Abstract:DNA methylation discrimination is often challenged by complicated pretreatment, insufficient sensitivity, and suboptimal accuracy. Here, single-molecule readout of DNA methylation is reported using single-layer MoS2 nanopores. By tuning pore dimension, the sensitivity of MoS2 nanopores is manipulated, empowering both labeling and labeling-free strategies for DNA methylation discrimination. With methyl-CpG-binding domain protein 1 (MBD1)-labeled methylated DNA translocation in customized nanopores, multiple methylated sites with distance as short as 70 bp in double strand DNA can be resolved. To further improve spatial resolution, small MoS2 nanopores are engineered with single-nucleotide sensitivity, realizing labeling-free methylation detection with single-nucleotide resolution to recognize two nucleotides with only one methyl difference. This study demonstrates the availability of engineered MoS2 nanopores in DNA methylation detection, underscoring their potential for epigenetic alteration research at the single-molecule level.

Abstract:Atomically thin nanopore membranes are considered to be a promising approach to achieve single base resolution with the ultimate aim of rapid and cheap DNA sequencing. Molybdenum disulfide (MoS2) is newly emerging as a material complementary to graphene due to its semiconductive nature and other interesting physical properties that can enable a wide range of potential sensing and nanoelectronics applications. Here, we demonstrate that monolayer or few-layer thick exfoliated MoS2 with subnanometer thickness can be transferred and suspended on a predesigned location on the 20 nm thick SiNx membranes. Nanopores in MoS2 are further sculpted with variable sizes using a transmission electron microscope (TEM) to drill through suspended portions of the MoS2 membrane. Various types of double-stranded (ds) DNA with different lengths and conformations are translocated through such a novel architecture, showing improved sensitivity (signal-to-noise ratio>10) compared to the conventional silicon nitride (SiNx) nanopores with tens of nanometers thickness. Unlike graphene nanopores, no special surface treatment is needed to avoid hydrophobic interaction between DNA and the surface. Our results imply that MoS2 membranes with nanopore can complement graphene nanopore membranes and offer potentially better performance in transverse detection.

Abstract:We report a novel MoS2-based fluorescent biosensor for DNA detection via hybridization chain reactions (HCRs). As an emerging nanomaterial, MoS2 has excellent fluorescence quenching ability and distinct adsorption properties toward single- and double-stranded DNA. In the sensing method, MoS2 nanosheets are used to suppress the background signal and control the "on" and "off" of fluorescence emission of the detection system with and without the presence of the target DNA. In addition, the signal generation is amplified through the target-triggered HCRs between two hairpin probes. The employment of MoS2 and HCRs guarantees the high sensitivity of the detection strategy with a detection limit of 15 pM. The biosensor also exhibits very good selectivity over mismatched DNA sequences. The detection takes place in solutions and requires only one "mix-and-detect" step. The high sensitivity, selectivity, and operational simplicity demonstrate that MoS2 can be a promising nanomaterial for versatile biosensing.

Abstract:MoS2 nanopores have recently been demonstrated to be capable of detecting translocation of proteins. In this work, we further investigate the effect of MoS2 nanopore size on the translocation process. The MoS2 nanopores are fabricated using TEM drilling that yields nanopore diameters of similar to 10 nm and similar to 20 nm. We use these nanopores to detect bovine serum albumin (BSA) proteins and analyze the translocation signal statistics as well as the shape of individual signals. It is found that larger MoS2 nanopores can reduce the threshold voltage for translocation and increase the translocation event rate. Translocation at reduced bias voltages may prolong individual translocation process, which can be beneficial for improving the sensing resolution.

Abstract:DNA-functionalized layered two-dimensional transition-metal dichalcogenides have attracted tremendous interest for constructing biosensors in recent years. In this work, we report diblock molecular beacons with poly-cytosine (polyC) tails anchored on molybdenum disulfide (MoS2) nanosheets as probes for microRNA detection. The polyC block is adsorbed on MoS2 and the molecular beacon block is available for hybridization to the target; duplex-specific nuclease provides signal amplification by target recycling. By changing the length of polyC, we regulate the density of probes on MoS2 and inhibit the adsorption of enzyme-cleaved oligonucleotides, thereby leading to higher quenching efficiency. PolyC-mediated molecular beacons on MoS2 have very low background signal, ultrahigh sensitivity (limit of detection ∼3.4 fM), specificity to detect a single nucleotide mismatch, and selectivity to detect target microRNA from serum samples. This detection platform holds great potential for quantitative analysis of miRNAs in clinical diagnosis and biomedical research.

Abstract:Two-dimensional layered transition-metal dichalcogenide nanosheets have shown great potential in biosensors owing to their unique properties. Here, we exfoliated ultrathin metallic and semiconductive MoS2 nanosheets based on a chemical exfoliation method. We compared the difference of fluorescence quenching efficiency between metallic and semiconductive MoS2 nanosheets. We found that the fluorescence quenching efficiency of MoS2 nanosheets is phase-dependent. The ultrathin metallic MoS2 nanosheets with larger contents of a 1T-phase structure show higher fluorescence quenching efficiency than semiconductive MoS2 nanosheets, which can be ascribed to the higher conductivity of metallic MoS2 nanosheets. On the basis of the excellent fluorescence quenching efficiency of metallic MoS2 nanosheets and their discriminative adsorption toward single-strand DNA and double-strand DNA, a fluorescent biosensor for multiplex detection of DNA was developed. This fluorescent biosensing platform allows simultaneous fluorescence quenching of two single-strand DNA probes labeled with different fluorophores, resulting in multiplex detection of different DNA sequences in one homogeneous solution with high sensitivity.

Abstract:MicroRNAs (miRNAs) play significant regulatory roles in physiologic and pathologic processes and are considered as important biomarkers for disease diagnostics and therapeutics. Simple, fast, sensitive, and selective detection of miRNAs, however, is challenged by their short length, low abundance, susceptibility to degradation, and homogenous sequence. Here, we report a novel design of nanoprobes for highly sensitive and selective detection of miRNAs based on MoS2-loaded molecular beacons (MBs) and duplex-specific nuclease (DSN)-mediated signal amplification (DSNMSA). We show that MoS2 nanosheets not only exhibit high affinity toward MBs but also act as an efficient quencher for absorbed MBs. The strong fluorescence-quenching ability of MoS2 in combination with cyclic DSNMSA contributes to the superior sensitivity of our method, with a limit of detection 4 orders of magnitude lower than that of traditional hybridization methods. Moreover, the nanoprobes also show high selectivity for discriminating homogenous miRNA sequences with one-base differences because of the discrimination ability of MBs and DSN. Furthermore, we demonstrate that the MoS2-loaded MB nanoprobes can be utilized for multiplexed detection of miRNAs. Given its high sensitivity and specificity, as well as the multiplexed function; this novel method as an effective tool shows a great promise for simultaneous quantitative analysis of multiple miRNAs in biomedical research and clinical diagnosis.
Size‐Dependent Optical Absorption of Layered MoS2 and DNA Oligonucleotides Induced Dispersion Behavior for Label‐Free Detection of Single‐Nucleotide Polymorphism

Abstract:Size‐dependent optical absorption of semiconductive (2H) layered molybdenum disulfide (MoS2), exhibiting great discrimination abilities to single‐ and double‐stranded DNA (ssDNA) and (dsDNA), is studied. In the presence of high concentration of salt, layered MoS2 trends to aggregate rapidly, leading to the increases of sizes in both vertical and lateral dimensions of the nanosheets, which results from the interplay between van der Waals attraction and electrical double‐layer repulsion. Meanwhile, the aggregation behavior of layered MoS2 is remarkably inhibited by the synergistic effects of DNA oligonucleotides. ssDNA can adsorb on the surface of layered MoS2, resulting in a great dispersion, even in the presence of high concentration of salt, while the dispersion behavior is weakened when ssDNA is replaced by dsDNA. Whereas compared to graphene with zero bandgap energy, layered MoS2, with semiconductive properties, exhibits great characteristic optical absorption in visible wavelength region devoted to exploring the aggregation behavior of layered MoS2. Therefore, DNA oligonucleotides induced size control of layered MoS2, contributing to the regular change of its characteristic absorption in visible region, is considered a label‐free bioassay for the detection of single‐nucleotide polymorphism. Due to its easy operation and high specificity, it is expected that the proposed assay holds great promise for further applications.

Abstract:The use of nanoprobes in sensors is a popular way to amplify their analytical performance. Coupled with two-dimensional nanomaterials, nanoprobes have been widely used to construct fluorescence, electrochemical, electrochemiluminescence (ECL), colorimetric, surface enhanced Raman scattering (SERS) and surface plasmon resonance (SPR) sensors for target molecules’ detection due to their extraordinary signal amplification effect. The MoS2 nanosheet is an emerging layered nanomaterial with excellent chemical and physical properties, which has been considered as an ideal supporting substrate to design nanoprobes for the construction of sensors. Herein, the development and application of molybdenum disulfide (MoS2)-based nanoprobes is reviewed. First, the preparation principle of MoS2-based nanoprobes was introduced. Second, the sensing application of MoS2-based nanoprobes was summarized. Finally, the prospect and challenge of MoS2-based nanoprobes in future were discussed.

Abstract:Transition metal ions, such as those generated through MoS2 material, possess an intrinsic fluorescence quenching property towards organic dye molecules; thus, they can be used to construct biosensors as quenchers. However, we found that the conventional bulk MoS2 blocks the view of fluorescence imaging, and is incapable of tracing and visualizing mucin 1-overexpression cancer cells. Herein, a FAM fluorophore-labeled ssDNA fluorescent probe (P-0-FAM) stacked on the surface of MoS2 quantum dots (QDs) was used to construct a MoS2 QDs-P-0-FAM biosensor. MoS2 QDs exhibit a high fluorescence quenching ability towards fluorescent dyes, possess large specific surface area and a large number of active sites to adsorb and quench more fluorescent probes, promoting sensitivity between quenching and the recovery signal. In addition, the lighter color of unstack-MoS2 QDs is beneficial to define the location of cancer cells compared to MoS2 nanosheets. The novel MoS2 QDs-based biosensor demonstrates high sensitivity to MUC1 with a detection limit of 0.5 nM, and may become an important tool toward the detection of cancer cells.

Abstract:Recently, molybdenum disulfide (MoS2) has attracted considerable attention in the field of biomolecular detection owing to its large surface area and remarkable optoelectronic properties. Here, we report on a novel Au-modified monolayer MoS2 sensor that allows rapid, sensitive, and selective detection of DNA molecules. The Au-thiol bond can effectively enhance the DNA adsorption on MoS2. With the increasing concentration of DNA solution, the photoluminescence peak shows a prominent blueshift induced by the decrease of dielectric constant around MoS2. The monolayer MoS2 nanosheet exhibits different photoluminescence properties toward single-strained DNA versus double strained DNA. Thus, the complementary target DNA could be distinguished from mismatched DNA through the photoluminescence spectra of MoS2. The density functional theory calculation of MoS2/DNA systems was performed to explore the detection mechanism. This work could promote the research of novel sensing platform by coupling nanomaterials with biomolecular recognition events.

Abstract:The combination of nanomaterials and DNA has captured extensive attention in the fields of sensor, catalysis, biomedicine and material science. The applications of DNA functionalized MoS2 nanosheets (NSs) in biosensor and biomedicine in turn present an fascinating topic about biointerface science between DNA and MoS2 NSs. The surface interaction between DNA and MoS2 NSs was explored by adsorption and desorption experiments. The results indicated that van der waals force played an crucial role in DNA adsorption by MoS2 NSs. Additionally, it was proved that the quenching of dye labelled DNA probe by MoS2 NSs was attributed to fluorescence resonance energy transfer. Based on the extraordinarily high quenching efficiency of MoS2 NSs and different adsorption affinity of MoS2 NSs toward aptamer and aptamer-targeted protein, an aptameric biosensor was fabricated to detect carbohydrate antigen 15-3. The fabricated biosensor not only displayed high sensitivity with the detection limit of 0.0039 U/mL (S/N = 3), but also exhibited good stability and selectivity. The proposed sensing platform provided an easy-constructed and cost-effective protocol to detect other aptamer targeted biomolecules. The fundamental study of biointerface science on DNA and MoS2 NSs will facilitate their potential applications in various fields.

Abstract:In this work, we use for the first time single layer MoS2 as the fluorescence quencher to design a detection method for Ag(+) with excellent robustness, selectivity and sensitivity. To maintain the ultrathin MoS2, bulk MoS2 materials have been exfoliated by intercalation with lithium followed by reaction with water. As-prepared two-dimensional MoS2 not only has good water-solubility but also obtains high fluorescence quenching efficiency within 5 min. Importantly, the detection limit of this assay for Ag(+) (1 nM) was lower than the maximum limitation guided by the United States Environmental Protection Agency (EPA) and the World Health Organization (WHO). Further, this new Ag(+) probe was demonstrated in monitoring Ag(+) in lake water samples with satisfactory results.

Abstract:A new MoS2 nanosheet-based fluorescent biosensor for protein detection is developed. This method combines the terminal protection of small-molecule-linked DNA (TPSMLD) and exonuclease III (Exo III)-aided DNA recycling amplification to convert protein assay into the highly sensitive detection of DNA. Taking the streptavidin (SA)-biotin system as a model, a detection limit of 0.67ngmL−1 SA is obtained with a good selectivity. The study demonstrated here not only offers simple, sensitive and selective detection method for protein assay, but also will expand the application of the emerging 2D nanomaterials into biological assay.

Abstract:A novel 2-dimensional graphene analog molybdenum disulfide/multi-walled carbon nanotube (MoS2/MWCNT) was synthesized by a simple hydrothermal method to achieve excellent electrochemical properties. An ultrasensitive electrochemical DNA biosensor was subsequently constructed by assembling a thiol-tagged DNA probe on a MoS2/MWCNT and gold nanoparticle (AuNP)-modified electrode that has already been coupled with glucose oxidase (GOD). In this work, GOD was used as a redox marker. The heteronanostructure formed on the biosensor surface appeared relatively good conductor for accelerating the electron transfer, while the modification of GOD and AuNPs provided multiple signal amplification for electrochemical biosensing. The multiple signal amplification strategy produced an ultrasensitive electrochemical detection of DNA down to 0.79fM with a linear range from 10fM to 107fM, and appeared high selectivity to differentiate three-base mismatched DNA and one-base mismatched DNA. The developed approach provided a simple and reliable method for DNA detection with high sensitivity and specificity, and would open new opportunities for sensitive detection of other biorecognition events.
