Abstract:The potato tuberworm, Phthorimaea operculella Zeller, is an oligophagous pest feeding on crops mainly belonging to the family Solanaceae. It is one of the most destructive pests of potato worldwide and attacks foliage and tubers in the field and in storage. However, the lack of a high-quality reference genome has hindered the association of phenotypic traits with their genetic basis. Here, we report on the genome assembly of P. operculella at the chromosomal level. Using Illumina, Nanopore and Hi-C sequencing, a 648.2 Mb genome was generated from 665 contigs, with an N50 length of 3.2 Mb, and 92.0% (596/648.2 Mb) of the assembly was anchored to 29 chromosomes. In total, 16619 genes were annotated, and 92.4% of BUSCO genes were fully represented. The chromosome-level genome of P. operculella will provide a significant resource for understanding the genetic basis for the biological study of this insect, and for promoting the integrative management of this pest in future.
Identification and Functional Characterization of Two Putative Pheromone Receptors in the Potato Tuber Moth, Phthorimaea Operculella

Abstract:Pheromones are a kind of signal produced by an animal that evoke innate responses in conspecifics. In moth, pheromone components can be detected by specialized olfactory receptor neurons (OSNs) housed in long sensilla trichoids on the male antennae. The pheromone receptors (PRs) located in the dendrite membrane of OSNs are responsible for pheromone sensing in most Lepidopteran insects. The potato tuber moth Phthorimaea operculella is a destructive pest of Solanaceae crops. Although sex attractant is widely used in fields to monitor the population of P. operculella, no study has been reported on the mechanism the male moth of P. operculella uses to recognize sex pheromone components. In the present study, we cloned two pheromone receptor genes PopeOR1 and PopeOR3 in P. operculella. The transcripts of them were highly accumulated in the antennae of male adults. Functional analysis using the heterologous expression system of Xenopus oocyte demonstrated that these two PR proteins both responded to (E, Z)-4,7–13: OAc and (E, Z, Z)-4,7,10–13: OAc, the key sex pheromone components of P. operculella, whilst they responded differentially to these two ligands. Our findings for the first time characterized the function of pheromone receptors in gelechiid moth and could promote the olfactory based pest management of P. operculella in the field.
Midgut microbiota diversity of potato tuber moth associated with potato tissue consumed

Abstract:The potato tuber moth (PTM), Phthorimaea operculella (Zeller), is a worldwide pest that feeds on both the leaves and tubers of potato plants. PTM larvae can digest leaves, or tubers, resulting in serious damage to potato plants in the field and potato tubers in storage. To understand how midgut bacterial diversity is influenced by the consumption of these two tissue types, the symbiotic bacteria in the potato-feeding PTM midgut and the endophytic bacteria of potato tissues were analyzed. At the genus level, the bacterial community composition in the PTM midgut was influenced by the tissues consumed, owing to their different nutrient contents. Escherichia_Shigella and Enterobacter were the most dominant genera in the midgut of leaf-feeding and tuber-feeding PTMs, respectively. Interestingly, even though only present in low abundance in leaves and tubers, Escherichia_Shigella were dominantly distributed only in the midgut of leaf-feeding PTMs, indicating that specific accumulation of these genera have occurred by feeding on leaves. Moreover, Enterobacter, the most dominant genus in the midgut of tuber-feeding PTMs, was undetectable in all potato tissues, indicating it is gut-specific origin and tuber feeding-specific accumulation. Both Escherichia_Shigella and Enterobacter abundances were positively correlated with the dominant contents of potato leaves and tubers, respectively. Enrichment of specific PTM midgut bacterial communities was related to different nutrient levels in different tissues consumed by the insect, which in turn influenced host utilization. We provide evidence that a portion of the intestinal microbes of PTMs may be derived from potato endophytic bacteria and improve the understanding of the relationship between potato endophytic bacteria and the gut microbiota of PTMs, which may offer support for integrated management of this worldwide pest.
Comparative life history and fecundity of Phthorimaea operculella (Lepidoptera: Gelechiidae) on leaves and tubers of different potato cultivars

Abstract:The potato tuberworm, Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae), is a noxious pest of the potato crop (Solanum tuberosum L.) in both field and storage in tropical and subtropical regions. Survival, development and reproduction of the potato tuberworm was compared on leaves and tubers of 10 common cultivated potato cultivars including Agria, Agata, Almera, Arinda, Baneba, Fiana, Marfona, Ramus, Satina, and Volvox at 25 +/- 1 degree C, 65 +/- 5% RH and a photoperiod of 14:10 (L:D) h. The results indicated that there were significant differences in the larval, pupal, and total developmental periods on the various potato cultivars. The total developmental periods varied from 26.6 (on Baneba) to 29.5 d (on Marfona),and ranged from 27.9 (on Marfona) to 30.5 d (on Agria) in the experiments on potato leaves and tubers, respectively. Immature survival rates were lowest on Agria (44.1%) and Marfona (40.6%) on potato leaves and tubers, respectively. The highest total fecundity was observed on Arinda (78.3 eggs) and Ramus (154.8 egg) on potato leaves and tubers, respectively. Significantly fewer eggs were laid on Marfona cultivar's leaves (44.6 eggs) and tubers (72.9 eggs) than any of the other tested potato cultivars. The female reproduction potential on potato leaves was significantly lower than on potato tubers. Cluster analysis of the biological parameters of P. operculella on different potato cultivars demonstrated that Marfona is partially resistant to potato tuberworm. The findings on the susceptibility or resistance of potato cultivars could be a fundamental component of integrated pest management programs for potato tuberworm.

Abstract:Abstract The potato tuber moth (PTM; caused by Phthorimaea operculella) is one of the most serious insect pests that causes damage to crop production and storage of potatoes. It causes damage to the potato, while larvae of P. operculella feed on and grow in the leaves and tubers of potatoes, resulting in direct product losses. The larvae construct tunnels through tubers and mine leaves, stems, and petioles, resulting in uneven galleries. Foliar damage to the potato crop does not normally result in major output losses, although contaminated tubers may have lower marketability and storage losses of up to 100%. Light watering every 4 days and mulching with neem leaves during the last 4 weeks before the harvest was found to be a successful treatment. Spraying a thin layer of testing natural oils also acts as a defensive layer, which causes confusion or disturbance to the searching neonate larvae. The pest is tough to eradicate, so producers rely heavily on insecticides and biological approaches. Potato tuberworm damage was reduced by rotating esfenvalerate and indoxacarb treatments before and after vine kill. At 5 g/kg potato tuber in farmer rustic storage, Acorus calamus dust exhibited great efficiency in preventing PTM tuber damage. In different regions of the world, parasitoids such as Copidosoma spp. and Apanteles spp. are significant in PTM management. Pheromone traps are used for pest population monitoring as well as pest management in the field and during storage.

Abstract:BACKGROUND:Potato, Solanum tuberosum, is one of the most important food crops in the world, playing a significant role in global food security. However, many potato industries and farms may suffer losses of tuber yield and quality in storage due to lepidopteran pests. Here, we evaluated the effectiveness of an ectoparasitic idiobiont mite Pyemotes zhonghuajia in the biological control of the potato tuber moth (PTM) Phthorimaea operculella by determining the lethal, sublethal (nonconsumptive) and transgenerational effects of P. zhonghuajia of various population densities and exposure durations on PTM survival, development and reproduction.RESULTS:Pyemotes zhonghuajia females were capable of killing all instar stages of PTM, while resistance to mite parasitism increased with the development of PTM life stage. The mortality of mature larvae (i.e., fourth instar) and pupae increased with increasing mite density and exposure duration. P. zhonghuajia imposed significant negative sublethal impacts on PTM pupation rate, female fecundity and adult longevity but not on immature development. The sublethal stress was transgenerational, resulting in lower reproduction in the offspring generation.CONCLUSION:P. zhonghuajia induces lethal, sublethal and transgenerational effects and significantly decreases PTM survival and reproductive out, demonstrating its high efficiency in the biological control of PTM. Our study provides insight into the mechanisms underlying the nonconsumptive effects of parasitism in an ectoparasite-host system and delivers critical information for the design and implementation of augmentative releases of P. zhonghuajia in the biological control of PTM in potato storage. © 2023 Society of Chemical Industry.

Abstract:The potato tuber moth (PTM), Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae), is a worldwide potato pest that thrives in warm temperate and tropical climates. The aim of this study was to evaluate the pathogenicity of indigenous isolates of three entomopathogenic fungi (EPFs) [ Beauveria bassiana (Bals.) Vuill.–3, Isaria fumosorosea (Wize) Brown & Smith–1, and Lecanicillium psalliotae (Treschew) Zare & Gams–1] against eggs, younger larvae (1st and 2nd instars), and older (3rd and 4th instars) larvae of PTM under laboratory conditions. Whole isolates were applied at three different conidial suspensions of (1 × 10 6 , 1 × 10 7 , and 1 × 10 8 conidia/mL), using a spray method. Pathogenicity tests revealed that all of the isolates tested had varying degrees of pathogenicity against the pest. The highest mortalities were seen in the larvae; however, younger larvae were more susceptible than the older ones. Eggs were generally less susceptible than larvae. Two B. basiana isolates (M49 and M48) were more lethal against PTM than the others, generating 100.0% and 96.1% mortalities in older larvae, respectively, and 100.0% mortalities in younger larvae. These two isolates likewise had the strongest egg-hatching inhibitory effects, preventing egg hatching in 0–48 h old eggs by 88.6% and 80.0%, respectively, 7 days after treatment. All tested isolates were also molecularly identified. According to the results of molecular phylogenetic analyses based on the ITS region sequence, the two most pathogenic B. bassiana isolates exhibit a high similarity (99%) with other B. bassiana isolates in GenBank.

Abstract:This study was conducted to determine some biological properties of the potato tuber moth Phthorimaea operculella, a new host of the egg-larval parasitoid Chelonus oculator at different temperatures. The emergence rate, development time, longevity, adult weight, and sex ratio of C. oculator were assessed at three different temperature levels (20±1 oC, 25±1 oC, 30±1 oC), 65±5% humidity and 16:8 light: dark conditions. Forty potato tuber moth eggs, aged 0-24 hours, adhered to the filter papers, were placed into the tubes, and presented to the parasitoids for parasitization for a day. At the end of this period, the parasitized eggs were allowed to develop at the specified temperatures. The study found the highest emergence rate was found at 30 °C, with 20.5%. The longest development period occurred at 20 °C, determined as 49.00±2.00 days in females and 52.27±1.64 days in males. The longest longevity of the parasitoid was found to be 41.71±2.29 and 45.73±3.75 days in males and females, respectively. The highest adult weight was observed at 25 °C for both males and females. The sex ratio was found to be in favor of males as the temperature decreased. It is thought that these results can be used for the biological control of potato tuber moth in the field and storage conditions for release studies.
Establishment of Beauveria bassiana as a fungal endophyte in potato plants and its virulence against potato tuber moth, Phthorimaea operculella (Lepidoptera: Gelechiidae)

Abstract:The potato tuber moth, Phthorimaea operculella, is the most damaging potato pest in the world and is difficult to control as the larvae are internal feeders in the foliage and tubers. Entomopathogenic fungi that colonize plants as endophytes, have lethal and sublethal pathological effects on insect pests. We show that Beauveria bassiana colonizes the aerial parts of potato plants endophytically after inoculation through soil drenching. Endophytic B. bassiana persisted in potato foliage for more than 50 days post inoculation. Bioassays indicated that foliage of B. bassiana‐inoculated potato plants were pathogenic against larvae of P. operculella. Sublethal experiments indicated that B. bassiana negatively affected the growth, development and reproduction of P. operculella. Development experiments showed that the weight of P. operculella pupae reared on B. bassiana‐colonized potato plants (4.25 mg) was significantly less than that of those reared on uninoculated control plants (8.89 mg). Compared with newly eclosed larvae fed on control plants, those fed on B. bassiana‐inoculated plants had significantly lower survivorship, with only 17.8% developing to the adult stage. Oviposition of P. operculella females reared on B. bassiana endophytically colonized plants was significantly lower (35 eggs/per female) than of those reared on uninoculated plants (115 eggs/per female). This study demonstrates that endophytic B. bassiana can be a potential biological control agent for the control and management of P. operculella. This article is protected by copyright. Abstract:Background: Potato (Solanum tuberosum L.) is a staple food crop globally, but its production is consistently threatened by diseases and arthropod pests like the potato tuber moth, Phthorimaea operculella Zeller (Lepidoptera: Gelechiidae). Phthorimaea operculella is often controlled by chemical applications. Sex pheromones have been used to detect, monitor, or control agricultural pests. Phthorimaea operculella sex pheromones mainly contain two chemical structures, 4E,7Z-trisadene-1-ol acetate and 4E,7Z,10Z-trisadene-1-ol acetate. However, the pheromone analogs are expected to act as mimics, synergists, antagonists, and inhibitors for pheromones and can be synthesized at a large scale. Result: In this study, a total of 11 sex pheromone analogs of P. operculella were designed and synthesized. Results showed that the antennae exhibited a concentration-dependent response to sex pheromones and their analogs. Different analogs and dosages had significant effects on the electrophysiological response of the antennae. In the field trials, when used alone, A6, B3 and B5 exhibited significant trapping effects in tobacco and potato fields. When used alternately with sex pheromone components, the analog B3 had significantly higher trapping effects in both tobacco and potato fields compared to the control, while combinations containing fluorinated analogs showed lower trapping effects. Conclusion: In summary, the use of sex pheromone analogs holds great promise for integration into pest management programs. Further investigation to fine-tune dosage and duration for optimal use is still needed. © 2024 Society of Chemical Industry. Published by John Wiley & Sons Ltd.
Characterization of a newly established potato tuber moth (Phthorimaea operculella Zeller) cell line

Abstract:Background & objective: Potato tuber moth (PTM), Phthorimaea operculella Zeller is a widely distributed, devastating pest of potatoes attacking the foliage and infest the tubers in both field and store causing serious economic damage. As application of PTM granulovirus (PTM-GV) has shown significant reduction in damage, attempts were made to develop a new cell line from this insect to grow PTM-GV for use as a biopesticide. Methods: Approximately 100 mg of insect eggs were collected, surface sterilized and crushed gently in a boiling tube aseptically. The tissues were washed with physiological saline, suspended in growth medium and incubated stationary at 28 degrees C. Morphology of cells was studied after staining with Giemsa. Besides karyological and growth curve studies, PCR amplification was also done for rapid amplified polymorphic DNA pattern. Results: A new cell line from the embryonic tissue of PTM was maintained in Mitsuhashi Maramorosch medium supplemented with 10 per cent foetal bovine serum. It is in the 78th passage level and designated as NIV-PTM-1095. Random amplified polymorphic DNA profile analysis indicated this as a new cell line from potato tuber moth and differed from the profiles of two other lepidopteran cell lines maintained in the laboratory. Three different cell types were observed at the 40th passage level and comprised of epithelial-like cells (77%), fibroblast-like cells (20%) and giant cells (3%). The chromosome number varied from 54-176. The cell line had a cell doubling time of approximately 42 h during the logarithmic phase of growth. The cell line did not support the multiplication of any of the baculoviruses used in the study. Interpretation & conclusion: Since the new cell line is found to replicate PTM-GV, it may be useful for the propagation of PTM-GV in large scale. Studies to scale up the production of the GV in the cell line and field trials may lead to its widespread use as an eco-friendly biopesticide.

Abstract:Potato ladybirds consist of at least two, and probably more, species of phytophagous lady beetles. Both adults and larvae skeletonize potato leaves and, while not as devastating as some other potato pests, can cause considerable crop losses. Potato ladybirds are native to Asia, but were recently introduced to Australia, New Zealand, and South America. Their biology is studied less compared to other major pests of potatoes, although the situation is gradually beginning to improve. Currently, potato ladybirds are controlled mostly by insecticides. However, there is also a good potential for using natural enemies and for breeding resistant potato cultivars.

Abstract:BACKGROUND: Phthorimaea operculella is a key pest of potato. The authors characterised the P. operculella olfactory system, selected the most bioactive host plant volatiles and evaluated their potential application in pest management. The electrophysiological responses of olfactory receptor neurons (ORNs) housed in long sensilla trichodea of P. operculella to plant volatiles and the two main sex pheromone components were evaluated by the single-cell recording (SCR) technique. The four most SCR-active volatiles were tested in a laboratory oviposition bioassay and under storage warehouse conditions.RESULTS: The sensitivity of sensilla trichodea to short-chained aldehydes and alcohols and the existence of ORNs tuned to pheromones in females were characterised. Male recordings revealed at least two types of ORN, each of which typically responded to one of the two pheromone components. Hexanal, octanal, nonanal and 1-octen-3-ol significantly disrupted the egg-laying behaviour in a dose-dependent manner. Octanal reduced the P. operculella infestation rate when used under storage conditions.CONCLUSIONS: This work provides new information on the perception of plant volatiles and sex pheromones by P. operculella. Laboratory and warehouse experiments show that the use of hexanal, octanal, nonanal and 1-octen-3-ol as host recognition disruptants and/or oviposition deterrents for P. operculella control appears to be a promising strategy.

Abstract:A survey for fungal pathogens of cereal aphids in the Pacific Northwest of the United States found several entomophthoraleans. A species of Entomophthora (in the strict sense) causing minor early-season mortality of aphids was of special taxonomic interest. Evaluation of this fungus indicated that it was best identified as E. chromaphidis Burger & Swain, a species long treated as a synonym of E. planchoniana Cornu. E. chromaphidis has primary conidia that are appreciably smaller than those of European collections of E. planchoniana, and can be isolated in culture whereas similar media and techniques have never yielded cultures of E. planchoniana. This study recognizes the aphidpathogenic species of Entomophthora to comprise a globally distributed species complex; Entomophthora planchoniana is a major pathogen whose distribution is primarily European whereas E. chromaphidis appears to be a comparatively minor pathogen with a wide distribution outside of Europe.

Abstract:Background Potato tuber worm (PTM) [ Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae)] is one of the most significant and widespread pests of potato. PTM damages potatoes both in the field and in potato storage areas. Control of the pest is getting harder as it is developing resistance to pesticides. Several entomopathogenic nematode (EPN) species have been reported to successfully control numerous agricultural pests worldwide. The main aim of the study was to isolate native nematode/s as a biological control agent against P. operculella . Morphometric measurements of the infective juvenile (IJ) and sequencing and characterization of the internal transcribed spacer (ITS) region was used to identify the nematode isolate to species level. The efficacy of EPN isolate Z-1 obtained from Zonguldak province, Turkey was tested against different life stages of the pest. Experiments were conducted in 150 ml plastic pots containing sterile soil mixture. Four EPN concentrations (i.e., 0, 250, 500 and 1000 IJs/ml) were applied to the soil. Data relating to the mortality of different life stages were collected daily till 6 days after inoculation. Results Molecular analyses based on the ITS sequence and morphometric data revealed that isolate Z-1 was Heterorhabditis bacteriophora . Mortality rates of PTM larvae exposed to 250, 500, and 1000 IJs/ml concentrations of native EPN were 62.9 ± 9.8, 74.0 ± 3.7, and 92.5 ± 3.7%, respectively. There were non-significant differences among tested EPN concentrations for pupal mortality and the highest concentration (i.e., 1000 IJs/ml) caused 25.6% mortality. Conclusions The results revealed that the native H. bacteriophora isolate was effective against late-stage larvae of PTM under laboratory conditions. Therefore, it can be used as an alternative management option of the pest.

Reference Genes for Expression Analyses by qRT-PCR in Phthorimaea operculella (Lepidoptera: Gelechiidae)

Abstract:Due to a lack of effective internal references, studies on functional genes in Phthorimaea operculella, a serious Lepidopteran pest attacking potatoes worldwide, have been greatly limited. To select suitable endogenous controls, ten housekeeping genes of actin (ACT), α-tubulin (α-TUB), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), elongation factor 1α (EF1α), 18S and 28S ribosomal RNA (18S, 28S), ribosomal protein genes RPL4, RPL13 and RPL27 and superoxide dismutase (SOD) were tested. Their expression levels were determined under three different experimental conditions (developmental stages, tissues/organs and temperatures) using qRT-PCR technology. The stability was evaluated with five methods (Ct value, geNorm, NormFinder, BestKeeper and RefFinder). The results clarified that RPL13, EF1α and RPL27 are ranked as the best reference gene combination for measuring gene expression levels among different developing stages and under various temperatures; EF1α and RPL13 are recommended to normalize the gene expression levels among diverse tissues. EF1α and RPL13 are the best reference genes in all the experimental conditions. To validate the utility of the selected reference pair, EF1α and RPL13, we estimated the tissue-biased expression level of chitin synthase A gene (PoChSA). As expected, PoChSA was abundantly expressed in ectodermally derived epidermal cells, and lowly transcribed in the midgut. These findings will lay the foundation for future research on the molecular physiology and biochemistry of P. operculella.

Abstract:Potatoes hold the distinction of being the largest non-cereal food crop globally. The application of insecticides has been the most common technology for pest control. The repeated use of synthetic insecticides of the same chemical class and frequent applications have resulted in the emergence of insecticide resistance. Two closely related pests that feed on potato crops are the potato tuber moth, Phthorimaea operculella, and the tomato leafminer, Phthorimaea absoluta (syn. Tuta absoluta). Previous studies indicated the existence of insecticide resistance to various classes of insecticides including organophosphates, carbamates, and pyrethroids in field populations of P. operculella and P. absoluta. However, the exact mechanisms of insecticide resistance in P. operculella and to a lesser extent P. absoluta remain still poorly understood. Detecting resistance genotypes is crucial for the prediction and management of insecticide resistance. In this study, we identified multiple genetic mutations related to insecticide resistance in two species of Phthorimaea. An unexpected genetic divergence on target-site mutations was observed between P. operculella and P. absoluta. Three mutations (A201S, L231V, and F290V) in Ace1 (acetylcholinesterase), four mutations (M918T, L925M, T928I, and L1014F) in VGSC (voltage-gated sodium channel), and one mutation (A301S) in RDL (GABA-gated chloride channel) have been detected with varying frequencies in Chinese P. absoluta field populations. In contrast, P. operculella field populations showed three mutations (F158Y, A201S, and L231V) in Ace1, one mutation (L1014F) in VGSC at a lower frequency, and no mutation in RDL. These findings suggest that pyrethroids, organophosphates, and carbamates are likely to be ineffective in controlling P. absoluta, but not P. operculella. These findings contributed to a deeper understanding of the presence of target-site mutations conferring resistance to commonly used (and cheap) classes of insecticides in two closely related potato pests. It is recommended to consider the resistance status of both pests for the implementation of resistance management strategies in potatoes.
Potential of Entomopathogenic Nematodes versus Alpha-cypermethrin Against Potato Tuber Moth, Phthorimaea operculella Zeller 1873 (Lep.: Gelechiidae) in Storage Conditions

Abstract:The potato tuber moth PTM, Phthorimaea operculella, is one of the most economically important potato pests worldwide. In the present study, the potential of Steinernema feltiae and Steinernema carpocapsae for controlling PTM in potato tubers was evaluated compared to alpha-cypermethrin. Steinernema carpocapsae in both concentrations (12.6×10IJs and 6.2×10IJs) showed a lower number of emerged insects than alpha-cypermethrin (10 mg ai l). Alpha-cypermethrin (20 mg ai l) showed the highest efficacy against PTM (81.17%), S.carpocapsae (12.6×10IJs), and alpha-cypermethrin (10 mg ai l) showed similar efficacy (72.53%) while S. feltiae (6.2×10IJs) showed the lowest efficacy (39.04%). The results showed that S.carpocapsae in both concentrations and S. feltiae (12.6×10IJs) were efficient the same as alpha-cypermethrin (10 mg ai l) having no environmental and health adverse impacts issued in the chemical insecticides usage. Both concentrations of alpha-cypermethrin and S. carpocapsae showed the least tuber damage with no significant differences, while it was as high as the control (59.26%) in both concentrations of S. feltiae. This promising finding introduces EPNs as a part of the potato tuber protection program in storage.
