Abstract:Human serum albumin (HSA) is an extracellular protein that has the highest concentration in blood plasma and is a carrier for many small molecules. HSA has an exceptional binding capacity for many endogenous and exogenous ligands, and contains two main binding sites with high affinity for diverse substances, named Site I and Site II. The binding cavity of Site II is more active and has greater ligand affinity than that of Site I. In this study, molecular simulation methods were used to investigate the molecular interactions between Site II and twelve Site II-specific ligands. The results showed that hydrophobic interactions were the main driving force for binding, with electrostatic interactions playing a secondary role. The key residues and binding mode on Site II were identified with the computational alanine-scanning approach. Three layers were found from the entrance to the interior of the binding pocket, contributing electrostatic interactions, hydrophobic interactions and mixed interactions, respectively. Finally, molecular docking and molecular dynamics were used to predict the binding mode of L-tryptophan on Site II. The results provided insights into the binding mode of Site II on HSA, which could guide the design of new Site II-specific drugs and ligands for the efficient separation of HSA.
Quantitation, Visualization, and Monitoring of Conformational Transitions of Human Serum Albumin by A Tetraphenylethene Derivative with Aggregation-Induced Emission Characteristics

Abstract:Human scrum albumin (HSA) is a major protein component of blood plasma, and its assay is of obvious value to biological research. We, herein, present a readily accessible fluorescent bioprobe for HSA detection and quantitation. A nonemissive tetraphenylethene derivative named sodium 1,2-bis[4-(3-sulfonatopropoxyl)phenyl]-1,2-diphenylethene (BSPOTPE) is induced to emit by IBA, showing a novel phenomenon of aggregation-induced emission (AIE). The AIE bioprobe enjoys a broad working range (0-100 nM), a low detection limit (down to 1 nM), and a superior selectivity to albumins. The fluorescent bioassay is unperturbed by the miscellaneous bioelectrolytes in the artificial urine. The AIE luminogen can also be used as a rapid and sensitive protein stain in gel electrophoresis for HSA visualization. Utilizing the AIE feature of BSPOTPE and the Forster resonance energy transfer from HSA to BSPOTPE, the unfolding process of HSA induced by guanidine hydrochloride is monitored, which reveals a multistep transition with the involvement of molten globule intermediates. Computational modeling suggests that the AIE luminogens dock in the hydrophobic cleft between subdomains IIA and IIIA of HSA with the aid of hydrophobic effect, charge neutralization, and hydrogen bonding interactions, offering mechanistic insight into the microenvironment inside the hydrophobic cavity.
Preparation and evaluation of mixed-mode resins with tryptophan analogues as functional ligands for human serum albumin separation☆

Abstract:Five tryptophan analogues with a hydrophobic indole ring and an amino group on each molecule were used as functional ligands of mixed-mode resins for human serum albumin (HSA) purification. Their adsorption performance was evaluated and the effects of pH and salt addition on HSA adsorption were studied. The resins prepared showed typical pH-dependent adsorption and the highest adsorption capacity and affinity were found at pH5.0 for all the resins tested. The saturated adsorption capacity was 138.02mg·g−1 with the tryptamine-functionalized resin, which significantly decreased at pH below 4.0 due to electrostatic repulsion between ligands and HSA. Moreover, the addition of NaCl or (NH4)2SO4 in media reduced HSA adsorption capacity, although the two salts showed different affecting profiles. The tryptamine-functionalized resin showed the best salt-tolerant performance, and its high adsorption capacity was maintained under high salt concentrations. In addition, the five resins prepared showed good adsorption selectivity for recombinant HSA from Pichia pastoris broth. Molecular docking results between tryptamine and HSA indicated that tryptamine was favorable to bind on Site II (indole-binding site) of HSA.

Abstract:Metanil yellow (MYL) is a toxic azo dye extensively used in food preparations and in textile industry as major colorant. It affects important biological processes leading to development of toxicity in different organ systems. Toxicological profile assessment has also revealed its potential for multi-organ toxicity. This work investigates the affinity, energetics, and structural dynamics of MYL binding with Human serum albumin (HSA) in Thyroxine binding site using multi-spectroscopic techniques and molecular simulations. Fluorescence spectroscopic studies of MYL-HSA complex at different temperatures revealed that MYL binds with HSA with stronger affinity (∼27 kJ/mole). The overall binding showed significant static quenching and was entropy driven. Circular dichroism studies showed that the secondary structure of HSA remained predominantly alpha helix. To accommodate within the Tr5 region of HSA, MYL underwent minor alterations in its conformation and position facilitated by the subtle expansion of the binding site (∼2 Å). The role of water in providing stability to the complex via water bridges and hydrogen bonds has also been investigated. Quantum Mechanics studies further revealed that the minor strain observed in the pyramidalamino nitrogen assisted MYL in the complexation.

Abstract:The binding behaviours of a transport protein, bovine serum albumin (BSA), in its native, unfolding and refolding states have been probed by monitoring the emission changes of two exogenous AIE-active fluorescent probes, M2 and M3, which are designed to be anionic and cationic, respectively. Due to their AIE properties, both M2 and M3 display emission enhancement when bound to the hydrophobic cavity of BSA. The binding site of M2 and M3 is found to be subdomain IIA. Then, the BSA + M2 and BSA + M3 systems are utilized to fluorescently signal the conformation changes of BSA caused by various external stimuli, including thermally or chemically induced denaturation. The data confirmed the multi-step unfolding process and the existence of a molten-globule intermediate state. The unfolding process consists of the rearrangement of subdomain IIA, the exposure of a negatively charged binding site in domain I that prefers interacting with cationic species, and the transformation of the molten-globule intermediate into the final random coil. The anionic and cationic modifications of the probes enable us to observe that electrostatic interactions play a role in the folding and unfolding of BSA.

Abstract:Spectroscopic approach and density functional theory (DFT) have been employed to investigate the interaction between melamine (MA) and bovine serum albumin (BSA). The UV absorption difference spectra show the MA–BSA complexes can form under physiological conditions. Furthermore, the B3LYP/6-311++G(d,p) calculations indicate that the protonated melamine (MAH+) bond to Asp, Glu, Asn, Gln, Ser, Thr amino acid residues or peptide groups via NH(MAH+)…OC(BSA) or NH(MAH+)…OC(BSA) hydrogen bonds, in agreement with the results of UV absorption difference spectra, and the interaction energies of them decrease in the order Asp, Glu>Pep>Asn, Gln>Ser, Thr. The fluorescence spectra, synchronous fluorescence spectra and FT-IR spectra demonstrate that this interaction has no obvious effect on the secondary structure of BSA.

Abstract:Human serum albumin (HSA) is among the blood plasma proteins crucial for transporting various natural and pharmaceutical substances throughout the body. This research examined how the structure of HSA changes when exposed to C m -E2O2-C m gemini surfactants, which possess various advantageous properties, utilizing a combination of multispectral analysis and computational modeling techniques. The aggregation behavior of gemini surfactants in the presence of HSA was also investigated to demonstrate the HSA-C m -E2O2-C m complexation. The intrinsic fluorescence findings showed that gemini surfactants caused HSA fluorescence to diminish due to static quenching. The binding constants ( K b ) for HSA-C m -E2O2-C m at varying temperatures were around 10 3 M −1 , with approximately one binding site detected. In addition, the effect of gemini surfactants on the conformation of HSA was analyzed by performing UV–vis, extrinsic, synchronous, RRS and 3-D fluorescence, and CD experiments. The interactions' binding sites and driving forces (also spontaneity) were calculated using molecular docking analysis, which agrees with site marker studies and thermodynamic parameter value results.

Abstract:The interaction of 2-mercaptobenzimidazole (MBI) with human serum albumin (HSA) was studied in vitro by equilibrium dialysis under normal physiological conditions. This study used fluorescence, ultraviolet-visible spectroscopy (UV-vis), Fourier transform infrared (FT-IR), circular dichroism (CD) and Raman spectroscopy, atomic force microscopy (AFM) and molecular modeling techniques. Association constants, the number of binding sites and basic thermodynamic parameters were used to investigate the quenching mechanism. Based on the fluorescence resonance energy transfer, the distance between the HSA and MBI was 2.495 nm. The ΔG(0), ΔH(0), and ΔS(0) values across temperature indicated that the hydrophobic interaction was the predominant binding Force. The UV, FT-IR, CD and Raman spectra confirmed that the HSA secondary structure was altered in the presence of MBI. In addition, the molecular modeling showed that the MBI-HSA complex was stabilized by hydrophobic forces, which resulted from amino acid residues. The AFM results revealed that the individual HSA molecule dimensions were larger after interaction with MBI. Overall, this study suggested a method for characterizing the weak intermolecular interaction. In addition, this method is potentially useful for elucidating the toxigenicity of MBI when it is combined with the biomolecular function effect, transmembrane transport, toxicological testing and other experiments.
Binding elucidation of azo dyes with human serum albumin via spectroscopic approaches and molecular docking techniques

Abstract:The large number of synthesized azo dyes is widely applied in the food, pharmaceutical, cosmetic, textile, and leather industries. In this study, the binding mechanism of two synthesized dyes with human serum albumin (HSA); as the most abundant protein in plasma; was elucidated by fluorescence spectroscopy, Fourier-transform infrared spectroscopy and molecular modeling methods. The fluorescence quenching measurements showed that each dye can quench the intrinsic fluorescence of HSA via a dynamic quenching mechanism with an increase in concentration. From the thermodynamic data observations, revealed that the binding process is a spontaneous molecular force for each dye with HSA due to hydrophobic interactions and hydrogen bonding. FT-IR spectra showed that the secondary structure of the protein changes due to interaction of each dye with HSA. Furthermore, docking simulation demonstrated that the probable binding location of both dyes is subdomain IIA of HSA (Sudlow site I) and that complex formed is stabilized by hydrophobic interactions and hydrogen bonding.Communicated by Ramaswamy H. Abstract:The interaction of dihydromyricetin (DMY) with human serum albumin (HSA) was investigated by spectroscopic methodologies and molecular docking.The fluorescence spectral results showed that HSA fluorescence was quenched regularly with the addition of DMY,the quenching mechanism may be a static fluorescence quenching procedue.All the magnitude of binding constants (KA) were larger than 105 L/mol and the number of binding sites (n) in the binary system were approximate to 1 in different temperature.According to thermodynamic parameters of Van't Hoff equation,it could be suggested the binding process of DMY with HSA was spontaneous and the main interaction force of DMY with HSA was electrostatic force.The binding distance (r) between the DMY and HSA was calculated to be about 3.32 nm based on the theory of Fisrster's nonradiation energy transfer,which indicated that the energy trasfer from HSA to DMY occurs with high possibility.The synchronous and 3D florescence spectroscopy demonstrated that the secondary conformation of HSA has been changed after interaction with DMY.From the result of site marker competitive experiments and the molecular docking,it could be deduced that DMY was inserted into the subdomain ⅡA (site Ⅰ) of HSA.
Unfolding of Human Serum Albumin by Gemini and Single-Chain Surfactants: A Comparative Study

Abstract:The interactions of human serum albumin (HSA) with four cationic gemini surfactants and four single chain surfactants have been investigated from the measurements of fluorescence spectroscopy, dynamic lights scattering (DLS), zeta potential, and circular dichroism (CD). The fluorescence results show the binding strength of surfactants to HSA, along with the blue shift under the maximum emission wavelength (lambda(max)) which is sensitive to the protein conformations. It has been found that the hydrodynamic diameters of the protein aggregates are enlarged and the electrokinetic potentials of them get positive with increasing the concentration of the surfactants. The conformational transformation of the secondary structures of surfactant-HSA aggregates has been confirmed through the quantitative analysis of the CD spectra. The isothermal titration microcalorimetry (ITC) has been employed to further analyze the binding process of single-chain surfactants to HSA. The comprehensive results obtained from various approaches have shown that both hydrophobic and electrostatic interactions are present in the cationic surfactant-HSA systems. Compared with the corresponding single-chain surfactants, the gemini ones have much stronger binding ability to induce the unfolding of HSA. These investigations on the interactions between HSA and surfactants with the altered chain architecture in different concentration regimes can facilitate the application of surfactant-protein systems in pharmaceutical, biotechnology and related fields. Abstract:The interaction between Ag+ and human serum albumin (HSA) has been intensively studied by means of equilibrium dialysis, ligand-to-metal charge transition (LMCT) bands, circular dichroism (CD) and Raman spectroscopy. Scatchard analysis of the results of equilibrium dialysis indicates the presence of two types of binding sites for Ag+ on HSA, and the orders of magnitude of binding stability constants are found to be 105 and 104, respectively. During the binding process, a gradual increase in absorbance values of LMCT bands is observed with time-scanning UV absorption spectra, implying the Ag(I) centers are continually formed in HSA. The time-scanning CD spectra provide evidence that the binding of Ag+ induces HSA to undergo a slow rearrangement of tertiary structure, and to change from the original conformation in the absence of Ag+ (B-state) to conformation binding with Ag+ (A-state). The rate constants and activation free energy of A–B transition are calculated. The Raman spectrum of Ag(I)–HSA system shows distinct vibration bands at 224 and 246 cm−1 in the low-frequency region, which significantly reveal the formation of Ag–S and Ag–N bonds. In addition, the electrostatic interaction between Ag+ and negatively charged oxygen is also detected with Raman spectroscopy.

Abstract:Human serum albumin (HSA) is the most abundant plasma protein in the human body with a plasma concentration of 0.6mM. HSA plays an important role in drug transport and metabolism. Enzymatic activity of HSA on different substrates or drugs has been studied and documented. The structural mechanism of this activity, however, is unknown. In this study, we have determined the crystal structures of HSA–myristate in a complex of aspirin and of salicylic acid, respectively. The crystal structure of HSA–myristate–aspirin illustrates that aspirin transfers acetyl group to Lys199 and is hydrolyzed into salicylic acid by HSA. The hydrolysis product, salicylic acid, remains bound to HSA at a similar location, but it shows a very different orientation when compared with the salicylic acid in the HSA–myristate–salicylic acid ternary complex. These results not only provide the structural evidence of esterase activity of HSA, and demonstrate the conformational plasticity of HSA on drug binding, but also may provide structural information for the modulation of HSA–drug interaction by computational approach based on HSA–drug structure.
Spectroscopic Studies on the Interaction of Murexide and Murexide-Copper Complex with Human Serum Albumin

Abstract:The interactions of human serum albumin (HSA) with murexide (Mx) and murexide-copper complex (Mx-Cu2+) were investigated by fluorescence spectroscopy and Fourier transform infrared spectroscopy FTIR) under simulative physiological conditions. The results show Mx and Mx-Cu2+ could quench the intrinsic fluorescence of HSA by static quenching and hydrophobic interaction was the predominant intermolecular force. The binding constant of Mx and HSA was increased in the presence of copper (H). According to fluorescence resonance energy transfer (FRET), the distancer between donor (HSA) and acceptor (Mx and MxCu(2+)) were calculated to be 2.82 and 2.53 nm, the ternary system energy transfer efficiency E' > binary system E, which showed the copper ion acts as the energy transfer agency. The synchronous fluorescence and FTIR study suggested that Mx and Mx-Cu2+ can cause almost the same changes in the secondary structure of HSA. Upon binding with Mx or Mx-Cu2+, complex, the a-helix structure was reduced more than 8%, the beta-sheet reduced 1%, while the beta-turn increased 6%, and the random coil increased 4%. The results show that the main factor of secondary structure changing is Mx.
Investigation on conformational variation and fibrillation of human serum albumin affected by molybdenum disulfide quantum dots

Abstract:In this work, binding interaction between molybdenum disulfide quantum dots (MoS2 QDs) and human serum albumin (HSA) was researched deeply to dissect the conformational variation and fibrillation of HSA affected by MoS2 QDs. The results revealed that MoS2 QDs bound strongly with HSA with molar ratio of 1:1 under the joint actions of hydrogen bond and van der Waals force, leading to the static fluorescence quenching of HSA. MoS2 QDs caused the secondary structure transition of HSA from α-helix stepwise to β-turn, β-sheet, and random coil gradually. MoS2 QDs reduced both the molar enthalpy change and the melting temperature of HSA, reducing the thermal stability of HSA significantly. It is worth noting that MoS2 QDs inhibited the fibrillation process of HSA according to the reduced hydrophobic environment and the disturbance of disulfide bonds in HSA network structure. These results reveal the precise binding mechanism of MoS2 QDs with HSA at molecular level, providing indispensable information for the potential application of MoS2 QDs in biological fields.

Abstract:Eugenol (EGL) and 2-methylpyrazine (2-MTP) are common food additives that are used as flavors in a variety of foods. To study the effect of food additives on human serum albumin (HSA), it is necessary to deeply study the interaction of EGL and 2-MTP with HSA. Fluorescence, UV–vis absorption, circular dichroism (CD) and computational techniques showed EGL and 2-MTP bind statically to HSA, inducing conformational changes of HSA. We calculated the quenching constants (k sv ), binding constants (k q ), binding distances (r) and thermodynamic constants (ΔH 0 , ΔS 0 , ΔG 0 ) at four different temperatures. Based on thermodynamic analysis and molecular dynamics (MD) simulations, the main forces of 2-MTP-HSA and EGL-HSA systems are hydrophobic interaction and π-π accumulation, respectively. In addition, the interaction alters the physiological activity of HSA, esterase activity and free radical scavenging ability. This study provides valuable insights into the molecular interactions of food additive compounds EGL and 2-MTP with HSA, the findings provide theoretical reference for the development of new food additives and drugs in the future.

Abstract:Sulfonamides are widely used antibiotics in agricultural production. However, the potential threat of these drugs to human health has increased global concern. Human serum albumin (HSA) is the main reservoir and transporter of exogenous small molecules in humans. In this study, the interaction between sulfadimethoxine (SMT) and human serum albumin (HSA) was studied using spectroscopy and computer simulation. Our results showed that the hydrogen bonding and van der Waals forces drove SMT to enter the binding site I of HSA spontaneously and resulted in the fluorescence quenching of HSA. The stability of the HSA–SMT complex decreased with an increase in temperature. The binding of SMT to HSA induced alterations in the secondary structure of HSA, where the content of α-helix decreased from 61.0% of the free state to 59.0% of the compound state. The π–π, π–σ, and π–alkyl interactions between HSA and SMT were found to play important roles in maintaining the stability of the complex.

Abstract:Raman spectra of human serum albumin (HSA) and HSA-3-picolylamine complex were obtained. The spectra indicate the configuration and structural transformations of HSA. The results show that the secondary structure is main alpha-helix, and the binding of 3-picolylamine doesn't change the secondary structure. Comparative study of the interaction of some meso-substituted anionic cyanine dyes with human serum albumin

Abstract:Spectral-fluorescent properties of the meso-substituted anionic cyanine dye 3,3'-di-(γ-sulfopropyl)-4,5,4',5'-dibenzo-9-methylthiacarbocyanine betaine (DMC) were studied in solutions and in the presence of human serum albumin (HSA). The properties of DMC were compared with those of the previously studied meso-substituted anionic dyes 3,3'-di(γ-sulfopropyl)-4,5,4',5'-dibenzo-9-ethylthiacarbocyanine betaine (DEC), 3,3'-di-(γ-sulfopropyl)-9-methylthiacarbocyanine betaine (MTC) and 3,3'-di-(γ-sulfopropyl)-5,5'-diphenyl-9-ethyloxacarbocyanine betaine (OCC), which were studied here in more detail. In aqueous solutions, DMC, like DEC, is prone to dimerization; it also forms H- and J-aggregates. The noncovalent interaction of DMC with HSA leads to decomposition of the dimers with a shift in the cis-trans isomeric equilibrium toward the trans-monomer complexed with HSA, which is accompanied by a significant increase in fluorescence. The spectral-fluorescent data were used to estimate the binding constants of the dyes with HSA and other characteristics of the dyes, which are important when used as probes for HSA. The effect of structural rearrangements of HSA upon denaturation by urea on the spectral-fluorescent properties of the dyes was studied. Molecular docking of the dye-HSA systems was performed. A comparative assessment of the prospects for the use of the dyes as spectral-fluorescent probes for HSA in vitro was carried out.

Abstract:The specific recognition of serum proteins by scavenger receptors is critical and fundamental in many biological processes. However, the underlying mechanism of scavenger receptor-serum protein interaction remains elusive. In this work, taking scavenger receptors class A1 (SR-A1) as an example, we systematically investigate its interaction with human serum albumin (HSA) at different states through a combination of molecular docking and all-atom molecular dynamics simulations. It is found that native HSA can moderately bind to collagen-like (CL) region or scavenger receptor cysteine-rich (SRCR) region, with both electrostatic (ELE) and van der Waals (VDW) interactions, playing important roles. After maleylation, the binding energy, particularly the ELE energy, between HSA and CL region is significantly enhanced, while the binding energy between HSA and SRCR region remains nearly unchanged. Additionally, we also observe that unfolding of the secondary structures in HSA leads to a larger contact surface area between denatured HSA and CL region, but has little impact on the HSA-SRCR region interaction. Therefore, similar to maleylated HSA, denatured HSA is also more likely to bind to the CL region of SR-A1.
