Abstract:Background Individuals who smoke exhibit lower body mass index (BMI) but face an elevated risk of obesity-related diseases. Emerging evidence suggests smoking may influence the composition of the microbiome. Thus, this study aims to investigate this paradox from the perspective of gut microbiota. Results Our findings contradict the conventional view that smoking primarily leads to weight loss. We observed a significant positive association between the smoking-related microbiota index and several obesity indicators. Longitudinal analysis revealed that smoking-related microbiota index is linked with an increased risk of diabetes, obesity-related cancers, and cardiovascular complications. Sibling comparison analyses further supported these results, demonstrating the robustness of the associations. Conclusion These findings challenge the conventional notion that smoking is merely associated with weight loss, uncovering instead that smoking-related microbiome changes significantly contribute to a higher risk of obesity and an augmented central fat distribution, ultimately leading to a higher risk of obesity-related diseases.

Abstract:The interplay among cigarette smoking status, oral microbiota, and cardiometabolic health is poorly understood. We aimed to examine the association of cigarette smoking status with oral microbiota and to assess the association of the identified microbial features with cardiometabolic risk factors in a Chinese population. This study included 587 participants within the Central China Cohort, including 111 smokers and 476 non-smokers, and their oral microbiota was profiled by 16S rRNA sequencing. Both oral microbial alpha- and beta-diversity were distinct between smokers and non-smokers ( p < 0.05). With adjustment for sociodemographics, alcohol and tea drinking, tooth brushing frequency, and body mass index, the relative abundance of nine genera and 26 pathways, including the genus Megasphaera and two pathways involved in inositol degradation which have potentially adverse effects on cardiometabolic health, was significantly different between two groups (FDR q < 0.20). Multiple microbial features related to cigarette smoking were found to partly mediate the associations of cigarette smoking with serum triglycerides and C-reactive protein levels ( p- mediation < 0.05). In conclusion, cigarette smoking status may have impacts on the oral microbial features, which may partially mediate the associations of cigarette smoking and cardiometabolic health.

Abstract:Introduction: Many studies identified that smokers had lower body mass index (BMI) than non-smokers, but the underlying mechanisms are not well understood. Hypothesis: An untargeted metabolomics study will identify novel mechanisms underlying smoking associated weight loss. Methods: We performed cross-sectional analyses among all 248 current smokers and 637 never smokers participated in the Bogalusa Heart Study 2016 survey. After stringent quality control and normalization, 1,202 plasma metabolites were assessed for mediation effects on smoking-BMI association after controlling for age, gender, race, education, drinking and physical activity. The collective mediation effects of metabolites were estimated by comparing the smoking-BMI associations before and after controlling for all significant metabolites after Bonferroni correction. Bidirectional Mendelian randomization analyses were then performed on a subset of participants (n=654) to delineate causal directions among smoking, BMI, and significant metabolites. We constructed multi-locus genetic risk scores (GRS) for smoking and BMI based on findings from published GWASs with a sample size of more than 250,000. Multivariable linear regression controlling for the same covariates as in the mediation analyses was used to estimate associations between smoking- and BMI-GRSs and the significant metabolites. Results: Compared to never smokers, current smokers had a 3.31 kg/m 2 (95% confidence interval [CI]: 2.15-4.47) lower BMI after adjusting for all covariables. A total of 40 metabolites significantly mediated and explained 90.7% of the smoking-BMI association. Seven of the significant metabolites were associated with smoking GRS but not BMI GRS, suggesting a true mediation role in the smoking-BMI association. The seven metabolites, including homoarginine (mediation beta=-0.49 kg/m 2 , 95% CI: -0.78 to -0.26), 1-oleoyl-GPE (18:1) (mediation beta=-0.44 kg/m 2 , 95% CI: -0.72 to -0.22), 1-linoleoyl-GPE (18:2)* (mediation beta=-0.59 kg/m 2 , 95% CI: -0.90 to -0.32), beta-cryptoxanthin (mediation beta=0.53 kg/m 2 , 95% CI: 0.27 to 0.83), 1-stearoyl-2-arachidonoyl-GPE (18:0/20:4) (mediation beta=0.39 kg/m 2 , 95% CI: 0.18 to 0.65), sphingomyelin (d18:2/21:0, d16:2/23:0)* (mediation beta=-0.54 kg/m 2 , 95% CI: -0.84 to -0.28), and sphingomyelin (d17:2/16:0, d18:2/15:0)* (mediation beta=-0.92 kg/m 2 , 95% CI: -1.30 to -0.59), mediated 36.3% of the association between current smoking and BMI. We also identified 6 metabolites significantly associated with BMI GRS but not with smoking GRS, and 2 significantly associated with both smoking- and BMI-GRSs. Conclusions: We identified 7 plasma metabolites that mediated a large proportion of the association between smoking and BMI.

Abstract:Background: Cigarette smoking is an important environmental risk factor for cardiovascular events of hypertension (HTN). Existing studies have provided evidence supporting altered gut microbiota by cigarette smoking, especially in hypertensive patients. Metabolic biomarkers play a central role in the functional potentials of the gut microbiome but are poorly characterized in hypertensive smokers. To explore whether serum metabolomics signatures and compositions of HTN patients were varied in smokers, and investigate their connecting relationship to gut microbiota, the serum metabolites were examined in untreated hypertensive patients using untargeted liquid chromatography-mass spectrometry (LC/MS) analysis. Results: A dramatic difference and clear separation in community features of circulating metabolomics members were seen in smoking HTN patients compared with the non-smoking controls, according to partial least squares discrimination analysis (PLS-DA) and orthogonal partial least squares discrimination analysis (OPLS-DA). Serum metabolic profiles and compositions of smoking patients with HTN were significantly distinct from the controls, and were characterized by enrichment of 12-HETE, 7-Ketodeoxycholic acid, Serotonin, N-Stearoyl tyrosine and Deoxycholic acid glycine conjugate, and the depletion of Tetradecanedioic acid, Hippuric acid, Glyceric acid, 20-Hydroxyeicosatetraenoic acid, Phenylpyruvic acid and Capric acid. Additionally, the metabolome displayed prominent functional signatures, with a majority proportion of the metabolites identified to be discriminating between groups distributed in Starch and sucrose metabolism, Caffeine metabolism, Pyruvate metabolism, Glycine, serine and threonine metabolism, and Phenylalanine metabolic pathways. Furthermore, the observation of alterations in metabolites associated with intestinal microbial taxonomy indicated that these metabolic members might mediate the effects of gut microbiome on the smoking host. Indeed, the metabolites specific to smoking HTNs were strongly organized into co-abundance networks, interacting with an array of clinical parameters, including uric acid (UA), low-denstiy lipoprotein cholesterol (LDLC) and smoking index. Conclusions: In conclusion, we demonstrated disparate circulating blood metabolome composition and functional potentials in hypertensive smokers, showing a linkage between specific metabolites in blood and the gut microbiome.

Abstract:BACKGROUND:Although the association between cigarette smoking and risk of type 2 diabetes is well established, its mechanisms are yet to be clarified. This study examined the possible mediating effects of adiponectin, leptin, and C-reactive protein (CRP) concentrations on the smoking-diabetes association.METHODS:Between 2002 and 2011, we followed 3338 Japanese workers, aged 35-66 years, who were enrolled in the second Aichi workers' cohort study. We used multivariable-adjusted Cox regression models to determine the hazard ratios and respective 95% confidence intervals (CIs) of the association between smoking status and risk of diabetes. A multiple mediation model with bootstrapping was used to estimate the magnitude and the respective bias-corrected (BC) 95% CIs of the indirect effects of smoking on diabetes through the three biomarkers.RESULTS:Relative to never smokers, the risk of diabetes was significantly elevated in current (hazard ratio 1.75, 95% CI 1.25-2.46) and ex-smokers (hazard ratio 1.54, 95% CI 1.07-2.22). The indirect effects of smoking on diabetes through adiponectin levels were statistically significant among light (point estimate 0.033, BC 95% CI 0.005-0.082), moderate (point estimate 0.044, BC 95% CI 0.010-0.094), and heavy smokers (point estimate 0.054, BC 95% CI 0.013-0.113). In contrast, neither the indirect effects of smoking on diabetes through leptin nor CRP levels were significant, as the corresponding BC 95% CIs included zero.CONCLUSIONS:In our analysis, adiponectin concentration appeared to partially mediate the effect of smoking on diabetes, while leptin and CRP levels did not.
From tobacco smoking to mutational signature: the role of epigenetic changes in human cancers

Abstract:Abstract Background Tobacco smoking is associated with a unique mutational signature in the human cancer genome. It is unclear whether tobacco smoking -altered DNA methylations and gene expressions affect smoking-related mutational signature.Methods We evaluated the smoking-related DNA methylation sites reported from five previous studies using peripheral blood cells to identify possible target genes. Using the mediation analysis approach, we evaluated whether the association of tobacco smoking with mutational signature was mediated through altered DNA methylation and expression of these target genes.Results Based on data obtained from 21,108 blood samples, we identified 374 smoking-related DNA methylation sites, annotated to 248 target genes. Using data from DNA methylations, gene expressions and smoking-related mutational signature generated from ~7,700 tumor tissue samples across 26 cancer types from The Cancer Genome Atlas (TCGA), we found 11 of the 248 target genes whose expressions were associated with smoking-related mutational signature at a Bonferroni-correction P < 0.001. This included four for head and neck cancer, and seven for lung adenocarcinoma. In lung adenocarcinoma, our results showed that smoking increased the expression of three genes, AHRR , GPR15 , and HDGF, and decreased the expression of two genes, CAPN8 , and RPS6KA1 , which were consequently associated with increased smoking-related mutational signature. Additional evidence showed that the elevated expression of AHRR (cg14817490) and GPR15 (cg19859270), were associated with smoking-altered hypomethylations. Lastly, we showed that the elevated expression of HDGF and decreases expression of RPS6KA1, were associated with poor survival of lung cancer patients.Conclusions Our findings provide novel insights into the contributions of tobacco smoking to carcinogenesis through complex molecular mechanisms of the elevated mutational signature by altered DNA methylations and gene expressions.

Abstract:Background Metabolomics helps to identify links between environmental exposures and intermediate biomarkers of disturbed pathways. We previously reported variations in phosphatidylcholines in male smokers compared with non-smokers in a cross-sectional pilot study with a small sample size, but knowledge of the reversibility of smoking effects on metabolite profiles is limited. Here, we extend our metabolomics study with a large prospective study including female smokers and quitters. Methods Using targeted metabolomics approach, we quantified 140 metabolite concentrations for 1,241 fasting serum samples in the population-based Cooperative Health Research in the Region of Augsburg (KORA) human cohort at two time points: baseline survey conducted between 1999 and 2001 and follow-up after seven years. Metabolite profiles were compared among groups of current smokers, former smokers and never smokers, and were further assessed for their reversibility after smoking cessation. Changes in metabolite concentrations from baseline to the follow-up were investigated in a longitudinal analysis comparing current smokers, never smokers and smoking quitters, who were current smokers at baseline but former smokers by the time of follow-up. In addition, we constructed protein-metabolite networks with smoking-related genes and metabolites. Results We identified 21 smoking-related metabolites in the baseline investigation (18 in men and six in women, with three overlaps) enriched in amino acid and lipid pathways, which were significantly different between current smokers and never smokers. Moreover, 19 out of the 21 metabolites were found to be reversible in former smokers. In the follow-up study, 13 reversible metabolites in men were measured, of which 10 were confirmed to be reversible in male quitters. Protein-metabolite networks are proposed to explain the consistent reversibility of smoking effects on metabolites. Conclusions We showed that smoking-related changes in human serum metabolites are reversible after smoking cessation, consistent with the known cardiovascular risk reduction. The metabolites identified may serve as potential biomarkers to evaluate the status of smoking cessation and characterize smoking-related diseases.

Abstract:OBJECTIVE:To confirm the effect of cigarette smoking on β-cell function, and further investigate the mediation effect of abdominal obesity.METHODS:Participants would include 1440 Chinese smokers who had participated in a community-based chronic disease screening project in Guangzhou and Zhuhai from 2006 to 2007. They were interviewed with structured questionnaire on their socio-demographic status and smoking behaviors. Waist-to-hip ratio (WHR) and fasting serum C-peptide concentration were also measured.RESULTS:After adjustment for the potential confounding factors, when compared with smokers with consumption 1 - 10 cigarettes/day, smokers with consumption of 11 - 20 cigarettes/day (adjusted OR = 1.53, 95%CI: 1.22 - 1.90) or > 20 cigarettes/day (adjusted OR = 1.92, 95%CI: 1.32 - 2.79) had significant higher risks to get C-peptide concentration larger than its median. Furthermore, 37.54% of the effect of cigarette smoking on C-peptide concentration was partially mediated by abdominal obesity.CONCLUSION:Cigarette smoking might be a risk factor for β-cell dysfunction and abdominal obesity.
Influence of Cigarette Smoke Exposure on Metabolic Profiling in Rats

Abstract:The influences of different cigarette smoke exposure on endogenous small molecular metabolome were evaluated by metabolomics research on the basis of liquid chromatography-mass spectrometry (LC/MS).Plasma and urine samples from normal control rats, ordinary cigarette smoked rats and a certain commercial cigarette with herbal additives smoked rats were analyzed respectively to obtain the metabolic profiling.The rats were exposed to cigarette smoke for one week, half a month and one month.The data were processed by the method of partial least squares discriminant analysis (PLS-DA).The potential biomarkers were screened out according to VIP (variable importance in the projection) and identified by database and standards.The PLS-DA results at different cigarette smoke exposing periods showed that the metabolism of rats was affected by both ordinary cigarette smoke exposed group and the certain commercial cigarette smoke exposed group, mainly including phospholipid metabolism, energy metabolism and oxidative damage.However, the damage of the commercial cigarette smoked rats was lower than that of ordinary cigarette smoked rats.The relative concentration changes of some important biomarkers among the three groups, especially in one month, further confirmed that the herbal additives in the cigarette may relieve the influence of cigarette smoke on the holistic metabolome of rats, and improve their interfered metabolism, such us phospholipid and energy metabolism.

Abstract:INTRODUCTION:A general detrimental effect of smoking during pregnancy on the health of newborn children is well-documented, but the detailed mechanisms remain elusive.OBJECTIVES:Beside the specific influence of environmental tobacco smoke derived toxicants on developmental regulation the impact on the metabolism of newborn children is of particular interest, first as a general marker of foetal development and second due to its potential predictive value for the later occurrence of metabolic diseases.METHODS:Tobacco smoke exposure information from a questionnaire was confirmed by measuring the smoking related metabolites S-Phenyl mercapturic acid, S-Benzyl mercapturic acid and cotinine in maternal urine by LC-MS/MS. The impact of smoking on maternal endogenous serum metabolome and children's cord blood metabolome was assessed in a targeted analysis of 163 metabolites by an LC-MS/MS based assay. The anti-oxidative status of maternal serum samples was analysed by chemoluminiscence based method.RESULTS:Here we present for the first time results of a metabolomic assessment of the cordblood of 40 children and their mothers. Several analytes from the group of phosphatidylcholines, namely PCaaC28:1, PCaaC32:3, PCaeC30:1, PCaeC32:2, PCaeC40:1, and sphingomyelin SM C26:0, differed significantly in mothers and children's sera depending on smoking status. In serum of smoking mothers the antioxidative capacity of water soluble compounds was not significantly changed while there was a significant decrease in the lipid fraction.CONCLUSION:Our data give evidence that smoking during pregnancy alters both the maternal and children's metabolome. Whether the different pattern found in adults compared to newborn children could be related to different disease outcomes should be in the focus of future studies.
Smoking induces coordinated DNA methylation and gene expression changes in adipose tissue with consequences for metabolic health

Abstract:Background: Tobacco smoking is a risk factor for multiple diseases, including cardiovascular disease and diabetes. Many smoking-associated signals have been detected in the blood methylome, but the extent to which these changes are widespread to metabolically relevant tissues, and impact gene expression or metabolic health, remains unclear. Methods: We investigated smoking-associated DNA methylation and gene expression variation in adipose tissue biopsies from 542 healthy female twins. Replication, tissue specificity, and longitudinal stability of the smoking-associated effects were explored in additional adipose, blood, skin, and lung samples. We characterized the impact of adipose tissue smoking methylation and expression signals on metabolic disease risk phenotypes, including visceral fat. Results: We identified 42 smoking-methylation and 42 smoking-expression signals, where five genes (AHRR, CYP1A1, CYP1B1, CYTL1, F2RL3) were both hypo-methylated and upregulated in current smokers. CYP1A1 gene expression achieved 95% prediction performance of current smoking status. We validated and replicated a proportion of the signals in additional primary tissue samples, identifying tissue-shared effects. Smoking leaves systemic imprints on DNA methylation after smoking cessation, with stronger but shorter-lived effects on gene expression. Metabolic disease risk traits such as visceral fat and android-to-gynoid ratio showed association with methylation at smoking markers with functional impacts on expression, such as CYP1A1, and at tissue-shared smoking signals, such as NOTCH1. At smoking-signals, BHLHE40 and AHRR DNA methylation and gene expression levels in current smokers were predictive of future gain in visceral fat upon smoking cessation. Conclusions: Our results provide the first comprehensive characterization of coordinated DNA methylation and gene expression markers of smoking in adipose tissue. The findings relate to human metabolic health and give insights into understanding the widespread health consequence of smoking outside of the lung.

Abstract:The aim of this study was to compare changes in the metabolite levels of ex-smokers and nonsmokers using a metabolomics approach, accounting for the weight gain in ex-smokers. Volunteer ex-smokers and nonsmokers were recruited from two cohorts Shijingshan (174) and Xishan (78), respectively, at a 1 : 1 ratio for age and sex. Nontargeted metabolomics was performed on the volunteers’ blood samples using liquid chromatography-mass spectrometry, and multivariate statistical analysis was performed using principal component analysis and orthogonal partial least squares discriminant analysis. Enrichment analysis was used to identify Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways associated with differential metabolites and weighted gene co-expression network analysis and maximal correlation coefficient (MCC) algorithms were used to identify key metabolites. The results revealed no significant differences between the distribution of blood metabolite levels in the ex-smokers and nonsmokers. The biosynthesis of valine, leucine, and isoleucine was determined to be associated with differential metabolites, and five key metabolites were identified. Further analysis revealed differences in weight gain and regained metabolite levels in ex-smokers, and 10 differential metabolites were identified that may be associated with weight gain in ex-smokers. These findings suggest that quitting smoking restores metabolites to almost normal levels and results in weight gain. The identified key metabolites and metabolic pathways may also provide a basis for clinical studies.
From Tobacco Smoking to Cancer Mutational Signature: a Mediation Analysis Strategy to Explore the Role of Epigenetic Changes

Abstract:Background Tobacco smoking is associated with a unique mutational signature in the human cancer genome. It is unclear whether tobacco smoking-altered DNA methylations and gene expressions affect smoking-related mutational signature. Methods We systematically analyzed the smoking-related DNA methylation sites reported from five previous casecontrol studies in peripheral blood cells to identify possible target genes. Using the mediation analysis approach, we evaluated whether the association of tobacco smoking with mutational signature is mediated through altered DNA methylation and expression of these target genes in lung adenocarcinoma tumor tissues. Results Based on data obtained from 21,108 blood samples, we identified 374 smoking-related DNA methylation sites, annotated to 248 target genes. Using data from DNA methylations, gene expressions and smoking-related mutational signature generated from ~ 7700 tumor tissue samples across 26 cancer types from The Cancer Genome Atlas (TCGA), we found 11 of the 248 target genes whose expressions were associated with smoking-related mutational signature at a Bonferroni-correction P < 0.001. This included four for head and neck cancer, and seven for lung adenocarcinoma. In lung adenocarcinoma, our results showed that smoking increased the expression of three genes, AHRR , GPR15 , and HDGF , and decreased the expression of two genes, CAPN8 , and RPS6KA1 , which were consequently associated with increased smoking-related mutational signature. Additional evidence showed that the elevated expression of AHRR and GPR15 were associated with smoking-altered hypomethylations at cg14817490 and cg19859270, respectively, in lung adenocarcinoma tumor tissues. Lastly, we showed that decreased expression of RPS6KA1 , were associated with poor survival of lung cancer patients. Conclusions Our findings provide novel insight into the contributions of tobacco smoking to carcinogenesis through the underlying mechanisms of the elevated mutational signature by altered DNA methylations and gene expressions.

Abstract:IMPORTANCE Chinese women have the highest rate of lung cancer among female never-smokers in the world, and the etiology is poorly understood. OBJECTIVE To assess the association between metabolomics and lung cancer risk among neversmoking women. DESIGN, SETTING, AND PARTICIPANTS This nested case-control study included 275 neversmoking female patients with lung cancer and 289 never-smoking cancer-free control participants from the prospective ShanghaiWomen's Health Study recruited from December 28, 1996, to May 23, 2000. Validated food frequency questionnaires were used for the collection of dietary information. Metabolomic analysis was conducted from November 13, 2015, to January 6, 2016. Data analysis was conducted from January 6, 2016, to November 29, 2018. EXPOSURES Untargeted ultra-high-performance liquid chromatography-tandem mass spectrometry and nuclear magnetic resonance metabolomic profiles were characterized using prediagnosis urine samples. A total of 39 416metabolites were measured. MAIN OUTCOMES AND MEASURES Incident lung cancer. RESULTS Among the 564 women, those who developed lung cancer (275 participants; median [interquartile range] age, 61.0 [52-65] years) and those who did not develop lung cancer (289 participants; median [interquartile range] age, 62.0 [53-66] years) at follow-up (median [interquartile range] follow-up, 10.9 [9.0-11.7] years) were similar in terms of their secondhand smoke exposure, history of respiratory diseases, and body mass index. A peak metabolite, identified as 5-methyl-2-furoic acid, was significantly associated with lower lung cancer risk (odds ratio, 0.57 [95% CI, 0.46-0.72]; P <.001; false discovery rate = 0.039). Furthermore, this peakwasweakly correlated with self-reported dietary soy intake (. = 0.21; P <.001). Increasing tertiles of this metabolite were associated with lower lung cancer risk (in comparison with first tertile, odds ratio for second tertile, 0.52 [95% CI, 0.34-0.80]; and odds ratio for third tertile, 0.46 [95% CI, 0.30-0.70]), and the association was consistent across different histological subtypes and follow-up times. Additionally, metabolic pathway analysis found several systemic biological alterations that were associated with lung cancer risk, including 1-carbon metabolism, nucleotide metabolism, oxidative stress, and inflammation. CONCLUSIONS AND RELEVANCE This prospective study of the untargeted urinarymetabolome and lung cancer among never-smoking women in China provides support for the hypothesis that soy-based metabolites are associated with lower lung cancer risk in never-smoking women and suggests that biological processes linked to air pollution may be associated with higher lung cancer risk in this population.

Abstract:The lung cancer rate among never-smokers is the highest among Asian women, however its etiology and any relevant non-smoking related biomarkers are still unclear. Pre-diagnostic lung cancer-related metabolic biomarkers may provide novel insights into lung cancer mechanisms, and may contribute to the discovery of etiologic factors for the high lung cancer prevalence among Asian women. We evaluated the role of the urinary metabolome in lung cancer development among female never-smokers in China by conducting a nested case-control study of 275 lung cancer cases and 289 healthy controls from the Shanghai Women9s Health Study, a prospective cohort comprised of 73,363 Chinese female never-smokers. Metabolic profiling of urinary chemical features was conducted using ultrahigh-performance liquid chromatography - tandem mass spectrometry (UPLC-MS) (39,409 spectral features) and 600 MHz 1 H nuclear magnetic resonance (NMR) spectroscopy (386 features). Unconditional logistic regression models were used to estimate the odds ratios (ORs) and 95% confidence intervals (CIs) for the association between each log-transformed metabolite level and lung cancer risk, adjusting for potential confounders such as age, body mass index, history of respiratory disease and passive smoking. Spearman correlation and linear regression were used to estimate associations between the most significant metabolites and pre-diagnosis dietary factors. Abstract:What's new? Environmental, occupational, lifestyle, and genetic factors have been implicated in lung cancer in never‐smokers. However, the exact etiology remains unclear. Within the Prospective Shanghai Women's Health Study, the authors conducted a nested case‐control study involving 395 never‐smoking women with lung cancer and 395 matched never‐scancer‐free individuals. A network of 121 metabolic features, notably including lyso‐glycerophospholipids, showed inverse associations with future risks of all lung cancers and lung adenocarcinoma. The features were enriched in pathways related to oxidative stress and nucleic acid and energy metabolism. Untargeted metabolomics in pre‐diagnostic samples can offer novel insights into lung cancer etiology in never‐smokers. The etiology of lung cancer in never‐smokers remains elusive, despite 15% of lung cancer cases in men and 53% in women worldwide being unrelated to smoking. Here, we aimed to enhance our understanding of lung cancer pathogenesis among never‐smokers using untargeted metabolomics. This nested case‐control study included 395 never‐smoking women who developed lung cancer and 395 matched never‐smoking cancer‐free women from the prospective Shanghai Women's Health Study with 15,353 metabolic features quantified in pre‐diagnostic plasma using liquid chromatography high‐resolution mass spectrometry. Recognizing that metabolites often correlate and seldom act independently in biological processes, we utilized a weighted correlation network analysis to agnostically construct 28 network modules of correlated metabolites. Abstract:Despite the fact that we have long understood that environmental exposures influence health, policies have been slow to reduce or eliminate those exposures. In an attempt to mitigate ongoing influences on health by environments, researchers are seeking molecular mechanisms that link exposure to outcome, with the goal of identifying targetable pathways and providing compelling evidence to drive policy change. Cigarette smoking in particular has a long list of adverse health effects, but the success of policies aimed at reducing exposure are complicated by tobacco dependence. Identification of specific genes or cellular functions that are dysregulated by cigarette smoke in physiologically relevant tissues both provides increased evidence for health impact and could provide the basis for novel treatments for those who struggle to stop smoking.
