Abstract:<p>LncRNA-cCSC1 is highly expressed in colorectal cancer (CRC). The study was designed to evaluate the function and mechanism of lncRNA-cCSC1 in cell proliferation of CRC. RT-PCR was used to measure the expression levels of lncRNA-cCSC1 in CRC cell lines. CCK-8, colony formation, EdU staining, flow cytometry and Western blot were performed to examine the effect of interference with lncRNA-cCSC1 expression on cell proliferation. miR-124-3p and the target genes of miR-124-3p were investigated using bioinformatics analysis and verified by dual-luciferase reporter, RT-PCR and Western blot. Rescue experiments were carried out to confirm the role of miR-124-3p in cell proliferation of CRC. Our results showed that cell proliferation of CRC was promoted by lncRNA-cCSC1 upregulation and inhibited by lncRNA-cCSC1 downregulation. In addition, miR-124-3p is predicted to be the target of lncRNA-cCSC1 and is negatively correlated with lncRNA-cCSC1. Moreover, the addition of miR-124-3p mimics or inhibitor reversed the effects induced by lncRNA-cCSC1 overexpression or silencing on cell proliferation of CRC. Additionally, lncRNA-cCSC1 regulated the expression level of CD44, a target gene of miR-124-3p. Finally, we studied the effects of the lncRNA-cCSC1/miR-124-3p axis on CD44. These results indicate that lncRNA-cCSC1 promotes cell proliferation of CRC through sponging miR-124-3p and upregulating CD44.</p>

What problem does this paper attempt to address?
The problem this paper attempts to address is to explore the function and mechanism of long non-coding RNA (lncRNA) cCSC1 in the proliferation of colorectal cancer (CRC) cells. Specifically, the researchers aim to experimentally verify whether lncRNA-cCSC1 promotes CRC cell proliferation by binding to miR-124-3p and upregulating the expression of CD44. ### Main Research Content: 1. **Expression of lncRNA-cCSC1 in CRC**: - The study found that lncRNA-cCSC1 is significantly overexpressed in colon adenocarcinoma (COAD) and rectal adenocarcinoma (READ), and its expression level in CRC tissues is higher than in adjacent normal tissues. - In four human CRC cell lines (SW480, SW620, HT29, and HCT116), the expression of lncRNA-cCSC1 is significantly higher than in the normal human colon epithelial cell line (NCM460). 2. **Effect of lncRNA-cCSC1 on CRC Cell Proliferation**: - Through experiments of overexpression and knockdown of lncRNA-cCSC1, researchers found that overexpression of lncRNA-cCSC1 promotes CRC cell proliferation, while knockdown inhibits cell proliferation. - Further experiments, including CCK-8, colony formation, cell cycle analysis, and EdU staining, all confirmed this conclusion. 3. **Interaction between lncRNA-cCSC1 and miR-124-3p**: - Bioinformatics analysis predicted that miR-124-3p is a downstream target gene of lncRNA-cCSC1. - Dual-luciferase reporter gene experiments validated the binding between lncRNA-cCSC1 and miR-124-3p and found that lncRNA-cCSC1 can negatively regulate the expression of miR-124-3p. - Rescue experiments showed that miR-124-3p mimics or inhibitors can reverse the effects of lncRNA-cCSC1 overexpression or knockdown on CRC cell proliferation. 4. **CD44 as a Target Gene of miR-124-3p**: - Bioinformatics analysis predicted that CD44 is a downstream target gene of miR-124-3p. - Experimental results showed that overexpression of lncRNA-cCSC1 upregulates the expression of CD44, while knockdown downregulates the expression of CD44. - Dual-luciferase reporter gene experiments further confirmed the binding relationship between miR-124-3p and CD44. ### Conclusion: This study reveals that lncRNA-cCSC1 promotes CRC cell proliferation by binding to miR-124-3p and upregulating the expression of CD44. This finding provides a new potential target for the diagnosis and treatment of CRC. However, further research is needed to fully elucidate the molecular mechanism of lncRNA-cCSC1 in CRC to provide more support for its role in CRC.
