Abstract:Brain metastasis is a major cause of cancer mortality, but its molecular mechanisms are severely understudied. In addition, little is known regarding the role of m 6A reader YTHDF3 in human diseases. Here, we show that YTHDF3 overexpression clinically correlates with brain metastases in breast cancer patients. YTHDF3 promotes cancer cell interactions with brain endothelial cells and astrocytes, blood-brain barrier extravasation, angiogenesis, and outgrow. Mechanistically, YTHDF3 enhances the translation of m 6A-enriched transcripts for ST6GALNAC5, GJA1, and EGFR, all associated with brain metastasis. Furthermore, overexpression of YTHDF3 in brain metastases is attributed to increased gene copy number and the autoregulation of YTHDF3 cap-independent translation by binding to m 6A residues within its own 5′ UTR. Our work uncovers an essential role of YTHDF3 in controlling the interaction between cancer cells and brain microenvironment, thereby inducing brain metastatic competence.

What problem does this paper attempt to address?
This paper aims to explore the role of YTHDF3 in breast cancer brain metastases and its molecular mechanisms. Specifically, the researchers found that the over - expression of YTHDF3 is clinically related to brain metastases in breast cancer patients and is associated with a reduced patient survival rate. Mechanistically, YTHDF3 promotes the interaction between cancer cells and the brain microenvironment and the occurrence of brain metastases by enhancing the translation of m6A - modified gene transcripts. In addition, the study also revealed the self - regulatory mechanism of YTHDF3 in the process of brain metastases, that is, the increase in the YTHDF3 gene copy number and the non - cap - dependent translation regulation of m6A modification in its own 5'UTR region. ### Main research questions 1. **The relationship between the over - expression of YTHDF3 and breast cancer brain metastases**: By analyzing gene expression data in public databases, the researchers found that the expression of YTHDF3 in breast cancer brain metastases was significantly higher than that in primary tumors and was associated with a reduced patient survival rate. 2. **The mechanism of action of YTHDF3 in the process of brain metastases**: Through experimental verification, YTHDF3 promotes the adhesion, extravasation, invasion of cancer cells to brain endothelial cells and the interaction with astrocytes by enhancing the translation efficiency of m6A - modified gene transcripts (such as ST6GALNAC5, GJA1 and EGFR). 3. **The self - regulatory mechanism of YTHDF3 in brain metastases**: The study found that the over - expression of YTHDF3 in brain metastases is due to the increase in gene copy number and the non - cap - dependent translation regulation of m6A modification in its own 5'UTR region. ### Key findings - **Clinical relevance**: The high expression of YTHDF3 is significantly associated with breast cancer brain metastases and low survival rate. - **Functional verification**: By knocking down YTHDF3, the researchers observed a reduction in brain metastasis formation in mouse models and an extension of the overall survival time of mice. - **Molecular mechanism**: YTHDF3 promotes various interactions between cancer cells and the brain microenvironment, including endothelial cell adhesion, extravasation, invasion and angiogenesis, by enhancing the translation efficiency of m6A - modified gene transcripts. - **Self - regulatory mechanism**: The over - expression of YTHDF3 is due to the increase in gene copy number and the non - cap - dependent translation regulation of m6A modification in its own 5'UTR region. ### Conclusion YTHDF3 plays a crucial role in breast cancer brain metastases. It promotes various interactions between cancer cells and the brain microenvironment by enhancing the translation efficiency of m6A - modified gene transcripts. These findings provide a new perspective for understanding the molecular mechanisms of breast cancer brain metastases and may provide potential targets for the development of new treatment strategies.
