Abstract:Herein, a highly sensitive detection platform for the dual-mode determination of sarcosine by fluorescence and colorimetry was constructed based on zirconium-based metal organic frameworks (Zr-MOFs) with multi-enzyme immobilization. Zr-MOFs with peroxidase-mimicking activity were firstly prepared by a solvothermal method, and sarcosine oxidase (SOX) and horseradish peroxidase (HRP) were simultaneously encapsulated in the Zr-MOFs to form the Zr-MOFs@SOX@HRP composite. Sarcosine could be catalyzed oxidation by the SOX in Zr-MOFs@SOX@HRP composite to generate hydrogen peroxide (H2O2). The generated H2O2 was decomposed through the synergistic catalysis of Zr-MOFs and HRP to generate free radicals that could oxidize the chromogenic substrate p-phenylenediamine (PPD) to produce oxPPD. The oxPPD generated could quench the fluorescence of the carbon dots at 510 nm via the internal filtering effect (IFE), The amount of sarcosine could be sensitively detected by monitoring the changes of colorimetric and fluorescence signals, with detection limits of 0.44 μM and 0.21 μM, respectively. Thus, the dual-mode detection of sarcosine by fluorescence and colorimetry through Zr-MOFs-mediated cascade reaction system was realized. The method was applied to the determination of sarcosine in the urine of healthy individuals as well as in the urine of prostate cancer patients with satisfactory results, which demonstrating the enormous potential for predicting prostate cancer disease.

2D Fe/Co-MOF/SOX cascade reactors for fast noninvasive detection of sarcosine level in prostate cancer urine

Abstract:Sarcosine plays a key role in screening for early prostate cancer. However, the several already reported peroxidase mimics immobilized with sarcosine oxidase (SOX) utilized to detect uriary sarcosine still have some limitations such as complex synthesis process, using of expensive heavy metals to mimic enzyme activity and long color development time. Herein, an inexpensive peroxidase-like 2D Fe/Co-MOF nanosheet was prepared by a simple solvent modulation method. The resultant 2D Fe/Co-MOF nanosheets have strong peroxidase activity, with its Vmax value for H2O2 of 15.3 × 10-8 M/s, being 1.76 times that of HRP. Then, using the 2D Fe/Co-MOF as a peroxidase model for anchoring natural SOX to construct 2D Fe/Co-MOF/SOX , which can act as a cascade reactor for detection of sarcosine. Considering the above properties, a platform for the detection of sarcosine was built based on a colorimetric method. Because of presence of the high ratio of Fe2+ caused by the electron transfer from Co2+ to Fe3+, large specific surface area and plentiful active sites, 2D Fe/Co-MOF/SOX with TMB (3,3',5,5'-tetramethylbenzidine) colorimetric reagent could have fast color development and can be applied conveniently and fastly in an early screening tool for prostate cancer patients. The sarcosine could be quantified by peroxidase activity with a detection range of 1-400 μM and a limit of detection (LOD) of 0.324 μM. More importantly, the average sarcosine concentration of 21.367 μM and 1.871 μM was detected in patient's and normal urine (n = 5), respectively, which showed an excellent screening effect and a great potential in early prostate cancer.

Abstract:It is critical to develop a highly efficient and sensitive method for detecting the biomarker sarcosine (SA) of prostate cancer due to its importance for men's health. In our work, a fluorescence (FL) and colorimetric dual-mode multienzyme cascade nanoplatform for SA detection was designed and constructed. CuNCs/FeMn-ZIF-8/PCN nanocomposites with high FL properties and peroxidase-like activity were successfully prepared by encapsulating copper nanoclusters (CuNCs) into FeMn-ZIF-8 and then loaded onto P-doped graphitic carbon nitride (PCN). Furthermore, the nanocomposites served as carriers for the immobilization of sarcosine oxidase (SOX) to construct a high-efficiency dual-mode multienzyme cascade nanoplatform CuNCs/SOX@FeMn-ZIF-8/PCN for the detection of SA. The intermediate H2O2 generated in the cascade caused the FL quenching of nanocomposites and the discoloration of 3,3',5,5'-tetramethylbenzidin. The linear ranges for SA detection in the dual-mode system were 1-100 μM (FL) and 1-200 μM (colorimetric), with detection limits of 0.34 and 0.59 μM, respectively. This nanoplatform exhibited notable repeatability, specificity, and stability, making it suitable for detecting sarcosine in real human urine samples. Therefore, this dual-mode multienzyme cascade nanoplatform would have a potential applicative prospect for detecting SA and other biomarkers in real clinical samples.

Abstract:Background: Early prostatic cancer (PCa) diagnosis significantly improves the chances of successful treatment and enhances patient survival rates. Traditional enzyme cascade-based early cancer detection methods offer efficiency and signal amplification but are limited by cost, complexity, and enzyme dependency, affecting stability and practicality. Meanwhile, sarcosine (Sar) is commonly considered a biomarker for PCa development. It is essential to develop a Sar detection method based on cascade reactions, which should be efficient, low skill requirement, and suitable for on-site testing. Results: To address this, our study introduces the synthesis of organic-inorganic self-assembled nanoflowers to optimize existing detection methods. The Sar oxidase (SOX)-inorganic hybrid nanoflowers (Cu3(PO4)2:Ce@SOX) possess inherent fluorescent properties and excellent peroxidase activity, coupled with efficient enzyme loading. Based on this, we have developed a dual-mode multi-enzyme cascade nanoplatform combining fluorescence and colorimetric methods for the detection of Sar. The encapsulation yield of Cu3(PO4)2:Ce@SOX reaches 84.5 %, exhibiting a remarkable enhancement in catalytic activity by 1.26-1.29 fold compared to free SOX. The present study employing a dual-signal mechanism encompasses 'turn-off' fluorescence signals ranging from 0.5 μM to 60 μM, with a detection limit of 0.226 μM, and 'turn-on' colorimetric signals ranging from 0.18 μM to 60 μM, with a detection limit of 0.120 μM. Significance: Furthermore, our study developed an intelligent smartphone sensor system utilizing cotton swabs for real-time analysis of Sar without additional instruments. The nano-platform exhibits exceptional repeatability and stability, rendering it well-suited for detecting Sar in authentic human urine samples. This innovation allows for immediate analysis, offering valuable insights for portable and efficient biosensors applicable to Sar and other analytes.

Abstract:High detection stability makes sarcosine oxidase (SOx) nanozyme as natural enzyme substitute, a promising sensor for prostate cancer biomarker Sarcosine (Sar). However, its limited catalytic capacity towards product (H 2 O 2 ) and the following weak signal response significantly restrict the detection sensitivity of Sar. Herein, a novel NH 2 -MIL88B(Fe)-porphyrin/riboflavin (NM88B-POR/Rf) electrochemical sensor with a hyperoxia microenvironment and cascade reaction functions is proposed to promote catalytic H 2 O 2 production and cascade signal amplification (from H 2 O 2 to •OH). Fe-porphyrin implantation in NM88B-POR/Rf as bionic heme enhanced the O 2 -carrying capacity, thus effectively improving the sensor's utilization rate of dissolved oxygen and the catalytic H 2 O 2 production ability of Rf. Based on enzymatic cascade strategy, NM88B in NM88B-POR/Rf as peroxidase nanozyme further catalyzed H 2 O 2 to generate •OH. Methyl blue was used to capture •OH and enhance Sar detection sensitivity. Detection results revealed that NM88B-POR/Rf presented as low limit of detection (LOD) as 3.31 pM with detection range of 10 pM∼1 mM. The synergistic effect of the hyperoxia microenvironment and enzyme cascade make LOD about 4 orders of magnitude lower than reported state-of-the-art biosensors. Moreover, NM88B-POR/Rf has good selectivity and stability and hence also be applied to the detection of Sar in real urine.

Abstract:As the well-known test-indicator for early prostate cancer (PCa), sarcosine (SA) is closely related to the differential pathological process, which makes its accurate determination increasingly significant. Herein, we for the first time expanded the peroxidase (POD)-like property of facile-synthesized Zn-TCPP(Fe) MOF to fluorescent substrates and exploited it to ratiometric fluorescent (RF) sensing. By harnessing the effective catalytic oxidation of MOF nanozyme toward two fluorescent substrates (Scopoletin, SC; Amplex Red, AR) with contrary changes, and target-responsive (SA + SOx)/MOF/(SC + AR) tandem catalytic reaction, we constructed the first MOF nanozyme-based RF sensor for the quantitative determination of SA. Superior to previous works, the operation of this RF sensor is under the guidance of AND-(ANDˆNAND) contrary logic circuit. The dual-channel binary output changes (from 1/0 to 0/1) not only enable the intelligent logical recognition of SA, bringing strengthened reliability and accuracy, but also manifest the proof-of-concept discrimination of PCa individuals and healthy ones. Through recording the fluorescence alterations of SC (F 465 ) and AR (F 585 ), two segments of linear relationships between ratiometric values (F 585 /F 465 ) and varied contents of SA are realized successfully. The LOD for SA could reach to as low as 39.98 nM, which outperforms all nanozyme-originated SA sensors reported till now. Moreover, this sensor also demonstrates high selectivity and satisfactory performance in human serum samples. Furthermore, the portable sensing of SA is realized under the assistance of smartphone-based RGB analysis, demonstrating the potential of point-of-care diagnostics of PCa in the future.

Abstract:Herein, we report a facile and label-free method for sensitive and specific determination of prostate cancer biomarker sarcosine via using photoluminescent bimetallic Au/Cu nanoplatelets (AuCu NPs) to construct an inner filter effect (IFE)-based photoluminescence (PL) sensing platform. The AuCu NPs were formed by the cysteine-induced co-reduction reaction, which displayed bright PL with an emission peak at 560 nm. Meanwhile, the Cu(I) doping caused a maximum 25-fold enhancement of quantum yield (QY), compared with the native Au(I) complexes, i.e., from 0.85 to 21.5%. By integrating the AuCu NPs with p-phenylenediamine (PPD) oxidation reaction, an IFE-based sensor for sarcosine detection was constructed. In this method, sarcosine is oxidized under the catalysis of sarcosine oxidase (SOx) to yield H2O2. The latter further oxidizes PPD to form 2,5-diamino-N,N'-bis(p-aminophenyl)-l,4-benzoquinone di-imine (PPDox) in the presence of horseradish peroxidase (HRP). The UV-vis absorption spectrum of the PPDox can overlap well with the excitation and emission spectra of the AuCu NPs, resulting in the efficient quenching of the AuCu NPs via the IFE effect. Therefore, this IFE-based AuCu NPs/SOx/PPD/HRP sensing platform can be used for highly sensitive and specific sensing of sarcosine. The sensing platform showed two linear regions of the PL intensity of the AuCu NPs versus the concentration of sarcosine in the range of 0.5-5 μM and 5-100 μM with a detection limit (LOD) of 0.12 μM (S/N = 3). Furthermore, this IFE-based sensing platform could be developed into a paper-based biosensor for simple, instrument-free, and visual detection of sarcosine.

Abstract:Background: Sarcosine is an amino acid that is formed by methylation of glycine and is present in trace amounts in the body. Increased sarcosine concentrations in blood plasma and urine are manifested in sarcosinemia and in some other diseases such as prostate cancer. For this purpose, sarcosine detection using the nanomedicine approach was proposed. In this study, we have prepared superparamagnetic iron oxide nanoparticles (SPIONs) with different modified surface area. Nanoparticles (NPs) were modified by chitosan (CS), and sarcosine oxidase (SOX). SPIONs without any modification were taken as controls. Methods and Results: The obtained NPs were characterized by physicochemical methods. The size of the NPs determined by the dynamic light scattering method was as follows: SPIONs/Au/NPs (100–300 nm), SPIONs/Au/CS/NPs (300–700 nm), and SPIONs/Au/CS/SOX/NPs (600–1500 nm). The amount of CS deposited on the NP surface was found to be 48 mg/mL for SPIONs/Au/CS/NPs and 39 mg/mL for SPIONs/Au/CS/SOX/NPs, and repeatability varied around 10%. Pseudo-peroxidase activity of NPs was verified using sarcosine, horseradish peroxidase (HRP) and 3,3′,5,5′-tetramethylbenzidine (TMB) as a substrate. For TMB, all NPs tested evinced substantial pseudo-peroxidase activity at 650 nm. The concentration of SPIONs/Au/CS/SOX/NPs in the reaction mixture was optimized to 0–40 mg/mL. Trinder reaction for sarcosine detection was set up at 510 nm at an optimal reaction temperature of 37 °C and pH 8.0. The course of the reaction was linear for 150 min. The smallest amount of NPs that was able to detect sarcosine was 0.2 mg/well (200 μL of total volume) with the linear dependence y = 0.0011x − 0.0001 and the correlation coefficient r = 0.9992, relative standard deviation (RSD) 6.35%, limit of detection (LOD) 5 μM. The suggested method was further validated for artificial urine analysis (r = 0.99, RSD 21.35%, LOD 18 μM). The calculation between the detected and applied concentrations showed a high correlation coefficient (r = 0.99). NPs were tested for toxicity and no significant growth inhibition was observed in any model system (S. cerevisiae, S. aureus, E. coli). The hemolytic activity of the prepared NPs was similar to that of the phosphate buffered saline (PBS) control. The reaction system was further tested on real urine specimens. Conclusion: The proposed detection system allows the analysis of sarcosine at micromolar concentrations and to monitor changes in its levels as a potential prostate cancer marker. The whole system is suitable for low-cost miniaturization and point-of-care testing technology and diagnostic systems. This system is simple, inexpensive, and convenient for screening tests and telemedicine applications.

Abstract:Cancer ranks as a leading cause of death in every country of the world. However, if they are discovered early, a lot of cancers can be prevented or cured. Discovering and monitoring cancer markers are the main methods for early diagnosis of cancer. To date, many fluorescent probes designed and used for early cancer diagnosis can only react with a single marker, which always causes insufficient accuracy in complex systems. Herein, a novel near-infrared (NIR) fluorescent probe (CyO-DNP) for the sequential detection of H2S and H+ is synthesized. In this probe, a heptamethine dye is selected as the fluorophore and a 2,4-dinitrophenyl (DNP) ether is chosen as recognition group. In the presence of H2S, CyO-DNP is transformed into CyO, which exhibits an intense fluorescence at 663 nm. Then, H+ induces the protonation of CyO to obtain CyOH, and the final fluorescence emission at 793 nm significantly enhances. Owing to the low cyt otoxicity and the NIR fluorescence emission, CyO-DNP can sequentially monitor endogenous H2S and H+ in cancer cells and image exogenous and endogenous H2S and H+ in mice. It is worth mentioning that CyO-DNP can effectively avoid the false positive signal caused by the liver and kidney and discriminate normal mice and tumor mice accurately. For all we know, CyO-DNP is the first fluorescent probe for early accurate diagnosis of cancer by sequentially detecting H2S and H+.

Abstract:As a biomarker of prostate cancer, sarcosine plays an important role in the diagnosis of cancer. The specificity of enzymatic reaction between sarcosine oxidase and sarcosine could be applied for the rapid and specific assay of sarcosine. Enzyme-based sarcosine quantification method has attracted our attention due to its high specificity and selectivity, but its wide use is restricted by the poor stability and challenging reusability. In this study, sarcosine oxidase was used to synthesize magnetic cross-linked enzyme aggregates (MCLEAs) with great stability and recyclability. The MCLEAs were characterized by transmission electron microscope, X-ray photoelectron spectroscopy, thermogravimetric analysis, X-ray diffractometer, and vibrating sample magnetometer, respectively. Subsequently, a colorimetric assay of sarcosine was developed based on the enzymatic reaction between MCLEAs and the substrate. The method can be used to quantify sarcosine with a linear range from 0.3125 to 10 μg mL−1 and recoveries ranging 87.50–97.75 %, suggesting its potential for reliable analysis of sarcosine in urine.

Abstract:As a common gaseous signaling molecule, hydrogen sulfide (H2S) plays a vital role in physiology and pathology. The development of fluorescent probes for detecting H2S has attracted widespread attention. However, most of the reported fluorescent probes with nitrobenzoxadiazole (NBD) as the recognition group have been widely used to simultaneously detect biothiols and H2S, instead of specifically detecting H2S. Herein, a novel NBD-based near-infrared (NIR) fluorescent probe named CX-N for the detection of H2S is synthesized. The selectivity of CX-N for H2S is significantly higher than that for biothiols and other potential interferences. After reacting with H2S, CX-N shows a significant increase in NIR fluorescence (75-fold), large Stokes shift (155 nm) and fast response (4 min). And the possible response mechanism of CX-N to H2S is given and confirmed by HPLC and HRMS. Based on the low cytotoxicity of CX-N, it has been used for H2S imaging in live cells and zebrafish. More importantly, CX-N has also been successfully applied for the real-time imaging of H2S in inflammatory and tumor mice based on its NIR emission, which provides a reliable platform for the specific recognition of H2S in complex biological systems.

Abstract:Rapid and intelligent prostate cancer (PCa) identification is crucial for early diagnosis. A strategy is proposed using sarcosine quantification in urine, achieving intelligent multicolor visualization platform. The RGB value can be extracted via mobile phone camera, enabling swift PCa diagnosis. This platform detects minute sarcosine concentration differences and can be expanded for measuring specific biomarkers in biological samples. Rapid and intelligent identification of prostate cancer (PCa) is critical for early diagnosis. Herein, a convenient, reliable, and intelligent strategy is proposed to screen PCa individuals through indirectly quantifying sarcosine (Sar), an early indicator of PCa, in clinical urine samples. Success is achieved by integrating sarcosine oxidase (SOX) as a specific recognition unit; nanozyme‐assisted multicolor intelligent visualization platform as a signal reporter. With the Fe‐MOFs and peroxidase, the synergetic action of SOX and response gold nanorods (Au NRs) is controlled etched to exhibit a multicolored signal. The sensor exhibits excellent linearity with Sar within 1–60 × 10−6 m, boasting a remarkable detection limit of 0.12 × 10−6 m. The RGB value of the display color can be directly extracted using a mobile phone camera. PCa diagnosis can be swiftly made (within 15 min) and directly by identifying two RGB colors (R 135). The enzyme‐assisted multicolor intelligent visualization platform is adept at detecting minute differences in Sar concentration in urine samples between PCa patients and healthy individuals. The concept of enzyme‐assisted multicolor sensing can be further expanded by modifying the type of immobilized enzymes, providing a valuable guideline for the rational design of multiple probes to measure specific biomarkers in biological samples.

Abstract:This manuscript explores the use of Carbon Quantum Dots and Graphene Quantum Dots for the detection of sarcosine. It covers the synthesis of these nanomaterials, and their fluorescent properties, including colorimetric, amperometric, and electrochemical methods. The study presents findings, discusses challenges, and provides future perspectives on improving sarcosine sensing technologies. Luminescent carbon nanoparticles, such as carbon quantum dots (CQDs) and graphene quantum dots (GQDs), are increasingly utilized for sarcosine detection due to their exceptional water permeability, sustained luminescence, and biocompatibility. Sarcosine as a biomarker holds significance for therapeutic purposes and disease identification. Carbon‐based sensors offer eco‐friendliness, recyclability, and affordability, making them sustainable for sensing applications across various domains. Despite the widespread use of standard sensors for sarcosine measurement, they face limitations. This paper presents a focused review of recent developments in biosensors utilizing luminescent carbon quantum dots and graphene quantum dots for sarcosine detection. Key processes are characterized by their selectivity and sensitivity to different forms of sarcosine and interfering substances like metallic ions and amino acids. The significant reaction mechanisms and detection methods for sarcosine are outlined and compared. Additionally, the challenges in developing functionalized CQDs and GQDs for further investigation are explored. In summary, luminescent carbon nanoparticles offer a promising avenue for sarcosine detection, addressing limitations of standard sensors through their cost‐effectiveness, sensitivity, and rapid response, thus holding potential for diverse sensing applications.

Abstract:In diagnosing prostate cancer and distinguishing it from other prostate diseases, the ratio of the concentration of free prostate-specific antigen (f-PSA) to total prostate-specific antigen (t-PSA), i.e., (f-PSA%) is more accurate than the concentration of t-PSA alone. Immunoassay based on surface-enhanced Raman scattering (SERS) frequency shift has been proven to be particularly suitable for detecting large biomolecules with high reproducibility. Along similar lines, the present study developed a SERS-based biosensor that simultaneously detects t-PSA and f-PSA. The 4-mercaptobenzoic acid (MBA) on the immunocapture substrate is coupled to the t-PSA antibody through the carboxyl group, and the combination of t-PSA induces the Raman frequency shifts of MBA. The immunocolloidal gold attached with f-PSA antibodies selectively capture the f-PSA that immobilized on the MBA-modified SERS substrates, allowing for f-PSA quantification according to the SERS intensities of the 5, 5'-Dithiobis (succinimidyl-2-nitrobenzoate) (DSNB) probe. The results show that f-PSA and t-PSA have good linear response in the concentration scale of 0.1-20 ng/mL, and 1-200 ng/mL, respectively. The biosensor combines Raman frequency shifts and intensities, which greatly simplifies traditional procedures for f-PSA% detection. All the results demonstrated the great potential of the proposed biosensor in highly reproducible and accurate diagnosis of prostate cancers.

Abstract:A metal–organic framework‐derived biomineral probe is guided by using enzyme‐embedded protein trap engineering for non‐invasive cancer monitoring. An all‐in‐one paper chromogenic device is developed and applied to a range of expanded bioanalytical platforms by modifying natural enzymes to introduce different cascade catalytic models to steer the construction of efficient bioprobes. A paper‐based naked‐eye recognition assay with enzyme‐encapsulated protein engineered metal–organic framework‐derived biominerals is developed for direct quantification of sarcosine in urine samples for screening of prostate cancer individuals. The detection strategy stems from the successful construction of a cascade response model, which involves the introduction of a cascade enzymatic catalytic reaction on Pt nanoparticles (NPs)‐loaded porous CeO2 by integrating a sarcosine oxidase as a special recognition unit and a chromogenic substrate as a signal molecule reporter. Pt NPs‐loaded CeO2 is subjected to a one‐step thermal treatment based on multilayered mesoporous Ce‐based metal–organic framework, and the calcined CeO2 exhibits the same distinct porous graded structure. Importantly, introduction of Pt NPs sharply enhances the peroxidase‐like activity of CeO2, which is considered to be caused by the difference in the adsorption behavior of hydrogen peroxide on the CeO2 surface and Pt/CeO2 obtained by density functional theory calculations. On the basis of this, the probe is used on a mass‐producible paper‐based working platform and 3D‐printed device to specifically screen for minor differences in sarcosine between urine samples from cancer patients and normal individuals. Enzyme‐assisted cascade catalytic reaction can be extended by replacing different recognition units for multiple analytes.

Abstract:An early diagnosis is the gold standard for cancer survival. Biosensors have proven their effectiveness in monitoring cancer biomarkers but are still limited to a series of requirements. This work proposes an integrated power solution, with an autonomous and self-signaling biosensing device. The biorecognition element is produced in situ by molecular imprinting to detect sarcosine, a known biomarker for prostate cancer. The biosensor was assembled on the counter-electrode of a dye-sensitized solar cell (DSSC), simultaneously using EDOT and Pyrrole as monomers for the biomimetic process and the catalytic reduction of triiodide in the DSSC. After the rebinding assays, the hybrid DSSC/biosensor displayed a linear behavior when plotting the power conversion efficiency (PCE) and the charge transfer resistance (RCT) against the logarithm of the concentration of sarcosine. The latter obtained a sensitivity of 0.468 Ω/decade of sarcosine concentration, with a linear range between 1 ng/mL and 10 μg/mL, and a limit of detection of 0.32 ng/mL. When interfacing an electrochromic cell, consisting of a PEDOT-based material, with the hybrid device, a color gradient between 1 ng/mL and 10 μg/mL of sarcosine was observed. Thus, the device can be used anywhere with access to a light source, completely equipment-free, suitable for point-of-care analysis and capable of detecting sarcosine within a range of clinical interest.
Non-enzymatic electrochemical detection of sarcosine in serum of prostate cancer patients by CoNiWBO/rGO nanocomposite

Abstract:Selective and sensitive sarcosine detection is crucial due to its recent endorsement as a prostate cancer (PCa) biomarker in clinical diagnosis. The reduced graphene oxide-cobalt nickel tungsten boron oxides (CoNiWBO/rGO) nanocomposite is developed as a non-enzymatic electrochemical sensor for sarcosine detection in PCa patients' serum. CoNiWBO/rGO is synthesized by the chemical reduction method via a one-pot reduction method followed by calcination at 500 °C under a nitrogen environment for 2 h and characterized by UV-Vis, XRD, TGA, and SEM. CoNiWBO/rGO is then deposited on a glassy carbon electrode, and sarcosine sensing parameters are optimized, including concentration and pH. This non-enzymatic sensor is employed to directly determine sarcosine in serum samples. Differential pulse voltammetry (DPV) and linear sweep voltammetry (LSV) are employed to monitor the electrochemical behavior where sarcosine binding leads to oxidation. Chronoamperometric studies show the stability of the developed sensor. The results demonstrate a wide linear range from 0.1 to 50 μM and low limits of detection, i.e., 0.04 μM and 0.07 μM using DPV and LSV respectivel. Moreover, the calculated recovery of sarcosine in human serum of prostate cancer patients is 78–96%. The developed electrochemical sensor for sarcosine detection can have potential applications in clinical diagnosis.

Abstract:A novel fluorescent immunosensor was developed based on the use of CuS nanoparticles (CuS NPs) as labels for the highly sensitive detection of human prostate cancer biomarker prostate specific antigen (PSA). In the presence of CuS NPs, the non-fluorescent substrate o-phenylenediamine could be oxidized into the stable fluorescent product 2,3-diamiophenazine at physiological pH. Throughout the reaction, no other oxidizing agents (e.g. hydrogen peroxide) were needed. The relatively mild oxidation conditions made the immunoassay robust, reliable and facile. The proposed immunoassay exhibited high sensitivity and specificity for the detection of PSA. A linear relationship between the fluorescent signals and the concentration of PSA was obtained in the range of 0.5 pg mL(-1) to 50 ng mL(-1), with a detection limit of 0.1 pg mL(-1) (S/N = 3). The proposed fluorescent immunoassay can be used as a promising platform for the detection of a variety of other biomarkers.
A Ti3 C2 TX /Pt-Pd based amperometric biosensor for sensitive cancer biomarker detection

Abstract:The detection of cancer biomarkers is of great significance for the early screening of cancer. Detecting the content of sarcosine in blood or urine has been considered to provide a basis for the diagnosis of prostate cancer. However, it still lacks simple, high-precision and wide-ranging sarcosine detection methods. In this work, a Ti3 C2 TX /Pt-Pd nanocomposite with high stability and excellent electrochemical performance has been synthesized by a facile one-step alcohol reduction and then used on a glassy carbon electrode (GCE) with sarcosine oxidase (SOx ) to form a sarcosine biosensor (GCE/Ti3 C2 TX /Pt-Pd/SOx ). The prominent electrocatalytic activity and biocompatibility of Ti3 C2 TX /Pt-Pd enable the SOx to be highly active and sensitive to sarcosine. Under the optimized conditions, the prepared biosensor has a wide linear detection range to sarcosine from 1 to 1000 µM with a low limit of detection of 0.16 µM (S/N = 3) and a sensitivity of 84.1 µA/mM cm2 . Besides, the reliable response in serum samples shows its potential in the early diagnosis of prostate cancer. More importantly, the successful construction and application of the amperometric biosensor based on Ti3 C2 TX /Pt-Pd will provide a meaningful reference for detecting other cancer biomarkers.

Abstract:In this study, an electrochemical aptasensor was developed for the specific detection of sarcosine using a covalent organic framework (COF). The imine-based two-dimensional COF was synthesized through a solvothermal process using terephthaldehyde and melamine. This resulted in the formation of a structure that is highly porous and has a unique surface area of 908 m2/g. The produced biosensor demonstrated a significant linear relationship between charge transfer resistance (Rct) and various concentrations of sarcosine in blood serum samples. The aptasensor had two linear ranges, spanning from 0.5 fM to 700 fM and 10 pM to 0.12 nM, respectively, with a detection limit of 0.15 fM. The incorporation of high surface area COFs in the aptasensor design offers a promising combination of sensitivity, stability, and specificity. This combination creates a valuable device for diagnosing and monitoring of prostate cancer and potentially other diseases.
