Abstract:Highly sensitive and selectivite detection of copper ions (Cu2+) and hydrogen sulfide (H2S) have become important research topics due to the potential harmful impacts of these chemicals to human health and the environment. In this study, we report the synthesis of a dual-functional peptide-based probe L (FITC-AhxSerSerHis), designed to mimic a copper-sulfur metalloprotein, and capable of continuous detection of Cu2+ and S2- based on colorimetric and fluorescent methods. The new probe L displayed excellent "turn off" fluorescence response and good selectivity for Cu2+ ions via a modification of the tripeptide and fluorescein isothiocyanate group, and produced an obvious color change visible to the naked eye. Furthermore, as an excitable probe, the L-Cu complex could continuously detect S2- with high selectivity and sensitivity in 100% aqueous buffered solutions. The detection limits for fluorescence titration measurements, calculated using the equation 3σ/k, were 76.7 nM (Cu2+) and 27.2 nM (S2-), which were well below U.S. EPA safety levels. In addition, L could be cycled to alternately detect Cu2+ and S2-, thereby making it a promising reversible probe. Moreover, L was successfully applied to monitoring Cu2+ and S2- in live RKO cells through fluorescence imaging, exhibiting low cytotoxicity and good cell permeability.

Abstract:The detection of copper ions and hydrogen sulfide with high sensitivity and selectivity has become a research priority as they play a crucial role in physiopathological processes both in the human body and in the environment. Herein, a fluorescent chemosensor named FGP1 (FITC-Ahx-Ser-Ser-His-Thr-Glu-Phe-NH2) was developed using novel peptides. This chemosensor solely recognizes Cu2+ without any disruption by other metal ions that may compete. The binding of FGP1-Cu leads to the quenching of FGP1 probe fluorescence with great sensitivity and rapid response time, resulting in a noticeable change of color observable to the naked eye. The association constant of FGP1-Cu is 7.34 × 105 M-1, thus when S2- is added to FGP1-Cu complexes, Cu2+ is removed and fluorescence is restored, resulting in the "off-on" process. The fluorescence titration measurements' detection limits, computed utilizing the 3σ/k equation, were 38.2 nM (Cu2+) and 37.9 nM (S2-). These values are considerably lower than the safe levels designated by the US EPA. Furthermore, on account of its favorable water solubility, high biocompatibility and low toxicity, FGP1 probe proved effective in tracking Cu2+ and S2- in Hela living cells using the fluorescence imaging technique.

Abstract:Cu2+ and S2− play important roles in our environment, so it is beneficial to develop sensors capable of detecting Cu2+ and S2− in biological samples. To accomplish this goal, a new peptide-based fluorescent chemical sensor L (FITC-Ahx-His-Glu-Phe-Cys-NH2) has been designed and synthesized. It can specifically recognize Cu2+ by forming a L-Cu complex which can quench the fluorescence of L. The subsequent addition of S2− can remove Cu2+ from the L-Cu complex, thereby restoring the fluorescence in a process described as an “On-Off-On” cycle. Cu2+ and S2− can be detected by L with extremely high sensitivity and the detection limits is 32 nM and 53 nM respectively. Additionally, the sensor L can detect Cu2+ and S2− at physiological pH conditions. The result of this research is the development of a peptide-based fluorescent chemical sensor L, which can specifically detect Cu2+ and S2− in the environment.

Abstract:Copper ions (Cu2+) and hydrogen sulfide (H2S) detection is of great significance in environmental monitoring and disease screening. Herein, a novel fluorescent probe (DAHK) was rationally designed and easily synthesized based on a copper peptide backbone labelled a dansyl fluorophore. DAHK exhibited large Stokes shift (210 nm), high sensitivity (38.9 nM) and high selectivity response to Cu2+ ions based on obvious fluorescence quenching in HEPES buffer (10.0 mM, pH 7.4) solutions. In addition, the in situ formed DAHK-Cu2+ ensemble was successfully used as a secondary probe for rapidly and sequentially detected H2S via fluorescence “off–on” response, and without interference by a range of other anions. The limit of detection (LOD) for 47.3 nM was determined, which was a much lower value than WHO and EPA guidelines (H2S) for drinking water. The reversibility of interaction of DAHK with Cu2+ and H2S was monitored using fluorescence emission spectra. Cells imaging experiments exhibited that DAHK had good membrane permeability and excellent low toxicity. Moreover, DAHK was successfully made into fluorescent test strips for visual detection and rapid analysis based on the exciting stability and excellent water solubility.

Abstract:In this study, a novel peptide-based fluorescent and colorimetric dual-functional probe (FAHK) was designed and synthesized by simulating the structure of copper peptide. FAHK exhibited high selectivity and rapid response to Cu2+ and Hg2+ with two non-fluorescence ensembles FAHK-Cu2+ and FAHK-Hg2+ formation by the 2:1 coordination mode, and the limits of detection for Cu2+ and Hg2+ were 37.1 nM and 20.8 nM, respectively. FAHK showed a naked eye colorimetric response for Cu2+ and Hg2+ in an aqueous solutions under visible light. As two new promising cascade probes, FAHK-Cu2+ and FAHK-Hg2+ ensemble were used for sequential selective recognition of S2− according to the displacement approach with fluorescence “turn-on” response and colorimetric change, and the limit of detection were 0.27 μM (FAHK-Cu2+ ensemble) and 0.12 μM (FAHK-Hg2+ ensemble) for S2−, which were well below the safety level the H2S standard in drinking waters (14.8 μM, WHO guidelines). What’ more, in view of low toxicity and biocompatibility, the potential application of FAHK in biological systems was demonstrated via living cells imaging. More importantly, FAHK could also be made into the paper test strips for visual detection of Cu2+, Hg2+ and S2− with the naked eye under 365 nm UV light and visible light, which suggested that better portability and operation simplicity that is favourable for on-site analysis.

Abstract:1. By applying the soft and hard acids and bases (HSAB) theory to amino acids, a novel peptide chemical sensor L has been designed that can simultaneously detect Zn2+, Cu2+, and S2−. 2. L exhibits minimal biological toxicity and holds potential as a sensor for detecting Zn2+, Cu2+, and S2− in biological systems. A novel peptide‐based chemical fluorescence sensor L (Dansyl‐His‐Pro‐Thr‐Cys‐NH2) was designed and synthesized. This sensor exhibits an "On–Off–On" detection cycle to detect Cu2+, Zn2+, and S2− in solution. According to the chelation‐enhanced fluorescence (CHEF) mechanism, when Zn2+ is present, the fluorescence is significantly enhanced and a blue shift occurs, representing a "Turn‐On" phase of the fluorescence detection mode. Because copper ions (Cu2+) have a paramagnetic quenching sensing mechanism, the fluorescence of L quenches rapidly with the formation of the L‐Cu system, representing the "Turn‐Off" phase. The subsequent introduction of S2− to the L‐Cu system results in the recovery of the L‐fluorescence, thereby representing the second "Turn‐On" phase. As a peptide molecule, the sensor L has several advantages over other types of sensors, including water solubility, high sensitivity, and good biocompatibility, with a very low detection limit. The detection lines of Zn2+ and Cu2+ are 97 nM (R = 0.993) and 75 nM (R = 0.995), respectively. Additionally, the sensor does not exhibit any obvious cell toxicity. These results indicate that this peptide chemiluminescent sensor has the potential to be applied in in vivo detection.

Abstract:L-cysteine (L-Cys) capped ZnS fluorescent probe (L-ZnS) were synthesized by binding ZnS nanoparticles in situ with L-Cys, the fluorescence intensity of L-ZnS increased more than 3.5 times than that of ZnS due to the cleavage of S-H bonds and the formation of Zn-S bonds between the thiol group of L-Cys and ZnS. The addition of copper ions (Cu2+) can effectively quench the fluorescence of L-ZnS to realize the rapid detection of trace Cu2+. The L-ZnS showed high sensitivity and selectivity to Cu2+. The LOD (limit of detection) of Cu2+ was as low as 7.28 nM and linearity in the concentration range of 3.5-25.5 μM. Meanwhile, for the first time, electron localization function (ELF), bond order density (BOD), and natural adaptive orbital (NAdO) analysis in the Multiwfn wavefunction program based on density functional theory were carried out to probe the binding sites and binding mode of L-Cys with Cu2+, it indicated that the deprotonated carboxyl oxygen atoms of L-Cys had the lowest electrostatic potential (ESP) and provided lone pair electrons to coordinate with Cu2+ to form non-luminescent ground state complexes, which led to fluorescence quenching of L-ZnS. From the microscopic point of view of atoms, the mechanism of fluorescence enhancement of L-Cys capped ZnS and the mechanism of fluorescence quenching after adding Cu2+ were revealed in depth, the theoretical analysis results were accordance with the experiments.

Abstract:Fluorescent sensor has demonstrated to be a facile and effective method to detect heavy metal ions owing to its unique characteristics such as simplicity, ease of operation and cost-effectiveness. In this work, a novel fluorescent probe with aggregation-induced emission (AIE) feature that contains an amide ligand base on the tetraphenylethylene (TPE) dye was designed and successfully synthesized. The utilization of the AIE-active fluorescent probe (named as FDPA) for detection of Cu2+ in aqueous solution has also been examined. We demonstrated that the resultant AIE-active molecule displays amphiphilic property and can self-assemble in aqueous solution with remarkable enhancement of fluorescent intensity owing to its AIE feature. The detection limit of probe for Cu2+ determination is 6.11 × 10-9 M and shows excellent selectivity in the presence of competitive ions. This work provides a useful route to overcome the fluorescence quenching of conventional fluorescent probes in aqueous solution and an elegant way to prepare fluorescent probes with better fluorescence properties.

Abstract:We report a new fluorescent probe named DGGH with dansyl-labeled tripeptide (Dan-Gly-Gly-His-NH2). DGGH showed obvious fluorescence quenching after the addition of Cu2+ ions and a 1:1 ensemble system (DGGH-Cu2+) was formed, the limit of detection was 37.9 nM. As expected, DGGH-Cu2+ ensemble system exhibited high selectivity and rapid response for S2− in 100% aqueous solutions, as a result of the addition of S2− immediately replaced Cu2+ from the DGGH-Cu2+ ensemble system. The limit of detection of the DGGH-Cu2+ ensemble for S2− was determined to be 13.7 nM. In addition, DGGH-Cu2+ has excellent detection performance within a wide pH range of 6 to 12, and its outstanding response time of less than 50s. Furthermore, we demonstrated the usability of the present approach for detecting Cu2+ and S2− in two water samples. What's more, DGGH was successfully applied to monitor Cu2+ and S2− in living LNcap cells based on good cells permeability and low toxicity.

Abstract:A novel highly selective and sensitive fluorescent probe (NJ1) had been designed and synthesized for Cu2+ detection by fluorescence quenching mechanism, and then the enhancement of fluorescence intensity with the addition of hydrogen sulfide in complex NJ1Cu aqueous solution at physiological conditions were described. This "on-off-on" type fluorescence recognition system was a reversible process, which could be utilized to monitor copper ion and hydrogen sulfide based on a complex Nj1Cu formation-Cu2+ displacement approach. Importantly, this real-time recognition process of Cu2+ and hydrogen sulfide exhibited excellent anti-interference ability. In addition, this new fluorescent probe also has potential utility for Cu2+ and hydrogen sulfide detection in living cells, providing a potential tool for investigating copper ion and hydrogen sulfide in living systems or environment. Abstract:A simple amide compound (QLBA), prepared from quinoline-2-carboxylic acid and 2-(1H-benzo[d]imidazol-2-yl)aniline, was synthesized and evaluated as an efficient fluorescence chemosensor for the selective recognition of copper ions (Cu2+). This sensor could detect Cu2+ in CH3OH/HEPPES buffer in a wide pH range (4.0-10.0), with obvious fluorescence quenching. The obtained QLBA-Cu2+ complex could be used as a new cascade sensor for detecting sulfide anions (S2-). The corresponding detection limits were found to be 2.2 x 10(-7) M for Cu2+ and 4.6 x 10(-7) M for S2-. The sensing mechanisms of QLBA toward Cu2+ were systematically investigated in detail by NMR, HR-MS analysis, and density functional theory (DFT) calculations. In addition, this sensor was verified to be of low cytotoxicity and to have good imaging characteristics for the detection of Cu2+ and further for the recognition of S2- in living cells, suggesting that QLBA was a useful tool for tracking Cu2+ and S2- ions in vivo.

Abstract:A new dual-mode ratiometric fluorescence and colorimetric probe for selective determination of Cu2+ was developed based on blue-emission sulfur quantum dots (SQDs) and yellow-emission carbon quantum dots (CQDs). The fluorescence and absorbance of CQDs increased in the presence of Cu2+ due to the Cu2+ -oxidized o-phenylenediamine group on the surface of the CQDs. Because of the inner filter effect between SQDs and CQDs-Cu2+, the fluorescence response of SQDs decreased following the introduction of Cu2+. Furthermore, in the presence of Cu2+, the dual-mode SQD–CQD probe showed visible color changes under both ultraviolet light and sunlight. Under optimal conditions, the dual-mode probe was used to quantitatively detect Cu2+ with a linear range of 0.1–5.0 μM for ratiometric fluorescence and colorimetry, with a limit of detection of about 31 nM and 47 nM, respectively. Finally, the dual-mode probe was used for the determination of Cu2+ in practical samples to expand the practical application, and the difference between ratiometric fluorescence and colorimetric methods was compared. The recovery results confirmed the high accuracy of the dual-mode probe, showing that it has immense potential for sensitive and selective detection of Cu2+ in practical samples.Graphical abstract

Abstract:The fluorescent probe has become an important method for the detection of heavy metal ions. In the present work, a new and simple fluorescent probe, Cu-P, for detecting copper ion (Cu^2+) was designed and synthesized. The probe has shown high sensitivity and selectivity toward Cu^2+. The detection limit was 13 nM (based on the 3o^-/slope). A significant color change from yellow to pink was observed; thus, the probe Cu-P could serve as a “naked-eye” indicator for Cu^2+. Furthermore, the proposed probe was used to detect Cu^2+ in real water and soil extract samples, with the result being satisfactory. Therefore, our proposed probe would provide a promising method for the detection of Cu^2+ in the environment.

Abstract:A new fluorometric and colorimetric dual-modalitye probe L for detection of Cu 2+ and L -His was constructed based on peptide skeleton via solid phase peptide synthesis (SPPS) technology. L exhibited highly selective recognition towards Cu 2+ through formation of L -Cu 2+ ensemble with a 2:1 binding stoichiometry, and the limit of detection (LOD) for Cu 2+ was determined to be 7.1 nM. Correspondingly, L -Cu 2+ ensemble was investigated by fluorescence technique which clearly established the high selectivity towards L -His based on fluorescence "off-on" response, and the limit of detection (LOD) for L -His was calculated to be approximately 15.3 nM. The selective sensing of Cu 2+ was efficiently reversible with L -His, which was no loss of fluorescence intensity in 10 cycles. Notably, the sensing behavior of L towards Cu 2+ and L -His detections was shown to be available within the wider pH interval (7.0-12.0). Cells imaging studies have shown that L was easily applied to the continuous imaging of Cu 2+ and L -His in living cells. Furthermore, the probe L -loaded test strips were also prepared for rapid and sensitive identification of Cu 2+ and L -His. Combining smartphones and color changes, the semi-quantitative relationship between the R/B and the concentration of Cu 2+ was established, and the limit of detection (LOD) reached up to 0.29 μM.

Abstract:Hydrogen sulfide (H2S) plays a vital role in biological systems, so it is important to develop new methods for monitoring H2S with high selectively and sensitivity. In this study, we report the efficient solid phase synthesis of a new peptide-based bioprobe L that mimics the Cu(II) binding sites of ceruloplasmin. L exhibits highly sensitive colorimetric and fluorescent "turn off" responses to Cu2+ with high selectively among various metal ions over a broad pH range in 100% aqueous solutions. Interestingly, as a new promising analytical bioprobe, the L-Cu ensemble can monitor H2S by both colorimetric and fluorescent changes, and the limit of detection (LOD) is found to be as low as 14.7 nM. The reversibility experiment indicated that L could be recycled many times as reversible bioprobe. It is worth noting that L could be used to detect Cu2+ and H2S specifically in two real samples. Moreover, L exhibited excellent water-solubility and low biotoxicity even at high concentrations based on MTT study, and cell imaging results indicated that it can be applied to detecting Cu(II) and H2S in living RKO cells monitored by confocal fluorescence microscopy imaging.

Abstract:Copper ions (Cu 2+ ) and sulfide (S 2− ) play essential roles in many physiologies and pathologic processes. Herein, a new "on–off-on" tryptanthrin-based probe TR-1 (TR-1) has been designed and synthesized in a facile and economical way. TR-1 exhibited highly selective and sensitive response to Cu 2+ without any interference over 14 competitive metal ions and the detection limit downs to 24 nM, which is far below the Chinese standard of fishery water quality (157 nM). The 'in situ' prepared complex TR-1 + Cu 2+ could also be applied to detect S 2− with the detection limit of 62 nM. Further, TR-1 was potentially applied for the analysis of copper ions in water samples.

Abstract:A novel salamo-based fluorescent chemical sensor l-Cu2+ was designed and prepared via the reaction of H2L and Cu(OAc)2·H2O. The whole research has found that Cu(II) atom in the complex crystal are located in the N2O2 cavity of H2L, forming a four-coordination system. Hirshfeld surface analysis confirmed the presence of hydrogen bonding and π···π stacking interactions. As a probe molecule, l-Cu2+ can selectively and sensitively detect S2- without interference from other ions, and can be effectively used within the biological pH range. In addition, DFT theoretical calculations and molecular electrostatic potential analysis (ESP) were used to investigate the electron cloud density and positive and negative electrostatic potentials inside the probe, as well as the mechanism for identifying sulfur ions. Finally, the probe l-Cu2+ has shown good practical application effects and can recognize S2− in three different water samples. It can also be used as a qualitative detection strip. l-Cu2+ complex possesses excellent application prospect in bacteriostasis.

Abstract:A fluorescent probe based on a glycyl-L-tyrosine-modified dansyl derivative (D-GT) is designed and synthesized. D-GT demonstrated great detection performance toward Cu2+ in an aqueous solution. Fluorescence quenching occurred due to the coordination of Cu2+ with D-GT. The sensitive detection of D-GT to Cu2+ was applied in aqueous solution within a wide pH span (6-12). A 1:1 coordinate stoichiometric way and an association constant of 6.47 × 104 M-1 between D-GT and Cu2+ were determined. The measured detection limit for Cu2+ in HEPES buffer solution (10 mM, pH 7.4) was 0.69 μM. The probe displayed an appropriate sensitivity toward Cu2+ in real drinking water samples and living cells, which reveals the potential applications of D-GT in complicated environments.

Abstract:A highly selective Schiff-type fluorescent probe (SY) with aggregation-induced emission (AIE) characteristics was synthesized based on salicylaldehyde derivatives. With increasing water content, it exhibits good sensitivity and interference resistance for the detection of Cu 2+ in mixed solvents (DMSO/H 2 O, v/ v = 3:2), and uses a fluorescence burst mechanism to rapidly analyze Cu 2+ ions in aqueous media with a detection limit as low as 3.98 × 10 −8 M, which is well below the permissible standard for Cu 2+ (∼20 μM) in drinking water (WHO). Importantly, the probe has been successfully applied to the determination of Cu 2+ in real water samples with good reversibility and recoveries ranging from 99.3% to 106.6% in tap water, and can also be used for the detection of Cu 2+ ions under 365 nm UV light. In addition, the coordination pattern of the probe with Cu 2+ was evaluated by mass spectrometry and working curves, and the stoichiometry of the probe and Cu 2+ ions was determined to be 2:1. In addition, density functional theory (DFT) was performed to help understand the electronic nature of SY and Cu 2+ complexation and chelation induced quenching mechanism.
