In Vitro Evaluation and Characterization of Double Emulsion Based on Polyvinylpyrrolidone/Agarose/ZIF-8 Hydrogel Nanocomposite Platform for Quercetin Delivery to Breast Cancer Cells

Abstract:Quercetin (QC) is a matchable option for cancer remedy, lessens the issues relevant to low solvability, and its quick release has become urgent. This research used a newfound nanocarrier based on a hydrogel nanocomposite, including polyvinylpyrrolidone (PVP), agarose (Aga), and ZIF-8 nanoparticles to load QC. Being of nanocomposite ingredients was authenticated by FTIR and XRD analysis. The surface correlation of the nanocomposite and stability of the nanocarrier produced was approved by FESEM images, Zeta, and DLS analysis, consecutively. With ZIF-8, drug loading is 47.50%, and encapsulation is 86.75%. Prolonged pH-sensitive release lasted about 100 h. Based on the R 2 amounts of the models employed to correspond to the drug release data, the Higuchi model can well explain release data at pH 5.4. The reduction in a living MCF-7 cell in the presence of PVP/Aga/ZIF-8 samples compared to PVP/Aga samples and the control sample portends the ZIF-8 in vitro cytotoxicity. The percent of late apoptotic cells in PVP/Aga/ZIF-8@QC (14.0%) is higher than in other samples (Control, PVP/Aga/ZIF-8), which may be owing to the controlled and slow release of QC from PVP/Aga/ZIF-8@QC.
