Abstract:Rh(III)-catalyzed C-H functionalization of 7-arylpyrazolo[1,5-a]pyrimidines was developed wherein the pyrazolo[1,5-a]pyrimidine moiety is reported for the first time to direct the C-H bond activation. Various 7-arylpyrazolo[1,5-a]pyrimidines underwent smooth C-H amidation with alkyl-, aryl-, and heteroaryl-substituted dioxazolones to afford the products in moderate to good yields. Mechanistic studies suggest that a six-membered rhodacycle intermediate involving N1 might play a key role in the regioselective catalytic cycle.

Abstract:A new, operationally simple one-pot method has been developed for the regioselective synthesis of a series of novel 4-olefinated dihydropyridines bearing various functional groups from the reaction of pyridine, acylacetamides, and sulfuryl chlorides under mild conditions. In this multicomponent process, subsequent the activation of pyridine by sulfuryl chloride/nucleophilic addition of acylacetonitrile formed in situ/enolization/sulfonylation leads to the formation of final products.

Abstract:Autocatalytic and DMSO-participating regioselective synthesis of N , N -disubstituted pyrimido[4,5- b ]quinoline-2,4(1 H ,3 H )-diones from anilines and barbituric acids has been achieved. In this newly developed one-pot tandem reaction, DMSO serves as a solvent cum methine source. Additionally, barbituric acid plays a dual role by acting as a substrate and a catalyst, making this reaction an environmentally benign approach to accessing valuable heterocycles. This method offers an auto-catalytic, additive-free, and operationally simple approach with a wide substrate scope and excellent tolerance for various functional groups. Furthermore, a few controlled experiments were conducted to gain insight into the reaction mechanism. Moreover, large-scale experiments have further enriched this methodology.

Abstract:A three-component reaction of one molecule of imine and two molecules of alkynoates is realized with the catalysis of Cu(I)–Fe(III) in a sequential manner to allow the direct synthesis of functionalized pyrroles, during which, one C–N bond and two C–C bonds are formed with high atom economy. This method benefits from easily available starting materials, low-cost catalysts, and convenient operations.

Abstract:A transition-metal- and oxidant-free amination/cyclization reaction to access 1,2,4-triazolo[1,5-a]pyridines was realized in water by using amino diphenylphosphinate as amino source. A broad array of readily accessible N-(pyridyl)amides could be converted into the products featuring a diverse set of functional groups. The sustainable methodology was successfully applied to the late-stage functionalization of natural products and drugs.

Abstract:A series of polysubstituted pyrroles were rapidly synthesized in moderate to good yields via catalyst-free one-pot three-component reaction of 1,3-dicarbonyl compounds with primary amines and nitrostyrenes by using liquid-assisted grinding. This protocol provides several advantages over the conventional chemical synthesis, such as simple work-up procedure, wide substrate scope, short reaction time, and environmental friendliness. Structures of polysubstituted pyrroles were confirmed by H-1 NMR, C-13 NMR, and ESI-MS studies.

Abstract:A novel synthesis method has been developed for the construction of functionalized 2,4-diarylpyridines. The reaction involves several key processes, including a reduction reaction, Michael reaction, decarboxylation, intramolecular cyclization, oxidation, ring-opening and aromatization reaction. All of these processes are facilitated by heating a mixture of the starting materials in the environmentally benign solvent, propylene carbonate (PC), with the oxidant (air) and the catalyst Cu(OAc)2. This cascade reaction not only forms two new bonds but also cleaves three existing bonds in a single pot. The protocol presented in this study is a valuable tool for synthesizing functionalized unsymmetrical 2,4-diarylpyridines.

Abstract:A facile, expeditious, user‐friendly, and convenient metal‐free synthesis employing base catalysis in a one‐pot procedure to construct 11H‐dipyrido[1,2‐a : 3′,2′‐d]pyrimidin‐11‐imine derivatives is reported. This protocol involves a domino process leading to the formation of double C−N bonds utilising KOtBu as the base and DMAc as the superior solvent at 25 °C for 2 h. We report a facile, expeditious, user‐friendly, and convenient metal‐free synthesis employing base catalysis in a one‐pot procedure to construct 11H‐dipyrido[1,2‐a : 3′,2′‐d]pyrimidin‐11‐imine derivatives. This protocol involves a domino process leading to the formation of double C−N bonds utilising KOtBu as the base and DMAc as the superior solvent at 25 °C for 2 h. The versatility of this methodology was demonstrated by its successful application to substrates with both electron‐withdrawing and electron‐donating functional groups, yielding novel functionalized stable 11H‐dipyrido[1,2‐a : 3′,2′‐d]pyrimidin‐11‐imine derivatives in good to excellent yields. Additionally, we have discussed a plausible reaction pathway for the synthesis.

Abstract:In this study, a novel functionalized metal–organic frameworks MIL-125(Ti)-N(CH 2 PO 3 H 2 ) 2 was designed and synthesized via post-modification methodology. Then, MIL-125(Ti)-N(CH 2 PO 3 H 2 ) 2 as a mesoporous catalyst was applied for the synthesis of a wide range of novel tetrahydropyrido[2,3- d ]pyrimidines as bioactive candidate compounds by one-pot condensation reaction of 3-(1-methyl-1 H -pyrrol-2-yl)-3-oxopropanenitrile, 6-amino-1,3-dimethylpyrimidine-2,4(1 H ,3 H )-dione and aromatic aldehydes at 100 °C under solvent-free condition. Interestingly, the preparation of tetrahydropyrido[2,3- d ]pyrimidine was achieved via vinylogous anomeric based oxidation mechanism with a high yield and short reaction time.

DMSO plays a dual role as the C<sub>1</sub> source and the reaction medium. Abstract:A biologically inspired organocatalytic one-pot synthesis of highly functionalized pyridazines, which are ubiquitous structural units in a number of biologically active compounds, has been developed by starting from readily available diazo compounds and Morita-Baylis-Hillman (MBH) carbonates. Under mild reaction conditions, this synthetic route tolerated significant substrate variation to deliver a broad range of substituted products, including CF3 -substituted pyridazines derivatives. Moreover, the introduction of trifluoromethyl groups into the ring of pyridazine could be completed conveniently from 2,2,2-trifluorodiazoethane.

Abstract:A microwave assisted tandem Heck-Sonogashira cross-coupling reaction between 6-N,N-di-Boc-amino-5-iodo-2-methyl pyrimidin-4-ol and various aryl alkynyl substrates has been developed. Abstract:An iron(III)-porphyrin-catalyzed cyclization of ketones with dimethyl sulfoxide (DMSO) and ammonium acetate for the synthesis of unsymmetrical and symmetrical pyridines by employing DMSO as C-4 or C-6 source has been developed. Various aryl ketone derivatives react readily with DMSO, producing the pyridines in yields of 30%similar to 85%. This method uses non-noble metals and proceeds under mild reaction conditions with operational simplicity, which thus allows the expedient assembly of pyridines from readily available ketones. Based on the preliminary experiments, a plausible mechanism of this transformation is disclosed.

Abstract:Ruthenium‐catalyzed and K2S2O8‐mediated synthesis of diverse sulfonylated 5‐aroyl‐2‐arylpyridines was developed through one‐pot stepwise annulation of two molecules of β‐ketosulfones and N,N‐dimethylformamide (DMF) under the sealed tube conditions. In the overall process, DMF acts as the synthon of one carbon and one imino moiety in the construction of pyridine skeleton via cascade formation of single (C‐C/C‐N) and double (C=C/C=N) bonds under refluxing DMF conditions. Plausible reaction mechanism is proposed and discussed.

Abstract:Fused pyrimidines composed of alternating heteroatoms and a pyrimidine moiety were synthesized efficiently using readily available starting material 4-hydroxycoumarin, aromatic aldehydes, and urea/thiourea at room temperature. Acid, metal salts, and surfactants were screened for their influence on catalytic activity in three-component reactions and sodium lauryl sulphate (SLS) was used as the best catalyst with different concentrations. Screening results of catalyst loading from our investigation showed that good to excellent yields were obtained with 10 mol%. Our method efficiently synthesized heterocycles and avoided the use of hazardous solvents and conventional organic solvents. Our procedure which involves a surfactant is operationally simple, environmentally benign, has excellent yields, short reaction times, and synthetically is as efficient as conventional procedures using organic solvents.
