Abstract:Water-soluble non-conjugated polymer dots (PDs) have been synthesized using hyperbranched polyethyleneimine (PEI) and dihydroxybenzaldehyde (DHB) for the first time via the Schiff base reaction at room temperature. The yielded non-conjugated PDs of PEI-DHB could display the well-defined spheric structure and good water solubility. In contrast to the common PDs otherwise showing blue emission, the PEI-DHB PDs could give out strong green fluorescence in aqueous media. Especially, the fluorescence of the PEI-DHB PDs could be specifically quenched by MnO2 nanosheets through the inner filter effects and further restored by the thiocholine that could reduce MnO2 nanosheets into Mn2+. Herein, thiocholine could be produced in hydrolysis reaction of acetylthiocholine catalyzed by the acetylcholinesterase (AChE), of which the catalytic activity could be irreversibly inhibitted by the introduction of organophosphates. A highly selective fluorimetric method was thereby been developed for the detection of organophosphorus pesticides using dimethyl-dichloro-vinyl phosphate as a model. The linear concentrations ranges from 0.050 to 2.5 μM. Importantly, the non-conjugated PDs probes with strong green fluorescence and high water solubility may promise the extensive applications in the environmental, food, and clinical analysis fields.
An Eco-Friendly, Simple, and Sensitive Fluorescence Biosensor for the Detection of Choline and Acetylcholine Based on C-dots and the Fenton Reaction.

Abstract:A simple and novel method is proposed for the preparation of Carbon dots (C-dots) with excellent properties. We firstly demonstrated that the fluorescence of C-dots decreased apparently in the presence of H2O2 and Fe2+. Based on the this finding, C-dots are successfully adopted as probes for the detection of H2O2. After the experimental conditions are optimized, the limit of detection (LOD) for H2O2 is found to be 0.1μM. Furthermore, we established an eco-friendly, simple and sensitive biosensor for the detection of choline and acetylcholine (ACh) based on the detection of H2O2 using C-dots as probes. The detection limit for choline is 0.1μM and the linear range is 0.1–40μM. The detection limit for ACh is found to be 0.5μM and the linear range is 0.5–60μM. The excellent performance of the proposed biosensor shows that this method possesses the potential for practical application.

Abstract:Polydiacetylene supramolecules (PDAs) are unique sensing materials. Upon environmental stimulation, blue PDAs can undergo a colorimetric transition from blue to red accompanied by fluorescence enhancement. In this paper, we report a new PDA system polymerized from a mixed liposome comprising 2-(2-(2-hydroxyethoxy)ethoxy)ethyl pentacosa-10,12-diynoate and pentacosa-10,12-diynoic acid at a 3:7 ratio. The PDA system provided new colorimetric and fluorometric assay methods for screening acetylcholinesterase and its inhibitors through three processes. First, myristoylcholine reacted with PDAs, which then underwent colorimetric and fluorometric transition. Second, acetylcholinesterase catalyzed the hydrolysis of myristoylcholine into tetradecanoic acid, which reduced the myristoylcholine concentration and led to faded color and fluorescence. Third and last, acetylcholinesterase inhibitors retarded the activity of acetylcholinesterase, thereby inducing the recovery of color and fluorescence.

Abstract:Well-distributed metals anchored on supports have attracted great attention owing to their potential application as cost-effective catalysts. Here, we demonstrate the successful preparation of Au-decorated N-rich carbon dot (Au-CD) nanocomposites, which possess excellent stability in aqueous media. The Au-CDs exhibit remarkable peroxidase-mimicking activity and high affinity to the substrate due to the fast electron-transfer process and ligand-free surface. Based on this, an Au-CD nanozyme-based colorimetric biosensor was successfully developed to assess the activity of acetylcholinesterase with high sensitivity and selectivity. More importantly, an ideal performance was achieved in clinical serum analysis. This work paves the way for the development of well-distributed metals anchored on supports as nanozymes for sensing applications.

Abstract:Amperometric biosensor is fabricated for the detection of carbaryl based on single walled carbon nanotubes (SWCNTs) and acetylcholine esterase (AchE). The dispersion of SWCNTs in positively charged polyelectrolyte, poly(diallyldimethylammonium chloride) (PDDA), possibly takes place due to weak supramolecular interaction between them, which then binds electrostatically to the negatively charged AchE at pH 7.4 using layer-by-layer (LbL) self-assembly technique. The optical intensity of UV/vis spectra increased with the number of layers, indicating the build up of a multilayer coating on the electrode. The activity of acetylcholine esterase on modified electrode of 3mm in diameter was found to be 0.2U. The biosensor showed good sensitivity and stability towards the monitoring of carbaryl pesticides in water with the detection limit of 10(-12)gL(-1) and recovery of 99.8 ± 2.7% to 10(-10)gL(-1). This protocol can be used for the immobilization of other enzymes to fabricate a range of biosensors.

Abstract:Acetylcholine (Ach) is an important neurotransmitter and plays a vital role in the diagnosis and treatment of many neurological diseases. A new acetylcholine optical fiber biosensor is reported based on the nitrogen doped fluorescent carbon quantum dots/ acetylcholinesterase (N-CQDs/AchE) complex, where AchE was immobilized on the surface of N-CQDs by the cross-linking method. N-CQDs were synthesized by the hydrothermal method as fluorescent probe and can be responsible for pH value. The optical properties, morphology, particle size distribution and surface groups of N-CQDs were characterized by UV-Vis spectrophotometer, Fluorescence spectrometer, High-resolution transmission electron microscope (TEM) and Fourier transform infrared spectrometer (FTIR), respectively. The relationship between acetylcholine chloride solution and fluorescence intensity of N-CQDs/AchE complex was detected in detailed. Good linear relationship between fluorescence intensity and the concentration of acetylcholine from 20-200 µM and 1-100 nM with the lower detection limits of 16.28 µM and 0.29 nM, respectively. Compared with other sensors, this new optic fiber sensor immobilized (N-CQDs/AchE) complex is more simple and operable.

Abstract:Three-component conjugated polymers of a strong donor-acceptor (D-A) type could be synthesized by Pd-catalyzed Suzuki coupling polymerization reaction of 1,2-bis(4-bromophenyl)-1,2-diphenylethene (M-1) with 9-octyl-3,6-bis(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)-9H-carbazole (M-2) and 4,6-bis((E)-4-bromostyryl)-2,2-difluoro-5-phenyl-2H-1l3,3,2l4-dioxaborinine (M-3). Among them, P-1 and P-2 with high TPE ratios at 0.95 and 0.9 showed obvious aggregation-induced emission (AIE) behavior; in contrast P-3 with a low TPE ratio at 0.8 showed an aggregation-caused quenching (ACQ) phenomenon. In particular, the three resulting polymer dots (P-1 to P-3 Pdots) exhibited a 200 mV lower electrochemiluminescence (ECL) potential due to their strong D-A electronic structure. Most importantly, the ECL signals of Pdots could be enhanced as high as 3 times by increasing their AIE-active TPE moiety ratios from 0.8 (P-3) to 0.95 (P-1) via the band gap emission process. Herein, P-1 Pdots with the strongest ECL signal were successfully used as ECL biosensors for the detection of catechol, epinephrine and dopamine with detection limits of 1, 7 and 3 nM, respectively. This work provides a new strategy for developing highly sensitive ECL biosensors by the smart structure design of the AIE-active Pdots.
Highly Efficient Electrochemiluminescence of Cyanovinylene-Contained Polymer Dots in Aqueous Medium and Its Application in Imaging Analysis

Abstract:Luminescent semiconducting polymer dots (Pdots) have attracted intense attention in the field of electrochemiluminescence (ECL) due to their nontoxic features. For utilizing the nontoxic Pdots to achieve sensitive ECL bioimaging detection, this work studied the ECL behaviors of poly[2-methoxy-5-(2-ethylhexyloxy)-1,4-(1-cyanovinylene-1,4-phenylene)] (CN-PPV) Pdots in aqueous solution, which introduced an electron-withdrawing cyano group to p-phenylenevinylene for enhancing the luminescent efficiency. The CN-PPV Pdots could be both electrochemically oxidized to a positively charged state and electrochemically reduced to a negatively charged state, which led to annihilation of ECL emission. The order of oxidation and reduction greatly influenced the annihilation behavior. In the presence of tri-n-propylamine (TPrA) or S2O82- as a coreactant, the CN-PPV Pdots showed strong band gap ECL emission at 602 nm, which followed two different routes and gave ECL efficiencies of 11.22% and 1.84% (vs Ru(bpy)32+/TPrA), respectively. The high ECL efficiency allowed CN-PPV Pdots/TPrA system for ECL imaging analysis. As a proof-of-methodology, an ECL imaging method was designed via the chelating interaction of metal ions and Pdots to achieve high selectivity. The proposed ECL imaging chip-based sensor exhibited excellent analytical performance for Fe3+ with a wide linear range from 100 pM to 100 μM and a detection limit of 67 pM. Compared with the ECL methods based on the direct intensity measurement, the developed ECL imaging method possesses the advantages of simplicity, rapid, and high-throughput and has application potential in monitoring water and food quality.

Abstract:In this paper, we developed a label-free and sensitive fluorescence sensor for acid phosphatase (ACP) and its inhibitor parathion-methyl (PM) detection based on glutathione-functionalized graphene quantum dots (GQDs@GSH). Upon addition of MnO2 nanosheets, the fluorescence of GQDs@GSH could be efficiently quenched via a fluorescence resonance energy transfer. ACP could easily catalyze the hydrolysis of L-Ascorbic acid-2-phosphate (AAP) to ascorbic acid (AA), which could reduce MnO2 nanosheets to Mn2+ in acidic environment, leading to dramatically increase of the fluorescence intensity of GQDs@GSH. Quantitative detection of ACP in a broad range from 0.1 to 9 mU mL-1 with a detection limit of 0.027 mU mL-1 could be achieved. The feasibility of the proposed sensor in real samples analysis was also studied and satisfactory results were obtained. Furthermore, the fluorescence assay strategy could also be used for the detection of parathion-methyl (PM) as ACP inhibitor.
Biosilicated CdSe/ZnS quantum dots as photoluminescent transducers for acetylcholinesterase-based biosensors

Abstract:CdSe/ZnS core/shell quantum dots (QDs) are functionalized with mercaptoundecanoic acid (MUA) and subsequently covered with poly-L-lysine (PLL) as the template for the formation of the silica outer shell. This nanocomposite is used as a transduction and stabilization system for optical biosensor development. The covalent immobilization of the enzyme acetylcholinesterase from Drosophila melanogaster (AChE) during the formation of the biomimetically synthesized silica is used here as a model, relatively unstable enzyme, as a proof of principle. The enzyme is successfully immobilized onto the QDs and then stabilized by the PLL capping and the subsequent formation of the outer nanoporous silica thin shell, giving rise to the QD/AChE/PLL/silica biosensor. It is shown that the poly-L-lysine templated silica outer shell does not modify the optical properties of the quantum dots, while it protects the enzyme from unfolding and denaturation. The small pores of the silica shell allow for the free diffusion of the analyte to the active center of the enzyme, while it does not allow for the proteases to reach the enzyme. The response of the QD/AChE/PLL/silica nano-biosensor to its substrate, acetylcholine chloride, is evaluated by monitoring the changes in the QDs' photoluminescence which are related to the changes in pH. These pH changes of the surrounding environment of the QDs are induced by the enzymatic reaction, and are associated with the analyte concentration in the solution. The biodetection system proposed is shown to be stable with a storage lifetime of more than 2 months. The data presented provides the grounds for the application of this nanostructured biosensor for the detection of AChE inhibitors.
Enzymatic reaction modulated DNA assembly on graphitic carbon nitride nanosheets for sensitive fluorescence detection of acetylcholinesterase activity and inhibition

Abstract:A novel fluorescent strategy has been developed by using an enzymatic reaction modulated DNA assembly on graphitic carbon nitride nanosheets (CNNS) for the detection of acetylcholinesterase (AChE) activity and its inhibitors. The two-dimensional and ultrathin-layer CNNS-material was successfully synthesized through a chemical oxidation and ultrasound exfoliation method. Because of its excellent adsorption selectivity to ssDNA over dsDNA and superior quenching ability toward the fluorophore labels, CNNS were employed to construct a sensitive fluorescence sensing platform for the detection of AChE activity and inhibition. The detection was based on enzymatic reaction modulated DNA assembly on CNNS, which involved the specific AChE-catalyzed reaction-mediated DNA/Hg 2+ conformational change and subsequent signal transduction and amplification via hybridization chain reaction (HCR). Under the excitation at 485 nm, the fluorescence signal from 500 to 650 nm ( λ max = 518 nm) of the developed sensing system was gradually increased with increasing concentration of AChE. The quantitative determination range of AChE is from 0.02 to 1 mU/mL and the detection limit was 0.006 mU/mL. The developed strategy was successfully applied to the assay of AChE in human serum samples, and can also be used to effectively screen AChE inhibitors, showing great promise providing a robust and effective platform for AChE-related diagnosis, drug screening, and therapy.

One-pot synthesis of fluorescent non-conjugated polymer dots for Fe3+ detection and temperature sensing

Abstract:The facile preparation of highly fluorescent polymer dots (PDs) still attracts substantial interest. Here, temperature/Fe3+ dual-responsive PDs are synthesized under mild conditions via the amidation reaction and self-assembly between hyperbranched polyethyleneimine and 5-aminosalicylic acid. The prepared PDs display strong green fluorescence with quantum yield of 15.5% and 53.3% in water and dimethylsulfoxide, respectively. The PDs also possess unique features, including excellent solubility, solvent polarity-dependent emission, remarkable photostability, as well as good salt-tolerance. Interestingly, the fluorescence intensity of PDs exhibits a reversible and sensitive response to temperature within 20–65 °C, which renders the PDs useful as a thermometer probe. Importantly, Fe3+ ion has the specific coordination ability toward the surface groups of PDs, leading to the aggregation and fluorescence quenching of PDs. Thus, the PDs are employed as a fluorescence probe for sensitive detecting Fe3+. The fluorescent intensity linearly decreases with increasing Fe3+ from 2 to 60 μM. Besides, Fe3+ concentration in river water samples is successfully assayed with this developed probe. The non-conjugated PDs with facile preparation, sensitive response to temperature and Fe3+ may hold potential applications in environmental monitoring.
Electrochemiluminescent Resonance Energy Transfer of Polymer Dots for Aptasensing

Abstract:This work designed a three-component polymer for the preparation of polymer dots (Pdots). The polymer contained 9-(diphenylmethylene)-9H-fluorene (DPF), 9,9-dioctyl-9H-fluorene (DOF) and 1,1'-binaphthyl moieties, and was synthesized via Pd-catalyzed Suzuki reaction. It exhibited obvious yellow-colored aggregation-induced emission (AIE) for fluorescence enhancement at 543nm via an intramolecular fluorescence resonance energy transfer from DOF moiety to DPF moiety. The Pdots prepared by nanoprecipitation could be conveniently cast on electrode surface and showed a stable anodic electrochemiluminescence (ECL) emission in the presence of triethylamine as a co-reactant. The ECL emission could be effectively quenched by rhodamine B via resonance energy transfer, which led to an "off-on" switch for the design of ECL sensing methodology. Using Pb2+ as a target model, an ECL aptasensor for the detection of trace Pb2+ was proposed, which showed a linear range of 100pM to 1.0μM with a detection limit down to 38.0pM This work demonstrated the first Pdots prepared with AIE-active polymer for highly efficient ECL sensing.

Abstract:Organophosphorus pesticide (OP) residues have gathered considerable attention because of their significant threat to society development and healthy life. Developing a sensitive and practical OPs sensor is highly urgent, whereas remains a huge challenge. To this end, we fabricated a high-performance fluorescence paper analytical device (PAD) for apparatus-free and visual sensing of OPs based on aggregation-induced emission (AIE) luminogen's bright emission in aggregated state, unique response of MnO2 to thiol compounds, and difference of MnO2 and Mn2+ in quenching fluorescence. AIE nanoparticles PTDNPs-0.10 and MnO2 respectively acted as core and shell to prepare 2, which possessed high stability and were dripped on cellulose paper's surface to fabricate AIE-PAD. The sensing mechanism is that OPs-treated acetylcholinesterase (AChE) prevents the formation of thiocholine, thereby minimizing the reduction of MnO2 into Mn2+ and changing the output signal. As a result, equipment-free and visual sensing of OPs was acquired with limit of detection of 1.60 ng/mL. This work justifies the feasibility of applying core-shell material to develop high-performance sensor and substituting complex/expensive solution-phase sensor with PAD, providing a new avenue to bring OPs analysis out of the lab and into the world.

Abstract:Acetylcholinesterase (AChE) plays a crucial role in biological neurotransmission. The aberrant expression of AChE is associated with various neurodegenerative diseases. Therefore, it is of great significance to develop a simple and highly sensitive AChE analysis platform. Herein, a simple colorimetric sensor was constructed for sensitive detection of AChE based on the peroxidase-like catalytic activity of Ni/Co layered double hydroxides (Ni/Co LDHs). In this sensor, the fabricated Ni/Co LDHs possess high peroxidase-like activity, enabling rapidly catalyze o-phenylenediamine (OPD) to produce yellow oxOPD in the presence of H2O2. This peroxidase-like activity of Ni/Co LDHs was found to be effectively inhibited by the presence of AChE. It is speculated that the combination of AChE on the outer surface of Ni/Co LDHs through non-covalent interaction may cover the active sites and hinder their adsorption to the substrates, leading to the failure of OPD oxidation. As a result, the yellow color from the oxOPD is related to the AChE concentration, enabling the direct AChE assay in an equipment-free manner. In addition, the fabricated Ni/Co LDHs could be modified on paper surface to obtain a paper-based analytical device for visualized colorimetric detection of AChE. The as-proposed sensor shows high sensitivity to AChE with a detection limit down to 6.6 μU mL-1. Therefore, this naked-eye paper-based sensor is capable of on-site and real-time detection of AChE, which has outstanding application prospects in clinical diagnosis and biomedical fields.

Abstract:Due to their great threat to human health and the environment, it is always of significance to develop efficient tools to monitor pesticide residues. Although the enzyme inhibition-based strategy is considered as a potential method and has been intensively explored for pesticide analysis, both vulnerable enzymatic activity and tedious operation greatly hinder its practical applications. Developing robust, easy-to-use and efficient devices for pesticide sensing is still challenging. Herein, we recommend an enzyme-nanozyme integrated hierarchically porous hydrogel (AChE-MnO2@HPH) for the convenient detection of pesticides. MnO2 microspheres were first employed as an emulsion stabilizer to fabricate MnO2@HPH by thermal polymerization, and AChE was then immobilized into the MnO2@HPH via a fast self-absorption process. With the unique porous structure and hydrophilic microenvironment, the constructed AChE-MnO2@HPH exhibited excellent enzymatic activity and stability as well as rapid response toward pesticides through inhibiting the activity of AChE. By applying a smartphone to read the color information originating from the chromogenic reaction of 3,3′,5,5′-tetramethylbenzidine (TMB) catalyzed by oxidase-mimicking MnO2, highly sensitive and convenient determination of fenthion, an organophosphorus pesticide model, was achieved with a detection limit down to 0.63 ng mL−1. Excellent performance robustness over 30 storage days and reliable detection of the target in real food samples were also demonstrated. With the advantages of easy operability, free of equipment, and good detection performance and long-term stability, the integrated AChE-MnO2@HPH will find promising use for pesticide residue in-field analysis.

Abstract:The optical transducer of CdTe semiconductor quantum dots (QDs) has been integrated with acetylcholinesterase enzyme (AChE) by the layer-by-layer (LbL) assembly technique, resulting in a highly sensitive biosensor for detection of organophosphorus pesticides (OPs) in vegetables and fruits based on enzyme inhibition mechanism. The detection limits of the proposed biosensors are as low as 1.05 × 10(-11) M for paraoxon and 4.47 × 10(-12) M for parathion, which are significantly better than those of the conventional GC/MS methods or amperometric biosensors (0.5 nM). These biosensors are used for quick determination of low concentrations of OPs in real vegetable and fruit samples and exhibit satisfactory reproducibility and accuracy. Moreover, the stock stability of the biosensors are very good due to the stabilizing environment for the enzyme in the nanostructures made by LbL technique. Many advantages provided by these biosensors, like fluorescent change recognized by naked eyes and mass production with low cost, will facilitate future development of rapid and high-throughput screening of OPs.
2D/3D hierarchical porous structure of mNPC/SMOH@C to construct an electrochemical sensor for the simultaneous determination of p-acetylaminophenol and p-aminophenol

Abstract:Background: As persistent organic pollutants (POPs), the accumulation of p-acetylaminophenol (PAT) and p-aminophenol (PAP) in water can seriously damage the health of plants and animals, ultimately leading to threats to human health and safety. Electrochemical sensors have the advantages of being fast, inexpensive, and accurate compared to the complex, expensive, and cumbersome conventional analytical methods. In this study, we designed and synthesized composites with two-dimensional/three-dimensional (2D/3D) porous structures to construct an efficient electrochemical platform for the simultaneous detection of PAT and PAP. Results: In this work, a novel 3D foamy birnessite Na0.55Mn2O4·1.5H2O@C (SMOH@C) was synthesized, which was composited with 2D ordered mesoporous nanosheets (mNPC) to construct electrochemical sensors detecting PAT and PAP simultaneously. The prepared 2D/3D porous structure of mNPC/SMOH@C increased the exposure of active sites due to its large specific surface area. The introduction of a 3D carbon skeleton altered the charge transfer rate of SMOH@C, and the rich pore structure and oxygen-rich vacancies created favorable conditions for the diffusion and adsorption of PAP and PAT, which enabled the sensitive detection of PAT and PAP. The constructed mNPC/SMOH@C electrochemical sensor could simultaneously detect PAT (1 × 10-7 - 1 × 10-4 M) and PAP (5 × 10-8 - 1 × 10-4 M) with detection limits of 20.4 nM and 30.1 nM, respectively. The sensor has good repeatability (RSD <4 %) and reproducibility (RSD <4 %), and satisfactory recoveries (96.7-102.8 %) were obtained in the analysis of natural water samples. Significance: In this paper, for the first time, we present the synthesis of 3D foam birnessite and its composite with mNPC for the electrochemical simultaneous detection of PAT and PAP. Our proposed strategy for fabricating 2D/3D porous composites lays the foundation for the design and synthesis of other porous materials. In addition, this study provides new ideas for developing efficient and practical electrochemical sensors for detecting pollutants in aquatic environments.
