PLA2G2A, a novel β-catenin/TCF target gene, inhibits gastric cancer invasion and metastasis

Abstract:B62 Carcinoma of the stomach (gastric cancer) is the second highest cause of worldwide cancer mortality. Expression of PLA2G2A , a secreted phospholipase, in gastric tumors has previously been shown to carry prognostic relevance, but the pathways regulating PLA2G2A , and how it mediates its pro-survival effect, are currently unknown. In this report, we demonstrate that PLA2G2A is a direct target of the Wnt signaling pathway, whose expression is reduced in late stage and metastatic gastric, colon, and prostate tumors compared to early stage carcinomas. We found evidence of PLA2G2A genomic deletion and promoter methylation in gastric cancer cell lines, and that PLA2G2A can inhibit the invasive ability of gastric cancer cells through the transcriptional down-regulation of metastasis mediator genes such as S100A4 and NEDD9 . Our results thus shed light on PLA2G2A’s mechanism of action in gastric cancer, and suggest that the induced expression of PLA2G2A , either by exogenous administration or endogenous reactivation, may represent a novel therapeutic approach to treat late stage and metastatic gastric cancers.
