Abstract:The relentless march of a highly pathogenic avian influenza virus (HPAIV) strain, known as H5N1, to become an unprecedented panzootic continues unchecked. The leap of H5N1 clade 2.3.4.4b from Eurasia and Africa to North America in 2021 and its further spread to South America and the Antarctic have exposed new avian and mammalian populations to the virus and led to outbreaks on an unrivaled scale. The virus has infected wild birds across vast geographic regions and caused wildlife deaths in some of the world's most biodiverse ecosystems. Hundreds of millions of poultry have died or been culled, affecting global food security in some of the world's poorest regions. Numerous mammalian species, including sea lions and fur animals, have been infected. Outbreaks in dairy cows in the United States have been occurring for months, seemingly unchecked in most affected states. Why is there not a greater sense of urgency to control these infections?

Abstract:We report highly pathogenic avian influenza A(H5N1) virus in dairy cattle and cats in Kansas and Texas, United States, which reflects the continued spread of clade 2.3.4.4b viruses that entered the country in late 2021. Infected cattle experienced nonspecific illness, reduced feed intake and rumination, and an abrupt drop in milk production, but fatal systemic influenza infection developed in domestic cats fed raw (unpasteurized) colostrum and milk from affected cows. Cow-to-cow transmission appears to have occurred because infections were observed in cattle on Michigan, Idaho, and Ohio farms where avian influenza virus–infected cows were transported. Although the US Food and Drug Administration has indicated the commercial milk supply remains safe, the detection of influenza virus in unpasteurized bovine milk is a concern because of potential cross-species transmission. Continued surveillance of highly pathogenic avian influenza viruses in domestic production animals is needed to prevent cross-species and mammal-to-mammal transmission.

Abstract:Highly pathogenic avian influenza (HPAI) viruses cross species barriers and have the potential to cause pandemics. In North America, HPAI A(H5N1) viruses related to the goose/Guangdong 2.3.4.4b hemagglutinin phylogenetic clade have infected wild birds, poultry, and mammals. Our genomic analysis and epidemiological investigation showed that a reassortment event in wild bird populations preceded a single wild bird-to-cattle transmission episode. The movement of asymptomatic cattle has likely played a role in the spread of HPAI within the United States dairy herd. Some molecular markers in virus populations were detected at low frequency that may lead to changes in transmission efficiency and phenotype after evolution in dairy cattle. Continued transmission of H5N1 HPAI within dairy cattle increases the risk for infection and subsequent spread of the virus to human populations.

Abstract:In February and March of 2024, an unusual illness began affecting dairy herds primarily in Texas and neighboring states. The causative agent of this illness was ultimately confirmed in late March 2024 to be a strain of highly pathogenic avian influenza H5N1 belonging to clade 2.3.4.4b. In the months following the emergence of this viral disease in cattle, infections have spread to at least 191 herds in 13 states at the time of this writing in August 2024, primarily through cattle and human movement. Surprisingly, early examination of raw milk samples from clinically affected animals indicated that the virus had an affinity for the mammary tissue, and viral shedding into raw milk occurred at high levels, exceeding 108 log10 50% tissue culture infectious dose (TCID50) in some cases. These high viral loads coupled with evidence that farm cats who consumed raw milk from clinically ill animals were infected and exhibited high mortality rates, raised concerns about the safety of the US milk supply for human consumption. To date, 4 cow-associated human infections have been reported, all from farm employees with direct contact with infected animals. Several parameters ultimately affect the theoretical public health risk from consumption of dairy products manufactured from a milk supply containing H5N1, namely (1) initial viral load, (2) persistence of H5N1 in raw milk, (3) viral inactivation through processing practices including pasteurization, and (4) human susceptibility and infectious dose. In the short period since the emergence of this disease in dairy cattle in the United States, research has begun to answer these critical questions, although our knowledge is still quite limited at this time. Here we review the literature available from the current H5N1 outbreak in US dairy cattle, as well as selected relevant literature from previous research in other animal agriculture sectors, that affect our current understanding of the parameters associated with the food safety risk of this disease in the US dairy supply chain.

Abstract:On March 24, 2024, the US Department of Agriculture (USDA) confirmed the first case of highly pathogenic avian influenza (HPAI) H5N1 clade 2.3.4.4b in dairy cattle. Following this initial detection, the virus was found to have spread to other species, including humans. Since then, at least 192 dairy herds across 13 states have tested positive for the virus. The emergence of HPAI H5N1 in dairy cattle poses a serious threat to the dairy industry, requiring an approach that integrates human, animal, and environmental health. Enhancing biosecurity, improving surveillance, and advancing research are critical to managing this challenge. Swift adaptation is needed to protect both public health and dairy production. Through this work, we discuss the epidemiological and clinical aspects of HPAI H5N1 in cattle by reviewing the spatiotemporal situation of the disease across the United States, the clinical signs and testing leading to a confirmed case, and the surveillance and control measures implemented at the national and state levels. Further, considering pathogenesis and risk factors, we discuss surveillance and control measures to implement within herds, and note areas where research is needed to understand how to control this emerging disease.

Abstract:KEYWORDS: Following a press release by the Minnesota Veterinary Medical Association (MVMA) on 26 March 2024 notifying its members of a juvenile goat on a Minnesota farm tested positive for the highly pathogenic avian influenza (HPAI) that was later confirmed to be H5N1 influenza virus by the National Veterinary Services Laboratories (NVSL) of the United States Department of Agriculture Animal and Plant Health Inspection Services (USDA-APHIS) [ Citation 1 ], the MVMA reported two days later that some dairy cattle in Texas and Kansas states in the USA were tested positive for the same (H5N1) virus [ Citation 2 ]. While the NVSL was working to confirm some presumptive positive test results from dairy herds in Texas, Idaho, and elsewhere [ Citation 3 ], H5N1 virus were again detected in a dairy herd in New Mexico as well as in 5 additional dairy herds in Texas on 2 April 2024 [ Citation 4 ]. It appears that this viral pathogen was spreading rapidly among dairy herds in several states in the USA. As of this writing (5 April 2024), H5N1 virus infections have been detected and confirmed in eight (08) dairy herds in Texas, three (03) in Kansas, two (02) in New Mexico, and one (01) herd each in Michigan and Ohio () [ Citation 5 ]. It is important to note that H5N1 viruses found thus far [i.e. HPAI A(H5N1) viruses of the genetic clade 2.3.4.4b, which is the same virus clade found among birds globally] in different states appear to be very similar to the viral strain of the same genetic clade 2.3.4.4b originally identified in dairy cattle in Texas and Kansas where they were first reported on 25 March 2024. This is partly because some dairy farms in Michigan and Idaho have recently received (or imported) cattle from Texas with previously identified cases of H5N1 in their cattle herds [ Citation 4 ].

Abstract:Concerns are mounting over the threat to humans from a highly pathogenic avian influenza virus, a threat first recognised in the late 1990s when a new clade of the H5N1 virus was detected in domestic waterfowl in China. The first human cases, all linked with exposure to infected poultry, were reported from Hong Kong in 1997,1 and the World Health Organization recorded a total of 463 deaths among 888 cases between January 2003 and March 2024.2 Infections are under-reported, but the high ratio of deaths to cases (52%) suggests that H5N1 could cause a major public health emergency if human exposure and viral evolution lead to sustained person-to-person transmission.We do not know if that will happen, but the likelihood seems to have risen over the past four years. Abstract:The rapid geographic spread of the highly pathogenic avian influenza (HPAI) A(H5N1) virus in poultry, wild birds, and other mammalian hosts, including humans, raises significant health concerns globally. The recent emergence of HPAI A(H5N1) in agricultural animals such as cattle and goats indicates the ability of the virus to breach unconventional host interfaces, further expanding the host range. Among the four influenza types—A, B, C, and D, cattle are most susceptible to influenza D infection and serve as a reservoir for this seven-segmented influenza virus. It is generally thought that bovines are not hosts for other types of influenza viruses, including type A. However, this long-standing viewpoint has been challenged by the recent outbreaks of HPAI A(H5N1) in dairy cows in the United States. To date, HPAI A(H5N1) has spread into fourteen states, affecting 299 dairy herds and causing clinical symptoms such as reduced appetite, fever, and a sudden drop in milk production. Infected cows can also transmit the disease through raw milk. This review article describes the current epidemiological landscape of HPAI A(H5N1) in US dairy cows and its interspecies transmission events in other mammalian hosts reported across the globe. The review also discusses the viral determinants of tropism, host range, adaptative mutations of HPAI A(H5N1) in various mammalian hosts with natural and experimental infections, and vaccination strategies. Finally, it summarizes some immediate questions that need to be addressed for a better understanding of the infection biology, transmission, and immune response of HPAI A(H5N1) in bovines.

Abstract:Avian influenza A (H5N1), a highly pathogenic virus, known as bird flu is emerging as a serious global health security risk. Avian flu subtypes H5N1 and H7N9 circulate in bird populations worldwide with high genomic diversity and the ability to reconstruct with the human influenza virus to introduce new strains capable of zoonotic (human-to-human) transmission. Influenza virus evolves and develops new pathogenic strains very rapidly. Around 50 million people died due to the 1918 'Spanish flu' pandemic caused by an H1N1 influenza virus [1]. In addition to the H1N1 pandemic (Swine flu) in 2009, there have been a few other human AIV pandemics in the past decades: the H2N2 pandemic (Asian flu) in 1957 and the H3N2 pandemic (Hong Kong flu) in 1968 [2]. The H5N1 outbreak not only infects birds but also breaks and crosses the interspecies barrier and affects mammals including animals and humans. H5N1 virus has limited human transmission because it binds to alpha-2, 3-linked sialic acid receptors (during its replication cycle) which are less abundant in the human respiratory system as compared to birds but its genetic reassortment ability makes it highly pathogenic and a health hazard for humans. The outbreak of the H5N1 virus has been reported currently in US dairy cows and workers. In humans, it can cause severe respiratory problems, multi-organ failure, and high mortality rates. Four humans have been affected since 2020 in the US [3]. The Centers for Disease Control and Prevention (CDC) is thoroughly monitoring the people with animal exposure, carefully inspecting the situation, and currently reporting a low public health risk. Experts are raising alarm about the transfer of viruses from birds to humans and are urging the implementation of new protocols for handling airborne infectious diseases. However, the worldwide spread of H5N1 and its ability to infect mammals causes major threats regarding the potential for human-to-human transmission, which may result in a pandemic. In addition to its potential impact on human health, bird flu poses an immense threat to agriculture and food security because of its economic implications. As the avian flu continues to pose a persistent and evolving threat to global health security, national, regional, and global vigilance, preparedness, and collaborative actions are needed. It is important to maintain robust surveillance, respond rapidly, and innovative prevention and control strategies due to sporadic outbreaks of avian flu. Management of avian flu demands a coordinated effort to protect human and animal health, mitigate economic impacts, and improve the global health security
