Abstract:Objectives: The femoral head is prone to osteonecrosis. This study investigated dynamic contrast-enhanced (DCE) MRI contrast washout features of the femoral head and compared the data with data from other bony compartments in normal rats.Methods: 7-month-old Wistar rats were used. DCE MRI of the right hip (n=18), right knee (n=12) and lumbar spine (n=10) was performed after an intravenous bolus injection of Gd-DOTA (0.3 mmol kg(-1)). Temporal resolution was 0.6 s for hip and spine, and 0.3 s for knee. The total scan duration was 8 min for hip and spine, and 4.5 min for knee. The regions of interest for enhancement measurement included femoral head, proximal femoral diaphysis, distal femoral diaphysis and epiphysis, proximal tibial epiphysis and diaphysis, and lumbar vertebrae L1-5.Results: Femoral head showed no enhancement signal decay during the DCE MRI period, while all other bony compartments showed a contrast washin phase followed by a contrast washout phase. In the knee joint, the contrast washout of the proximal tibia diaphysis was slower that of other bony compartments of the knee.Conclusion: Based on the evidence of delayed contrast washout, this study showed that blood perfusion in the femoral head could be compromised in normal rats.
Longitudinal in Vivo Intrinsic Optical Imaging of Cortical Blood Perfusion and Tissue Damage in Focal Photothrombosis Stroke Model.

Abstract:A thorough understanding of the spatiotemporal dynamics of blood supply and tissue viability is of great importance in stroke researches. In the current study, vascular and cellular responses to focal ischemia were monitored with optical coherence tomography on chronic rat photothrombotic stroke model. The 3D mapping of blood perfusion and cellular scattering were achieved by analyzing the temporal dynamics and depth attenuation of intrinsic backscattered light respectively. Optical coherence tomography revealed that vessels of different types presented various spatial and temporal dynamics during the photothrombotic occlusion and the later recovery period. The large distal middle cerebral arteries presented a spontaneous recanalization and the small pial microvessels presented a reperfusion along with newly appeared vessels from the peripheral into the core area. The cortical capillary perfusion presented a weak recovery. Compared to the male group, the female rats showed a faster vascular recovery after photothrombotic. Moreover, the dynamic changes of the cellular scattering signal showed a high spatial and temporal correlation with the cortical capillary perfusion. Combined with well-designed photothrombotic stroke model and chronic optical window, optical coherence tomography imaging offers a unique approach to improve the understanding of stroke procedure and evaluate the treatment outcomes.
Brain Microvessels: Factors Altering Their Patency after the Occlusion of a Middle Cerebral Artery (wistar Rat).

Abstract:The progression from ischemic injury to pannecrosis that occurs in the rat brain several hours after occluding a large artery may be partly attributable to a worsening of the circulation through the microvessels. The objective of this study was to quantitate selected structural changes involving astrocytes and endothelial cells within an area of focal brain ischemia created by the occlusion of a middle cerebral artery. The magnitude of these structural changes was correlated with alterations in the patency to a circulating macromolecule through the microvessels (< or = 15 mu in diameter) located within the territory of the occluded artery. One hundred eighty-five adult male Wistar rats had the right middle cerebral artery occluded after threading a nylon monofilament through the external carotid artery. Experiments were terminated by either cardiovascular perfusion or decapitation and immersion fixation at intervals ranging between 30 minutes and 7 days after the arterial occlusion. Randomly selected animals from each experimental subgroup were injected intravenously with horseradish peroxidase (molecular weight 44 kd) approximately 20 minutes before death. The progressive decline in the area fraction comprised by the vessels filled with horseradish peroxidase was preceded at 30 to 60 minutes by an increase in the surface area occupied (on a cross-section of a microvessel) by endothelial cells (both nucleus and cytoplasm). This was followed by an increase of 23.7% in the mean diameter of astrocytes nuclei and a decrease of approximately 35% in lumenal surface of the microvessels. These observations suggest that the occlusion of a large cerebral artery causes prompt swelling of endothelial cells and astrocytes; both of these early biological responses may interfere with erythrocyte circulation and oxygen delivery, which (after the arterial occlusion) are entirely dependent on the circulation provided by the collateral arterial connections. Through its interference with microvascular patency and oxygen delivery, cell swelling may influence the rate at which neurons become necrotic. In this model of brain infarct the number of necrotic neurons peaks approximately 72 hours after middle cerebral artery occlusion.(ABSTRACT TRUNCATED AT 400 WORDS)

Abstract:To study initial thrombogenesis in vivo, a model has been developed by irradiation of filtered light and intravascular administiation of fluorescein sodium. The procedure of thrombogenesis in mesenteric microvessels of rat was monitored and recorded with a video system. The recorded video images were converted into the digital images and analyzed by microcomputer. Using the image processing, we could measure the size of thrombogenesis and microvessel diameter. Abstract:Background. New X-ray microangiography and third-generation synchrotron radiation-based micro-computed tomography have opened new perspectives for microvascular imaging of extremity. Here we aimed to visualize deep-level microvascular structure in rat hind limb by microangiographic technique, and compare images with those by conventional method.Materials and Methods. A total of 10 Sprague Dawley rats were used for in vivo and ex vivo study (five rats/group). Microangiography in vivo and ex vivo was performed and images were compared with those by conventional method. Synchrotron radiation-based micro-computed tomography (SR mu CT) was also performed to reveal three-dimensional (3D) morphology of the blood vessel in rat hind limb.Results. By microangiographic technique, blood vessels in the rat limb could be clearly depicted with the minimum visualized blood vessel about 9 mm in diameter, and higher angiographic scores were achieved than those by conventional X-ray. In addition, the vascular network could be defined and analyzed at the micrometer scale from the 3D renderings of limb vessel as shown by SR mu CT.Conclusions. Synchrotron radiation-based microangiography and SR mu CT thus provided a practical and effective means to observe the microvasculature of limbs, which might be useful in assessment of angiogenesis in lower limbs. Abstract:A better understanding of functional changes in the cerebral microvasculature following ischemic injury is essential to elucidate the pathogenesis of stroke. Up to now, the simultaneous depiction and stereological analysis of 3D micro-architectural changes of brain vasculature with network disorders remains a technical challenge. We aimed to explore the three dimensional (3D) microstructural changes of microvasculature in the rat brain on 4, 6 hours, 3 and 18 days post-ischemia using synchrotron radiation micro-computed tomography (SRμCT) with a per pixel size of 5.2 μm. The plasticity of angioarchitecture was distinctly visualized. Quantitative assessments of time-related trends after focal ischemia, including number of branches, number of nodes, and frequency distribution of vessel diameter, reached a peak at 6 h and significantly decreased at 3 days and initiated to form cavities. The detected pathological changes were also proven by histological tests. We depicted a novel methodology for the 3D analysis of vascular repair in ischemic injury, both qualitatively and quantitatively. Cerebral angioarchitecture sustained 3D remodeling and modification during the healing process. The results might provide a deeper insight into the compensatory mechanisms of microvasculature after injury, suggesting that SRμCT is able to provide a potential new platform for deepening imaging pathological changes in complicated angioarchitecture and evaluating potential therapeutic targets for stroke.

Abstract:Objective To visualize deep-level microvascular structure in rat hind limb by microangiographic technique,and compare images with those by conventional method.Methods Eight Sprague Dawley rats were used for ex vivo study.Either normal saline or barium sulfate solution was infused through the iliac artery catheter into the left hind limb.The specimens of different thickness were then imaged by synchrotron radiation source,and images were compared with those by conventional method.In synchrotron radiation imaging,in-line phase-contrast imaging technique was used,and phase contrast images were compared at different energy levels of monochromatic synchrotron radiation and object-image distances.One rat was used for synchrotron radiation imaging of the liver,lung,and limb without using any contrast.Results Using synchrotron radiation,blood vessel could be detected in the liver without using any contrast,and the hind limb infused with normal saline or barium sulfate solution.In addition,vessels in the hind limb infused with barium sulfate solution could be clearly depicted with more detail by synchrotron radiation imaging compared with conventional method,with the minimum visualized blood vessel about 9 μm in diameter.Conclusion Synchrotron radiation-based microangiography provided a practical and effective mean to observe the microvasculature of limbs,which might be useful in assessment of angiogenesis in lower limbs.
Effects Of High-Fat Diet On The Morphological Characteristics Of Cerebral Microvasculature Without Hyperlipidemia In Wistar Rat

Abstract:A high-fat diet (HFD) is associated with cerebrovascular disorders, however; its effect on the microvasculature in the brain is unknown. The aim of this study was to investigate the cerebral microvessel pathology by scanning electron microscope. Ten male Wistar rats were randomly divided into two groups: standard diet (n= 5) and HFD (n= 5). All of the rats were fed either standard rodent chow diet or HFD for 10 weeks and the brains were examined for microvascular pathology at the end of experiment. Coronal sections of a region of the cortex were observed by employing scanning electron microscope. Cerebral microvessels in the standard diet group exhibited the luminal endothelial surface with microvilli, indentations, and occasional vacuoles. The microvessels in the HFD group showed varying degrees of endothelial cell pathology including altered nuclear swelling as well as degenerative changes in the luminal wall. There was also thickening of the vascular basal laminae and expanded perivascular spaces often filled with amorphous debris. On a HFD, the luminal endothelial surface had more microvilli, indentations, and vacuoles. A HFD can affect the morphology of cerebral microvasculature and arteries in the Wistar rat in the absence of hyperlipidemia and lead to a series of pathological changes and microvascular remodeling, which can increase the likelihood of blood flow retardation and platelet aggregation, and ultimately thrombosis. The effects of HFD on microvasculature is more serious than that on large and medium- sized artery. Scanning electron microscope is a tool for studying microvasculature and artery pathology.

Abstract:The brain experiences alterations including cerebral ischemia and tissue damage after focal ischemic stroke. A thorough understanding of the spatiotemporal dynamics of blood perfusion and tissue damage is of great importance in stroke research. In chronic rat photothrombotic (PT) stroke model, a parallel study of both vascular and cellular responses to regional ischemia was performed with optical coherence tomography (OCT) in a label-free and depth-resolved manner. OCT revealed that vessels of different types and depth presented various spatial and temporal dynamics. In the ischemic core area, the distal middle cerebral arteries (dMCAs) were blocked gradually with laser irradiation and a spontaneous recanalization was observed at Day 5. In the chronic recovery period, the blocked small pial microvessels presented an apparent neovascularization progressing from the peripheral into the core area, with the final blood flow volume exceeding the baseline before PT. While the cortical capillary perfusion of the core area totally disappeared at Day 3 after PT and never recovered in the core area till the end of observation. The results demonstrated that blood reperfusion mainly occurred in the dMCAs vessels and pial microvessels of the superficial layer, but not in capillaries located deep in the cortex. The response of the cellular scattering and tissue damage showed a high spatial and temporal correlation with the capillary perfusion. On the whole, ischemic area and lesion area from attenuation coefficient are not exactly the same but complentary, with great help in understanding stroke mechanism comprehensively.
Cerebral Perfusion Characteristics and Dynamic Brain Structural Changes in Stroke-Prone Renovascular Hypertensive Rats: A Preclinical Model for Cerebral Small Vessel Disease

Abstract:Hypertension is a leading cause of cerebral small vessel disease (CSVD) and vascular dementia in elderly individuals. We aimed to assess cerebral perfusion and dynamic changes in brain structure in stroke-prone renovascular hypertensive rats (RHRSPs) with different durations of hypertension and to investigate whether they have pathophysiological features similar to those of humans with CSVD. The RHRSP model was established using the two-kidney, two-clip (2k2c) method, and the Morris water maze (MWM) test, MRI, immunohistochemistry, and biochemical analysis were performed at multiple time points for up to six months following the 2k2c operation. Systolic blood pressure was significantly greater in the RHRSP group than in the sham-operated group at week 4 post-surgery and continued to increase over time, leading to cognitive decline by week 20. Arterial spin labeling revealed cerebral hypoperfusion in the RHRSP group at 8 weeks, accompanied by vascular remodeling and decreased vessel density. Diffusion tensor imaging and Luxol fast blue staining indicated that white matter disintegration and demyelination gradually progressed in the corpus callosum and that myelin basic protein levels decreased. Eight weeks after surgery, blood-brain barrier (BBB) leakage into the corpus callosum was observed. The albumin leakage area was negatively correlated with the myelin sheath area ( r =-0.88, p <0.001). RNA-seq analysis revealed downregulation of most angiogenic genes and upregulation of antiangiogenic genes in the corpus callosum of RHRSPs 24 weeks after surgery. RHRSPs developed cerebral hypoperfusion, BBB disruption, spontaneous white matter damage, and cognitive impairment as the duration of hypertension increased. RHRSPs share behavioral and neuropathological characteristics with CSVD patients, making them suitable animal models for preclinical trials related to CSVD.

[Breast sarcoma. Diagnostic and therapeutic problems].

Abstract:From 1981 to 1988 six patients, five females and one male, with breast sarcoma were treated in the Department of Surgery, University Hospital, Bergen. Four of them had phyllodes tumour, one a malignant myoepithelioma and one a liposarcoma. Metastases to axillary lymph nodes or distant metastases were not observed. Sarcomas account for about 1% of malignant breast tumours in our department. The cases demonstrate diagnostic and therapeutical problems in dealing with breast sarcomas.
In Vivo Morphological Observation of Cortical Vessel in Mice after Chronic Cerebral Hypoperfusion

Abstract:Objective To dynamically observe the morphological changes of the cortical vessel in mice after chronic cerebral hypoperfusion through a cranial window. <br> Methods The left common carotid artery ligation and transient right common carotid artery ligation operations were performed on 7 Tie2-GFP transgenic mice. The cortical vessels were observed using confocol microscopy through a cranial window before brain ischemia (D0) and 7 days after brain ischemia (D7). The diameter and density of capillaries, along with the diameter and morphological change of arterioles and venules, were analyzed. <br> Results①The diameters of capillary before and 7 days after the brain ischemia were (6.62±0.75)μm and (12.50±3.29)μm, which increased significantly after brain ischemia (P<0.001).②The capillary densities before and 7 days after the brain ischemia were (11.67±1.72) segments per region of interest (ROI) and (11.08±2.06) segments per ROI. The difference was not signiifcant (P=0.0583).③The diameters of the precapillary arteriole, post capillary venule, true capillary and thoroughfare channel before and 7 days after the brain ischemia were (6.33±0.94)μmvs (12.36±3.20)μm, (6.87±1.10)μmvs (12.37±2.78)μm, (6.37±0.52)μmvs (11.41±3.10)μm and (6.35±0.92)μm vs (13.91±6.17)μm, respectively, all increased significantly after brain ischemia (P<0.01, <0.01,<0.01, and <0.05, respectively). The most signiifcant dilation was observed in thoroughfare channel.④The diameters of venules on D0 and D7 were (19.59±8.74)μm and (24.81±6.25)μm, which increased signiifcantly after brain ischemia (P=0.0054). However, the diameters of arterioles on D0 and D7 were (12.71±2.10)μm and (13.20±3.09)μm, the difference was not signiifcant (P=0.2947);⑤Capillary remodeling, tortuosity and new anastomosis between venules were observed after brain ischemia. <br> Conclusion The dilation of capillaries and venules were observed after brain ischemia. These changes may contribute to the regulation of cerebral blood flow after chronic cerebral hypoperfusion.
Targeted photothrombotic subcortical small vessel occlusion using in vivo real-time fiber bundle endomicroscopy in mice

Abstract:The development of an accurate subcortical small vessel occlusion model for pathophysiological studies of subcortical ischemic stroke is still insignificant. In this study, in vivo real-time fiber bundle endomicroscopy (FBEµ) was applied to develop subcortical photothrombotic small vessel occlusion model in mice with minimal invasiveness. Our FBFµ system made it possible to precisely target specific blood vessels in deep brain and simultaneously observe the clot formation and blood flow blockage inside the target blood vessel during photochemical reactions. A fiber bundle probe was directly inserted into the anterior pretectal nucleus of the thalamus in brain of live mice to induce a targeted occlusion in small vessels. Then, targeted photothrombosis was performed using a patterned laser, observing the process through the dual-color fluorescence imaging. On day one post occlusion, infarct lesions are measured using TTC staining and post hoc histology. The results show that FBEµ applied to targeted photothrombosis can successfully generate a subcortical small vessel occlusion model for lacunar stroke.

Abstract:Deviation of blood flow from an optimal range is known to be associated with the initiation and progression of vascular pathologies. Important open questions remain about how the abnormal flow drives specific wall changes in pathologies such as cerebral aneurysms where the flow is highly heterogeneous and complex. This knowledge gap precludes the clinical use of readily available flow data to predict outcomes and improve treatment of these diseases. As both flow and the pathological wall changes are spatially heterogeneous, a crucial requirement for progress in this area is a methodology for co-mapping local data from vascular wall biology with local hemodynamic data. In this study, we developed an imaging pipeline to address this pressing need. A protocol that employs scanning multiphoton microscopy was designed to obtain 3D data sets for smooth muscle actin, collagen and elastin in intact vascular specimens. A cluster analysis was developed to objectively categorize the smooth muscle cells (SMC) across the vascular specimen based on SMC density. In the final step in this pipeline, the location specific categorization of SMC, along with wall thickness was co-mapped with patient specific hemodynamic results, enabling direct quantitative comparison of local flow and wall biology in 3D intact specimens.

Methods Seven OLETF diabetic rats were used as animal models and diabetic stage was determined by oral glucose tolerance test. Seven LETO rats were taken as control group.When the rats were 58 weeks old, serum lipoproteins, insulin,C-peptide were tested by kit. Blood pressure,left ventricular pressure were determined separately by femoral artery and carotid artery catheterization. The myocardial tissues stained with HE,PAS,VG were observed by light microscope. The myocardial ultrastructure was observed with electron microscope and capillary basement membrane thickness was measured in 3 rats from both groups. Results Compared with normal control group, levels of TG,CHO in OLETF diabetic rats were higher (P0.05), the maximal rate of the left ventricular pressure fall (peak negative dp/dt) was lower (P0.05). HE staining showed cardiomyocyte hypertrophy and contraction; PAS staining showed excessive accumulation of glycogen and VG staining showed an accumulation of extracellular collagen. Transmission electron microscope showed cardiomyocyte contraction,formation of myofibril contraction bands, accumulation of extracellular matrix, thickening of capillary basement membrane.The basement membrane thickness in diabetes group ( 64.33±4.38 ) nm was thicker than that of the control group (46.09±3.62) nm (P0.001). Conclusions The pathologic changes of myocardium are cardiomyocye contraction,myocardial interstitial fibrosis and thickening of basement membrane, coexisting with left ventricular diastolic dysfunction.
A novel model of traumatic femoral head necrosis in rats developed by microsurgical technique

Abstract:Abstract Background Clinical angiography and vascular microperfusion confirmed that the femoral head retains blood supply after a collum femur fracture. However, no animal model accurately mimics this clinical situation. This study was performed to establish a rat model with retained viability of the femoral head and partial vasculature deprivation-induced traumatic caput femoris necrosis by surgery. Methods Thirty rats were randomly divided into three groups ( n = 10 per group): normal group, sham-operated group (Control), and ischemic osteonecrosis group. The femoral head of the normal group of rats underwent a gross anatomy study and microangiography to identify femoral head blood supply. Microsurgical techniques were used to cauterize the anterior-superior retinacular vessels to induce osteonecrosis. Hematoxylin and Eosin (H&E) staining were used for femoral head histologic assessment. Morphologic assessments of the deformity in and trabecular bone parameters of the femoral head epiphysis were performed using micro-CT. Results The blood supply of the femoral head in rats primarily came from the anterior-superior, inferior, and posterior retinacular arteries. However, anterior-superior retinacular vasculature deprivation alone was sufficient in inducing femoral head osteonecrosis. H&E showed bone cell loss in nuclear staining, disorganized marrow, and trabecular structure. The bone volume (BV) decreased by 13% and 22% in the ischemic group after 5 and 10 weeks, respectively. The mean trabecular thickness (Tb.Th) decreased from 0.09 to 0.06 mm after 10 weeks. The trabecular spacing (Tb.Sp) increased from 0.03 to 0.05 mm after 5 weeks, and the epiphyseal height-to-diameter (H/D) ratio decreased. Conclusions We developed an original and highly selective rat model that embodied femoral head traumatic osteonecrosis induced by surgical anterior-superior retinacular vasculature deprivation.

Tracking elemental changes in an ischemic stroke model with X-ray fluorescence imaging

Abstract:Stroke is a leading cause of long-term disability in adults and a leading cause of death in developed nations. The cascade of cellular events and signalling that occur after cerebral ischemia are complex, however, analyzing global element markers of metabolic state affords the means to monitor stroke severity, status of injury, and recovery. These markers provide a multi-parameter method for assessing changes through the post-stroke time course. We employ synchrotron-based elemental mapping to follow elemental changes in the brain at 1 h, 1-, 2-, and 3-days, and at 1-, 2-, 3-, and 4-weeks post-stroke in a photothrombotic stroke model in mice. Our analysis reveals a highly consistent metabolic penumbra that can be readily identified based on the level of dysregulated potassium and other key elements. Maps of elemental distributions are also useful to demarcate events in the cellular response to the inflammatory cascade, including ion dysregulation, recruitment of cells to the lesion, and glial scar formation.
