributors between the tea infusion and the headspace over the infusion tended to equilibrate during the tea brewing process. This study provides a theory-based reference method by analyzing the real-time aromatic characteristics in green tea. The optimal time was determined for aromatic quality control, and the complementary relationship between the volatiles in the headspace and its counterpart, tea infusion, was primarily elucidated.

Abstract:To reveal the characteristic chemical profiles of Pu-erh raw tea (PRT) and traditional green tea (TGT), a high-throughput analytical method based on UPLC-Q-Orbitrap-MS/MS was proposed. 145 components were characterized with a three-level qualitative strategy and the integrated filtering strategy combining nitrogen rule, mass defect, and diagnostic ions information. 124 components were quantified using an internal standard method. The total contents of flavan-3-ols and derivatives, phenolic acids and derivatives were higher in PRT than TGT, while flavonoids were reversed. Furthermore, partial least squares-discriminant analysis (PLS-DA) models were established to classify TGT and PRT. 23 characteristic components were revealed by variable importance in projection method. Their difference in content is between 1.5 and 7.3 times for PRT and TGT. The results showed the chemical characteristics of TGT and PRT clearly and comprehensively. The high-throughput method demonstrated considerable potential in the analysis of complex chemical system, such as food and herbs.

Parameters that influenced the extraction were optimized, and the optimal conditions for the proposed method were as follows: the saturation of sodium chloride of 100%, the ratio of tea to water of 1:5.92, and a total extraction time of 60 min comprising 11.9 min at 30 °C, 17.4 min at 50 °C, and 30.7 min at 70 °C. Under these optimal conditions, the total peak area of the volatile compounds and the number of the identified compounds using the proposed method were both more than those by using the conventional method, which indicated that temperature affects the extraction efficiency of compounds with different volatilities. Using combined temperatures in a single HS-SPME procedure could obtain more information including contents and number of the volatile compounds identified in the determination of volatile compounds with different volatilities of black tea and other complex substrates.

Abstract:A static headspace-gas chromatography-mass spectrometry(HS-GC-MS) method was used to analyze aroma components from Pu-erh tea. The separated peaks were identified by mass spectral library searching combined with retention index comparison. Thirty aroma components were identified from Pu-erh tea, mainly including alco hols(47.73%) and esters(40.97%). The precision expressed as relative standard deriva tion(RSD, n = 6) of the proposed method was in the range of 4.11%–6.59%. This method can be widely applied for the qualitative analysis of aroma components from Pu-erh tea.
Evaluation of Roasting Effect on Selected Green Tea Volatile Flavor Compound and Pyrazine Content by HS-SPME GC-MS

Abstract:The present study aims at the development and validation of a quali-quantitative headspace solid-phase microextraction gas chromatography–mass spectrometry (HS-SPME GC-MS) method for the analysis of odorants in different Chinese and Japanese green teas. A 65 μm PDMS/DVB fiber was used, and GC-MS was performed on a ZB-5 (30 m × 0.25 mm ID, 0.25 μm film) column. The main odorants in green tea samples were identified and the method was fully validated. Linearity (r2 0.981–0.999), sensitivity (LOQ 0.005–0.04 μg/mL), reproducibility (CV% 3.12–10.37), accuracy (recovery% 79.08–99.17) and matrix effect (ME% −9.5 to +4.5) were determined. Quantitation of 2,4-heptadienal, β-damascenone, β-ionone, linalool, indole, 2-ethyl-6-methyl-pyrazine, 2-ethyl-5-methyl-pyrazine, 2-ethyl-3,6-dimethylpirazine, 2-ethyl-3,5-dimethyl-pyrazine and 2-acetyl-pyrazine was carried out in the presence of isotopically labeled compounds as the internal standards. The proposed method was applied to the comparison of the profile of the volatile flavor compounds (VFCs) of green tea subjected to roasting treatment at three different temperatures (160, 180 and 200 °C for 30 min). In particular, the roasting process was monitored by following the quantitative variations of the selected odorant content, considered as the most important contributory components to the Hojicha or black tea (roasted tea) flavor. A temperature of 160 °C was found to be the best roasting temperature.
