Abstract:A hexafluoroisopropanol（HFIP）-catalyzed highly diastereoselective formal [4 + 2] cyclization between ortho-hydroxyphenyl para-quinone methides and difluoroenoxysilanes is developed. This tandem protocol provides a simple and straightforward approach to assemble diverse multiply functionalized difluorinated chromans with high to excellent diastereoselectivity by employing difluoroenoxysilane as a new C2 synthon.

Abstract:The first enantioselective total synthesis of (+)-alsmaphorazine E has been achieved through a traceless chirality transfer strategy, which also enabled structural reassignment of the natural product. Abstract:A newly designed thiazoline iminopyridine ligand for enantioselective cobalt-catalyzed sequential Nazarov cyclization/electrophilic fluorination was developed. Various chiral α-fluorocyclopentenones were prepared with good yields and diastereo- and enantioselectivities. Further derivatizations could be easily carried out to provide chiral cyclopentenols with three contiguous stereocenters. Furthermore, a direct deesterification of fluorinated products could afford chiral α-single fluorine-substituted cyclopentenones.

Abstract:The construction of fluorinated architectures has been a topic of interest to medicinal chemists due to their unique ability to improve the pharmacokinetic properties of bioactive compounds. However, the stereoselective synthesis of fluoro-organic compounds with vicinal stereogenic centers is a challenge. Herein, we present a directing-groupfree nickel-hydride catalyzed hydroalkylation of fluoroalkenes to afford fluorinated motifs with two adjacent chiral centers in excellent yields and stereoselectivities. Our method provides expedient access to biologically relevant, highly enantioenriched organofluorine compounds. Furthermore, the strategy can be used for the diastereo- and enantioselective synthesis of vicinal difluorides, which have recently gained attention in the fields of organocatalysis and peptide mimics.

Abstract:We have developed an efficient method for the preparation of chiral fluorinated compounds by sequential organocatalyzed desymmetrization of 3-substituted glutaric anhydrides and photoredox-catalyzed decarboxylic fluorination. Chiral fluorides can be prepared in yields of up to 58% and with excellent enantioselectivities of up to 98% ee.

Abstract:An approach to construct tetrasubstituted carbon stereocenters bearing both a trifluoromethylthio (SCF3) group and a cyano group has been realized, through asymmetric organophosphine‐catalyzed Mannich‐type reactions. The products were obtained in high yields and moderate to high enantioselectivities. Two diastereoisomers could be isolated from each reaction (overall 25 examples), rendering this approach a viable opportunity for molecular diversity generation and improvement of the bioactivity of related drug candidates.

Abstract:The new reactions between o-hydroxyphenyl enaminones and Langlois reagent (CF3SO2Na) for the tunable synthesis of 3-(trifluoromethylthio) chromones and 3-trifluoromethylsulfinyl chromones are reported herein. Both type of reactions proceed under transition metal-free conditions. In addition, the conditions for the synthesis of 3-trifluoromethylsulfinyl chromones have also been found to be applicable for the synthesis of 3-alkyl/arylsulfinyl chromones.

Abstract:An enantioselective intramolecular allylic C-H oxidation to generate optically active chromans has been accomplished under the cooperative catalysis of a palladium complex of chiral phosphoramidite ligand and 2-fluorobenzoic acid. Mechanistic studies suggest that this reaction commences with a Pd-catalyzed allylic C-H activation event and then undergoes asymmetric allylic alkoxylation. The synthetic significance of the method has been embodied by concisely building up a key chiral intermediate to access (+)-diversonol.

Abstract:A novel chiral spirocyclic amide (SPA)-derived triazolium organocatalyst has been designed and demonstrated to effect asymmetric homo- and heterodialkylations of various bisoxindoles, enabling enantioselective construction of vicinal all-carbon quaternary stereocenters. These reactions feature excellent enantioand diastereoselectivities (up to 99% ee and >20:1 dr) as well as good to high yields (up to 89% over two steps). As an application of this methodology, the first asymmetric total synthesis of (-)-chimonanthidine has been achieved.

Abstract:An enantioselective [5 + 1] annulation of α-hydroxyl aryl ketones with cyclohexadienone-tethered enones via chiral dinuclear zinc catalysts is reported. A Brønsted base and Lewis acid cooperative activation model is the critical factor for the chirality transfer process, giving access to a wide array of enantioenriched bicyclic pyrans scaffolds bearing two oxa-quaternary carbon centers with good yields and high optical purities. Notably, this reaction offers a new reaction mode of α-hydroxyl ketones as C,C-bisnucleophiles in catalytic asymmetric transformation.

Abstract:Introduction of the trifluoromethyl group (CF3 ) into organic molecules in an enantioselective manner has attracted significant attention, but still remains a challenging problem. We herein report a catalytic asymmetric trifluoromethylation of cyclic ketones via a ScIII /chiral bisoxazoline-catalyzed homologation reaction by employing 2,2,2-trifluorodiazoethane (CF3 CHN2 ) as the CF3 source. This desymmetrization process is highly efficient and generates two chiral centers with excellent diastereoselectivity and enantioselectivity, affording chiral α-trifluoromethyl cyclic ketones in a straightforward manner.

Abstract:A novel intramolecular Smiles rearrangement of α-fluoro-β-keto-pyrimidylsulfones (usually used as a carbon nucleophile) was developed, providing a versatile avenue for synthesis of tri/tetra-substituted monofluorinated pyrimidyl enol ethers. Among these, diverse (Z)-monofluorovinylsulfones and sulfinates were efficiently assembled by adding extra electrophile and fine-tuning reaction conditions. The process is triggered by a keto-enol tautomerism from enol oxyanion to pyrimidine 2-carbon, completely different from the classical carbon nucleophilic addition reaction approach.

Abstract:A fast and efficient Selectfluor-triggered fluorination/cyclization reaction ofo-hydroxyarylenaminones has been successfully developed. The reaction successfully provides an expedient method for the synthesis of 3-fluoro-chromones promoted by potassium carbonate, which shows readily available starting materials and is easy to operate. In addition, a plausible mechanism of this tandem cyclization reaction was proposed where 4H-chromen-4-one, 2-(dimethylamino)-3,3-difluorochroman-4-one, and 3,3-difluoro-2-hydroxychroman-4-one were not found to be the reactive intermediates. Moreover, these novel compounds have been obtained in moderate to good yields, and their structures have been confirmed by 1H NMR, 13C NMR, and high-resolution mass spectrometry.

Abstract:Chiral phosphoric acid-catalyzed enantioselective aza-Friedel-Crafts reaction of trifluoromethyl benzoxazinones with pyrroles is reported. Under mild conditions, a range of enantioenriched dihydrobenzoxazinones bearing trifluoromethylated quaternary stereocenters could be obtained in good to excellent yield and ee. A remarkable fluorine effect is observed, and preliminary mechanistic studies combined with theory calculations suggest that triple-hydrogen-bonding interactions hold the transition structure rigidly and allow the bulky substituents of the catalyst to influence the enantioselectivity.
