Abstract:Plants generally accumulate free proline under osmotic stress conditions. Upon removal of the osmotic stress, the proline levels return to normal. In order to understand the mechanisms involved in regulating the levels of proline, we cloned and characterized a proline dehydrogenase (PDH) cDNA from Arabidopsis thaliana (AtPDH). The 1745 bp cDNA contains a major open reading frame encoding a peptide of 499 amino acids. The deduced amino acid sequence has high homology with both Saccharomyces cerevisiae and Drosophila melanogaster proline oxidases and contains a putative mitochondrial targeting sequence. When expressed in yeast, the AtPDH cDNA complemented a yeast put1 mutation and exhibited proline oxidase activity. We also determined the free proline contents and the delta 1-pyrroline-5-carboxylate synthetase (P5CS) and PDH mRNA levels under different osmotic stress and recovery conditions. The results demonstrated that the removal of free proline during the recovery from salinity or dehydration stress involves an induction of the PDH gene while the activity of P5CS declines. The reciprocal regulation of P5CS and PDH genes appears to be a key mechanism in the control of the levels of proline during and after osmotic stress. The PDH gene was also significantly induced by exogenously applied proline. The induction of PDH by proline, however, was inhibited by salt stress.
Overexpression of AtHsp90.2, AtHsp90.5 and AtHsp90.7 in Arabidopsis Thaliana Enhances Plant Sensitivity to Salt and Drought Stresses

Abstract:Three AtHsp90 isoforms, cytosolic AtHsp90.2, chloroplast-located AtHsp90.5, and endoplasmic reticulum (ER)-located AtHsp90.7, were characterized by constitutive overexpressing their genes in Arabidopsis thaliana . Both types of the transgenic plants overexpressing cytosolic and organellar AtHsp90s showed reduced tolerance to salt and drought stresses with lower germination rates and fresh weights, but improved tolerance to high concentration of Ca 2+ comparing with the wild type plants. Transcriptional analysis of ABA-responsive genes, RD29A , RD22 and KIN2 under salt and drought stresses, indicated that the induction expression of these genes was delayed by constitutive overexpression of cytosolic AtHsp90.2, but was hardly affected by that of organellar AtHsp90.5 and AtHsp90.7. These results implied that Arabidopsis different cellular compartments-located Hsp90s in Arabidopsis might be involved in abiotic stresses by different functional mechanisms, probably through ABA-dependent or Ca 2+ pathways, and proper homeostasis of Hsp90 was critical for cellular stress response and/or tolerance in plants.

Abstract:Plants respond to conditions of severe environmental changes or stresses, such as dehydration, low temperature, high-salinity with a number of physiological and developmental changes. We have isolated more than thirty genes that respond to dehydration stresses at transcriptional level. In the present study, we will report the expression and function of differentArabidopsis genes involved in the kinase cascade and in the proline biosynthesis. MAP kinases play key roles in the signalling pathway activated by growth factors and in stress response in animals. The mRNA levels of three protein kinases, ATMEKK1 (MAP kinase kinase kinase), ATMPK3 (MAP kinase) and ATPK19 (ribosomal S6 kinase), increased markedly and simultaneously in response to touch, cold and salinity stresses. These results suggest that MAP kinase cascades, which are thought to respond to a variety of extracellular signals, are regulated not only at the post-translational level but also at the transcriptional level in plants. It is also suggested that MAP kinase cascades in plants may function in transducing signals under environmental stresses. Proline is thought to be a compatible osmolyte like glycine betaine, which plays a role in counteracting the effects of osmotic stress in plants. We have isolated two cDNAs fromArabidopsis (cAtPSCS andcAtPSCR) encoding P5CS (Δ1-pyrroline-5-carboxylate synthetase) and P5CR (Δ1-pyrroline-5-carboxylate reductase) which play key roles in the biosynthesis of proline in plants under water stress. Northern blot analysis revealed that theAtP5CS gene was induced by dehydration, high salt and treatment with ABA. Moreover, a simultaneous accumulation of proline was observed as a result of these treatments ofArabidopsis plants. By contrast, theAtPSCR gene was not induced to a significant extent by dehydration or high-salt stress. These observations suggest that theAtP5CS gene plays a principal role in the biosynthesis of proline inArabidopsis under osmotic stress.

Abstract:Orange (OR) is a DnaJ-like zinc finger protein with both nuclear and plastidial localizations. OR, and its orthologs, are highly conserved in flowering plants, sharing a characteristic C-terminal tandem 4× repeats of the CxxCxxxG signature. It was reported to trigger chromoplast biogenesis, promote carotenoid accumulation in plastids of non-pigmented tissues, and repress chlorophyll biosynthesis and chloroplast biogenesis in the nucleus of de-etiolating cotyledons cells. Its ectopic overexpression was found to enhance plant resistance to abiotic stresses. Here, we report that the expression of OR in Arabidopsis thaliana was upregulated by drought treatment, and seedlings of the OR-overexpressing (OE) lines showed improved growth performance and survival rate under drought stress. Compared with the wild-type (WT) and OR-silencing (or) lines, drought-stressed OE seedlings possessed lower contents of reactive oxygen species (such as H2O2 and O2−), higher activities of both superoxide dismutase and catalase, and a higher level of proline content. Our enzymatic assay revealed a relatively higher activity of Δ1-pyrroline-5-carboxylate synthase (P5CS), a rate-limiting enzyme for proline biosynthesis, in drought-stressed OE seedlings, compared with the WT and or lines. We further demonstrated that the P5CS activity could be enhanced by supplementing exogenous OR in our in vitro assays. Taken together, our results indicated a novel contribution of OR to drought tolerance, through its impact on proline biosynthesis.

Light Control of Salt-Induced Proline Accumulation Is Mediated by ELONGATED HYPOCOTYL 5 in Arabidopsis

Abstract:Plants have to adapt their metabolism to constantly changing environmental conditions, among which the availability of light and water is crucial in determining growth and development. Proline accumulation is one of the sensitive metabolic responses to extreme conditions; it is triggered by salinity or drought and is regulated by light. Here we show that red and blue but not far-red light is essential for salt-induced proline accumulation, upregulation of Δ1-PYRROLINE-5-CARBOXYLATE SYNTHASE 1 (P5CS1) and downregulation of PROLINE DEHYDROGENASE 1 (PDH1) genes, which control proline biosynthetic and catabolic pathways, respectively. Chromatin immunoprecipitation and electrophoretic mobility shift assays demonstrated that the transcription factor ELONGATED HYPOCOTYL 5 (HY5) binds to G-box and C-box elements of P5CS1 and a C-box motif of PDH1. Salt-induced proline accumulation and P5CS1 expression were reduced in the hy5hyh double mutant, suggesting that HY5 promotes proline biosynthesis through connecting light and stress signals. Our results improve our understanding on interactions between stress and light signals, confirming HY5 as a key regulator in proline metabolism.

Abstract:Many plants synthesize and accumulate proline in response to osmotic stress. Despite the importance of this pathway, however, the exact metabolic route and enzymes involved in the synthesis of proline in plants have not been unequivocally identified. We report here the isolation of a mothbean (Vigna aconitifolia) cDNA clone encoding a bifunctional enzyme, delta 1-pyrroline-5-carboxylate synthetase (P5CS), with both gamma-glutamyl kinase and glutamic-gamma-semialdehyde dehydrogenase activities that catalyzes the first two steps in proline biosynthesis. The two enzymatic domains of P5CS correspond to the ProB and ProA proteins of Escherichia coli and contain a leucine zipper in each domain, which may facilitate inter- or intramolecular interaction of this protein. The Vigna P5CS enzyme activity is feedback regulated by proline but is less sensitive to end-product inhibition than is the E. coli gamma-glutamyl kinase. The P5CS gene is expressed at high levels in Vigna leaves and is inducible in roots subjected to salt stress, suggesting that P5CS plays a key role in proline biosynthesis, leading to osmoregulation in plants.
Enhanced drought tolerance with artificial microRNA-mediated StProDH1 gene silencing in potato

Abstract:Proline is a multifunctional molecule, which especially is associated with response to abiotic stress. The proline dehydrogenase (ProDH) is the rate-limiting enzyme in the process of proline catabolism. In the present study, two potato ProDH genes, StProDH1 and StProDH2, were cloned from potato (Solanum tuberosum L.) cultivar 'Favorita'. Gene expression analysis indicated that these two genes exhibited tissue-specific expression and responded differently to various stress conditions, and that the StProDH1 plays a predominant role in most of these conditions. An artificial microRNA expression vector, under the control of CaMV 35S promoter, was constructed to generate transgenic potato plants by Agrobacterium-mediated transformation, thereby silencing the StProDH1 gene. The result showed that the artificial microRNA caused a decline in StProDH1 expression, which was accompanied by a concomitant increase in proline content in transgenic plants under drought stress. The transgenic potato plants showed normal phenotypes under drought stress, while the non-transgenic plants appeared withered. Results of physiological parameters assays showed that malondialdehyde, relative water content, and chlorophyll content in the transgenic potato plants were higher than that of non-transgenic plants. Upon rehydration after two weeks of drought, proline and soluble sugar contents of the transgenic potato plants were significantly higher than those of non-transgenic plants. These results combined demonstrate that the StProDH1 is a key player in the process of potato response to drought stress.
Functional Characterization of the Stipa purpurea P5CS Gene under Drought Stress Conditions

Abstract:Free proline has multiple functions in plant cells, such as regulating osmotic potential and protecting both proteins and cell membranes. The expression of Δ1-Pyrroline-5-carboxylate synthase (P5CS), a key enzyme in the proline biosynthetic pathway, increases under drought, salt and cold stress conditions, causing plant cells to accumulate large amounts of proline. In this study, we cloned and identified the P5CS gene from Stipa purpurea, which has a full-length of 2196 bp and encodes 731 amino acids. A subcellular localization analysis indicated that SpP5CS localized to the cytoplasm. The ectopic overexpression of SpP5CS in Arabidopsis thaliana resulted in higher proline contents, longer roots, higher survival rates and less membrane damage under drought stress conditions compared with wild-type controls. SpP5CS-overexpressing A. thaliana was more resistant to drought stress than the wild type, whereas the deletion mutant sp5cs was less resistant to drought stress. Thus, SpP5CS may be a potential candidate target gene for increasing plant resistance to drought stress.

Rhamnogalacturonan lyase 1 (RGL1), as a suppressor of E3 ubiquitin ligase Arabidopsis thaliana ring zinc finger 1 (AtRZF1), is involved in dehydration response to mediate proline synthesis and pectin rhamnogalacturonan-I composition

Abstract:Proline metabolism plays a crucial role in both environmental stress responses and plant growth. However, the specific mechanism by which proline contributes to abiotic stress processes remains to be elucidated. In this study, we utilized atrzf1 (Arabidopsis thaliana ring zinc finger 1) as a parental line for T-DNA tagging mutagenesis and identified a suppressor mutant of atrzf1, designated proline content alterative 31 (pca31). The pca31 mutant suppressed the insensitivity of atrzf1 to dehydration stress during early seedling growth. Using Thermal Asymmetric Interlaced-PCR, we found that the T-DNA of pca31 was inserted into the promoter region of the At2g22620 gene, which encodes the cell wall enzyme rhamnogalacturonan lyase 1 (RGL1). Enzymatic assays indicated that RGL1 exhibited rhamnogalacturonan lyase activity, influencing cell wall pectin composition. The decrease in RGL1 gene expression suppressed the transcriptomic perturbation of the atrzf1 mutant. Silencing of the RGL1 gene in atrzf1 resulted in a sensitive phenotype similar to pca31 under osmotic stress conditions. Treatment with mannitol, salt, hydrogen peroxide, and abscisic acid induced RGL1 expression. Furthermore, we uncovered that RGL1 plays a role in modulating root growth and vascular tissue development. Molecular, physiological, and genetic experiments revealed that the positive modulation of RGL1 during abiotic stress was linked to the AtRZF1 pathway. Taken together, these findings establish that pca31 acts as a suppressor of atrzf1 in abiotic stress responses through proline and cell wall metabolisms.
Effect of HbDHN1 and HbDHN2 Genes on Abiotic Stress Responses in Arabidopsis

Abstract:Dehydrin is a type of late embryogenesis abundant (LEA) protein. The dehydrin genes, HbDHN1 and HbDHN2, in Hevea brasiliensis were previously found to be induced at the wounding site of epicormic shoots, with local tissue dehydration identified as the key signal for laticifer differentiation. However, the exact role of the HbDHNs remains unknown. In this study, HbDHN1 and HbDHN2 expression was examined under multiple abiotic stresses; namely, cold, salt, drought, wounding, abscisic acid (ABA), ethylene (ET), and jasmonic acid (JA) treatment. Although, both HbDHNs were defined as SK2-type dehydrin, they showed different cellular localizations. Overexpression of the HbDHNs in Arabidopsis thaliana further revealed a significant increase in tolerance to salt, drought and osmotic stresses. Increased accumulation of proline and a reduction in electrolyte leakage were also observed under salt and drought stress, and a higher water content was indicated under osmotic stress. The transgenic plants also showed higher activity levels of ascorbate peroxidase (APX), superoxide dismutase (SOD) and catalase, and accumulated less hydrogen peroxide (H2O2) and superoxide ([Formula: see text]). Given that reactive oxygen species (ROS) are thought to be a key signal for laticifer differentiation, these findings suggest that HbDHNs act as ROS scavengers, directly or indirectly affecting laticifer differentiation. Both HbDHNs therefore influence physiological processes, improving plant tolerance to multiple abiotic stresses.

Analysis of two L-Galactono-1,4-lactone-responsive genes with complementary expression during the development of Arabidopsis thaliana

Abstract:Unraveling the role of genes annotated as protein of unknown function is of importance in progression of plant science. l-Galactono-1,4-lactone (l-GalL) is the terminal precursor for ascorbic acid (AsA) biosynthesis in Arabidopsis thaliana, and a previous study showed two DUF (domains of unknown function) 642 family genes (At1g80240 and At5g25460, designated as DGR1 and DGR2, respectively) to be sensitive to it. In this work, leaves from wild-type Arabidopsis were fed with d-glucose, l-galactose, l-GalL and AsA, and the expression level of the At1g80240 and At5g25460 genes showed a specific response to l-GalL, but not to the other supplements despite the increases of the tissue AsA contents. Analysis of promoter-β-glucuronidase (GUS) transgenic plants showed the two genes to be complementarily expressed at the root apex and in the rest of the root excluding the apex, respectively, in both young and old seedlings, and to be expressed at the leaf primordia. The GUS activity under the control of the At5g25460 promoter was high in the cotyledon and leaf veins of young seedlings. These findings were consistent with the results of quantitative real-time PCR. Interestingly, the T-DNA insertion mutant of At5g25460 (SALK_125079) displayed shorter roots and smaller rosettes than Col-0; however, no phenotypic difference was observed between the T-DNA insertion mutant of At1g80240 and the wild type. This is the first report on the expression and functional analysis of these two DUF642 family genes, with the results revealing the contribution of DGR genes to the development of Arabidopsis.
Characterization of Glutamate-Mediated Hormonal Regulatory Pathway of the Drought Responses in Relation to Proline Metabolism in Brassica napus L.

Abstract:Proline metabolism influences the metabolic and/or signaling pathway in regulating plant stress responses. This study aimed to characterize the physiological significance of glutamate (Glu)-mediated proline metabolism in the drought stress responses, focusing on the hormonal regulatory pathway. The responses of cytosolic Ca2+ signaling, proline metabolism, and redox components to the exogenous application of Glu in well-watered or drought-stressed plants were interpreted in relation to endogenous hormone status and their signaling genes. Drought-enhanced level of abscisic acid (ABA) was concomitant with the accumulation of ROS and proline, as well as loss of reducing potential, which was assessed by measuring NAD(P)H/NAD(P)+ and GSH/GSSG ratios. Glu application to drought-stressed plants increased both salicylic acid (SA) and cytosolic Ca2+ levels, with the highest expression of calcium-dependent protein kinase (CPK5) and salicylic acid synthesis-related ICS1. The SA-enhanced CPK5 expression was closely associated with further enhancement of proline synthesis-related genes (P5CS1, P5CS2, and P5CR) expression and a reset of reducing potential with enhanced expression of redox regulating genes (TRXh5 and GRXC9) in a SA-mediated NPR1- and/or PR1-dependent manner. These results clearly indicate that Glu-activated interplay between SA- and CPK5-signaling as well as Glu-enhanced proline synthesis are crucial in the amelioration of drought stress in Brassica napus.

Cotton GhHyPRP3 encoding a hybrid proline-rich protein is stress inducible and its overexpression in Arabidopsis enhances germination under cold temperature and high salinity stress conditions

Abstract:In this study, the cDNA coding for a hybrid proline-rich protein (HyPRP) was isolated from cotton cDNA libraries and designated GhHyPRP3. Analysis of the deduced amino acid sequence revealed that it contained an N-terminal signal peptide, a central proline-rich domain, and a C-terminal cysteine-rich domain highly homologous to other hybrid proline-rich group B proteins. RNA gel blot analysis showed that GhHyPRP3 mRNA was most abundant in petals and 10 DPA ovules indicating that expression of GhHyPRP3 was petal-preferential and ovule developmentally regulated. In addition, GhHyPRP3 transcription in roots was up-regulated by salt stress, cold stress, and osmotic stress, but down-regulated by GA3. A promoter-GUS reporter revealed that the GhHyPRP3 promoter directed gene expression in root–shoot junction, roots, and petals of transgenic Arabidopsis plants. Subcellular localization results showed that GhHyPRP3 was localized to the plasma membrane. Transgenic lines overexpressing GhHyPRP3 had a higher germination rate under cold temperature and high salinity stress conditions compared with wild type. Overall, GhHyPRP3 may function in flower and ovule development and participate in the defense responses to low temperature and salt stress.

Promoter of the TmHKT1;4-A1 gene of Triticum monococcum directs stress inducible, developmental regulated and organ specific gene expression in transgenic Arbidopsis thaliana

Abstract:HKT transporters which mediate Na+-specific transport or Na+-K+ co-transport, play an important role in protecting plants from salinity stress by preventing Na+-over-accumulation in leaves. In this work, a 1508-bp genomic fragment upstream of the TmHKT1;4-A1 translated sequence from Triticum monococcum has been isolated, cloned, and designated as the ''PrTmHKT1;4-A1'' promoter. Sequence analysis of ''PrTmHKT1;4-A1'' revealed the presence of cis-regulatory elements which could be required for abiotic stress and abscisic acid (ABA) responsiveness. The PrTmHKT1;4-A1 sequence was fused to the β-glucuronidase gene and the resulting construct was transferred into Arabidopsis plants. Histochemical assays of stably transformed Arabidopsis plants showed that PrTmHKT1;4-A1 is active in this heterologous system. Under control conditions, GUS histochemical staining was observed significantly only in leaves of 20-day-old plants. Histological sections prepared at this stage and in leaves revealed activity localized in leaf veins (phloem and bundle sheath). In flowers, GUS activity was detected only in sepals. After 3 days of challenging the plants with salt, dehydration or ABA treatments, the PrTmHKT1;4-A1 transformed Arabidopsis plants showed a substantial increase in the GUS staining in leaves, compared to untransformed plants under the same conditions. Real time qPCR expression analysis of the uidA gene, showed that GusA transcripts were up-regulated by salt, dehydration, and ABA treatments. All together, these results showed that PrTmHKT1;4-A1 is an age-dependent, abiotic-stress-inducible, organ-specific and tissue-specific promoter in a heterologous dicot system.
OsProDH Negatively Regulates Thermotolerance in Rice by Modulating Proline Metabolism and Reactive Oxygen Species Scavenging

Abstract:Background Global warming threatens rice growth and reduces yields. Proline plays important roles in plant abiotic stress tolerance. Previous research demonstrated that engineering proline metabolism-related genes can enhance tolerance to freezing and salinity inArabidopsis.OsProDHencodes a putative proline dehydrogenase and is a single copy gene in rice. However, whetherOsProDHplays roles in abiotic stress in rice remains unknown. Findings Quantitative RT-PCR analysis revealed thatOsProDHtranscript contents were relatively higher in leaf blade and root tissues and the high temperature treatment repressed expression ofOsProDH. The predicted OsProDH protein localized in mitochondria. Using theOryza sativa ssp. japonicacultivar KY131, we generatedOsProDHoverexpression (OE) lines and knockout mutant lines using the CRISPR/Cas9 (CRI) system. Overexpression ofOsProDHdecreased proline content, while mutation ofOsProDHincreased proline content compared with that of KY131. The CRI and OE lines were respectively more resistant and sensitive to heat stress than KY131. Heat stress induced proline accumulation and mutation ofOsProDHled to proline overproduction which reduced H(2)O(2)accumulation in the seedlings. Conclusions OsProDHnegatively regulates thermotolerance in rice. Our study provides a strategy to improve heat tolerance in rice via manipulating proline metabolism.

Abstract:BACKGROUND A new device, the CCS-30 Contour Transtar, was recently launched for the treatment of obstructed defecation syndrome (ODS). OBJECTIVES To evaluate the efficacy of the Contour Transtar in resection of true rectal prolapse in relation to age and concomitant urogynecologic procedures. METHODS During a 50 (median) month period 15 women with rectal prolapse of ≥ 5 cm and complaints of obstructed defecation underwent perineal resection of rectal prolapse with the Contour Transtar. RESULTS In 3 of the 15 patients (20%) rectal prolapse recurred. Amelioration of ODS symptoms and improved continence were noted in 82% and 75%, respectively, following surgery. CONCLUSIONS The Contour Transtar procedure for full-thickness rectal prolapse is a safe and promising procedure and is likely suitable for elderly poor risk patients.

Abstract:Polychlorinated biphenyl (PCB) concentrations in the blood of mothers who were occupationally exposed to PCBs in a capacitor manufacturing facility, and their children, were analyzed from 1975 to 1979. The factory terminated PCB use in 1972, and environmental PCBs were eliminated artificially. Despite this, blood PCB levels of the mothers were very high, i.e., 10-100 times higher than that of non-occupationally exposed persons. In a previous report, we discussed the relationship between breast-feeding and PCB levels in the blood. This relationship is confirmed further by the present investigation which continued during a 5-yr period. The PCB levels in blood of children were influenced greatly by the duration of breast-feeding, but showed little relationship to the PCB levels in maternal blood. The PCB exposure period of the mothers, time of birth, and age of the children were less clearly related to the PCB levels in the children. The PCB concentration in the children's blood decreased at a constant rate and was independent of their blood PCB levels. A similar result was noted for their mothers' PCB levels. A slight difference, however, was found in the rate of decrease of blood PCB levels between the mothers and children. The results of questionnaire research on the health conditions and the medical examinations for the children showed that frequency of complaints, e.g., red eye, fever, itchy skin, and carious teeth, related favorably to the duration of breast-feeding. Less severe findings, i.e., decay of nails, pigmentation, mottled enamel, carious teeth, which were typical symptoms in yusho patients, were observed in some children. They were not, however, diagnosed as having PCB poisoning inasmuch as medical treatments were unnecessary.

Abstract:Soybean is one of the important oil crops and a major source of protein and lipids. Drought can cause severe soybean yields. Dehydrin protein (DHN) is a subfamily of LEA proteins that play an important role in plant responses to abiotic stresses. In this study, the soybean GmDHN9 gene was cloned and induced under a variety of abiotic stresses. Results showed that the GmDHN9 gene response was more pronounced under drought induction. Subcellular localization results indicated that the protein was localized in the cytoplasm. The role of transgenic Arabidopsis plants in drought stress response was further studied. Under drought stress, the germination rate, root length, chlorophyll, proline, relative water content, and antioxidant enzyme content of transgenic Arabidopsis thaliana transgenic genes were higher than those of wild-type plants, and transgenic plants contained less O 2 − , H 2 O 2 and MDA contents. In short, the GmDHN9 gene can regulate the homeostasis of ROS and enhance the drought resistance of plants.

Abstract:Lysine synthesis in prokaryotes, some phycomycetes and higher plants starts with the condensation of L-aspartate-beta-semialdehyde (L-ASA) and pyruvate into dihydrodipicolinic acid. The enzyme that catalyses this step, dihydrodipicolinate synthase (DHDPS), is inhibited by the end-product lysine and is therefore thought to have a regulatory control on lysine synthesis. We have cloned and sequenced an Arabidopsis thaliana DNA fragment containing 900 bases upstream of the dhdps coding sequence. A transcriptional fusion of this fragment with the beta-glucuronidase reporter gene (uidA. Gus) was used to study the transcription properties of this promoter fragment (DS). No lysine-induced repression on transcription could be detected. Expression of DS-Gus activity in transformed Arabidopsis thaliana and Nicotiana tabacum was found to be cell type-specific. In the vegetative parts of the plant, GUS activity was located in meristems and young vasculature of roots, in vasculature of stem and leaves and in the meristems of young shoots. In flowers, high expression was found in the carpels, style, stigma, developing embryos, tapetum of young anthers and pollen. We demonstrated that the Arabidopsis DS promoter can direct its cell type-specific expression in a heterologous plant, Nicotiana tuabacum. The importance of transcriptional regulation of the dhdps gene, and in more general genes involved in amino acid biosynthesis, is discussed.
