Abstract:Highly emissive inorganic-organic nanoparticles with core-shell structures are fabricated by a one-pot, surfactant-free hybridization process. The surfactant-free sol-gel reactions of tetraphenylethene- (TPE) and silole-functionalized siloxanes followed by reactions with tetraethoxysilane afford fluorescent silica nanoparticles FSNP-1 and FSNP-2, respectively. The FSNPs are uniformly sized, surface-charged and colloidally stable. The diameters of the FSNPs are tunable in the range of 45-295 nm by changing the reaction conditions. Whereas their TPE and silole precursors are non-emissive, the FSNPs strongly emit in the visible vision, as a result of the novel aggregation-induced emission (AIE) characteristics of the TPE and silole aggregates in the hybrid nanoparticles. The FSNPs pose no toxicity to living cells and can be utilized to selectively image cytoplasm of HeLa cells.

Abstract:Harnessing the power of mesoporous silica to encapsulate organic fluorescent dyes has led to the creation of an extraordinary class of nanocomposite photonic materials. These materials stand out for their ability to produce the brightest fluorescent particles known today, surpassing even the luminosity of quantum dots of similar spectrum and size. The synthesis of these materials offers precise control over the shape and size of the particles, ranging from the nano to the multi-micron scale. Just physical encapsulation of the dyes opens new possibilities for mixing different dyes within individual particles, paving the way for nearly limitless multiplexing capabilities. Moreover, this approach lays the groundwork for the development of highly sensitive sensors capable of detecting subtle changes in temperature and acidity at the nanoscale, among other parameters. This mini-review highlights the mechanism of synthesis, explains the nature of ultrabrightness, and describes the recent advancements and future prospects in the field of ultrabright fluorescent mesoporous silica particles, showcasing their potential for various applications.

Abstract:Fluorescent, radio-opaque, and paramagnetic silica nanoparticles 100 ± 10 nm in size are developed by incorporating fluorescent tris(2,2′-bipyridyl) dichlororuthenium(II) hexahydrate and paramagnetic gadolinium ions. The nanoparticles are radio-opaque due to the presence of electron-dense Ru and Gd atoms. It is estimated that each nanoparticle carries about 16 000 Gd3+ ions. The multimodality of the silica nanoparticles is shown by fluorescence (Figure, left) and X-ray (right) imaging.

Abstract:Modified fluorescent nanoparticles continue to emerge as promising candidates for drug delivery, bioimaging, and labeling tools for various biomedical applications. The ability of nanomaterials to fluorescently label cells allow for the enhanced detection and understanding of diseases. Silica nanoparticles have a variety of unique properties that can be harnessed for many different applications, causing their increased popularity. In combination with an organic dye, fluorescent nanoparticles demonstrate a vast range of advantageous properties including long photostability, surface modification, and signal amplification, thus allowing ease of manipulation to best suit bioimaging purposes. In this study, the Stöber method with tetraethyl orthosilicate (TEOS) and a fluorescent dye sulfo-Cy5-amine was used to synthesize fluorescent silica nanoparticles. The fluorescence spectra, zeta potential, quantum yield, cytotoxicity, and photostability were evaluated. The increased intracellular uptake and photostability of the dye-silica nanoparticles show their potential for bioimaging.

Abstract:We report a novel fluorescent polyfluorene-containing methacrylate macromonomer (PFMA) and its well-defined copolymers with 2-(dimethylamino) ethyl methacrylate (DMAEMA) [P(PFMA-r-DMAEMA)s] via reversible addition-fragmentation chain transfer (RAFT) polymerization. These copolymers selfassemble into photoluminescent nanoparticles in aqueous solutions and act as an excellent scaffold for the incorporation of various pigments to emit tunable colors, including white, through Forster resonance energy transfer with high quantum yield up to 0.80. The directly cast films of these NPs after evaporation of water exhibit strong colorful emission on both smooth quartz and rough plaster surfaces. The NP aqueous solutions are ready for ink-jet printing to produce exquisite fluorescent pictures.

Abstract:<p>Water-soluble phosphorescent nanoparticles are highly desirable for biomedical applications. However, the phosphorescence of nanoparticles is usually quenched in aqueous solutions due to the nonradiative deactivation of triplet excitons by the oxygen in water, thus it is a huge challenge to realize water-soluble phosphorescent nanoparticles. Herein, ultralong and efficient phosphorescence has been achieved from water-soluble carbon nanodots (CNDs) by confine CNDs in a nanospace. Silica capsulation layer is used to ensure the solublity of the CNDs, isolate the CNDs from the surrounding oxygen in the aqueous solution. Furthermore, the motion and vibration of covalent bonds in the CNDs are limited. In this way, the nonradiative deactivation rates of triplet excitons has been decreased, and efficient phosphorescence is achieved from water-soluble CNDs. The lifetime and phosphorescence quantum yield of the CNDs are 1.86 s and 11.6 %, both of which are the best values ever reported for water-soluble phosphorescent nanoparticles. In terms of the efficient phosphorescence of the CNDs in aqueous solution, <em>in vivo/vitro</em> afterglow imaging is also been demonstrated.</p>

Abstract:Ultrasmall sub-10 nm nanoprobes and carriers are of significant interest due to their favorable biodistribution characteristics in in vivo experiments. Here we describe the one-pot synthesis of PEGylated mesoporous silica nanoparticles with a single pore, tunable sizes around 9 nm and narrow size distributions that can be labeled with near-infrared dye Cy5.5. Particles are characterized by a combination of transmission electron microscopy, dynamic light scattering, fluorescence correlation spectroscopy, optical spectroscopy, nuclear magnetic resonance spectroscopy, and nitrogen sorption/desorption measurements. The possibility to distinguish an "inside" and "outside" may render these particles an interesting subject for further studies in sensing, drug delivery, and theranostics applications.

Abstract:The problem of functionalization of recently reported ultrabright fluorescent mesoporous silica nanoparticles while preserving their fluorescent brightness is solved. This is a serious issue because of the open geometry of mesoporous channels and physical encapsulation of fluorescent dye inside those channels. Amine modification of mesoporous nanoparticles is described to preserve the brightness comparable to that of earlier reported ultrabright silica nanoparticles. Scaling to 40 nm sized particles, amine‐functionalized nanoparticle have fluorescent brightness equivalent to the one of 630 free rhodamine 6G (R6G) dye molecules in water. To demonstrate further most challenging functionalization, which relies on using organic‐solvent‐based chemistry, folic acid conjugation is developed. Two different methods are used to conjugate folites to the amine functionalities. Both methods result in a decrease of fluorescence intensity, which can nonetheless still be called ultrabright. The brightness can drop to either 310 or 80 R6G dye molecules per particle of nominal diameter of 40 nm.

Abstract:The development of novel fluorescent nanoprobes with remarkable optical properties, suitable particle size, high water dispersibility and good biocompatibility has recently attracted increasing interest for various biomedical applications. In this work, a novel type of luminescent polymeric nanoparticle based on polymerizable dyes with aggregation induced emission (AIE) properties and a zwitterionic monomer were prepared via reversible addition fragmentation chain transfer polymerization. Due to their amphiphilic properties, these copolymers could facilely self-assemble into AIE dye containing luminescent zwitterionic polymeric nanoparticles, which were characterized by a series of characterization techniques including transmission electronic microscopy, Fourier transform infrared spectroscopy, fluorescence spectroscopy and X-ray photoelectron spectroscopy. The results showed that these luminescent polymeric nanoparticles with diameters of tens of nanometers showed high water dispersibility and strong fluorescence in aqueous solution. To explore their potential for biomedical applications, biocompatibility and cell uptake behavior of these luminescent polymeric nanopartilces were further evaluated. We demonstrated that these polymeric nanoparticles are biocompatible with A549 cells and promising for bioimaging applications. Taken advantage of these merits of the AIE dye based zwitterionic polymeric nanoparticles, which could elegantly avoid the aggregation induced quenching of conventional organic dyes and nonbiodegradability of fluorescent inorganic nanoparticles, the ultrabright and biocompatible luminescent polymeric nanoparticles described in this work should be of highly potential for various biomedical applications.

Abstract:We report herein a straightforward and label-free approach to prepare luminescent mesoporous silica nanoparticles. We found that calcination at 400 °C can grant mesoporous organosilica nanoparticles with strong fluorescence of great photo- and chemical stability. The luminescence is found to originate from the carbon dots generated from the calcination, rather than the defects in the silica matrix as was believed previously. The calcination does not impact the particles' abilities to load drugs and conjugate to biomolecules. In a proof-of-concept study, we demonstrated that doxorubicin (Dox) can be efficiently encapsulated into these fluorescent mesoporous silica nanoparticles. After coupled to c(RGDyK), the nanoconjugates can efficiently home to tumors through interactions with integrin αvβ3 overexpressed on the tumor vasculature. This calcination-induced luminescence is expected to find wide applications in silica-based drug delivery, nanoparticle coating, and immunofluorescence imaging.

Abstract:Fluorescent nanoparticles have a variety of biomedical applications as diagnostics and traceable drug delivery agents. Highly fluorescent porous silica nanoparticles were synthesized in a water/oil phase by a microemulsion method. What is unique about the resulting porous silica nanoparticles is the combination of a single-step, efficient synthesis and the high stability of its fluorescence emission in the resulting materials. The key of the success of this approach is the choice of a lipid dye that functions as a surrogate surfactant in the preparation. The surfactant dye was incorporated at the interface of the inorganic silica matrix and organic environment (pore template), and thus insures the stability of the dye–silica hybrid structure. The resulting fluorescent silica materials have a number of properties that make them attractive for biomedical applications: the availability of various color of the resulting nanoparticle from among a broad spectrum of commercially dyes, the controllablity of pore size (diameters of ~5 nm) and particle size (diameters of ~40 nm) by adjusting template monomer concentration and the water/oil ratio, and the stability and durability of particle fluorescence because of the deep insertion of surfactant’s tail into the silica matrix.

Photophysical properties and fluorescence lifetime imaging of exfoliated near-infrared fluorescent silicate nanosheets

Abstract:The layered silicates Egyptian Blue (CaCuSi4O10, EB), Han Blue (BaCuSi4O10, HB) and Han Purple (BaCuSi2O6, HP) emit as bulk materials bright and stable fluorescence in the near-infrared (NIR), which is of high interest for (bio)photonics due to minimal scattering, absorption and phototoxicity in this spectral range. So far the optical properties of nanosheets (NS) of these silicates are poorly understood. Here, we exfoliate them into monodisperse nanosheets, report their physicochemical properties and use them for (bio)photonics. The approach uses ball milling followed by tip sonication and centrifugation steps to exfoliate the silicates into NS with lateral size and thickness down to ≈ 16-27 nm and 1-4 nm, respectively. They emit at ≈ 927 nm (EB-NS), 953 nm (HB-NS) and 924 nm (HP-NS), and single NS can be imaged in the NIR. The fluorescence lifetimes decrease from ≈ 30-100 μs (bulk) to 17 μs (EB-NS), 8 μs (HB-NS) and 7 μs (HP-NS), thus enabling lifetime-encoded multicolor imaging both on the microscopic and the macroscopic scale. Finally, remote imaging through tissue phantoms reveals the potential for bioimaging. In summary, we report a procedure to gain monodisperse NIR fluorescent silicate nanosheets, determine their size-dependent photophysical properties and showcase the potential for NIR photonics.
Synthesis of Highly Luminescent Silica-Coated Upconversion Nanoparticles from Lanthanide Oxides or Nitrates Using Co-Precipitation and Sol–Gel Methods

Abstract:Upconversion nanoparticles (UCNPs) are under consideration for their use as bioimaging probes with enhanced optical performance for real time follow-up under non-invasive conditions. Photostable and core-shell NaYF4:Yb3+,Er3+-SiO2 UCNPs obtained by a novel and simple co-precipitation method from lanthanide nitrates or oxides were herein synthesized for the first time. The sol–gel Stöber method followed by oven or supercritical gel drying was used to confer biocompatible surface properties to UCNPs by the formation of an ultrathin silica coating. Upconversion (UC) spectra were studied to evaluate the fluorescence of UCNPs upon red/near infrared (NIR) irradiation. ζ-potential measurements, TEM analyses, XRD patterns and long-term physicochemical stability were also assessed and confirmed that the UCNPs co-precipitation synthesis is a shape- and phase-controlling approach. The bio- and hemocompatibility of the UCNPs formulation with the highest fluorescence intensity was evaluated with murine fibroblasts and human blood, respectively, and provided excellent results that endorse the efficacy of the silica gel coating. The herein synthesized UCNPs can be regarded as efficient fluorescent probes for bioimaging purposes with the high luminescence, physicochemical stability and biocompatibility required for biomedical applications.

Abstract:Optical near-infrared (NIR) nanomaterials provide a unique opportunity for applications in bioimaging and medical diagnosis. A kind of hydrophilic NIR fluorescent core-shell structured silica nanoparticle containing NIR cyanine chromophore, named as CyN-12@NHs, for in vivo bioimaging is developed through a facile one-pot strategy. The hydrophobic CyN-12 molecules can be successfully encapsulated into the core via the self-assembly of the amphiphilic block copolymer PS-b-PAA and subsequent shell cross-linking of silane. The as-prepared CyN-12@NHs exhibits typically spherical core-shell structure, which has a uniform size of 35 nm with a narrow size distribution, and excellent dispersity in aqueous solution. Moreover, NIR absorption (690 nm) and bright fluorescence (800 nm) of CyN-12@NHs with a large Stokes shift (110 nm) in aqueous system make it an amenable high quality bioimaging contrast agent. The core-shell nanostructure significantly enhances the chemical and photo-stability of CyN-12 via the encapsulation, which possesses a 50-times longer half-life period than free CyN-12, along with a better resistance to reactive oxygen species (ROS). Furthermore, in living cell imaging, CyN-12@NHs shows nearly no cytotoxicity and is able to outline the HepG2 cells. The in vivo imaging on a tumor-bearing mouse model indicates that CyN-12@NHs selectively accumulates in the liver after intravenous injection, and has a long retention in tumor after intra-tumor injection without decrease in fluorescence activity. Overall, the excellent photo-properties of CyN-12@NHs could meet the intricate requirements for tumor imaging, such as high sensitivity, sufficient tissue penetration, and high spatial resolution. The strategy of the silica-cyanine hybrid nanoparticles paves a desirable and efficient route to fabricate highly hydrophilic NIR fluorescent contrast agents for tumor imaging and therapy, especially with a breakthrough in photo-stability, bright fluorescence as well as large Stokes shift.

Abstract:The synthesis of ultrabright fluorescent mesoporous silica nanoparticles (UFSNPs) of various sizes loaded with different amounts of fluorescent dye (Rhodamine 6G) is reported here. The dye is physically entrapped inside the nanochannels of the silica matrix created during templated sol–gel self assembly. Due to the specific nanoenvironment, the fluorescence of the encapsulated dye molecules remains unquenched up to very high concentrations, which results in relatively high fluorescence. The particle size (ranging from 20–50 nm) and dye loading (0.8–9.3 mg dye per g particles) are controlled by the timing of the synthesis and the concentration of several organotriethoxysilanes, which are coprecursors of silica. The quantum yields of the encapsulated dye range from 0.65 to 1.0. The relative brightness of a single particle is equivalent to the fluorescence of 30–770 free nondimerized R6G dye molecules in water, or to that of 1.5–39 CdSe/ZnS quantum dots. Despite the presence of some hydrophobic groups on the particles' surfaces, colloidal suspensions of the particles are relatively stable (as monitored for 120 days). Ultrabright fluorescent mesoporous silica nanoparticles of various sizes (20–50 nm) with different amounts of encapsulated fluorescent dye (Rhodamine 6G) are reported. Here, a water-soluble, stable, highly-luminescent nanosystem, Ir-Eu-MSN (MSN = mesoporous silica nanoparticles, Ir-Eu = [Ir(dfppy)(2)(pic-OH)](3)Eu center dot 2H(2)O, dfppy = 2-(2,4-difluorophenyl)pyridine, pic-OH = 3-hydroxy-2-carboxypyridine), was developed by an in situ coordination reaction to form an insoluble dinuclear iridium(III) complex-sensitized-europium(III) emissive complex within mesoporous silica nanoparticles (MSNs) which had high loading efficiency. Compared with the usual approach of physical adsorption, this in-situ reaction strategy provided 20-fold the loading efficiency (43.2%) of the insoluble Ir-Eu complex in MSNs. These nanoparticles in solid state showed bright red luminescence with high quantum yield of 55.2%, and the excitation window extended up to 470 nm. These Ir-Eu-MSN nanoparticles were used for luminescence imaging in living cells under excitation at 458 nm with confocal microscopy, which was confirmed by flow cytometry. Furthermore, the Ir-Eu-MSN nanoparticles were successfully applied into high-contrast luminescent lymphatic imaging in vivo under low power density excitation of 5 mW cm(-2). This synthetic method provides a universal strategy of combining hydrophobic complexes with hydrophilic MSNs for in vivo bioimaging. (C) 2014 Elsevier Ltd. All rights reserved.
Development of Mesoporous Silica-Based Nanoprobes for Optical Bioimaging Applications

Abstract:A mesoporous silica nanoparticle (MSN)-based nanoplatform has attracted growing attention in the biomedical field due to the unique characteristics of MSNs including a high surface area, tunable pore sizes, colloidal stability, ease of functionalization, and desirable biocompatibility. Typically, MSNs are designed as nanocarriers for the incorporation of a variety of contrast agents for bioimaging, which can address the intrinsic drawbacks of contrast agents, including poor solubility in water, rapid photobleaching, and low stability. This review summarizes the recent advances in the field of MSN-based nanoprobes for fluorescence imaging and photoacoustic (PA) imaging applications. The approaches for the incorporation of contrast agents into MSN-based nanoplatforms including encapsulating contrast agents within MSNs, covalently conjugating contrast agents on the surface or pores of MSNs, physically absorbing contrast agents in the pores of MSNs, and doping contrast agents in the framework of MSNs are introduced. MSN-based nanoprobes for fluorescence imaging and PA imaging are discussed. The enhanced fluorescence imaging and PA imaging performances of MSN-based nanoprobes relative to the bare contrast agents are introduced and the underlying mechanisms are discussed in detail. Finally, current challenges and perspectives of MSN-based nanoprobes in the bioimaging field are discussed.
