Abstract:A phosgene- and metal-free synthesis of unsymmetrical arylurea derivatives utilizing 3-substituted dioxazolones and commercially available amines has been developed. The 3-substituted dioxazolones serve as the precursors of thein situgenerated isocyanate intermediates in the presence of non-toxic sodium acetate as a base and methanol as a solvent under mild heating conditions. A series of unsymmetrical phenylureas includingN, N'- mono-, di-, and trisubstituted derivatives are chemoselectively obtained in moderate to excellent yields from a broad scope of both 3-substituted dioxazolones and amines without a significant amount of the symmetrical byproducts. In many cases, the resulting unsymmetrical phenylureas are easily separated by filtration allowing no chromatographic purification. These investigations provide an opportunity for facile, practical, and eco-friendly synthesis of unsymmetrical diarylureas. Application of this method is demonstrated toward the gram-scale preparation of anti-cancer drugs, sorafenib and the 3-cycle continuous synthesis in conjugation with the recycle of sodium acetate and methanol process of known herbicide, daimuron.

Abstract:The preparation of a series of 2-pyridinamines and their alkyl derivatives is described.The target compounds are synthesized starting from corresponding 2-cyanopyridines, via incomplete hydrolysis in the presence of hydrogen peroxide in the dilute alkaline mixture of acetone-2% sodium hydroxide solution, and Hoffmann degradation reaction with freshly made NaBrO.This route is of industrial value because of cheap and readily available materials, moderate reaction conditions and convenient operations.

Abstract:A general and facile one-pot protocol for the preparation of a broad range of alkyl and aryl isothiocyanates has been developed from their corresponding primary amines under aqueous conditions. This synthetic process involves an in situ generation of a dithiocarbamate salt from the amine substrate by reacting with CS(2) followed by elimination to form the isothiocyanate product with cyanuric acid as the desulfurylation reagent. The choice of solvent is of decisive importance for the successful formation of the dithiocarbamate salt particularly for highly electron-deficient substrates. This novel and economical method is suitable for scale-up activities.

Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:An efficient and straightforward procedure toward the synthesis of amides-substituted 2H-benzopyrans H-benzopyrans has been explored through palladium-catalyzed one-pot aminocarbonylation reaction. By using nitro compounds as attractive nitrogen sources, with Mo(CO)6 6 as both CO source and reductant, a variety of amides-containing 2H- H- benzopyrans were obtained in moderate to high yields with good functional group tolerance. This protocol has advantages including atom-economy, no manipulation of toxic CO gas, and broad substrate scope.

Abstract:The nucleophilic difluoroalkylation of isocyanates with difluoromethyl 2-pyridyl sulfone furnished 2,2-difluoro-2-pyridinylsulfonylacetamides with good to excellent yield. Abstract:A novel tandem reductive amination/intermolecular nucleophilic aromatic substitution (SNAr) sequence has been established for the synthesis of amine containing pyrimidine in formation of one carbon-oxygen and one carbon-nitrogen bonds in a one-pot fashion. Treatment of aldehyde with arylamine, 2-methanesulfonyl-4,6-dimethoxypyrimidine and sodium borohydride provides good overall yield. The p-toluenesulfonic acid (PTSA) can be used as activator and is generally needed in the reaction. Dioxane is the preferred reaction solvent, but reactions can also be carried out in tetrahydrofuran (THF), MeCN, toluene and dichloromethane. The procedure is carried out effectively in the presence of K2CO3. The reaction proceeds smoothly with aromatic aldehydes and arylamines possessing electron-donating or -withdrawing groups. This method can be applied to the synthesis of the oilseed rape herbicide and is superior to the classical one in several aspects: cutting out several purification steps, minimizing solvent use and chemical waste, and saving time. Its advantages such as operational convenience, high-efficient synthesis, and starting material availability make it a desirable method for preparing amines with molecular diversity and biological activity.

Abstract:Tandem reactions for the efficient synthesis of multifunctionalized 1,2,3,4-tetrahydropyridines, 2,3-dihydropyridin-4(1H)-ones, and pyridine derivatives have been developed and reaction mechanisms have been investigated. Synthetic cascades are initiated by the Zn(OTf)2-mediated [5+1] cycloaddition of N-formylmethyl-substituted tertiary enamides to isocyanides, thus leading to the versatile heterocyclic enamino imine intermediates. Interception of the intermediates by diastereoselective reduction of imine functionality with Me4NBH(OAc)3 afforded 1,6-disubstituted trans-3-hydroxy-4-arylamino- or -alkylamino-1,2,3,4-tetrahydropyridines, whereas acylation of the imino group followed by acidic hydrolysis produced 1,6-disubstituted 3-acyloxy-2,3-dihydropyridin-4(1H)-ones. Aerobic oxidation led to the aromatization followed by intermolecular acyl-group transfer from the pyridinium nitrogen to the 3-hydroxy moiety, thereby yielding substituted 3-acyloxy-4-aminopyridines. Synthetic potentials of the resulting products have been demonstrated by expedient and highly stereoselective synthesis of cis,cis-4,5-dihydroxy-2-phenylpiperidine and trans,trans-4-amino-5-hydroxy-2-phenylpiperidine compounds, which are important in medicinal chemistry, through simple and practical reduction reactions.

Abstract:Herein, we introduce a novel method for the synthesis of S‐substituted isothiourea compounds via the selective 1,2‐carboimination between oxime esters as bifunctional reagents and the C=S bond of isothiocyanates under visible‐light catalysis. This approach deviates from conventional methods by precisely modulating the substrate electronics to selectively functionalize the C=S bond of isothiocyanates over the C=N bond, eliminating the need for strong bases and high temperatures and bypassing the formation of thiourea intermediates. Consequently, this protocol enables the efficient one‐step synthesis of S‐alkyl isothioureas, with featuring mild reaction conditions, operational simplicity and broad substrate scope.

Abstract:An efficient procedure for the synthesis of 7‐(aryl)‐8‐nitro‐2,3,6,7‐tetrahydroimidazo[1,2‐a]pyridinones, 8‐(aryl)‐9‐nitro‐3,4,7,8‐tetrahydropyridone[1,2‐a]pyrimidines and 9‐(aryl)‐10‐nitro‐2,3,4,5,8,9‐hexahydropyridone[1,2‐a]diazepine via one‐pot three component reaction of diamine, nitroketene dithioacetal (1,1‐bis(methylsulfanyl)‐2‐nitroethene), and coumarine‐3‐ carboxylic acid derivatives in EtOH under reflux conditions is reported. The advantages of this procedure are simplicity, easy purification, good yields, and catalyst‐free conditions. All products were confirmed by 1H‐ and 13C‐NMR, IR, MS, and X‐ray crystal structure analyses.

Abstract:This article presents an efficient one-step synthesis of tetrahydropyrazolo[1,5- a ]pyrimidines through the recyclization of N -arylitaconimides with aminopyrazoles. The heterocyclic system of pyrazolo[3,4- b ]pyrimidine is known for its diverse biological properties, making its derivatives significant in pharmaceutical and medicinal chemistry. The study focuses on the regio- and chemoselective cascade reaction of N -arylitaconimides with 5-aminopyrazoles, demonstrating a new approach to synthesizing pyrazolo[1,5- a ]pyrimidines and pyrazolo[3,4- b ]pyridines. The methodology, involving boiling in isopropyl alcohol with acetic acid, yields selectively either pyrazolo[3,4- b ]pyridines or pyrazolo[1,5- a ]pyrimidines based on the substituents in the aminopyrazoles. The study elucidates the reaction mechanism, structural characterization using NMR spectroscopy, and confirms the structures via high-performance liquid chromatography and mass spectrometry. The simplicity and synthetic potential of this approach make it a valuable method for the preparation of these heterocyclic frameworks. Abstract:Previously synthesized ethyl 2-benzamido-4-(1,4-benzodioxan-2-yl)thiophene-3-carboxylates were cyclized to 2-aryl-3-aminothieno[2,3- d ]pyrimidin-4(1 H )-ones by the action of hydrazine hydrate. The condensation of ethyl 2-amino-4-(1,4-benzodioxan-2-yl)thiophene-3-carboxylate with chloroacetyl chloride gave the corresponding N-substituted 2-chloroacetamide which reacted with piperidine and morpholine to produce 2-aminoacetamides, and the latter were also subjected to cyclization with hydrazine hydrate to obtain 3-aminothienopyrimidin-4-one derivatives. The synthesized compounds were tested for antihypoxic activity.

Abstract:The multicomponent reaction of 2-nitroacetophenone (or nitroacetone), acetaldehyde diethyl acetal, β-dicarbonyl compound, and ammonium acetate in an acetic acid solution allowed the acquisition of previously undescribed 4-methyl-substituted derivatives of 5-nitro-1,4-dihydropyridine in satisfactory yields. The oxidation of the obtained 5-nitro-1,4-dihydropyridine derivatives resulted in the corresponding 2,4-dimethyl-5-nitropyridines. In addition, for the first time in the synthesis of unsymmetrical 1,4-dihydropyridines by the Hantzsch reaction acetaldehyde, diethyl acetal was used as a source of acetaldehyde. The use of more volatile and sufficiently reactive acetaldehyde in this reaction did not lead to a controlled synthesis of unsymmetrical 5-nitro-1,4-dihydropyridines. The proposed multicomponent approach to the synthesis of 4-methyl-substituted 5-nitro-1,4-dihydropyridines and their subsequent aromatization into pyridines made it possible to obtain previously undescribed and hardly accessible substituted 5(3)-nitropyridines.

Abstract:In this paper, a novel synthetic method for 4-N-substituted 6-bromopyrido[2,3-d]pyrimidines was reported. Starting from 2-aminonicotinonitrile, following by bromination of aromatic ring, condensation, cyclization, and Dimroth rearrangement, a series of 21 new pyrido[2,3-d]pyrimidine derivatives was prepared in high yields (up to 98%). In the last two steps, microwave irradiation was used. The structures of synthesized compounds were determined by NMR, IR, and HRMS techniques. The results showed that application of microwave irradiation has a beneficial effect for the preparation of desired products, as it is simple and efficient method for improving the yields and reducing the formation of undesirable by-products.

Abstract:We report an efficient method for the synthesis of C(1)-carboxamide derivatives by adding isocyanides to 3,4-dihydroisoquinoline N-oxides and 3,4-dihydro-β-carboline 2-oxide in the presence of TMSOAc. 3,4-Dihydroisoquinoline-1-carboxylamide derivatives and 9-dihydro-3H-pyrido[3,4-b]indole-1-carboxamide derivatives were obtained in reasonable yields. The method could be used to synthesize alangiobussine I, an alkaloid, in 61% yield.
