Abstract:This paper presents a segmentation method for human locomotion. The research on the rules of human locomotion plays an important role for the analysis of human movement strategy. We collected locomotion data from eighteen volunteers by motion capture system(MCS), and found that the measured stride length and frequency with speed had a mutation in the critical point between walking and running. To solve this problem, a segmentation method is proposed to describe the two gaits motion while the continuous formula is not suitable for using again. Especially, a running formula is proposed by introducing a compression of virtual leg based on the elastic linear inverted pendulum model(ELIPM), which can accurately describe the relationship between stride length, frequency and speed. By experimental comparisons, the proposed method can reduce 77% of the statistical error than Alexander's one, which indicates this method is an effective approach and can uniform the motion rules for human walking and running.

Abstract:BACKGROUND:Accurate measurements of in-vivo knee joint kinematics are essential to elucidate healthy knee motion and the changes that accompany injury and repair. Although numerous experimental measurements have been reported, the accurate non-invasive analysis of in-vivo knee kinematics remains a challenge in biomechanics. RESEARCH QUESTION:The study objective was to investigate in-vivo knee kinematics before, at, and after contact during walking and running using a combined high-speed dual fluoroscopic imaging system (DFIS) and magnetic resonance (MR) imaging technique. METHODS:Three-dimensional (3D) knee models of ten participants were created using MR images. Knee kinematics during walking and running were determined using high-speed DFIS. The 3D knee models were then related to fluoroscopic images to obtain in-vivo six-degrees-of-freedom knee kinematics. RESULTS:Before contact knee flexion, external femoral rotation, and proximal-distal distance were 11.9°, 3.4°, and 1.0 mm greater during running compared to walking, respectively. Similar differences were observed at initial contact (9.9°, 7.9°, and 0.9 mm, respectively) and after contact (6.4°, 2.2°, and 0.8 mm, respectively). Posterior femoral translation at initial contact was also increased during running compared to walking. SIGNIFICANCE:This study demonstrated accurate instantaneous in-vivo knee kinematic characteristics that may further the understanding of the intrinsic biomechanics of the knee during gait.
Experimental Comparison between 4D Stereophotogrammetry and Inertial Measurement Unit Systems for Gait Spatiotemporal Parameters and Joint Kinematics

Abstract:(1) Background: Traditional gait assessment methods have limitations like time-consuming procedures, the requirement of skilled personnel, soft tissue artifacts, and high costs. Various 3D time scanning techniques are emerging to overcome these issues. This study compares a 3D temporal scanning system (Move4D) with an inertial motion capture system (Xsens) to evaluate their reliability and accuracy in assessing gait spatiotemporal parameters and joint kinematics. (2) Methods: This study included 13 healthy people and one hemiplegic patient, and it examined stance time, swing time, cycle time, and stride length. Statistical analysis included paired samples t-test, Bland–Altman plot, and the intraclass correlation coefficient (ICC). (3) Results: A high degree of agreement and no significant difference (p > 0.05) between the two measurement systems have been found for stance time, swing time, and cycle time. Evaluation of stride length shows a significant difference (p < 0.05) between Xsens and Move4D. The highest root-mean-square error (RMSE) was found in hip flexion/extension (RMSE = 10.99°); (4) Conclusions: The present work demonstrated that the system Move4D can estimate gait spatiotemporal parameters (gait phases duration and cycle time) and joint angles with reliability and accuracy comparable to Xsens. This study allows further innovative research using 4D (3D over time) scanning for quantitative gait assessment in clinical practice.

Abstract:Objective: This study aimed at quantifying the difference in kinematic and joint moments calculation for lower limbs during gait utilizing a markerless motion system (TsingVA Technology, Beijing, China) in comparison to values estimated using a marker-based motion capture system (Nokov Motion Capture System, Beijing, China). Methods: Sixteen healthy participants were recruited for the study. The kinematic data of the lower limb during walking were acquired simultaneously based on the markerless motion capture system (120 Hz) and the marker-based motion capture system (120 Hz). The ground reaction force was recorded synchronously using a force platform (1,200 Hz). The kinematic and force data were input into Visual3D for inverse dynamics calculations. Results: The difference in the lower limb joint center position between the two systems was the least at the ankle joint in the posterior/anterior direction, with the mean absolute deviation (MAD) of 0.74 cm. The least difference in measuring lower limb angles between the two systems was found in flexion/extension movement, and the greatest difference was found in internal/external rotation movement. The coefficient of multiple correlations (CMC) of the lower limb three joint moments for both systems exceeded or equaled 0.75, except for the ad/abduction of the knee and ankle. All the Root Mean Squared Deviation (RMSD) of the lower limb joint moment are below 18 N·m. Conclusion: The markerless motion capture system and marker-based motion capture system showed a high similarity in kinematics and inverse dynamic calculation for lower limbs during gait in the sagittal plane. However, it should be noted that there is a notable deviation in ad/abduction moments at the knee and ankle.

Comparison of two kinematic trunk models in five activities of daily living

Abstract:Objective:To compare the kinematic difference of two trunk models during five activities of daily living.The two models are trunk model composed of thoracic segment,upper lumbar segment and lower lumbar segment and simplified trunk model composed of thoracic segment and the whole lumbar segment.Method:Twenty-six young healthy adults performed level walking,stair climbing,trunk flexion,flexing to pick up an object in front of the ipsilateral feet and flexing to pick up an object in front of the contralateral feet.The kinematic data were collected using motion capture system NDI.This study applied principal component analysis to investigate variance accounted for(VAF degree of contribution) and load coefficient of the two trunk models.Result:The flexion angle and the lateral bending angle of the whole lumbar segment are both significantly higher than thoracic segment in simplified trunk model while the flexion angle and the lateral bending angle of lower lumbar segment are both significantly higher than the others in subdivided trunk model.In addition,the load coefficient of two lumbar segments in subdivided trunk model are opposite in sign.Conclusion:The main motion of trunk is lumbar and the main motion segment of lumbar is lower lumbar segment.During some activities of daily living,the two lumbar segments have intersegmental counteraction.Therefore,it is essential to subdivide trunk segment,especially lumbar segment,into multiple segments when analyze the kinematic character of trunk motion.

Abstract:Current literature lacks a comparative analysis of different motion capture systems for tracking upper limb (UL) movement as individuals perform standard tasks. To better understand the performance of various motion capture systems in quantifying UL movement in the prosthesis user population, this study compares joint angles derived from three systems that vary in cost and motion capture mechanisms: a marker-based system (Vicon), an inertial measurement unit system (Xsens), and a markerless system (Kinect). Ten healthy participants (5F/5M; 29.6 ± 7.1 years) were trained with a TouchBionic i-Limb Ultra myoelectric terminal device mounted on a bypass prosthetic device. Participants were simultaneously recorded with all systems as they performed standardized tasks. Root mean square error and bias values for degrees of freedom in the right elbow, shoulder, neck, and torso were calculated. The IMU system yielded more accurate kinematics for shoulder, neck, and torso angles while the markerless system performed better for the elbow angles. By evaluating the ability of each system to capture kinematic changes of simulated upper limb prosthesis users during a variety of standardized tasks, this study provides insight into the advantages and limitations of using different motion capture technologies for upper limb functional assessment.

Concurrent assessment of gait kinematics using marker-based and markerless motion capture

Abstract:Abstract Kinematic analysis is a useful and widespread tool used in research and clinical biomechanics for the estimation of human pose and the quantification of human movement. Common marker-based optical motion capture systems are expensive, time intensive, and require highly trained operators to obtain kinematic data. Markerless motion capture systems offer an alternative method for the measurement of kinematic data with several practical benefits. This work compared the kinematics of human gait measured using a deep learning algorithm-based markerless motion capture system to those of a common marker-based motion capture system. Thirty healthy adult participants walked on a treadmill while data were simultaneously recorded using eight video cameras (markerless) and seven infrared optical motion capture cameras (marker-based). Video data were processed using markerless motion capture software, marker-based data were processed using marker-based capture software, and both sets of data were compared. The average root mean square distance (RMSD) between corresponding joints was less than 2.5 cm for all joints except the hip, which was 3.6 cm. Lower limb segment angles indicated pose estimates from both systems were very similar, with RMSD of less than 5.5° for all segment angles except those that represent rotations about the long axis of the segment. Lower limb joint angles captured similar patterns for flexion/extension at all joints, ab/adduction at the knee and hip, and toe-in/toe-out at the ankle. These findings demonstrate markerless motion capture can measure similar 3D kinematics to those from marker-based systems.

Abstract:Two experiments were conducted to verify the feeding value of NutriDense (ND) and Nutri-Dense Low-Phytate (NDLP) corn (Exseed Genetics LLC, BASF Plant Science, Research Triangle Park, NC) relative to that of yellow dent (YD) corn in swine diets. NutriDense corn is a high-protein, high-oil variety, and NDLP is a high-protein, high-oil, low-phytate variety. In Exp. 1, 315 nursery pigs that initially weighed 15.2 kg were used in a 21-d growth assay. Dietary treatments were arranged in a 3 x 3 factorial; main effects were corn source (YD, ND, and NDLP) and added fat (0, 3, or 6%, as-fed basis). Diets were formulated to contain 3.83 g of lysine/Mcal using calculated nutrient values. There were no corn source x fat interactions observed. Pigs fed YD, ND, and NDLP had ADG of 750, 734, and 738 g/d and G:F of 0.64, 0.66, and 0.65, respectively. No differences (P > 0.10) in ADG were observed among the three corn sources; however, pigs fed diets containing either ND or NDLP corn had decreased ADFI (P < 0.02) and improved G:F (P < 0.05) compared with pigs fed diets containing YD corn. Increasing dietary fat increased ADG (727, 746, and 748 g/d; linear, P < 0.04) and G:F (0.62, 0.66, and 0.68; linear, P < 0.01) and decreased ADFI (linear, P < 0.01). Using the NRC (1998) value for ME in YD corn, we calculated the energy value for ND and NDLP based on G:F differences compared with pigs fed YD corn. These data indicated the ME values for ND and NDLP corn are 4.5 and 2.5% greater (3,575 and 3,505 Kcal/kg), respectively, than for YD corn (3,420 Kcal/kg). In Exp. 2, 1,144 gilts (initial BW = 50.1 kg) were used in a commercial research facility to evaluate the effects of corn source (ND and YD) and added fat (0, 3, or 6%, as-fed basis) in a 2 x 3 factorial on pig performance and carcass traits. There was a corn source x fat interaction for ADFI and G:F. Increasing added fat resulted in greater changes in ADFI and G:F in pigs fed YD corn diets compared with those fed ND corn. Feeding ND corn increased ADG (main effect, P < 0.04), and greater percentages of added fat increased ADG (main effect; linear, P < 0.01). Results of Exp. 2 suggest that ND corn has 5.3% more ME than YD corn. The additional energy provided by ND corn improves G:F in both nursery and grow-finish pigs, and ND corn offers a means of formulating diets more concentrated in energy than YD corn.

Abstract:To date, there are several measurement methods for evaluating human kinematics based on inertial sensors or vision systems. However, a comprehensive comparison has not been undertaken to determine which of these systems offers the most appropriate accuracy for clinical or sports evaluations. This study conducted a comparative analysis of different motion measurement systems: optoelectronic system (OS), inertial measurement units (IMUs), and vision-based methods, including deep neural network (DNN) and non-DNN approaches. Ten healthy subjects were involved, performing walking (W.) and running (R.) tests at various speeds (3.5, 5.0, and 7.0 km/h). The measurement of human kinematics was conducted by taking video images via two RGB cameras, together with an IMU-based system and an OS as the gold standard. Comparative analysis was conducted on a set of measurement methods, including IMU, a method based on blob analysis (BA), and DNN algorithms: Alphapose (AP), TC former (TC), RTMPose (RTM), and MediaPipe (MP). Data analysis involved triangulation and measurement of lower limb joint angles. Results showed that vision systems do not allow ankle joint measurement, and IMUs outperformed other methods in terms of RMSE and absolute error of range of motion ( . RTM and MP exhibited results similar to IMUs, especially for the hip and knee joints, with the minimum absolute error reporting values of ( and ( for the hip joint and ( and ( for the knee joint, respectively.

Abstract:Objective: Patients with chronic stroke capable of independent gait were classified into functional ambulation category (FAC) 4 or 5, and the kinetic and kinematic data on their lower limb joints on the affected and unaffected sides were compared with that of healthy individuals. Finally, the qualitative changes in the gait of patients with stroke were investigated based on the differences in FAC scores. Methods: Twelve healthy participants and 19 patients with stroke capable of independent gait were included. The three-dimensional (3D) motion analysis and conventional assessment were conducted for all patients with stroke. Results: The FAC 5 group exhibited a larger range of motion (ROM) than the FAC 4 group in knee and hip joints on the affected side and only in the hip on the unaffected side. In the FAC 5 group, ROM differences in the healthy group on either the affected or unaffected side were absent. The peak of the hip flexion moment on the affected side in both the FAC 4 and 5 groups was smaller than that in the healthy group and in the FAC 4 group on the unaffected side. The absorption power minimum on the affected side was smaller only in the FAC 4 group than that in the healthy group and was larger in the FAC 5 group than that in the FAC 4 group. On the unaffected side, the absorption power minimum was smaller only in the FAC 4 group than that in the healthy group. Conclusion: Functional differences in gait were found in patients classified based on conventional evaluation capable of independent gait after post-stroke rehabilitation. Patients may not exhibit complete recovery in the kinetic indices even if they are judged to be normal in the conventional evaluation, and the kinematic gait indices indicate recovery. Evaluating kinetic indices in addition to kinematic indices is necessary, and joint power may be an especially useful index.

Abstract:In clinical settings, examining the range of motion (ROM) of trunk rotation and functional tasks is commonly employed as part of musculoskeletal assessments to diagnose low back pain (LBP) cases. This study compared trunk kinematics during functional activities between individuals with and without non-specific LBP. A total of 45 LBP patients and 45 healthy participants were recruited for this research. The Xsens wearable motion capture system was utilized to record movements during axial trunk rotations and activities such as single-leg standing, walking, hurdle steps, squats, and box lifting. During functional tasks, individuals with LBP displayed larger trunk ROMs, except for rotation in the transverse plane (TP) during single-leg standing. Statistically significant differences were observed between the two groups in trunk extension-flexion in the frontal plane (FP) and TP, trunk bending in TP, trunk rotation in FP and TP, single-leg standing in the sagittal plane (SP), walking in SP and FP, hurdle step in TP, and squat in the FP plane (p < 0.05). Individuals with LBP exhibited smaller trunk ROMs during simple trunk rotation movements, whereas larger ROMs were observed during functional tests. These findings shed light on the particular alterations observed in trunk movement and function experienced by individuals with LB.

Abstract:Quantifying lumbar back motion during functional activities in real-life environments may contribute to a better understanding of common pathologies such as spinal disorders. The current study therefore aimed at the comparative evaluation of the Epionics SPINE system, a portable device for measuring sagittal lumbar back motion during functional activities. Twenty healthy participants were therefore evaluated with the Epionics SPINE and a Vicon motion capture system in two identical separate research visits. They performed the following activities: standing, sitting, chair rising, box lifting, walking, running and a counter movement jump (CMJ). Lumbar lordosis angles were extracted as continuous values as well as average and range of motion (ROM) parameters. Agreement between the systems was evaluated using Bland-Altman analyses, whereas within- and between-session reliability were assessed using intraclass correlation coefficients (ICC) and minimal detectable changes (MDC). The analysis showed excellent agreement between the systems for chair rising, box lifting and CMJ with a systematic underestimation of lumbar lordosis angles during walking and running. Reliability was moderate to high for all continuous and discrete parameters (ICC≥0.62), except for ROM during running (ICC=0.29). MDC values were generally below 15°, except for CMJ (peak values up to 20° within and 25° between the sessions). The Epionics SPINE system performed similarly to a Vicon motion capture system for measuring lumbar lordosis angles during functional activities and showed high consistency within and between measurement sessions. These findings can serve researchers and clinicians as a bench mark for future investigations using the system in populations with spinal pathologies.

Abstract:Introduction: Analysis of running gait is necessary for prevention of hip and knee injuries. Two dimensional (2D) motion analysis systems are being widely used at a clinical level for quantication of kinematic parameters during running gait analysis. Observational methods of assessing gait are largely dependent on clinicians' ability to quantify and analyze the kinematic data in a reliable manner. Under these circumstances, use of motion analysis software with inbuilt running gait analysis protocols help to improve the reliability of data. To investigate the convergent validity, Objective: inter- and intra-rater reliability of a 2-D motion analysis system (GaitON®) for quantication of hip and knee kinematics at initial contact during running. 21 healthy male recreational runners were Methods: recorded using a clinical 2D video setup during over-ground running. Two blinded raters quantied joint angles on video using GaitON® and Kinovea software. The intra-rater reliability [intra-class correlation coefcient (ICC 3, 1)], interrater reliability, the limits of agreement (LOA) in the Bland-Altman plot were also determined. The a Results: ngles quantied using GaitON® were signicantly correlated with the angles quantied in Kinovea with r = 0.997; p = 0.01 for the hip and r = 0.982; p = 0.01 for the knee. The ICC values (intra-rater reliability) for the hip and knee were 0.998 and 0.990 respectively whereas ICC values (inter-rater reliability) of 0.996 and 0.978 were computed for the hip and knee respectively. The st Conclusion: rong validity and excellent inter- and intra-rater reliability results establish a concrete foundation for use of GaitON® as a tool for evaluating the running gait of a patient in a clinical setup.
Alteration of movement patterns in low back pain assessed by Statistical Parametric Mapping

Abstract:Changes in movement pattern in low back pain (LBP) groups have been analysed by reporting predefined discrete variables. However, this approach does not consider the full kinematic data waveform and its dynamic information, potentially exposing the analysis to bias. Statistical Parametric Mapping (SPM) has been introduced and applied to 1 dimensional (D) kinematic variables allowing the assessment of data over time. The aims of this study were to assess differences in 3D kinematics patterns in people with and without LBP during functional tasks by using SPM and to investigate if SPM analysis was consistent with standard 3D range of motion (RoM) assessments. 3D joints kinematics of the spine and lower limbs were compared between 20 healthy controls and 20 participants with non-specific LBP during walking, sit-to-stand and lifting. SPM analysis showed significant differences in the 3Dkinematics of the lower thoracic segment, upper and lower lumbar segment and knee joint during walking and lifting mostly observed at the beginning and/or towards the end of the tasks. ROMs differed between groups in the lower thoracic segment (walking/sit-to-stand), upper and lower lumbar segments (walking/sit-to-stand/lifting), hip and knee (sit-to-stand/lifting). Based on these results, the two approaches can yield different data interpretations. SPM analysis allows the identification of differences in movement that occur over time. This adds value to LBP movement analysis as it allows an understanding of the LBP strategies adopted during motion that may not be conveyed by simple discrete parameters such as ROMs.

Statistical parametric mapping and linear mixed models were used to compare kinematic variables between and within timepoints. ResultsBoth continuous and discrete data analyses identified changes in pelvis and forelimb kinematics. Discrete data analyses showed significant changes in hindlimb and back kinematics, where such differences were not found to be significant by continuous data analysis. In contrast, continuous data analysis provided additional information on the timing and duration of the differences found. Main limitationsA limited number of ponies were included. ConclusionsThe use of continuous data provides additional information regarding gait adaptations to bilateral lameness that is complementary to the analysis of discrete variables. The main advantage lies in the additional information regarding time dependence and duration of adaptations, which offers the opportunity to identify functional adaptations during all phases of the stride cycle, not just the events related to peak values.

Abstract:Measuring motion of the human foot presents a unique challenge due to the large number of closely packed bones with congruent articulating surfaces. Optical motion capture (OMC) and multi-segment models can be used to infer foot motion, but might be affected by soft tissue artifact (STA). Biplanar videoradiography (BVR) is a relatively new tool that allows direct, non-invasive measurement of bone motion using high-speed, dynamic x-ray images to track individual bones. It is unknown whether OMC and BVR can be used interchangeably to analyse multi-segment foot motion. Therefore, the aim of this study was to determine the agreement in kinematic measures of dynamic activities. Nine healthy participants performed three walking and three running trials while BVR was recorded with synchronous OMC. Bone position and orientation was determined through manual scientific-rotoscoping. The OMC and BVR kinematics were co-registered to the same coordinate system, and BVR tracking was used to create virtual markers for comparison to OMC during dynamic trials. Root mean square (RMS) differences in marker positions and joint angles as well as a linear fit method (LFM) was used to compare the outputs of both methods. When comparing BVR and OMC, sagittal plane angles were in good agreement (ankle: R2 = 0.947, 0.939; Medial Longitudinal Arch (MLA) Angle: R2 = 0.713, 0.703, walking and running, respectively). When examining the ankle, there was a moderate agreement between the systems in the frontal plane (R2 = 0.322, 0.452, walking and running, respectively), with a weak to moderate correlation for the transverse plane (R2 = 0.178, 0.326, walking and running, respectively). However, root mean squared error (RMSE) showed angular errors ranging from 1.06 to 8.31° across the planes (frontal: 3.57°, 3.67°, transverse: 4.28°, 4.70°, sagittal: 2.45°, 2.67°, walking and running, respectively). Root mean square (RMS) differences between OMC and BVR marker trajectories were task dependent with the largest differences in the shank (6.0 ± 2.01 mm) for running, and metatarsals (3.97 ± 0.81 mm) for walking. Based on the results, we suggest BVR and OMC provide comparable solutions to foot motion in the sagittal plane, however, interpretations of out-of-plane movement should be made carefully.

Abstract:Digital quantification of gait can be used to measure aging- and disease-related decline in mobility. Gait performance also predicts prognosis, disease progression, and response to therapies. Most gait analysis systems require large amounts of space, resources, and expertise to implement and are not widely accessible. Thus, there is a need for a portable system that accurately characterizes gait. Here, depth video from two portable cameras accurately reconstructed gait metrics comparable to those reported by a pressure-sensitive walkway. 392 research participants walked across a four-meter pressure-sensitive walkway while depth video was recorded. Gait speed, cadence, and step and stride durations and lengths strongly correlated (r > 0.9) between modalities, with root-mean-squared-errors (RMSE) of 0.04 m/s, 2.3 steps/min, 0.03 s, and 0.05-0.08 m for speed, cadence, step/stride duration, and step/stride length, respectively. Step, stance, and double support durations (gait cycle percentage) significantly correlated (r > 0.6) between modalities, with 5% RMSE for step and stance and 10% RMSE for double support. In an exploratory analysis, gait speed from both modalities significantly related to healthy, mild, moderate, or severe categorizations of Charleson Comorbidity Indices (ANOVA, Tukey's HSD, p < 0.0125). These findings demonstrate the viability of using depth video to expand access to quantitative gait assessments.

Abstract:Trabectedin and its N12-demethylated analogue ET729 bind covalently to the central guanine of selected DNA triplets. Although both drugs equally target several sites, including AGA, we show that covalent modification of CGA is only achieved by ET729. By means of molecular dynamics simulations of the precovalent complexes, we explain in atomic detail how such a simple structural modification brings about this notable change in the DNA-binding selectivity profiles of these two drugs.

Abstract:Gait analysis for patients with orthopedic joint diseases is crucial to understand their functional status. Inertial measurement unit (IMU) systems, as alternatives to optical motion capture (OMC) systems, enable gait analysis outside the laboratory. However, their accuracy requires validated before widespread clinical use. Therefore, this study evaluated the validity of the Xsens IMU system for lower-limb joint angle measurements during walking in patients with hip and knee pathology and compared the error metrics among patients with hip pathology, patients with knee pathology and healthy controls. We used OMC system and Xsens IMU systems to simultaneously collect lower-limb kinematic data of 130 patients with knee pathology, 110 patients with hip pathology and 25 healthy individuals during walking at self-selected speed. Validity was assessed using root mean square error (RMSE), amplitude difference and coefficient of determination (R 2 ). The RMSEs for the patient groups ranged from 2.5° to 8.8°, with half of the joint angles showing acceptable accuracy and the rest tolerable accuracy. The joint angles measured by both systems were more consistent in the sagittal plane (R 2 : 0.62–0.96), with lower consistency in the frontal and transverse planes (R 2 : 0.44–0.63). Compared to the control group, both patient groups had higher RMSEs and lower R 2 across most joint angles. Our results suggest that the Xsens IMU system offers highly comparable sagittal plane kinematic waveforms in patients with hip and knee pathology. Caution should be taken when interpreting the frontal and transverse plane kinematics and assessing patients with severe joint deformities
