Abstract:Abstract The golden camellia, a highly valued ornamental plant, belongs to the Chrysantha section, the sole group in the Camellia genus that produces yellow flowers. The cultivation and broader application of this unique species have been significantly hindered by the scarcity of golden camellia and the limited knowledge of the molecular mechanisms governing yellow flower development. To investigate the dynamics of metabolite accumulation and gene expression during the yellow flower formation of C. perpetua, a continuously flowering golden camellia, a comprehensive metabolomic and transcriptomic analysis was performed across five distinct developmental stages. This analysis identified 1,160 metabolites, with flavonoids constituting the largest proportion at 21.6%. Comparative transcriptomic profiling indicated that differentially expressed genes (DEGs) associated with photosynthesis were predominantly active during the early stages of flower development, while DEGs involved in the flavonoid biosynthesis pathway showed a marked increase as the flowers matured. The integrated metabolomic and transcriptomic data highlighted the pivotal regulatory role of the flavonoid biosynthesis pathway in the formation of yellow flowers in C. perpetua, identifying 50 key genes and 17 crucial metabolites and mapping their interaction networks. Furthermore, weighted gene co-expression network analysis pinpointed several hub genes, such as flavonol synthase, which are likely critical to flavonoid production due to their elevated expression levels, particularly in the early bud and yellowing phases. This study provides valuable insights into the molecular pathways that underlie yellow flower development in golden camellia species.
Elucidation of the Key Pathway for Flavonol Biosynthesis in Golden Camellia and Its Application in Genetic Modification of Tomato Fruit Metabolism

Abstract:Abstract Golden Camellia refers to a group of species in the genus Camellia that display yellow petals. The secondary metabolites in these petals hold ornamental significance and potential health benefits. However, the biosynthetic mechanisms governing the synthesis of these metabolites in golden petals remain elusive, and the exploitation of their bioactive components is not fully realized. This research involved the collection and analysis of 23 species of golden Camellia, leading to the discovery that flavonols, particularly quercetin 3-O-glucoside and quercetin 7-O-glucoside, are the primary contributors to the golden flower pigmentation. Integrative transcriptomics and co-expression network analyses pinpointed CnFLS1 as a crucial gene in the biosynthetic pathway, which, in conjunction with CnCHS, CnF3'H, and CnUFGT, orchestrates the specific pathway for flower color development. The enzyme assays revealed a high affinity and catalytic efficiency of CnFLS1 for DHQ, and transient expression of CnFLS1 in tobacco was shown to enhance the biosynthesis of quercetin flavonols, highlighting the pathway specificity in golden Camellia. Moreover, strategic transformations of cultivated tomatoes with various biosynthetic genes yielded transgenic lines exhibiting yellow fruit and quercetin-enriched flesh. These modified lines not only contained distinct flavonol components characteristic of golden Camellia but also demonstrated markedly improved antioxidant capabilities and enhanced resistance. The outcomes of this study not only elucidate the metabolic processes underlying the pigmentation of golden Camellia flowers but also provide a foundation for the development of novel tomato breeds through synthetic biology.
Population Genetic Structure of Camellia Nitidissima (theaceae) and Conservation Implications

Abstract:Camellia nitidissima Chi (Theaceae), with its golden-yellow flowers, is a famous ornamental species. Due to deforestation and collection of seedlings, its natural populations have receded greatly in recent decades. Genetic diversity and genetic differentiation of the twelve extant natural populations and one ex situ conserved population of C. nitidissima in China were analyzed using inter-simple sequence repeats (ISSR) markers. We found a low level of genetic diversity at both the species (P = 63.22%, Nei's genetic diversity H-T = 0.1561 and Shannon diversity H-sp = 0.2490) and population levels (P = 18.77%, H-E = 0.0831 and H-pop = 0.1188) and a relatively high degree of differentiation among populations (AMOVA analysis: 41.85%; Hickory theta(B) : 0.4056) in naturally occurring populations. In contrast, the ex situ population contained higher genetic variability compared to each natural population. No significant correlation was found between genetic diversity and population size. Based on the results, we suggest that all the wild C. nitidissima populations should be protected in situ. For the ex situ conservation of the species in Guilin Botanical Garden, samples from Long'an County should be added to the existing collections.

Abstract:Whether ex situ populations constructed in the limited nursery resources of botanical gardens can preserve enough genetic diversity of endangered plants in the wild remains uncertain. Here, a case study was conducted with Camellia tunghinensis, which is one of the species with the lowest natural distribution area in the sect. Chrysantha (golden camellia) of the family Theaceae. We investigated the genetic diversity and population structure of 229 samples from wild and ex situ populations using genotyping by sequencing (GBS). Core germplasm was constructed from these samples. The results showed that wild C. tunghinensis exhibited high genetic diversity, with observed heterozygosity of 0.257–0.293 and expected heterozygosity of 0.247–0.262. Compared with wild populations, the genetic diversity of ex situ populations established by transplanting wild seedlings was close to or even higher. However, the genetic diversity of those established by seed or cuttings of a few superior trees was lower. The Admixture analysis revealed that the structure of the ex situ populations derived from seeds and cuttings was relatively simple compared with the ex situ populations derived from transplanted wild seedlings and wild populations. These results suggested that direct transplanting of wild seedlings was more conducive to preserving the genetic diversity of endangered plants in the wild. In addition, wild populations demonstrated a small differentiation (mean FST = 0.044) among themselves, possibly due to long‐term and frequent gene flow between the wild populations. In contrast, moderate differentiation (mean FST > 0.05) was detected among ex situ populations and between ex situ and wild populations. This may be the combined result of the absence of gene flow pathways and strong selection pressure in various ex situ environments. Finally, 77 core germplasms were extracted from 229, likely representing the genetic diversity of C. tunghinensis. This study provides future strategies for the ex situ conservation and management of the golden camellia species and other rare and endangered plants.

Abstract:An investigation of wild germplasm resources of Camellia japonica L. was conducted. The remaining natural distribution of the species is restricted in Changmeiyan Island, Daguan Island and the top of Laoshan Mountain. Only 506 trees were observed and their habitat has been severely damaged. However, contributed by protection measures taken in recent years, these remaining wild trees are growing very well.

Abstract:Abstract Camellia reticulata Lindl., also known as Yunnan Camellia, is an important ornamental plant in China, especially for its large and stunning flowers. A comprehensive understanding of their coloration mechanisms can aid breeders in developing new cultivars and improving their ornamental value; however, it is still unclear in Yunnan Camellia, especially in mixed-color flowers. In this study, we conducted metabolic and transcriptomic comparison analyses to investigate the coloration differences in three Yunnan Camellia cultivars: C. reticulata ‘Shizitou’ (SZT), C. reticulata ‘Damanao’ (MN), and C. reticulata ‘Tongzimian’ (TZM). Our results revealed that the initial flowering stage may play a critical role in the color change of MN. Metabolome analysis demonstrated that cyanidin was the primary anthocyanin in SZT and MN’s red region, while its content was low in TZM and MN’s white region. According to the transcriptome analysis, the anthocyanins biosynthesis pathway was reconstructed in Yunnan Camellia , and the low expression of CHS was detected in TZM and MN’s white region, while ANR maintained a high expression level, which may lead to the low content of cyanidin in them. Transcription factors MYBs, bHLH, and bZIP may play a key role in regulating anthocyanin-structural genes. The co-expression analysis showed that the meristem tissue may play a crucial role in the formation of the mixed white-red color in MN. Our study enriched the genetic basis of flower coloration differences in Yunnan Camellia which will be a valuable genomic resource to understanding the biology of coloration formation and for breeding the Camellia cultivars.

Abstract:Background: Cha-hua (Camellia reticulata) is one of China's traditional ornamental flowers developed by the local people of Yunnan Province. Today, more than 500 cultivars and hybrids are recognized. Many ancient camellia trees still survive and are managed by local peopl. A few records on cha-hua culture exist, but no studies expound the interaction between C. reticulata and traditional culture of ethnic groups. The contribution of traditional culture of different nationalities and regions to the diversity of Camellia reticulate is discussed. Methods: Ethnobotanical surveys were conducted throughout Central and Western Yunnan to investigate and document the traditional culture related to Camellia reticulata. Five sites were selected to carry out the field investigation. Information was collected using participatory observation, semi-structured interviews, key informant interviews, focus group discussions, and participatory rural appraisal (PRA). Results: Most of the ancient camellia trees were preserved or saved in the courtyards of old buildings and cultural or religious sites. Religion-associated culture plays an important role in C. reticulata protection. In every site we investigated, we found extensive traditional culture on C. reticulata and its management. These traditional cultures have not only protected the germplasm resources of C. reticulata, but also improved the diversity of Camellia cultivars. Conclusions: There are abundant and diverse genetic resources of cha-hua, Camellia reticulata in Yunnan. Cha-hua is not only an ornamental flower but also has been endowed with rich spiritual connotation. The influence of traditional culture had improved the introduction and domestication of wild plants, breeding and selection of different varieties, and the propagation and dissemination of the tree in Yunnan. However, either some ancient cha-hua trees or their associated traditional culture are facing various threats. The old cha-hua trees and the ethnic camellia culture should be respected and protected since they have made great contributions in the history, and will make more contributions in the future.
Genetic diversity and population structure of yellow camellia (Camellia nitidissima) in China as revealed by RAPD and AFLP markers

Abstract:Camellia nitidissima, a rare plant but a useful genetic resource for commercial cultivation of ornamental camellias, is distributed in a narrow region of South China and North Vietnam. In this study, RAPD and AFLP markers were used to assess the genetic diversity and population structure of six natural populations of C. nitidissima from Guangxi in South China. Twenty RAPD primers amplified 183 bands, of which 143 bands were polymorphic, and 8 AFLP primer pairs produced 502 bands, of which 364 were polymorphic. Independent as well as combined analyses of the cluster analyses of the RAPD and AFLP fragments showed that the six populations could be classified into two major genetic groups corresponding to the Nanning and Fangcheng areas. The Mantel test revealed significant correlation between the genetic and geographic distances of C. nitidissima populations (r = 0.953, p = 0.036). AMOVA analysis allowed the partitioning of the genetic variation between groups (36.09%), among populations within groups (25.78%), and within populations (38.14%). An understanding of both the genetic diversity and the population structure of C. nitidissima in China can also provide insight into the conservation and management of this endangered species.

Abstract:Camellia nitidissima Chi (Theaceae) is a famous ornamental species with golden-yellow flowers. Due to deforestation and destructive collection of seedlings, its natural population size has been reduced greatly in recent decades. To provide basic information for its conservation, we studied the morphological and physiological characteristics of seeds and the environmental factors influencing germination. The seed production of C. nitidissima was of low yield, which caused lack of seed resource. The seeds were big and heavy, so they did not spread far from the mother trees and did not enter soil after falling down. Optimal temperatures for seed germination were 25–30°C. The fresh-harvested seeds had high moisture content and weak desiccation-tolerance, so they tended to be recalcitrant. Soaking for 24h in 10–30% PEG solution could significantly reduce the seed germination percentage. The imbibed seeds placed on the soil surface mostly could germinate, but some of them could not develop into seedlings because the embryo roots did not grow into soil. When the seeds were sowed at the depth of 1cm, the seedling emergence reached over 70%, but then decreased with increased sowing depth, and reached about 50% when at the depth of 5cm. The seeds sowed in sandy soil had higher emergence than those in clayed soil. It is feasible for seedling nursing to harvest seeds in time, harvesting prematurely or postmaturely may reduce seed quality.

Abstract:Introduction: Camellia , the largest genus of Theaceae, is well-known for having high economic values. Camellia granthamiana demonstrates large beautiful flowers with some primitive characters, such as multiple large and persistent bracteoles and sepals, was listed as Vulnerable species on the IUCN Red List. Methods: In this study, we investigated all possible records of the species, and sampled four natural populations and five cultivated individuals. By applying shallow-genome sequencing for nine individuals and RAD-seq sequencing for all the sampled 77 individuals, we investigated population genetic diversity and population structure of the species. Results and discussion: The results showed that the population sampled from Fengkai, previously identified as C. albogigias , possessed different plastid genome from other species possibly due to plastid capture; the species possesses strong population structure possibly due to the effect of isolation by distance, habitat fragmentation, and self-crossing tendency of the species, whose effective population size declined quickly in the past 4,000 years. Nevertheless, C. granthamiana maintains a medium level of genetic diversity within population, and significant differentiation was observed among the four investigated populations, it is anticipated that more populations are expected to be found and all these extant populations should be taken into instant protection.

Distribution and conservation status of Camellia longzhouensis (Theaceae), a critically endangered plant species endemic to southern China

Abstract:Camellia longzhouensis (Theaceae) is an endemic evergreen shrub or small tree with a distribution restricted to South China. It is listed as Grade-II in the National Key Protected Wild Plants List. In this study, we surveyed its distribution, examined its population structure, identified factors affecting its survival, and reassessed its extinction risk. We found that C. longzhouensis was only distributed in the Nonggang National Nature Reserve and the surrounding area. Its individuals only grew under the secondary forest canopy in the karst mountain. A total of 58 individuals of C. longzhouensis in three sub-populations were found. Soil fertility and understory light availability were the main habitat factors influencing the survival of C. longzhouensis. Anthropogenic disturbances and reproductive obstacles have caused a low seed-setting rate, poor seedling survival, and a lack of adult plants of C. longzhouensis in the natural habitat. The population structure of C. longzhouensis is spindle-shaped, indicating poor natural regeneration and inhibited seedling recruitment. Cleistanthus petelotii had a significant positive interspecific interaction with C. longzhouensis in the community. Based on the information obtained here and IUCN criterion C2ai, we recommend that C. longzhouensis be categorized in the International Union for Conservation of Nature (IUCN) Red List as Critically Endangered. We also developed a comprehensive protection strategy, consisting of in situ conservation, ex situ conservation, reintroduction, and commercial utilization. This strategy can be readily applied to protect other endangered plants with economic value in karst poor regions.

Abstract:Camellia nitidissima is an endangered species whose flowers are bright yellow. In the past 50 years, habitat changes caused by climate and human activity have greatly reduced the extent and size of natural populations of C. nitidissima. Here, to provide a fundamental basis for this plant's protection and utilization, the genetic diversity and genetic structure of C. nitidissima natural populations were studied using simple sequence repeats (SSR) markers. Based on a comparative analysis of multiple transcriptomes from different Camellia species, a group of consensus transcript sequences were obtained for marker development. We screened 237 predicted transcript loci, and identified 21 highly polymorphic SSR markers for population analysis. In total, 82 alleles were obtained to assess the genetic diversity of four natural populations in Guangxi Zhuang autonomous region, China. The results revealed that the mean observed heterozygosity (Ho) and expected heterozygosity (He) of the populations were 0.624 and 0.546, respectively, and the genetic diversity was mostly among populations. Further, we uncovered a significant positive correlation between the genetic distance and geographic distance of C. nitidissima populations. The high genetic diversity found in this study holds promise for the future conservation and restoration of C. nitidissima populations based on efforts from in situ and ex situ strategies implemented in a carefully monitored way.
Genotypic and Phenotypic Diversity of Endemic Golden Camellias Collected from the North of Vietnam

Abstract:Golden Camellias have recently been used as a food, cosmetic, and traditional medicine in China and Vietnam. Forty-two species have natural distribution in Vietnam, of which thirty-two species were considered endemic species of this country. The morphology of leaves and flowers of these species were similar; therefore, their taxonomic identification usually needed experts and the authentication has often been confused among species. Our study aims to describe the genetic diversity and the relationship of six species Camellia phanii, Camellia tamdaoensis, Camellia tienii, Camellia flava, Camellia petelotii and Camellia euphlebia by using three chloroplast DNA-barcodes: matK, rbcL and trnH-psbA. We also clarified the significant differences in anatomical characteristics of midvein and blade of their leaves, which suggested the possibility to use these criteria in taxonomy. In addition, preliminary chemical profiles of the methanolic extracts of leaves from six Golden Camellias such as total phenolic content (TPC), total flavonoid content (TFC), total anthocyanin content (TAC) and chlorogenic acids content (TCGAs) also showed the diversity among them. Interestingly, the discrimination on the catechins profile among six species followed the same tendency with the genetic distance on the phylogeny tree suggesting that catechins (i. e., discriminative catechins) can be biomarkers for the chemotaxonomy of these six Golden Camellias.

Abstract:Ex situ conservation in urban areas is an essential complementary approach to in situ conservation in wild areas. In this study, we combined multiple approaches, including vegetation investigations for both natural habitats and urban green spaces, eco-physiological experiments and remote sensing investigations for urban green spaces, to identify potential habitats for introducing the endangered plant species Calycanthus chinensis to urban green spaces. The results showed that (1) C. chinensis prefer living under sparse forests with canopy densities of 20-60%; (2) C. chinensis was not a shade tolerant species due to its chlorophyll a/b ratio (2.58) being higher than the threshold (2.3) for shade tolerant species; (3) the large and thin leaves of C. chinensis are easily damaged by strong wind, so this species can only live under moderate canopy cover; (4) to maintain a sparse crown for the well-being of C. chinensis, the upper layer trees in urban green spaces need to be thinned slightly; and (5) introducing this endangered species increases biodiversity and ecosystem services of urban green spaces. Finally, this study provides a framework and a case study for using urban green spaces as micro-refuges for endangered species.

Abstract:Background: Camellia nitidissima is a rare, prized camellia species with golden-yellow flowers. It has a high ornamental, medicinal, and economic value. Previous studies have shown substantial flavonol accumulation in C. nitidissima petals during flower formation. However, the mechanisms underlying the golden flower formation in C. nitidissima remain largely unknown. Results: We performed an integrative analysis of the transcriptome, proteome, and metabolome of the petals at five flower developmental stages to construct the regulatory network underlying golden flower formation in C. nitidissima. Metabolome analysis revealed the presence of 323 flavonoids, and two flavonols, quercetin glycosides and kaempferol glycosides, were highly accumulated in the golden petals. Transcriptome and proteome sequencing suggested that the flavonol biosynthesis-related genes and proteins upregulated and the anthocyanin and proanthocyanidin biosynthesis-related genes and proteins downregulated in the golden petal stage. Further investigation revealed the involvement of MYBs and bHLHs in flavonoid biosynthesis. Expression analysis showed that flavonol synthase 2 (CnFLS2) was highly expressed in the petals, and its expression positively correlated with flavonol content at all flower developmental stages. Transient overexpression of CnFLS2 in the petals increased flavonol content. Furthermore, correlation analysis showed that the jasmonate (JA) pathways positively correlated with flavonol biosynthesis, and exogenous methyl jasmonate (MeJA) treatment promoted CnFLS2 expression and flavonol accumulation. Conclusions: Our findings showed that the JA-CnFLS2 module regulates flavonol biosynthesis during golden petal formation in C. nitidissima.

Abstract:Golden camellia is defined as a species of the Camellia genus with yellow flowers, which have long been used as a medicine, food, and cosmetic in many Asian countries. To date, more than 50 golden camellia species are considered endemic in Vietnam; however, more information is needed about its chemical constituents and biological activity. This work aims to unveil the potential of Camellia tienii Ninh, a golden camellia species, as an herbal beverage by examining the presence and abundance of chemical components in flowers and leaves. A comprehensive strategy has been developed using both liquid and gas chromatography coupled with mass spectrometry. Specifically, LC-MS-based widely targeted analyses were opted to characterize 158 polar metabolites belonging mainly to flavonoids, catechins, and amino acids classes, and an untargeted approach using GC-MS annotated 42 major volatile compounds such as terpenes and fatty acids. The extensive profile revealed by these techniques could help understand the significant discrimination between two organs. C. tienii flowers accumulated more flavonoids, amino acids, and fatty acids, while leaves contain more terpenes, suggesting different pharmacological properties of these materials. Overall, this pipeline can be applied for other Camellia species and valorization of these valuable resources for health benefits purposes.

Abstract:Camellia flava (Pit.) Sealy 1949 is a rare and precious species with golden flowers, which hold important ornamental and breeding values. In this study, the complete chloroplast genome of C. flava is reported for the first time. The chloroplast genome exhibits a typical quadripartite structure with a total length of 156,670 bp and a GC content of 37.32%, including a large single-copy region (86,250 bp), a small single-copy region (18,292 bp), and a pair of inverted repeat regions (26,064 bp). A total of 133 genes, including 88 protein-coding genes, 37 tRNA genes, and 8 rRNA genes were annotated. The phylogenetic analysis revealed a close relationship between C. flava and C. tamdaoensis. The chloroplast genome sequence of C. flava serves as a valuable resource for further breeding research and genetic phylogenetic studies.
