Abstract:Fluorescent carbon dots (CDs) have been synthesized via the calcination method using natural gynostemma as the precursor, without any toxic ingredients or surface passivation chemicals. CDs have a narrow size distribution, and the mean particle size is about 2.5 nm. CDs exhibit good water dispersibility and can emit intense blue fluorescence under 365 nm UV light in an aqueous solution, which can be stable in different conditions. The biotoxicity of CDs on the embryonic development of zebrafish is evaluated, the hatch rate and survival rate of embryos are around 90%, and the malformation rate is less than 10%. Because of the excellent fluorescence stability and biocompatibility, CDs can be used in zebrafish for bioimaging. In addition, the antioxidative stress property of CDs is investigated both in vitro and in vivo, and the presence of CDs can promote the mRNA expression of related genes to encode more antioxidant proteins in zebrafish. Therefore, fluorescent CDs would be a potential candidate for bioimaging and treating diseases caused by excessive oxidation damage, such as cancer, senility, and other diseases associated with aging.

Abstract:Fluorescent carbon dots (CDs) with advantages including excellent biocompatibility, low cost, and easy modification make them highly promising for applications in biological imaging. Here, we successfully synthesized dark green fluorescent CDs (g-CDs) using trinitropyrene as a carbon source and 2,4-diaminobenzenesulfonic acid as the reactant in one-pot synthesis. The results revealed that the g-CDs had an average uniform particle size of 4.93 nm, with a structure similar to graphite. The g-CDs imparted excellent water-solubility due to the presence of hydrophilic groups. The g-CDs exhibited an optimal fluorescence emission wavelength of 512 nm in acetone solvent, with a maximum quantum yield of 38.4%. When transferred into a normal saline solution, the g-CDs retained their green fluorescence, with the optimal fluorescence emission wavelength shifting to 505 nm. Notably, we observed a significant increase in fluorescence intensity and maximum quantum yield of up to 79.4% when the g-CDs were dissolved in a polar solvent. Furthermore, in vitro fluorescence imaging experiments demonstrated rapid cell entry, excellent fluorescence performance, high biocompatibility, and universality of the g-CDs. These findings underscore the high hydrophobicity and biocompatibility of our newly designed g-CDs, thus highlighting their immense potential as fluorescent probes in bioimaging and disease tracing applications.

Abstract:Carbon dots (CDs) synthesized from biological sources have attracted much interest in bioimaging and biomedical applications due to their excellent biocompatibility, and thus, a facile synthesis of CDs with high fluorescence quantum yield (QY) is requisite for practical applications. In this work, we report a simple, rapid, and green approach to synthesize photoluminescent CDs using eutrophic algal blooms as the carbon source. This method offers a possibility for large scale production of highly luminescent CDs (QY = 13%) with the average particle size ∼8 nm. These CDs are highly water-soluble and exhibit nanosecond fluorescence lifetime with high photostability, luminescence stability in different environments, low cytotoxicity, and excellent cell permeability. Laser scanning confocal microscopy shows the uptake of CDs by MCF-7 cells, and the destined application of these CDs as a potential biomarker is demonstrated.

Abstract:A simple and green approach was developed to synthesize highly fluorescent carbon dots (CDs) using albumin as a carbon source in aqueous solution at room temperature. The CDs were characterized by excellent monodispersion, superior photostability, pH-independent emission, long fluorescence lifetime and high quantum yield (QY). The photoluminescent (PL) mechanism of CDs was explored by means of time-resolved PL decay, and the results revealed that PL originated from the emission of both defect state and intrinsic state. In addition, biological imaging with the application of CDs was carried out in human breast cancer Bcap-37 cell, which demonstrated that CDs were provided with an excellent biocompatiblity, low cytotoxicity and good transmembrane ability. Besides, CDs could be considered as a potential substitute for organic dyes or semiconductor quantum dots (SQDs) in biological imaging.

Abstract:The high-value-added exploitation of biomass is a critical way to supply new carbon nanomaterials for “green resources."Biomass-derived carbon dots (CDs) are regarded as highly promising fluorescent nanomaterials. Carbon dots (FS-CDs) were prepared in this study using a simple solvothermal process, with fish scales serving as the carbon source. The FS-CDs were found to be evenly distributed, displaying an average diameter of 2.52 nm. Additionally, they demonstrated excitation-dependent emission and exhibited a notably high quantum yield of 31.71%. The high photoluminescence quantum yield in FS-CDs was shown to be associated with the (N, O)-codoped and high content of doped nitrogen, as determined by spectroscopic and microscopic examination in conjunction with the theoretical lattice model. Furthermore, it was discovered that FS-CDs exhibit a notable capacity for fluorescence imaging in zebrafish. This study demonstrates the significant utilization value of fish scales, which expands their potential applications and offers an environmentally friendly, cost-effective, and scalable approach for producing biomass-derived CDs.

Abstract:AbstractCarbon quantum dots (C-QDs) are becoming a desirable alternative to metal-based QDs and dye probes owing to their high biocompatibility, low toxicity, ease of preparation and unique photophysical properties. Herein, we describe fluorescence bioimaging of zebrafish using C-QDs as probe in terms of the preparation of C-QDs, zebrafish husbandry, embryo harvesting and introduction of C-QDs into embryos and larvae by soaking and microinjection. The multicolor of C-QDs was validated with their imaging for zebrafish embryo. The distribution of C-QDs in zebrafish embryos and larvae were successfully observed from their fluorescence emission. the bio-toxicity of C-QDs was tested with zebrafish as model and C-QDs do not interfere to the development of zebrafish embryo. All of the results confirmed the high biocompatibility and low toxicity of C-QDs as imaging probe. The absorption, distribution, metabolism and excretion route (ADME) of C-QDs in zebrafish was revealed by their distribution. Our work provides the useful information for the researchers interested in studying with zebrafish as a model and the applications of C-QDs. The operations related zebrafish are suitable for the study of the toxicity, adverse effects, transport and biocompatibility of nanomaterials as well as for drug screening with zebrafish as model.

Abstract:Carbon dots (CDs) have been extensively studied owing to their fascinating optical properties and wide potential applications. Here, we report an easy-to-perform and organic-solvent-free synthesis strategy for green-emissive CDs (G-CDs) possessing high photoluminescence (PL) quantum yield (QY). The G-CDs are synthesized by heating the homogeneous precursors of citric acid and cyanamide in an open vessel, circumventing the use of organic solvents, complex operations, high-pressure reactors, and expensive instruments in the synthesis process. The effect of various reaction variables on the formation and the optical properties of G-CDs are systematically investigated. The resultant G-CDs show bright PL emission at 521 nm with PL QY up to 73%. Then a white light-emitting diode (LED) with Commission Internationable de L'Eclairage (CIE) coordinates of (0.33, 0.34) and color rendering index (CRI) of 92 is constructed based on G-CDs/thermoplastic polyurethane (TPU) composite. Moreover, a visual microfluidic detection platform is designed by using G-CDs as fluorescent probes for rapid quantitative detection of Fe3+, Cu2+, and Mn2+ metal ions, which can realize synchronized testing of multiple samples. This study might promote the development and preparation methods of high-performance CDs with various optical applications.

Abstract:High fluorescence quantum yield graphene quantum dots (GQDs) have showed up as a new generation for bioimaging. In this work, luminescent GQDs were prepared by an ameliorative photo-Fenton reaction and a subsequent hydrothermal process using graphene oxide sheets as the precursor. The as-prepared GQDs were nanomaterials with size ranging from 2.3 to 6.4 nm and emitted intense green luminescence in water. The fluorescence quantum yield was as high as 24.6% (excited at 340 nm) and the fluorescence was strongest at pH 7. Moreover, the influences of low-concentration (12.5, 25 μg/mL) GQDs on the morphology, viability, membrane integrity, internal cellular reactive oxygen species level and mortality of HeLa cells were relatively weak, and the in vitro imaging demonstrated GQDs were mainly in the cytoplasm region. More strikingly, zebrafish embryos were co-cultured with GQDs for in vivo imaging, and the results of heart rate test showed the intake of small amounts of GQDs brought little harm to the cardiovascular of zebrafish. GQDs with high quantum yield and strong photoluminescence show good biocompatibility, thus they show good promising for cell imaging, biolabeling and other biomedical applications.

Abstract:Fluorescent red-emissive carbon dots (RCDs) were synthesized using the solvothermal method with citric acid as a carbon source, N , N -dimethylformamide as a nitrogen source, and formamide as a solvent. The as-synthesized RCDs show red fluorescence in an aqueous solution and have an excellent stability towards photobleaching as well as extremely low cytotoxicity and are successfully used for cell and zebrafish imaging. The results indicate that RCDs have potential applications in both in vitro and in vivo bioimaging.

Abstract:A facile, green, and high-output hydrothermal synthesis was proposed for the fabrication of highly fluorescent nitrogen-doped carbon quantum dots (N-doped CDs). The nitrogen content in N-doped CDs reached 19.2% and demonstrated strong blue fluorescence emission was obtained with fluorescence quantum yield (QY) of up to 32.9%, which exhibit high fluorescence quantum yield, high photostability, and excellent biocompatibility. The N-doped CDs possess high photostability, low toxicity, and excellent biocompatibility, based on which the N-doped CDs were successfully applied as a fluorescence probe for cell imaging. Moreover, it was then successfully demonstrated for sensitive and selective detection of Fe 3+ in serum.

Abstract:Biomass-based carbon quantum dots (CQDs) have become a significant carbon materials by their virtues of being cost-effective, easy to fabricate and low in environmental impact. However, there are few reports regarding using cyanobacteria as a carbon source for the synthesis of fluorescent CQDs. In this study, the low-cost biomass of cyanobacteria was used as the sole carbon source to synthesize water-soluble CQDs by a simple hydrothermal method. The synthesized CQDs were mono-dispersed with an average diameter of 2.48 nm and exhibited excitation-dependent emission performance with a quantum yield of 9.24%. Furthermore, the cyanobacteria-derived CQDs had almost no photobleaching under long-time UV irradiation, and exhibited high photostability in the solutions with a wide range of pH and salinity. Since no chemical reagent was involved in the synthesis of CQDs, the as-prepared CQDs were confirmed to have low cytotoxicity for PC12 cells even at a high concentration. Additionally, the CQDs could be efficiently taken up by cells to illuminate the whole cell and create a clear distinction between cytoplasm and nucleus. The combined advantages of green synthesis, cost-effectiveness and low cytotoxicity make synthesized CQDs a significant carbon source and broaden the application of cyanobacteria and provide an economical route to fabricate CQDs on a large scale.

Abstract:Abstract Carbon dots (CDs) are emerging as versatile fluorescent nanoprobes for bioimaging applications due to advantages like tunable emissions, excellent biocompatibility, facile surface functionalization, and ease of synthesis. This review summarizes recent advances in applying biosynthesized CDs for sensitive bioimaging. CDs derived from sustainable biomass sources through green techniques like hydrothermal and microwave synthesis demonstrate bright, excitation-tunable photoluminescence spanning visible to near-infrared spectra. Careful control of synthesis parameters and surface passivation strategies enhance quantum yields above 50% comparable to toxic semiconductor dots. Conjugation with polymers, peptides, and recognition elements like antibodies impart solubility and selectivity towards cancer cells and biomarkers. In vitro validation in standard lines shows targeted organelle imaging abilities. In vivo administration reveals renal clearance pharmacokinetics with preferential tumor accumulation via enhanced permeability effects. Average tumor growth inhibition around 50-80% was achieved in mouse xenografts using CDs-drug formulations through combined therapeutic effects of chemotherapy and photothermal ablation under imaging guidance. However, concerns regarding toxicity from chronic exposures, large-scale reproducible manufacturing, and multimodal imaging capabilities need redressal prior to further clinical translation.

Abstract:Carbon dots (CDs) are nanoparticles that have gained widespread attention due to their unique properties including multicolored fluorescence, photochemical stability, and biocompatibility. However, the conventional approaches for the preparation of CDs typically require high energy input and toxic reagents. Herein, a sustainable, high throughput approach is reported for producing uniform CDs. Specifically, spent coffee grounds (SCGs) are used to produce coffee ground-derived carbon dots (CFCDs) via a simple wet ball-milling process. The versatility of our approach can be extended to the synthesis of heteroatom-doped CDs and the preparation of CDs from various SCG sources. Also, the functional groups of the CFCDs are manipulated by the addition of citric acids during the ball-milling process to produce carboxylic acid-functionalized carbon dots (CA-CFCDs). CA-CFCDs exhibit highly selective and sensitive detection of Fe3+ ions in aqueous solution via fluorescence quenching, and the relevant quenching mechanism is investigated. Finally, we demonstrate the fluorescent microgel sensor exhibiting a rapid, ratiometric fluorescence change upon the addition of Fe3+ ion by encapsulating the CA-CFCDs within the microgel matrix.

Abstract:Carbon dots (CDs) provides unprecedented opportunities for optical applications due its unique properties, but the energy-extensive consumption, high-risk factor and time-consuming synthesis procedure greatly hinders its industrialization process. Herein, we proposed an ultra-low energy consumption solvent-free synthetic strategy for fast preparing green/red fluorescence carbon dots (G-/R-CDs) using m-/o-phenylenediamine and primary amine hydrochloride. The involvement of primary amine hydrochloride can improve the formation rate of G-CDs/R-CDs through effectively absorbing microwave energy and providing acid react environment. The developed CDs exhibit good fluorescence efficiency, optical stability and membrane permeability for dexterous bioimaging in vivo. Based on inherently high nitrogen content, the G-CDs/R-CDs possess excellent nuclear/nucleolus targeting ability, and were successfully applied for screening cancer and normal cells. Furthermore, the G-CDs/R-CDs were further applied for fabricating high-safety and high-color rendering index white light-emitting diodes, providing a perfect candidate for indoor lighting. This study opens up new horizons for advancing practical applications of CDs in related fields of biology and optics.

Abstract:Carbon dots (CDs) are a class of highly efficient and low-toxicity fluorescent nanomaterials. In this study, using cordyceps militaris as a carbon source, we prepared green-emitting CDs with excellent properties, such as good photobleaching resistance, salt resistance and chemical stability, as well as high quantum yield. Further exploration of the versatile applications of G-CDs in chemical detection and information anti-counterfeiting fields revealed that G-CDs can not only be used as detection agent for analyzing alcohol content in alcoholic beverages but also serve as waterproof fluorescent ink for painting and writing due to their water-resistant property. This work is based on natural biomass as the main precursor to prepare fluorescent CDs with excellent luminescent characteristics and stable physicochemical properties. It is significant for reducing reliance on chemical reagents, lowering the cost of synthesizing CDs, and expanding the application range of CDs.

Abstract:Carbon dots (CDs) are attracting increasing research attention due to their exceptional attributes, including their biocompatibility, water solubility, minimal toxicity, high photoluminescence, and easy functionalization. Green CDs, derived from natural sources such as fruits and vegetables, present advantages over conventionally produced CDs, such as cost-effectiveness, stability, simplicity, safety, and environmental friendliness. Various methods, including hydrothermal and microwave treatments, are used to synthesize green CDs, which demonstrate strong biocompatibility, stability, and luminescence. These properties give green CDs versatility in their biological applications, such as bioimaging, biosensing, and drug delivery. This review summarizes the prevalent synthesis methods and renewable sources regarding green CDs; examines their optical features; and explores their extensive biological applications, including in bioimaging, biosensing, drug/gene delivery, antimicrobial and antiviral effects, formatting of mathematical components, cancer diagnosis, and pharmaceutical formulations.
Zebrafish: A Promising Model for Evaluating the Toxicity of Carbon Dot-Based Nanomaterials

Abstract:Carbon dots (CDs) exhibit a wide range of desirable properties including excellent photoluminescence, photostability, and water solubility, making them ideally suitable for use in the context of drug delivery, bioimaging, and related biomedical applications. Before these CDs can be translated for use in humans, however, further research regarding their in vivo toxicity is required. Owing to their low cost, rapid growth, and significant homology to humans, zebrafish (Danio rerio) are commonly employed as in vivo model systems in the toxicity studies of nanomaterials. In the present report, our group employed a hydrothermal approach to synthesize CDs and then assessed their toxicity in zebrafish. The resultant CDs were roughly 2.4 nm spheroid particles that emitted strong blue fluorescence in response to the excitation at 365 nm. These CDs did not induce any evident embryonic toxicity or did cause any apparent teratogenic effects during hatching or development when dosed at 150 μg/mL. However, significant effects were observed in zebrafish embryos at CD concentrations >200 μg/mL, including pericardial and yolk sac edema, delayed growth, spinal cord flexure, and death. These high CD concentrations were further associated with the reduction in zebrafish larval locomotor activity and decreased dopamine levels, reduced frequencies of tyrosine hydroxylase-positive dopaminergic neurons, and multiple organ damage. Abstract:Due to their unique optical and biochemical properties, the water-soluble fluorescent carbon dots (CDs) have attracted a lot of attention recently. Here, strong fluorescent carbon dots with excellent quality have been synthesized by the hydrothermal refluxing method using lactose as carbon source and tris(hydroxymethyl) aminomethane (i.e. Tris) as surface passivation reagent. This facile approach was simple, efficient, economical, green without pollution, and allows large-scale production of CDs without any post-treatment. TEM measurements showed that the resulting particles exhibited an average diameter of 1.5 nm. The obtained CDs possess small particle sizes, good stability in a wide range of pH values (pH 3.5-9.5), high tolerance of salt concentration, strong resistibility to photobleaching, and a fluorescent quantum yield up to 12.5%. The CDs were applied to optical bioimaging of HeLa cells, showing low cytotoxicity and excellent biocompatibility.
