Abstract:Biofilms are central to some of the most urgent global challenges across diverse fields of application, from medicine to industry to the environment and exert considerable economic and social impact.To combat biofilm growth on surfaces, chemical-based approaches using immobilization of antimicrobial agents, such as antibiotics or silver particles, can trigger antimicrobial resistance.Alternative, physical-based approaches (e.g.nanostructures) can be used, but their effects may not last due to initial adherent bacteria spreading laterally and masking surfaces within a couple of days.We have demonstrated that a multiscale surface structure can further delay biofilm formation but it is still not effective over longer periods of time.The innovation of lubricant impregnated surfaces (LIS) as surfaces slippery to liquids has been attracting significant interest in anti-icing, most recently, as a new anti-biofilm strategy.However, the potential loss of lubricant through repeated usage or shear remains a key limiting factor to wider adoption as a practical solution.In the present work, we report an anti-biofilm surface strategy using liquid-like nanocoating where the risk of lubricant loss is removed.

Abstract:Macromolecular antimicrobial agents such as cationic polymers and peptides have recently been under an increased level of scrutiny because they can combat multi-drug-resistant microbes. Most of these polymers are non-biodegradable and are designed to mimic the facially amphiphilic structure of peptides so that they may form a secondary structure on interaction with negatively charged microbial membranes. The resulting secondary structure can insert into and disintegrate the cell membrane after recruiting additional polymer molecules. Here, we report the first biodegradable and in vivo applicable antimicrobial polymer nanoparticles synthesized by metal-free organocatalytic ring-opening polymerization of functional cyclic carbonate. We demonstrate that the nanoparticles disrupt microbial walls/membranes selectively and efficiently, thus inhibiting the growth of Gram-positive bacteria, methicillin-resistant Staphylococcus aureus (MRSA) and fungi, without inducing significant haemolysis over a wide range of concentrations. These biodegradable nanoparticles, which can be synthesized in large quantities and at low cost, are promising as antimicrobial drugs, and can be used to treat various infectious diseases such as MRSA-associated infections, which are often linked with high mortality.
Self-Standing Bioinspired Polymer Films Doped with Ultrafine Silver Nanoparticles as Innovative Antimicrobial Material

Abstract:Thin self-standing films with potential antimicrobial synergistic activity have been produced by a simple green chemical synthesis with overnight thermal treatment. Their properties have been studied by scanning electron microscopy, X-ray photoelectron spectroscopy and other techniques to understand their potential range of applications. In this work, the focus was set on the development of a potential novel and effective alternative to conventional antimicrobial materials. By creating an antimicrobial polymer blend, and using it to develop and immobilize fine (~25 nm) silver nanophases, we further aimed to exploit its film-forming properties and create a solid composite material. The resulting polymer matrix showed improved water uptake percentage and better stability in the presence of water. Moreover, the antimicrobial activity of the films, which is due to both organic and inorganic components, has been evaluated by Kirby-Bauer assay against common foodborne pathogens (Staphylococcus aureus and Salmonella enterica) and resulted in a clear inhibition zone of 1.2 cm for the most complex nanocomposition. The excellent performance against bacteria of fresh and 6-month-old samples proves the prospects of this material for the development of smart and biodegradable food packaging applications.

Abstract:Antimicrobial active films based on poly (lactic acid) (PLA) were prepared with nano-silver (nano-Ag) and nano-zinc oxide (nano-ZnO) using a solvent volatilizing method. The films were characterized for mechanical, structural, thermal, physical and antimicrobial properties. Scanning electron microscopy (SEM) images characterized the fracture morphology of the films with different contents of nano-Ag and nano-ZnO. The addition of nanoparticles into the pure PLA film decreased the tensile strength and elasticity modulus and increased the elongation of breaks-in other words, the flexibility and extensibility of these composites improved. According to the results of differential scanning calorimetry (DSC), the glass transition temperature of the PLA nano-composite films decreased, and the crystallinity of these films increased; a similar result was apparent from X-ray diffraction (XRD) analysis. The water vapor permeability (WVP) and opacity of the PLA nano-composite films augmented compared with pure PLA film. Incorporation of nanoparticles to the PLA films significantly improved the antimicrobial activity to inhibit the growth of Escherichia coli. The results indicated that PLA films with nanoparticles could be considered a potential environmental-friendly packaging material.

Abstract:Microbial colonization on various surfaces is a serious problem. Biofilms from these microbes pose serious health and economic threats. In addition, the recent global pandemic has also attracted great interest in the latest techniques and technology for antimicrobial surface coatings. Incorporating antimicrobial nanocompounds into materials to prevent microbial adhesion or kill microorganisms has become an increasingly challenging strategy. Recently, many studies have been conducted on the preparation of nanomaterials with antimicrobial properties against diseases caused by pathogens. Despite tremendous efforts to produce antibacterial materials, there is little systematic research on antimicrobial coatings. In this article, we set out to provide a comprehensive overview of nanomaterials-based antimicrobial coatings that can be used to stop the spread of contamination to surfaces. Typically, surfaces can be simple deposits of nanomaterials, embedded nanomaterials, as well as nanotubes, nanowires, nanocolumns, nanofibers, nanoneedles, and bio-inspired structures.

Abstract:This study presents a scalable solgel method to fabricate Fe2O3/AgBr nanoimprints for antimicrobial thin films. The Fe2O3/AgBr heterojunction exhibits strong antibacterial and biofilm‐inhibiting properties under light and dark conditions while maintaining biocompatibility, enabling selective reactive oxygen species production, bacterial targeting, and cell recovery for self‐cleaning, antimicrobial applications in cell culture and beyond. The resuscitation of bacteria through biofilms presents a critical challenge in controlling microbial pathogenesis and addressing antimicrobial resistance. Continuous antibiofilm activity, particularly on frequently contacted surfaces, is therefore critical. In this study, a scalable is introduced, one‐step fabrication of Fe2O3/AgBr nanoimprints using a polymerizable sol–gel (PSG) approach to create functional nanostructured thin films with strong antimicrobial properties. Fe2O3, a visible‐light photocatalyst, is coupled with AgBr, a photosensitizer and dark‐active antimicrobial, forming a heterojunction that demonstrated potent antibacterial activity against Escherichia coli and Pseudomonas putida under both dark and light conditions. The heterojunctions exhibit significant biofilm inhibition in the dark, particularly against the robust biofilm‐forming P. putida, while visible light irradiation ensures complete biofilm clearance. These surfaces also achieve optimal reactive oxygen species (ROS) production, selectively targeting bacteria without compromising the integrity of mammalian cells. The biocompatibility is confirmed through MTT, TBARS, and apoptosis assays, demonstrating the non‐cytotoxic nature of the substrates. Moreover, the surfaces enable cell patterning and recovery of mammalian cells from microbial contamination, highlighting their potential in creating bacterial‐free environments for cell culture. This innovative method offers a promising route to next‐generation, self‐cleaning antimicrobial coatings, combining continuous biofilm inhibition with excellent biocompatibility and scalability.

Sustainable and effective antimicrobial surface based on cellulose thin films

Abstract:In the present work, we developed a sustainable and effective antimicrobial surface film based on Micro-Fibrillated Cellulose. The resulting porous cellulose thin film is barely noticeable to human eyes due to its sub-micron thickness, of which the coverage, porosity and microstructure can be modulated by the formulations developed. Using goniometers and a quartz crystal microbalance (QCM), we observed a threefold reduction in water contact angles and accelerated (more than 50% faster) water evaporation kinetics on the cellulose film. The thin film exhibits not only a rapid inactivation effect against SARS-CoV-2 in 5 minutes, following deposition of the virus loaded droplets, but also an exceptional ability to reduce contact transfer of liquid, e.g. respiratory droplets, onto surfaces such as artificial skin by more than 90%. It also exhibits excellent antimicrobial performance in inhibiting the growth of both gram-negative and gram-positive bacteria (<a class="link-external link-http" href="http://E.coli" rel="external noopener nofollow">this http URL</a> and <a class="link-external link-http" href="http://S.epidermidis" rel="external noopener nofollow">this http URL</a>) due to the excellent porosity and hydrophilicity. Additionally, the cellulose film shows nearly 100% resistance to skin scraping in dry condition thanks to its strong attachment to the substrate, whilst good removability once wetted, suggesting its practical suitability for daily use. Importantly, the coating can be formed on solid substrates readily by spraying and requires solely a simple formulation of a plant-based cellulose material with no additives, rendering it a scalable, affordable and green solution for antimicrobial surfaces. Implementing such cellulose films could thus play a significant role in controlling future pan- and epidemics, in particularly during the first phase when appropriate medication needs to be developed.

Abstract:Physical topography modification is an approach to fabricate nanostructures surfaces with antimicrobial properties. Lithography-based technologies offer an effective technique to develop the desired sizes and geometry. The replica molding technique was employed to fabricate the PDMS nanostructures using the PMMA imaging layer and characterized using a FESEM and AFM. The cell viability of gram-positive bacteria on structural diminished by almost 80% and the cells were deformed and ruptured once attached to the structured surface. Thus, the PDMS structured surface enhanced the bactericidal properties of the film, which effectively inhibit bacterial attachment.

Abstract:Globally, fungal infections have evolved as a strenuous challenge for clinicians, particularly in patients with compromised immunity in intensive care units. Fungal co-infection in Covid-19 patients has made the situation more formidable for healthcare practitioners. Surface adhered fungal population known as biofilm often develop at the diseased site to elicit antifungal tolerance and recalcitrant traits. Thus, an innovative strategy is required to impede/eradicate developed biofilm and avoid the formation of new colonies. The development of nanocomposite-based antibiofilm solutions is the most appropriate way to withstand and dismantle biofilm structures. Nanocomposites can be utilized as a drug delivery medium and for fabrication of anti-biofilm surfaces capable to resist fungal colonization. In this context, the present review comprehensively described different forms of nanocomposites and mode of their action against fungal biofilms. Amongst various nanocomposites, efficacy of metal/organic nanoparticles and nanofibers are particularly emphasized to highlight their role in the pursuit of antibiofilm strategies. Further, the inevitable concern of nanotoxicology has also been introduced and discussed with the exigent need of addressing it while developing nano-based therapies. Further, a list of FDA-approved nano-based antifungal formulations for therapeutic usage available to date has been described. Collectively, the review highlights the potential, scope, and future of nanocomposite-based antibiofilm therapeutics to address the fungal biofilm management issue.

Abstract:The advances in nanocomposites incorporating bioactive substances have the potential to transform the food packaging sector. Different nanofillers have been incorporated into polymeric matrixes to develop nanocomposite materials with improved mechanical, thermal, optical and barrier properties. Nanoclays, nanosilica, carbon nanotubes, nanocellulose, and chitosan/chitin nanoparticles have been successfully included into polymeric films, resulting in packaging materials with advanced characteristics. Nanostructured antimicrobial films have promising applications as active packaging in the food industry. Nanocomposite films containing antimicrobial substances such as essential oils, bacteriocins, antimicrobial enzymes, or metallic nanoparticles have been developed. These active nanocomposites are useful packaging materials to enhance food safety. Nanocomposites are promising materials for use in food packaging applications as practical and safe substitutes to the traditional packaging plastics.

Abstract:Present worldwide difficulties in healthcare and the environment have motivated the investigation and research of novel materials in an effort to find novel techniques to address the current challenges and requirements. In particular, the use of nanomaterials has demonstrated a significant promise in the fight against bacterial infections and the problem of antibiotic resistance. Metal nanoparticles and carbon-based nanomaterials in particular have been highlighted for their exceptional abilities to inhibit many types of bacteria and pathogens. In order for these materials to be as effective as possible, synthetic techniques are crucial. Therefore, in this review article, we highlight some recent developments in the design and synthesis of various nanomaterials, including metal nanoparticles (e.g., Ag, Zn, or Cu), metal hybrid nanomaterials, and the synthesis of multi-metallic hybrid nanostructured materials. Following that, examples of these materials’ applications in antimicrobial performance targeted at eradicating multi-drug resistant bacteria, material protection such as microbiologically influenced corrosion (MIC), or additives in construction materials have been described.

Abstract:The second part of our review describing new achievements in the field of biofilm prevention and control, begins with a discussion of the active antibiofilm nanocoatings. We present the antibiofilm strategies based on antimicrobial agents that kill pathogens, inhibit their growth, or disrupt the molecular mechanisms of biofilm-associated increase in resistance and tolerance. These agents of various chemical structures act through a plethora of mechanisms targeting vital bacterial metabolic pathways or cellular structures like cell walls and cell membranes or interfering with the processes that underlie different stages of the biofilm life cycle. We illustrate the latter action mechanisms through inhibitors of the quorum sensing signaling pathway, inhibitors of cyclic-di-GMP signaling system, inhibitors of (p)ppGpp regulated stringent response, and disruptors of the biofilm extracellular polymeric substances matrix (EPS). Both main types of active antibiofilm surfaces, namely non-leaching or contact killing systems, which rely on the covalent immobilization of the antimicrobial agent on the surface of the coatings and drug-releasing systems in which the antimicrobial agent is physically entrapped in the bulk of the coatings, are presented, highlighting the advantages of each coating type in terms of antibacterial efficacy, biocompatibility, selective toxicity, as well as drawbacks and limitations. Developments regarding combined strategies that join in a unique platform, both passive and active elements are not omitted. In such platforms with dual functionality, passive and active strategies can be applied either simultaneously or sequentially. We especially emphasize those systems that can be reversely and repeatedly switched between the non-fouling status and the bacterial killing status, thereby allowing several bacteria-killing/surface regeneration cycles to be performed without significant loss of the initial bactericidal activity. Eventually, smart antibiofilm coatings that release their antimicrobial payload on demand, being activated by various triggers such as changes in local pH, temperature, or enzymatic triggers, are presented. Special emphasis is given to the most recent trend in the field of anti-infective surfaces, specifically smart self-defensive surfaces for which activation and switch to the bactericidal status are triggered by the pathogens themselves.
Synergistic antimicrobial nanofiber membranes based on metal incorporated silica nanoparticles as advanced antimicrobial layers

Abstract:In this post-new-normal era, the public prioritizes preventive measures over curing, which is a constructive approach to staying healthy. In this study, an innovative antimicrobial membrane material has been developed, showcasing the promising potential for various applications. The metal-doped silica nanoparticles (Ag, Cu, and Co) were incorporated into a cellulose acetate (CA) polymer-based nanofiber membrane using the electrospinning technique. The metal nanoparticles were doped into a silanol network of silica nanoparticles. The fabricated membranes underwent detailed characterization using a wide range of techniques including PXRD, FTIR, Raman, SEM, TEM, TGA, and tensile testing. These analyses provided compelling evidence confirming the successful incorporation of metal-doped silica nanoparticles (Ag, Cu, and Co) into cellulose-based nanofibers. The band gap energies of the fabricated CA mats lie below 3.00 eV, confirming that they are visible light active. The trimetallic silica nanohybrid exhibited the lowest band gap energy of 2.84 eV, proving the self-sterilizing ability of the CA mats. The DPPH assay further confirmed the best radical scavenging activity by the trimetallic silica nanohybrid incorporated nanofiber mat (91.77 ± 0.88%). The antimicrobial activity was assessed by using the bacterial ATCC strains of Staphylococcus aureus , Streptococcus pneumoniae , MRSA (Methicillin-resistant Staphylococcus aureus ), Escherichia coli , Klebsiella pneumoniae , and Pseudomonas aeruginosa and fungal strains; quality control samples of Trichophyton rubrum , Microsporum gypsium , and Aspergillus niger , as well as the ATCC strain of Candida albicans . The trimetallic silica nanohybrid-incorporated CA membranes demonstrated the most significant inhibition zones. The reported findings substantiate the self-sterilizing mat's viability, affordability, efficacy against a broad spectrum of microbial strains, cost-effectiveness, and biodegradability. Furthermore, the mat serves as a dual-purpose physical and biological barrier against microbes, affirming its potential impact.

Abstract:Nanotechnology makes it possible to intentionally modify the properties of surfaces and endow them with any desired function.Nanostructured surfaces, coatings, and films have represented important advances in control of wetting, adhesion, mechanical, thermal, magnetic, electrical, and optical properties and ability to direct cell behavior.A number of promising approaches to fabricate functional micro-and nanostructured surfaces, coatings, and films have been stimulated by opportunities to enhance the properties of surfaces and interfaces via the combination of surface structure, morphology, and physical and chemical properties.Such functional micro-and nanostructured surfaces, coatings, and films are playing an increasingly important part in a broad range of novel applications, such as energy, electronics, photonics, as well as sensor systems, advanced materials, and medical devices.This special issue features articles that cover a wide range of recent progress in design, fabrication, and characterization of innovative nanostructured surfaces with the aim of improving their surface properties and functional performance, as well as new insights on physical principles underlying their properties and enormous potential applications.Highlighted below are important contributions from this special issue.C. Abstract:In this work, cellulose-based nanocomposite films having trimetallic (Bi, Fe, and Sn) nanoparticles were prepared by green adaptive methodology using Terminalia arjuna leaf extract as a reducing and stabilizing agent. Then, they were characterized by FTIR and SEM. The color change of microcrystalline cellulose films revealed the formation of the trimetallic (Bi, Fe, Sn) nanoparticles. Characteristics absorption peaks for reducing functional groups indicated the presence and role of the plant material used; moreover, the presence of various bands in FTIR spectra below 1000 cm−1 was indicative of the formation of (Bi, Fe, and Sn) nanocomposites. These synthesized nanomaterials were also tested for their antimicrobial potency against Escherichia coli and Pseudomonas aeruginosa. Positive outcomes designated their potential to be adopted for biomedical applications and in food packaging as an alternative of synthetic plastics to control pollution.

Abstract:CONSPECTUS: Self-disinfecting surfaces are a current pressing need for public health and safety in view of the current COVID-19 pandemic, where the keenly felt worldwide repercussions have highlighted the importance of infection control, frequent disinfection, and proper hygiene. Because of its potential impact upon real world translation into downstream applications, there has been much research interest in multiple disciplines such as materials science, chemistry, biology, and engineering. Various antimicrobial technologies have been developed and currently applied on surfaces in public spaces, such as elevator buttons and escalator handrails. These technologies are mainly based on conventional methods of grafting quaternary ammonium salts (QACs) such as benzalkonium chloride or the immobilization of metal species of silver or copper. However, neither the long-term efficacy nor the fast-killing properties have been proven, and the future repercussions from extended use, such as environmental hazards and the induction of MDR development, is unknown. Nanostructured surfaces with excellent antimicrobial activities have been claimed to be the next generation of self disinfecting surfaces with various promising applications and passive antimicrobial mechanisms, without the potential repercussions of active ingredient overuse. In this Account, we briefly introduce the concept of mechanobactericidal action realized by these nanostructured surfaces first discovered on cicada wings. The elimination of microbes on the surface was actualized by the physical rupture of the microbe cell wall by nanoprotusions, without any involvement of chemical species. By mimicking the physical features of naturally occurring biocidal surfaces, the fabrication of nanostructures on various substrates such as titania, silicon, and polymers has been well described. Observations of the dependence of their antimicrobial efficacy on physical characteristics such as height, density, and rigidity have also been documented. However, the complex fabrication of such nanostructures remains the main drawback preventing its widespread application. We outline our efforts in fabricating a series of zinc-based nanostructured materials with facile and generally applicable wet chemistry methods, including nanodaggered zeolitic imidazolate frameworks (ZIF-L) and ZnO nanoneedles. In our investigations, we discovered that there were additional modes of action that contributed to the excellent biocidal activities of our materials. The impact of surface chemistry and charge was partially responsible for the selectivity and efficacy of ZIF-L-coated surfaces, where the positively charged surfaces were able to attract and adhere negatively charged bacteria to the surface. The combination of semiconductor ZnO nanoneedles on electron-donating substrates allowed for the generation of reactive oxygen species (ROS), realizing the remote killing of bacteria unadhered to the nanostructured surface. Additionally, we demonstrate several real-life applications of the synthesized materials, underscoring the importance of materials development suited for scale-up and eventual translation to potential applications and commercial end products.
