Abstract:Glioma is the most common malignant primary brain tumor, and the accurate diagnosis of glioma has always been a challenge. Moreover, cerebellar glioma, which is difficult to be detected by magnetic resonance imaging, is not usually diagnosed until after the appearance of clinical symptoms. In this study, TRZD, a near-infrared (NIR) persistent luminescence (PL) nanoparticle with a dual function of imaging and therapy, was synthesized based on ZnGa2O4:Cr3+,Sn4+. TRZD showed excellent rechargeable NIR PL for more than 30 hours in vivo with good tissue penetration for long-term autofluorescence-free imaging. The tumor growth of both the subcutaneous and orthotropic glioma models was significantly inhibited by TRZD. This is the first-time approach using NIR PL nanoprobes for both diagnosis and therapy of glioma. This is also the first-time report of nanotechnology-based diagnosis and therapy of cerebellar gliomas. This study offers a highly promising multifunctional nanoparticle for theranostics of a wide range of brain diseases.

Abstract:Glioblastoma Targeted Therapy In article number 2209579, Wang and co-workers report a near-infrared persistent luminescence nanoparticle for the diagnosis and targeted therapy of glioblastoma (GBM). GBM cell membrane (GCM) coating and chlorotoxin enhanced the blood-brain barrier (BBB) penetration and the targeting and tumor-homing of GBM. This study offers an example of the highly promising drug carriers for precise diagnosis and therapy of central nervous system diseases.

Abstract:Brain diseases with high mortality cause severe threats to human health and life. Non-invasively high-resolution imaging technologies contribute to obtaining detailed brain structures to figure out the pathogenesis. In addition, timely visualization benefits the early warning and diagnosis of brain diseases. Rare-earth-based nanoparticles(RENPs) with excellent optical properties are highlighted for the diagnosis and treatment of brain diseases in recent years. Their tunable down-conversion fluorescence enables the high-resolution NIR-II fluorescence imaging of the brain. Besides, the excellent up-conversion luminescence can be applied to designing fluorescent probes to visualize brain diseases. Importantly, the surface of RENPs can be tailored to cross the blood-brain barrier, target the disease site in the brain and deliver drugs specifically, providing an effective tool for treating brain diseases. Excitingly, RENPs can be combined with emerging therapies, such as phototherapy, gas therapy and sonodynamic therapy, significantly improving the therapeutic effects of glioblastoma. This review summarizes the rational design, functionalization strategies and application of RENPs in brain imaging, diagnosis and therapy. Furthermore, a brief perspective on existing challenges and further development of RENPs is presented.

Abstract:Cell-intrinsic reporters such as luciferase (LUC) and red fluorescent protein (RFP) have been commonly utilized in preclinical studies to image tumor growth and to monitor therapeutic responses. While extrinsic reporters that emit near infrared I (NIR-I: 650-950 nm) or near-infrared II (NIR-II: 1000 -1700 nm) optical signals have enabled minimization of tissue autofluorescence and light scattering, it has remained unclear as to whether their use has afforded more accurate tumor imaging in small animals. Here, we developed a novel optical imaging construct comprised of rare earth lanthanide nanoparticles coated with biodegradable diblock copolymers and doped with organic fluorophores, generating NIR-I and NIR-II emissive bands upon optical excitation. Simultaneous injection of multiple spectrally-unique nanoparticles into mice bearing tumor implants established via intraperitoneal dissemination of LUC+/ RFP+ OVCAR-8 ovarian cancer cells enabled direct comparisons of imaging with extrinsic vs. intrinsic reporters, NIR-II vs. NIR-I signals, as well as targeted vs. untargeted exogenous contrast agents in the same animal and over time. We discovered that in vivo optical imaging at NIR-II wavelengths facilitates more accurate detection of smaller and earlier tumor deposits, offering enhanced sensitivity, improved spatial contrast, and increased depths of tissue penetration as compared to imaging with visible or NIR-I fluorescent agents. Our work further highlights the hitherto underappreciated enhancements in tumor accumulation that may be achieved with intraperitoneal as opposed to intravenous administration of nanoparticles. Lastly, we found discrepancies in the fidelity of tumor uptake that could be obtained by utilizing small molecules for in vivo as opposed to in vitro targeting of nanoparticles to disseminated tumors. Abstract:Rare earth-doped nanoparticles have been widely used in disease diagnosis, drug delivery, tumor therapy, and bioimaging. Among various bioimaging methods, the fluorescence imaging technology based on the rare earth-doped nanoparticles can visually display the cell activity and lesion evolution in living animals, which is a powerful tool in biological technology and has being widely applied in medical and biological fields. Especially in the band of near infrared (700–1700 nm), the emissions show the characteristics of deep penetration due to low absorption, low photon scattering, and low autofluorescence interference. Furthermore, the rare earth-doped nanoparticles can be endowed with the water solubility, biocompatibility, drug-loading ability, and the targeting ability for different tumors by surface functionalization. This confirms its potential in the cancer diagnosis and treatment. In this review, we summarized the recent progress in the application of rare earth-doped nanoparticles in the field of bioimaging and tumor treatment. The luminescent mechanism, properties, and structure design were also discussed.

Abstract:Brain diseases pose a great threat to human health worldwide, thus it is of great importance to explore new materials for their diagnosis and therapy. Compared with the ultraviolet (UV) and visible light, near infrared (NIR) light has better biosafety, lower tissue auto-fluorescence, and stronger penetration into skull. Hence, NIR light responsive materials have attracted great attention recently which can not only realize the high spatial resolution and signal-to-noise ratio imaging for diagnosis, but also achieve effective NIR light triggered treatment noninvasively. Besides, the brain lesions can be visualized clearly by NIR imaging, which can provide more accurate information for guiding treatments. Therefore, this work systematically summarizes the recent advances of NIR light responsive materials for diagnosis and treatment of brain diseases. Firstly, this work briefly reviews the pathological features of blood brain barrier (BBB) and the rational design of materials to target brain disease. Then, the emerging approaches for diagnosis and treatment of various brain diseases with NIR light responsive materials are introduced. Subsequently, the imaging-guided therapies are elaborated, such as the fluorescence imaging guided surgery and sonodynamic therapy. Finally, some challenges and prospects on the design of NIR light responsive materials for precise diagnosis and therapy of brain diseases are put forward.

Abstract:Nowadays, cancer poses a significant hazard to humans. Limitations in early diagnosis techniques not only result in a waste of healthcare resources but can even lead to delays in diagnosis and treatment, consequently reducing cure rates. Therefore, it is crucial to develop an imaging probe that can provide diagnostic information precisely and rapidly. Here, we used a simple hydrothermal method to design a multimodal imaging probe based on the excellent properties of rare-earth ions. Calcium fluoride co-doped with ytterbium, gadolinium, and neodymium (CaF2:Y,Gd,Nd) nanoparticles (NPs) is highly crystalline, homogeneous in morphology, and displays a high biosafety profile. In addition, in vitro and ex vivo experiments explored the multimodal imaging capability of CaF2:Y,Gd,Nd and demonstrated the efficient performance of CaF2:Y,Gd,Nd during NIR-II fluorescence/photoacoustic/magnetic resonance imaging. Collectively, our novel diagnosis nanoparticle will generate new ideas for the development of multifunctional nanoplatforms for disease diagnosis and treatment.

Abstract:Biomedicine is continuously demanding new luminescent materials to be used as optical probes for the acquisition of high resolution, high contrast and high penetration in vivo images. These materials, in combination with advanced techniques, could constitute the first step towards new diagnosis and therapy tools. In this work, we report on the synthesis of long lifetime rare-earth-doped fluoride nanoparticles by adopting different strategies: core/shell and dopant engineering. The here developed nanoparticles show intense infrared emission in the second biological window with a long luminescence lifetime close to 1 millisecond. These two properties make the here presented nanoparticles excellent candidates for time-gated infrared optical bioimaging. Indeed, their potential application as optical imaging contrast agents for autofluorescence-free in vivo small animal imaging has been demonstrated, allowing high contrast real-time tracking of gastrointestinal absorption of nanoparticles and transcranial imaging of intracerebrally injected nanoparticles in the murine brain.

Abstract:A comprehensive approach for the construction of NIR-I/NIR-II nanofluorophores with exceptional brightness and excellent chemo- and photostability has been developed. This study first confirmed that the amphiphilic molecules with stronger hydrophobic moieties and weaker hydrophilic moieties are superior candidates for constructing brighter nanofluorophores, which are attributed to its higher efficiency in suppressing the intramolecular charge transfer/aggregation-caused fluorescence quenching of donor-acceptor-donor type fluorophores. The prepared nanofluorophore demonstrates a fluorescence quantum yield exceeding 4.5% in aqueous solution and exhibits a strong NIR-II tail emission up to 1300 nm. The superior performance of the nanofluorophore enabled the achievement of high-resolution whole-body vessel imaging and brain vessel imaging, as well as high-contrast fluorescence imaging of the lymphatic system in vivo. Furthermore, their potential for highly sensitive fluorescence detection of tiny tumors in vivo has been successfully confirmed, thus supporting their future applications in precise fluorescence imaging-guided surgery in the early stages of cancer.

Abstract:Abstract Rare earth upconversion nanoparticles (UCNPs) with the superior performance in the biomedical field have attracted great attention. Due to the good light stability, low toxicity, deep tissue penetration and excellent bio-compatibility, UCNPs display great potential for development in the biomedical field. Excellent related reports are increasing year by year, like molecular sensing, bioimaging and therapeutics. In this paper, the preparation, modification, application of upconversion fluorescent nanoparticles and the latest research results in the field of biomedical materials have been reported in detail. Graphical abstract : In this paper, we systematically summarize and describe the preparation, modification, application and latest discovery in biomedical field of rare earth upconversion nanoparticles (UCNP). Due to the good light stability, low toxicity, deep tissue penetration and excellent biocompatibility, UCNPs display superior performance for the development of biomedical field in cancer therapy. Our work will certainly inspire researchers in related fields to create more meaningful upconversion nanomaterials and do more contributions to human cancer treatment research.

Abstract:Nanotheranostics, which can provide great insight into cancer therapy, has been deemed as a promising technology to settle the unmet medical needs. The rational design of high performance nanotheranostics with multiple complementary imaging features and satisfactory therapeutic efficacy is particularly valuable. Herein, versatile nanotheranostic agents DPPB-Gd-I NPs were fabricated by using gadolinium-diethylenetriaminepentaacetic acid chelates and an iodine-decorated copolymer as encapsulation matrixes to encapsulate a polymer DPPB through one-step nanoprecipitation. We have demonstrated that such nanoagents are able to efficiently damage tumors under single dose injection and NIR laser illumination conditions due to the enhanced photodynamic therapy and enhanced photothermal therapy (the tumor inhibition rate was as high as 94.5%). Moreover, these nanoagents can be utilized as dual-modal NIR-II fluorescence/magnetic resonance imaging probes for tumor diagnosis with high sensitivity, deep tissue penetration, and excellent spatial resolution. Overall, this work offers a powerful tactic to fabricate high performance nanotheranostics for clinical application.

Abstract:Multifunctional nanoprobes, such as nanocrystals, nanoshells, and luminescent nanomaterials, have been developed for imaging biological processes; such as cell signaling, neuroimaging, protein conformation probing, DNA conformation probing, gene transcription, virus infection and replication in cells, protein dynamics, tumor diagnosis, and therapy evaluation. With the application of nanotechnology for CNS-active agents' delivery, nanostructured materials are emerging as a powerful means for diagnosis of CNS disorders, including brain tumors, because of their unique optical size, and surface properties. This review summarizes the recent patents on imaging nanoprobes for brain tumor diagnosis and therapy. The future development in this active cross-disciplinary field will be discussed as well.

Abstract:Most hypoxic tumors are insensitive to radiation, which is a major obstacle in the development of conventional radiotherapy for tumor treatment. Some drugs, such as cisplatin (CDDP), have been extensively used both as an anticancer drug and clinically as a radiosensitizer to enhance radiotherapy. Herein, we develop rattle-structured multifunctional up-conversion core/porous silica shell nanotheranostics (UCSNs) for delivering CDDP to tumors for synergetic chemo-/radiotherapy by CDDP radiosensitization and magnetic/luminescent dual-mode imaging. UCSNs had a dynamic light scattering diameter of 79.1 nm and excellent water dispersity and stability. In vitro studies showed that CDDP loaded in UCSNs (UCSNs-CDDP) was more effective than free CDDP as a radiosensitizer. After injection, UCSNs-CDDP also demonstrated unambiguously enhanced radiotherapy efficacy in vivo. Our report aims at presenting a novel strategy in biomedical nanotechnology that allows simultaneous dual-mode imaging and localized therapy via synergetic chemo-/radiotherapy, which may achieve optimized therapeutic efficacy in cancer treatment.

Abstract:Brain research is a cutting-edge topic but it remains challenging to achieve noninvasive sensitive diagnosis of brain diseases. Lanthanide nanocrystals (NCs) with near-infrared II (NIR-II) photoluminescence (PL) have shown great promise for noninvasive through-skull high-resolution imaging. Currently, it still lacks a systematic review discussing the emerging spectroscopic and nanoprobe engineering of NIR-II luminescent lanthanide NCs for specific imaging of brain diseases. Herein, we comprehensively review recent development of NIR-II lanthanide nanoprobes for brain disease imaging in vivo. We first summarize luminescence regulation strategies aiming to enhance the NIR-II PL efficiency of lanthanide NCs. Then, the design strategies of lanthanide nanoprobes are introduced with the effort to improve their brain delivery efficacy and NIR-II PL imaging specificity and sensitivity in the brain. In particular, we comprehensively detail the latest tailoring of lanthanide nanoprobes for NIR-II PL imaging of brain diseases such as brain injuries and tumors. Finally, we propose future opportunities and challenges in the exploration of NIR-II lanthanide nanoprobes and practical imaging studies of brain diseases. This review is anticipated to motivate future engineering of lanthanide nanoprobes for the diagnosis and treatment of brain diseases.

Abstract:Fluorescence imaging in the second near-infrared region(900–1700 nm, NIR-II) with a high resolution and penetration depth due to the significantly reduced tissue scattering and autofluorescence has emerged as a useful tool in biomedical fields. Recently, many efforts have been devoted to the development of fluorophores with an emission band covering the long-wavelength end of NIR-II region(1500–1700 nm) to eliminate the autofluorescence. Alternatively, we believe imaging with a narrow bandwidth could also reduce the autofluorescence. As a proof of concept, NaYF4:Yb, Nd@NaYF4 downconversion nanoparticles(DCNPs) with sharp NIR-II emission were synthesized. The luminescence of DCNPs showed a half-peak width of 49 nm centered at 998 nm, which was perfectly matched with a (1000±25) nm bandpass filter. With this filter, we were able to retain most of the emissions from the nanoparticles, while the autofluorescence was largely reduced. After PEGylation, the DCNPs exhibited great performance for blood vessel and tumor imaging in living mice with significantly reduced autofluorescence and interference signals. Abstract:Abstract Lanthanides have unique electronic configurations and stepped energy states of [Xe]4f N , which endows them with advantages including stable luminescence, large stokes shift, wide excitation spectral bands, and long fluorescence lifetime. NIR‐II fluorescence imaging overcomes the strong tissue absorption, scattering, and spontaneous fluorescence interference of traditional fluorescence and can achieve deeper tissue penetration and higher spatial and temporal resolution in vivo imaging. Lanthanides hold great potential for in vivo biosensing and bioimaging. In this work, the design and preparation of nanomaterials doped with different lanthanide elements are reviewed in this paper. Applications of lanthanide‐doped nanomaterials to different tumor therapeutics through NIR‐II fluorescence imaging and the unique potential of NIR lanthanide nanomaterials in tumor diagnosis and therapy are also discussed.

Abstract:Fluorescent proteins are investigated extensively as markers for the imaging of cells and tissues that are treated by gene transfection. However, limited transfection efficiency and lack of targeting restrict the clinical application of this method rooted in the challenging development of robust fluorescent proteins for in vivo bioimaging. To address this, a new type of near-infrared (NIR) fluorescent protein assemblies manufactured by genetic engineering is presented. Due to the formation of well-defined nanoparticles and spectral operation within the phototherapeutic window, the NIR protein aggregates allow stable and specific tumor imaging via simple exogenous injection. Importantly, in vivo tumor metastases are tracked and this overcomes the limitations of in vivo imaging that can only be implemented relying on the gene transfection of fluorescent proteins. Concomitantly, the efficient loading of hydrophobic drugs into the protein nanoparticles is demonstrated facilitating the therapy of tumors in a mouse model. It is believed that these theranostic NIR fluorescent protein assemblies, hence, show great potential for the in vivo detection and therapy of cancer.
