Abstract:Superoxide dismutases (SODs) are a group of metalloenzymes that catalyze the dismutation of superoxide radicals (O2˙-) into hydrogen peroxide (H2O2) and oxygen (O2). As the first line of defense against reactive oxygen species (ROS)-mediated damage, SODs are expected to play an important role in the treatment of oxidative stress-related diseases. However, the clinical applications of SODs have been severely limited by their structural instability and high cost. Compared with natural enzymes, nanozymes, nanomaterials with enzyme-like activity, are more stable, and economical, can be easily modified and their activities can be adjusted. Due to their excellent characteristics, nanozymes have attracted widespread attention in recent years and are expected to become effective substitutes for natural enzymes in many application fields. Importantly, some nanozymes with SOD-like activity have been developed and proved to have a mitigating effect on diseases caused by oxidative stress. These studies on SOD-like nanozymes provide a feasible strategy for breaking through the dilemma of SOD clinical applications. However, at present, the specific catalytic mechanism of SOD-like nanozymes is still unclear, and many important issues need to be resolved. Although there are many comprehensive reviews to introduce the overall situation of the nanozyme field, the research on SOD-like nanozymes still lacks a systematic review. From the structure and mechanism of natural SOD enzymes to the structure and regulation of SOD-like nanozymes, and then to the measurement and application of nanozymes, this review systematically summarizes the recent progress in SOD-like nanozymes. The existing shortcomings and possible future research hotspots in the development of SOD-like nanozymes are summarized and prospected. We hope that this review would provide ideas and inspirations for further research on the catalytic mechanism and rational design of SOD-like nanozymes.

Abstract:Superoxide dismutase (SOD) is a common antioxidant enzyme found majorly in living cells. The main physiological role of SOD is detoxification and maintain the redox balance, acts as a first line of defence against Reactive nitrogen species (RNS), Reactive oxygen species (ROS), and other such potentially hazardous molecules. SOD catalyses the conversion of superoxide anion free radicals (O 2 -.) into molecular oxygen (O 2) and hydrogen peroxide (H 2O 2) in the cells. Superoxide dismutases (SODs) are expressed in neurons and glial cells throughout the CNS both intracellularly and extracellularly. Endogenous oxidative stress (OS) linked with enlarged production of reactive oxygen metabolites (ROMs), inflammation, deregulation of redox balance, mitochondrial dysfunction and bioenergetic crisis are found to be prerequisite for neuronal loss in neurological diseases. Clinical and genetic studies indicate a direct correlation between mutations in SOD gene and neurodegenerative diseases, like Amyotrophic Lateral Sclerosis (ALS), Huntington's disease (HD), Parkinson's Disease (PD) and Alzheimer's Disease (AD). Therefore, inhibitors of OS are considered as an optimistic approach to prevent neuronal loss. SOD mimetics like Metalloporphyrin Mn (II)-cyclic polyamines, Nitroxides and Mn (III)- Salen complexes are designed and used as therapeutic extensively in the treatment of neurological disorders. SODs and SOD mimetics are promising future therapeutics in the field of various diseases with OS-mediated pathology.

Abstract:Abstract Nanozymes with superoxide dismutase (SOD)-like activity have attracted increasing interest due to their ability to scavenge superoxide anion, the origin of most reactive oxygen species in vivo. However, SOD nanozymes reported thus far have yet to approach the activity of natural enzymes. Here, we report a carbon dot (C-dot) SOD nanozyme with a catalytic activity of over 10,000 U/mg, comparable to that of natural enzymes. Through selected chemical modifications and theoretical calculations, we show that the SOD-like activity of C-dots relies on the hydroxyl and carboxyl groups for binding superoxide anions and the carbonyl groups conjugated with the π-system for electron transfer. Moreover, C-dot SOD nanozymes exhibit intrinsic targeting ability to oxidation-damaged cells and effectively protect neuron cells in the ischemic stroke male mice model. Together, our study sheds light on the structure-activity relationship of C-dot SOD nanozymes, and demonstrates their potential for treating of oxidation stress related diseases.

Abstract:Superoxide dismutase (SOD) is a class of enzymes that restrict the biological oxidant cluster enzyme system in the body, which can effectively respond to cellular oxidative stress, lipid metabolism, inflammation, and oxidation. Published studies have shown that SOD enzymes (SODs) could maintain a dynamic balance between the production and scavenging of biological oxidants in the body and prevent the toxic effects of free radicals, and have been shown to be effective in anti-tumor, anti-radiation, and anti-aging studies. This research summarizes the types, biological functions, and regulatory mechanisms of SODs, as well as their applications in medicine, food production, and cosmetic production. SODs have proven to be a useful tool in fighting disease, and mimetics and conjugates that report SODs have been developed successively to improve the effectiveness of SODs. There are still obstacles to solving the membrane permeability of SODs and the persistence of enzyme action, which is still a hot spot and difficulty in mining the effect of SODs and promoting their application in the future.

Abstract:The Cu,Zn superoxide dismutase (SOD1) is an ubiquitary cytosolic dimeric carbohydrate free molecule, belonging to a family of isoenzymes involved in the scavenger of superoxide anions. This effect certainly represents the main and well known function ascribed to this enzyme. Here we highlight new aspects of SOD1 physiology that point out some inedited effects of this enzyme in addition to the canonic role of oxygen radical enzymatic dismutation. In the last two decades our research group produced many data obtained in in vitro studies performed in many cellular lines, mainly neuroblastoma SK-N-BE cells, indicating that this enzyme is secreted either constitutively or after depolarization induced by high extracellular K+ concentration. In addition, we gave many experimental evidences showing that SOD1 is able to stimulate, through muscarinic M1 receptor, pathways involving ERK1/2, and AKT activation. These effects are accompanied with an intracellular calcium increase. In the last part of this review we describe researches that link deficient extracellular secretion of mutant SOD1G93A to its intracellular accumulation and toxicity in NSC-34 cells. Alternatively, SOD1G93A toxicity has been attributed to a decrease of Km for H2O2 with consequent OH radical formation. Interestingly, this last inedited effect of SOD1G93A could represent a gain of function that could be involved in the pathogenesis of familial Amyotrophic Lateral Sclerosis (fALS).

Abstract:Superoxide dismutases (SODs) are universal enzymes of organisms that live in the presence of oxygen. They catalyze the conversion of superoxide into oxygen and hydrogen peroxide. Superoxide anions are the intended product of dedicated signaling enzymes as well as the byproduct of several metabolic processes including mitochondrial respiration. Through their activity, SOD enzymes control the levels of a variety of reactive oxygen species (ROS) and reactive nitrogen species, thus both limiting the potential toxicity of these molecules and controlling broad aspects of cellular life that are regulated by their signaling functions. All aerobic organisms have multiple SOD proteins targeted to different cellular and subcellular locations, reflecting the slow diffusion and multiple sources of their substrate superoxide. This compartmentalization also points to the need for fine local control of ROS signaling and to the possibility for ROS to signal between compartments. In this review, we discuss studies in model organisms and humans, which reveal the dual roles of SOD enzymes in controlling damage and regulating signaling.

Abstract:Superoxide dismutases (SODs) are metalloenzymes that play a major role in antioxidant defense against oxidative stress in the body. SOD supplementation may therefore trigger the endogenous antioxidant machinery for the neutralization of free-radical excess and be used in a variety of pathological settings. This paper aimed to provide an extensive review of the possible uses of SODs in a range of pathological settings, as well as describe the current pitfalls and the delivery strategies that are in development to solve bioavailability issues. We carried out a PubMed query, using the keywords "SOD", "SOD mimetics", "SOD supplementation", which included papers published in the English language, between 2012 and 2020, on the potential therapeutic applications of SODs, including detoxification strategies. As highlighted in this paper, it can be argued that the generic antioxidant effects of SODs are beneficial under all tested conditions, from ocular and cardiovascular diseases to neurodegenerative disorders and metabolic diseases, including diabetes and its complications and obesity. However, it must be underlined that clinical evidence for its efficacy is limited and consequently, this efficacy is currently far from being demonstrated.

Abstract:Redox equilibria and the modulation of redox signalling play crucial roles in physiological processes. Overproduction of reactive oxygen species (ROS) disrupts the body’s antioxidant defence, compromising redox homeostasis and increasing oxidative stress, leading to the development of several diseases. Manganese superoxide dismutase (MnSOD) is a principal antioxidant enzyme that protects cells from oxidative damage by converting superoxide anion radicals to hydrogen peroxide and oxygen in mitochondria. Systematic studies have demonstrated that MnSOD plays an indispensable role in multiple diseases. This review focuses on preclinical evidence that describes the mechanisms of MnSOD in diseases accompanied with an imbalanced redox status, including fibrotic diseases, inflammation, diabetes, vascular diseases, neurodegenerative diseases, and cancer. The potential therapeutic effects of MnSOD activators and MnSOD mimetics are also discussed. Targeting this specific superoxide anion radical scavenger may be a clinically beneficial strategy, and understanding the therapeutic role of MnSOD may provide a positive insight into preventing and treating related diseases.

Tin Porphyrin-Based Nanozymes with Unprecedented Superoxide Dismutase-Mimicking Activities

Abstract:As the oxidative stress is related to human aging and many diseases, a diversity of antioxidant biomimetic enzymes to eliminate reactive oxygen species in vivo and maintain the redox balance has attracted intensive attention. Of particular interest are superoxide dismutase (SOD)-mimicking artificial enzymes that bear inherent characteristics of natural counterparts but overcome their deficiencies in thermal and acidic stability. Inspired by the metallized active center of natural SODs, here, we engineered different groups of metalloporphyrins and found that Sn-metallized porphyrins can act as novel SOD mimics, in which Sn-metallized meso-tetra(4-carboxyphenyl) porphine (Sn-TCPP) can more effectively catalyze the disproportionation of superoxide radical anions (•O2-) into hydrogen peroxide and oxygen. Especially, Sn-TCPP-based metal-organic frame nanozyme (Sn-PCN222) displays an unusually high catalytic activity that remarkably exceeds those of commonly used counterparts. Such unprecedented catalytic behaviors are proposed to depend on the Sn(IV)/Sn(II) transition at the center of Sn-TCPP. In addition, the metal-organic framework (MOF) nanozymes also display higher thermal and acidic stability than natural SODs. Interestingly, we find that Sn-complexed methylated tetra-(4-aminophenyl) porphyrin shows an aggregation-induced SOD activity in an acidic environment, whereas conventional SOD mimics do not function well in this case. Given these unique features, our reported Sn-porphyrin-based nanozymes would be potent alternatives for natural SODs to be widely used in clinical treatments of oxidative stress-related diseases.

Abstract:Oxidative stress emerges as a critical mechanism underlying the development and progression of various brain diseases, arising from either increased generation of reactive oxygen species (ROS) or dysfunction of the antioxidant defense system. Nanozymes, possessing enzymatic catalytic capabilities alongside the physicochemical characteristics of nanosized materials, have showcased extraordinary promise in regulating the production or scavenging of ROS. Consequently, nanozymes have been utilized to detect the basic level and dynamic changes of neurochemical biomarkers closely related to physiological and pathological brain conditions, as well as to mitigate oxidative damage and facilitate the penetration of the blood -brain barrier. As such, nanozymes have emerged as a promising tool for developing diagnostic and therapeutic approaches for various neurological disorders. This review offers a comprehensive overview of the catalytic mechanisms exhibited by nanozymes, which have demonstrated tremendous potential in the effective diagnosis and treatment of brain disorders. Through summarizing the emerging methods for applying nanozymes, this review aims to provide directions on the rational design of nanozymes for the theranostics of brain diseases. Additionally, this review seeks to address the current challenges and delineate future directions for the advancement of this field.

Abstract:Many short-lived and highly reactive oxygen species, such as superoxide anion (O2-) and hydrogen peroxide (H2O2), are toxic or can create oxidative stress in cells, a response involved in the pathogenesis of numerous diseases depending on their concentration, location, and cellular conditions. Superoxide dismutase (SOD) activities as an endogenous and exogenous cell defense mechanism include the potential use in treating various diseases, improving the potential use in treating various diseases, and improving food-stuffs preparation dietary supplements human nutrition. Published work indicates that SOD regulates oxidative stress, lipid metabolism, inflammation, and oxidation in cells. It can prevent lipid peroxidation, the oxidation of low-density lipoprotein in macrophages, lipid droplets' formation, and the adhesion of inflammatory cells into endothelial monolayers. It also expresses antioxidant effects in numerous cancer-related processes. Additionally, different forms of SOD may also augment food processing and pharmaceutical applications, exhibit anticancer, antioxidant, and anti-inflammatory effects, and prevent arterial problems by protecting the proliferation of vascular smooth muscle cells. Many investigations in this review have reported the therapeutic ability and physiological importance of SOD. Because of their antioxidative effects, SODs are of great potential in the medicinal, cosmetic, food, farming and chemical industries. This review discusses the findings of human and animal studies that support the advantages of SOD enzyme regulations to reduce the formation of oxidative stress in various ways.

Abstract:Superoxide dismutases (SODs) are necessary antioxidant enzymes that protect cells from reactive oxygen species (ROS). Decreased levels of SODs or mutations that affect their catalytic activity have serious phenotypic consequences. SODs perform their bio-protective role by converting superoxide into oxygen and hydrogen peroxide by cyclic oxidation and reduction reactions with the active site metal. Mutations of SODs can cause cancer of the lung, colon, and lymphatic system, as well as neurodegenerative diseases such as Parkinson's disease and amyotrophic lateral sclerosis. While SODs have proven to be of significant biological importance since their discovery in 1968, the mechanistic nature of their catalytic function remains elusive. Extensive investigations with a multitude of approaches have tried to unveil the catalytic workings of SODs, but experimental limitations have impeded direct observations of the mechanism. Here, we focus on human MnSOD, the most significant enzyme in protecting against ROS in the human body. Human MnSOD resides in the mitochondrial matrix, the location of up to 90% of cellular ROS generation. We review the current knowledge of the MnSOD enzymatic mechanism and ongoing studies into solving the remaining mysteries.

Abstract:Superoxide dismutase 3 (SOD3) is a type of antioxidant enzyme, which plays an important role in converting superoxide anion into hydrogen peroxide through its extracellular activity. This enzyme has been widely studied and evaluated from various points of view, including maintaining cellular redox balance, protecting against oxidative damage, and enhancing overall cellular resilience. The current paper focuses on SOD3 expression from a functional perspective. In addition to a detailed examination of the gene and protein structure, we found ample evidence indicating that the expression level of SOD3 undergoes alterations in response to various transcription factors, signaling pathways, and diverse conditions. These fluctuations, by disrupting the homeostasis of SOD3, can serve as crucial indicators of the onset or exacerbation of specific diseases. In this regard, significant efforts have been dedicated in recent years to the treatment of diseases through the regulation of SOD3 expression. The ultimate goal of this review is to extensively highlight and demonstrate the immense potential of SOD3 as a therapeutic target, emphasizing its profound impact on health outcomes.

Abstract:With high catalytic activity and stability, nanozymes have huge advantage in generating or eliminating the reactive oxygen species (ROS) due to their intrinsic enzyme-mimicking abilities, therefore attracting wide attention in ROS-related disease therapy. To better design nanozyme-based platforms for ROS-related biological application, we firstly illustrate the catalytic mechanism of different activities, and then introduce different strategies for using nanozymes to augment or reduce ROS level for the applications in cancer therapy, pathogen infection, neurodegeneration, etc. Finally, the challenges and future opportunities are proposed for the development and application of nanozymes.

Abstract:Although nanozyme has become an emerging area of research attracting extensive attention recently, the activity and specificity of currently reported nanozymes are generally lower than those of natural enzymes. Developing highly active and specific nanozymes is therefore extremely necessary and also remains a great challenge. Superoxide dismutase (SOD) catalyzes the disproportionation of cytotoxic O2− into hydrogen peroxide and oxygen, and plays an important role in reducing human oxidative stress. In this work, we prepare Cu single-atom catalysts (Cu/GO SACs, GO = graphene oxide) through a simple and low-cost strategy at room temperature using Cu foam and graphene oxide. Cu/GO SACs can maintain excellent catalytic activity under harsh environment. Compared with the natural enzyme, SOD-like Cu/GO SAC nanozyme has higher catalytic activity and meanwhile, it does not possess the common properties of other mimic enzymes often existing in nanomaterials. Based on the excellent SOD-like enzyme activity of Cu/GO SACs, it successfully eliminates the active oxygen in cigarette smoke. This work not only provides a new idea for the design and synthesis of nanozymes with excellent SOD mimetic properties, but also is promising in the treatment of lung injury and inflammatory diseases related to free radical production.

Abstract:Abstract Nanozymes are nanomaterials with enzyme-like characteristics. As a new generation of artificial enzymes, nanozymes have the advantages of low cost, good stability, simple preparation, and easy storage, allowing them to overcome many of the limitations of natural enzymes in enzymatic therapy. Currently, most reported nanozymes exhibit oxidoreductase-like activities and can regulate redox balance in cells. Nanozymes with superoxide dismutase and catalase activity can be used to scavenge reactive oxygen species (ROS) for cell protection, while those with peroxidase and oxidase activity can generate ROS to kill harmful cells, such as tumor cells and bacteria. In this review, we summarize recent progress in nanozyme-based medicine for enzymatic therapy and highlight the opportunities and challenges in this field for future study.

These nanomaterials can regulate the redox levels of cells by mimicking the catalytic processes of natural enzymes such as peroxidase,I catalase, and superoxide dismutase. This paper describes the intracellular ROS regulation 1 and oxidative stress-related diseases treatment effects of metal compounds nanozymes, noble metal nanozymes amid carbon-based nanozymes from the perspective of their enzyme-like activities. As a new type of enzyme mimic, nanozymes are expected to provide a new strategy for the treatment of diseases in the future biomedical field.

Abstract:Oxidative stress plays a critical role in the onset and progression of neurodegenerative diseases. Traditional methods for regulating oxidative stress using drugs or enzyme molecules often face limitations in efficacy, potential side effects and the ability to fully meet clinical needs. The emergence of nanozymes offers a novel approach to overcoming these challenges and exploring therapeutic mechanisms. This article reviews the latest advancements in using nanozymes for treating neurodegenerative diseases, with a focus on the interaction between reactive oxygen species (ROS) and the nervous system. It begins by analyzing the characteristics of ROS and summarizing the mechanisms of ROS interaction with neurons and glial cells within the complex nervous network. Next, it introduces and summarizes application examples and mechanistic insights into different types of ROS-related nanozymes in various neurodegenerative diseases. Additionally, the current state and future prospects of combining nanozymes with advanced technologies, such as in vitro detection and artificial intelligence, for disease treatment are discussed. This approach has the potential to significantly advance the development of therapies for neurodegenerative diseases.

Abstract:With in-depth research over the past decade, impressive progress of nanozymes has been made in exploration of their activity, mechanism, and advanced applications. Among them, nanozymes with catalase-, superoxide dismutase-, glutathione peroxidase-like, and other activities can catalyze reactive oxygen species into water or oxygen and relieve oxidative stress on cells, thus attracting special attention for their potential application in antioxidant therapy. This chapter aims to focus on the main classifications and synthesis methods of antioxidant nanozymes, their enzymatic mechanisms and activity modulation approaches, and their current progress, challenges, and future directions for antioxidant therapy.
