Abstract:The parameters for complete hydrolysis of L-phenyl acetyl phenylglycine (L-PAPG) using immobilized penicillin G acylase (IMEPGA) were investigated. IMEPGA exhibited maximum activity at pH 8.5 and 50 degrees C. The apparent Km value observed was 10 mM. Quantitative hydrolysis (>97%) of the L-PAPG was achieved within 45 min, at pH 7.8 and 37 degrees C, when 0.5% (w/v) of DL-PAPG was used and the concentration of IMEPGA was 133 IU/gm of DL-PAPG. The IMEPGA was used for 50 cycles.

Abstract:Chemical investigation of the endophytic fungus Penicillium sp. isolated from Limonium tubiflorum growing in Egypt afforded four new compounds of polyketide origin, including two macrolides, penilactone (1) and 10,11-epoxycurvularin (2), a dianthrone, neobulgarone G (7), and a sulfinylcoumarin, sulfimarin (14), along with twelve known metabolites (3-6, 8-13, 15 and 16). The structures of all compounds were assigned by comprehensive spectral analysis (1D and 2D NMR) and mass spectrometry. Compounds 3, 4, 13 and 16 showed pronounced antitrypanosomal activity with mean MIC values ranging from 4.96 to 9.75μM. Moreover, when tested against a panel of three human tumor cell lines compounds 3, 4, 6 and 12 showed selective growth inhibition against Jurkat and U937 cell lines with IC(50) values ranging from 1.8 to 13.3μM. The latter compounds also inhibited TNFα-induced NF-κB activity in K562 cells with IC(50) values ranging from 1.6 to 10.1μM, respectively.

Abstract:A significant minority of parents are concerned about adolescents engaging in risky sexual behavior following human papillomavirus (HPV) vaccination. The way the HPV vaccine is reported in the media has the potential to influence public understanding and vaccination decisions. The present study examined the content of articles published between 2003 and 2008 in British national newspapers that addressed the issue of adolescents engaging in risky sexual behavior following HPV vaccination. We used mixed methods to analyze 92 articles in which the issue was mentioned. Qualitative framework analysis highlighted three main types of discussion: news stories proposing that adolescents will engage in risky sexual behavior following HPV vaccination, counterarguments insisting that adolescents will not engage in risky sexual behavior after HPV vaccination, and parents' views of the issue of risky sexual behavior. The results indicated that newspapers provide parents with broadly positive descriptive norms about vaccination; however, the issue that adolescents will engage in risky sexual behaviors following HPV vaccination is regularly discussed in the national press and has the potential to increase parents' concerns about vaccination.
Phytochemical Profile of the Methanolic Leaf Extract of Vernonia amygdalina Del. and In Vitro Anticoccidial Effect of Its Fractions against Eimeria tenella

Abstract:Phytochemical screening of the crude methanolic leaf extract of Vernonia amygdalina was carried out. The extract and its fractions were tested for in vitro anticoccidial efficacy against the unsporulated oocysts of Eimeria tenella by the oocyst sporulation inhibition model in a 96-well microtitre plate. Phytochemical analysis of the extract revealed the presence of alkaloids, tannins, saponins, glycosides, and triterpenoids. Different concentrations of the extracts, including 250 mg/ml, 125 mg/ml, 62.5 mg/ml, 31.25 mg/ml, 15.625 mg/ml, and 7.8 mg/ml, were tested against freshly prepared unsporulated oocysts of E. tenella. Triplicate assays were conducted for each concentration of the extract to screen for anticoccidial activity. The methanol extract and fractions showed activity against the oocysts of E. tenella in a dose-dependent manner. The sporulation inhibition rate was determined as effective when the result showed more than 50% inhibition at each concentration level of treatment. Only the methanol extract and its butanol fraction at the lowest treatment concentration of 7.8 mg/ml showed an inhibition rate of 55% and 68%, respectively. All treatments: methanol extract (100%), hexane fraction (100%), butanol fraction (100%), and aqueous residue fraction (58%) were active at the highest concentration of 250 mg/ ml. It is concluded that the methanol leaf extract of Vernonia amygdalina and its butanol fraction had better oocyst sporulation inhibition activity against Eimeria tenella and may further be explored for their anticoccidial properties.

Abstract:In our continuing search for novel natural products with antiplasmodial activity, an extract of Aniba citrifolia was found to have good activity, with an IC50 value less than 1.25 μg/mL. After bioassay-directed fractionation, the known indolizinium alkaloid anibamine (1) and the new indolizinium alkaloid anibamine B (2) were isolated as the major bioactive constituents, with antiplasmodial IC50 values of 0.170 and 0.244 μM against the drug-resistant Dd2 strain of Plasmodium falciparum. The new coumarin anibomarin A (3), the new norneolignan anibignan A (5), and six known neolignans (7-12) were also obtained. The structures of all the isolated compounds were determined based on analyses of 1D and 2D NMR spectroscopic and mass spectrometric data, and the absolute configuration of anibignan A (5) was assigned from its ECD spectrum. Evaluation of a library of 28 anibamine analogues (13-40) indicated that quaternary charged analogues had IC50 values as low as 58 nM, while uncharged analogues were inactive or significantly less active. Assessment of the potential effects of anibamine and its analogues on the intraerythrocytic stages and morphological development of P. falciparum revealed substantial activity against ring stages for compounds with two C-10 side chains, while those with only one C-10 side chain exhibited substantial activity against trophozoite stages, suggesting different mechanisms of action.

Abstract:Phytochemical analysis of the leaves of Vernonia amygdalina yielded two known sesquiterpene lactones: vernolide and vernodalol. The two compounds were tested by agar dilution method against 10 bacteria strains and 5 fungi species. Both compounds exhibited a significant bactericidal activity against five Gram positive bacteria while lacking efficacy against the Gram negative strains. In the antifungal test, while vernolides exhibited high activity with LC50 values of 0.2, 0.3 and 0.4mg/ml against Penicillium notatum, Aspergillus flavus, Aspergillus niger and Mucor hiemalis, respectively, vernodalol showed moderate inhibitions against Aspergillus flavus, Penicillium notatum and Aspergillus niger with LC50 values of 0.3, 0.4 and 0.5mg/ml, respectively. Both compounds were ineffective against Fusarium oxysporum, a microbe known to be highly resistant to chemical agents. However, the antimicrobial results of this study correspond positively with the claimed ethnomedical uses of the leaves of Vernonia amygdalina in the treatment of various infectious diseases.

Abstract:This study aimed to produce linalool loaded zinc oxide nanocomposite (LZNPs) and assess its in vitro and in vivo antileishmanial effects against Leishmania major. LZNPs was produced through the synthesis of an ethanolic solution containing polyvinyl alcohol. The average size of LZNPs was determined to be 105 nm. The findings indicated that LZNPs displayed significant (p < 0.01) antileishmanial effects on promastigotes and amastigotes. Following exposure of promastigotes to LZNPs, there was a notable rise in the percentage of early and late apoptotic cells from 9.0 to 57.2 %. The gene expression levels of iNOS, IFN-γ, and TNF-α in macrophages were upregulated in a dose-dependent approach following exposure to LZNPs. LZNPs alone and in conjunction with glucantime (Glu) resulted in a reduction in the diameter and parasite load of CL lesions in infected mice. Treatment of the CL-infected mice with LZNPs at 25 and 50 mg/kg mainly in combination with Glu-reduced the tissue level of malondialdehyde (MDA), increased both gene and protein expression of the antioxidant enzymes as well as raised the expression level of IFN-γ and IL-12 cytokines, whereas caused a significant reduction in the expression level of IL-4. The present study shows that LZNPs has potent antileishmanial effects and controls CL in a mice model through its antioxidant and immunomodulatory properties. Further investigation, especially in clinical trials, could explore the potential use of this nanocomposite in managing and treating CL.

Abstract:The strongly hydrophobic C(60) fullerene is a carbon allotrope of huge interest in materials science and in pharmaceutical chemistry that can be solubilized in water either by extensive chemical functionalization or by inclusion in appropriate carriers such as the cyclodextrins with formation of host-guest complexes. Here we report a molecular dynamics study of the complexes formed in solution by C(60) with gamma- and delta-cyclodextrins. The most stable host-guest complex stoichiometry is determined to be 2:1 through simulations in vacuo and in explicit water by the stepwise addition of the cyclodextrins to C(60). No a priori assumption about the inclusion stoichiometry and geometry is made. The equilibrium fluctuations of the complexes that can affect the system stability are also investigated within the molecular dynamics runs.

Abstract:Leishmaniases comprise a group of infectious parasitic diseases caused by various species of Leishmania protozoa and are considered a significant public health problem worldwide. Current therapies face serious limitations, including low efficacy, high costs, and routes of administration that require hospital support and are already exhibiting resistance. Additionally, only a limited number of drugs, such as meglumine antimonate, amphotericin B, and miltefosine, are available, all of which are associated with severe side effects. In this context, the need for new effective drugs with fewer adverse effects is evident. Therefore, this study investigated the anti- Leishmania activity of a dichloromethane fraction (DCMF) extracted from Arrabidaea brachypoda roots. This fraction inhibited the viability of L. infantum , L. braziliensis , and L. Mexicana promastigotes, with IC 50 values of 10.13, 11.44, and 11.16 μg/mL, respectively, and against L. infantum amastigotes (IC 50 = 4.81 μg/mL). Moreover, the DCMF exhibited moderate cytotoxicity (CC 50 = 25.15) towards RAW264.7 macrophages, with a selectivity index (SI) of 5.2. Notably, the DCMF caused damage to the macrophage genome only at 40 μg/mL, which is greater than the IC 50 found for all Leishmania species. The potential mechanisms of action of brachydins were also investigated. The results suggest that DCMF demonstrates similar antileishmanial effectiveness to isolated brachydin B, without causing genotoxic effects on mammalian cells. This finding is crucial because the isolation of the compounds relies on several steps and is very costly while obtaining the DCMF fraction is a simple and cost-effective process. Furthermore, the computational analysis indicates that brachydin compounds bind to the Triosephosphate isomerase (TIM) enzyme via two main mechanisms: destabilizing the interface between the homodimers and interacting with catalytic residues situated at the site of binding. Based on all the results, DCMF exhibits promise as a therapeutic agent for leishmaniasis due to its significantly reduced toxicity in comparison to the adverse effects associated with current reference treatments.

Abstract:Toxoplasma gondii (T. gondii) poses significant risks, especially to those with weakened immune systems, highlighting the need for new, less toxic treatments. Laurus nobilis L. ( L. nobilis ), known for its beneficial properties including antiparasitic and antioxidant effects, offers potential for innovative therapies. This study assessed the anti-Toxoplasma, cytotoxic, antioxidant, and anti-hemolytic effects of L. nobilis 's aqueous and ethanolic extracts both in vitro and in silico , including a phytochemical analysis. HPLC analysis identified a range of phenolic compounds, notably syringic acid, ferulic acid, gallic acid, caffeine, and quercetin. Both extracts exhibited an inhibitory effect on the growth of T. gondii tachyzoites in vitro and caused less cytotoxicity to host cells. In silico studies revealed that rutin and quercetin were the most potent compounds, with binding affinity scores for the DHFR enzyme of −10.6 and −9.3 kcal/mol, respectively. Antioxidant studies demonstrated that both extracts displayed antioxidant effects in the DPPH • , ABTS •+ , NO • , β-carotene-linoleic acid system, FRP, and TAC assays. Additionally, they provided significant protection to erythrocytes against H 2 O 2 -induced hemolysis. These findings, suggests that L. nobilis exhibits significant potential as a source of therapeutic agents for combating T. gondii infection and managing its associated adverse effects.

Abstract:Work-related injuries, often classified as occupational injuries (OI), stand out among visits due to external causes (accidents and violence) in health services. To describe the characteristics and factors associated with emergency room visits for OI, a cross-sectional study was conducted using data from the Survey of Violence and Injuries in Emergency Services (VIVA Inquérito 2011) in 24 state capitals and the Federal District. The prevalence of treatment for OI and prevalence ratios (PR) with confidence intervals of 95% (95%CI) were calculated. There were 29,463 emergency room visits due to accidental injuries in the population above 18 years of age. The prevalence of OI was 33.4% and was positively and significantly associated with the male gender, age 30-59 years old, industrial workers, agricultural sector or repair and maintenance services. The occurrence of OI was significantly higher in attendance for objects falling on people (PR = 3.37, 95% CI 2.80 to 4.05) and injuries due to perforating object (PR = 3.01, 95% CI 2.50-3.65). The results support the surveillance of external causes and direct public policies to promote occupational health.
Fate of labeled choline administered intraruminally to pregnant ewes given manganese-deficient or -supplemented rations.

Abstract:Tissue samples were taken from pregnant ewes and their 3-month fetuses 96 hours after intraruminal dosing of the ewes with tritiated choline. Starting 5 months before mating, one group of ewes had been given a manganese (Mn)-deficient ration and a second group had been given a Mn-supplemented (normal) ration. The results indicate that Mn depletion decreased the overall uptake of choline; the radioactivity of all tissues from the Mn-depleted group was lower than the radioactivity of the corresponding tissues from the group given a Mn-supplemented (normal) ration. The radioactivity of fetal tissues was also lower in the Mn-deficient group than it was in the Mn-supplemented group. Lower radioactivity concentrations were also observed in the whole plasma (sampled at various times after radioisotope dosing) and its lipid extract of Mn-depleted ewes than in the plasma and its lipid extract of Mn-supplemented ewes. There was no significant difference in the fatty acid composition of hepatic phosphatidyl choline between the two groups. However, the proportions of linoleic, stearic, and arachidonic acids were lower in fetal hepatic phosphatidyl choline than in maternal hepatic phosphatidyl choline.
Anti-parasitic activity and computational studies on a novel labdane diterpene from the roots of Vachellia nilotica

Abstract:Abstract A new labdane diterpene characterized as 18α- O - trans - p -feruloyl-15-methyl-8(17)-labdanoate has been isolated from the roots of Vachellia nilotica . Also isolated were p -coumaric acid, ferulic acid, stearic acid, lupeol, and a mixture of β-sitosterol and stigmasterol. The compounds were obtained after a series of column chromatography on silica gel, and their structures were elucidated using NMR and LC-MS analyses. The new diterpene showed good anti-parasitic activity with an IC 50 of 0.0177 μM against Trypanosoma brucei and 0.0154 μM against Leishmania major using an Alamar Blue assay. The compound also displayed very good inhibitory activity against Leishmania major compared to Trypanosoma brucei rhodesiense with a binding energy of −10.5 and −7.8 kcal/mol, respectively. Density functional theory analysis showed that the studied compound has low LUMO–HOMO energy, signifying a high chemical reactivity with the ability to donate electrons to electron-accepting species.

Abstract:Abstract Background : Malaria is a disease that is caused by Plasmodium parasite that has resulted in death of so many persons in the world especially children below 5 years. Plasmodium falciparum is one of the most widespread etiological agent for human malaria and has become increasingly resistant to standard antimalarial drugs. This study was therefore aimed at evaluating the in vitro antiplasmodial efficacy and cytotoxicity of Allanbackia floribunda stem bark, leaf and oil. Methods : Trager and Jensen method was used to culture Plasmodium falciparum and maintained in fresh O + human erythrocytes at 3% hematocrit in complete medium (Roswell Park Memorial Institute-RPMI 1640). Mark III test developed by WHO was used to assess the antiplasmodial activity of the plant’s crude extract and fractions against the ring stage of P. falciparum strains Pf3D7. Cytotoxicity was determined against Vero cell line using microculture tetrazolium (MTT). GC-FID was employed to identify bioactive constituents in the most active fraction. Results : The plant extracts showed varied degrees of inhibition of parasitaemia with IC 50Pf3D7 values ranging between 4.0 to 1000 μg/ mL. The methanol stem bark extract of A. floribunda exhibited very active antiplasmodial activity (IC 50Pf3D7 = 4.3 ± 0.17μg / mL), the leaf extract showed active antiplasmodial activity (IC 50Pf3D7 = 8.0 ± 0.28 μg / mL) while the oil extract was inactive against the parasite (IC 50 > 100 μg / mL). Both the leaf and stem bark extracts were found to be non-cytotoxic in contrast to the standard cytotoxic drug, doxorubicin. The selectivity indices (S.I.) of the extracts against the parasites were 13.32 and 8.18 for the stem bark and leaf, respectively. Among the stem bark fractions, dichloromethane (DCM) had the best inhibition against the P. falciparum parasite (DCM IC 50Pf3D7 of 1.51 μg/ mL) and was closely followed by the hydromethanol (HMet) fraction (HMet IC 50Pf3D7 of 5.0 μg/ mL) while that of ethylacetate (EAct) and hexane (Hxn) fractions were almost similar (IC 50 Pf3D7 of 6 and 6.25 μg/ mL, respectively).GC-FID analysis of the most active fraction, DCM, showed high presence antiplasmodial flavonoids (naringenin, luteolin, kaemferor and myricetin) and terpenes (α-pinene and β-caryophyllene) which may have been responsible for the excellent antiplasmodial action of the most active fraction. Conclusion: This investigation confirmed that A. floribunda stem bark has potent activity against P. falciparum and it is relatively safe to normal cell. The antiplasmodial action is credited to the presence of bioactive flavonoids and terpenes that may have acted singly or synergistically.

Abstract:A new flavanonol, 3-hydroxyerythrisenegalone (1), and four known compounds (2–5) were isolated from the extract of Mundulea sericea leaves. Investigation of the roots of this plant afforded an additional three known compounds (6–8). The structures were elucidated using NMR spectroscopic and mass spectrometric analyses. The absolute configuration of 1 was established using ECD spectroscopy. In an antiplasmodial activity assay, compound 1 showed good activity with an IC50 of 2.0 μM against chloroquine-resistant W2, and 6.6 μM against the chloroquine-sensitive 3D7 strains of Plasmodium falciparum. Some of the compounds were also tested for antileishmanial activity. Dehydrolupinifolinol (2) and sericetin (5) were active against drug-sensitive Leishmania donovani (MHOM/IN/83/AG83) with IC50 values of 9.0 and 5.0 μM, respectively. In a cytotoxicity assay, erythrisenegalone (3) showed significant activity on BEAS-2B (IC50 4.9 μM) and to HePG2 (IC50 10.8 μM) human cell lines. All the other compounds showed low cytotoxicity (IC50 > 30 μM) against human lung adenocarcinoma cells (A549), human liver cancer cells (HepG2), lung/bronchus cells (epithelial virus transformed) (BEAS-2B) and immortal human hepatocytes (LO2)

Abstract:This study aimed to assess the antileishmanial activity of biomolecules obtained from Olea europaea L. leaves and twigs recovered from eight Tunisian cultivars. The extraction was first carried out with 80% methanol, and then the obtained extract was fractionated using three solvents of increasing polarity: cyclohexane (CHX), dichloromethane (DCM) and ethyl acetate (EtOAc). The antileishmanial activity was determined against leishmanial strains responsible for cutaneous, visceral, and mucocutaneous leishmaniasis. The cyclohexane fraction of the leaves of cv. Chemlali from the region of Sidi-Bouzid exhibited the strongest leishmanicidal activity against all the tested leishmanial strains. The inhibition concentrations (IC 50 ) were 16.5, 14.5, and 7.4 μg mL −1 for Leishmania mexicana (cutaneous), Leishmania braziliensis (mucocutaneous), and Leishmania donovani (visceral), respectively. Interestingly, low cytotoxicity was observed on THP-1 cells with selective indexes (SI) ranging from 22.8 to 50.5. HPLC-HRMS and full-house NMR analysis allowed the identification of three triterpenic compounds, oleanolic acid (IC 50 = 64.1 μg mL −1 ), erythrodiol (IC 50 = 52.0 μg mL −1 ), and uvaol (IC 50 = 53.8 μg mL −1 ). Antileishmanial activity of uvaol and oleanolic acid has been previously reported. However, this work constitutes the first report of the antileishmanial activity of erythrodiol which showed combinatorial interaction with uvaol (IC 50 = 26.1 μg mL −1 ) against Leishmania tropica. The mixture of the three compounds, as major ones, exhibited an enhanced activity against Leishmania tropica (IC 50 = 16.3 μg mL −1 ) compared to erythrodiol alone or the combination of uvaol and erythrodiol. This finding is of great importance and needs further investigation.

Abstract:Bioassay guided fractionation of the stem barks of Isolona cooperi led to the isolation of a new lactone, apoprunellelactone (APL, 1), and two known compounds, 5-[1-hydroxyhexyl]-2H-furan-2-one (2) and oleic acid (3). Their structures were elucidated by spectral analysis including MS, UV, IR, 1D and 2D-NMR spectroscopy. Evaluated for its antiprotozoal activities, APL (1) was found to be the most active on Leishmania donovani and L. major promastigotes with EC50 values of 16.3 and 8.2 µM, respectively. Against Trypanosoma brucei brucei trypomastigote forms, the activity of APL was moderated (MEC = 38.0 µM). Its hemisynthetic ester acetic derivative (1c) was 2-42 times more active than that of the APL and reference drugs, justifying further in vivo evaluation of the two compounds (1 and 1c) on Leishmania sp and Trypanosoma brucei brucei/mice models.

Abstract:The past decade has witnessed the cloning of a new family of ion channels that are responsive to temperature. Six of these transient receptor potential (TRP) channels are proposed to be involved in thermosensation and are located in sensory nerves and skin. The TRPV1, TRPV2, TRPV3, and TRPV4 channels have incompletely overlapping functions over a broad thermal range from warm to hot. Deletion of the individual TRPV1, TRPV3, and TRPV4 channels in mice has established their physiological role in thermosensation. In all cases thermosensation is not completely abolished – suggesting some functional redundancy among the channels. Notably, the TRPV2 channel is responsive to hot temperatures in heterologous systems, but its physiological relevance in vivo has not been established. Cool and cold temperatures are sensed by TRPM8 and TRPA1 family members. Currently, the pharmaceutical industry is developing agonists and antagonists for the various TRP channels. For instance, TRPV1 receptor agonists produce hypothermia, while antagonists induce hyperthermia. Recent investigations have found that different regions of the TRPV1 receptor are responsive to temperature, nociceptive stimuli, and various chemical agents. With this information, it has been possible to develop a TRPV1 compound that blocks responses to capsaicin and acid while leaving temperature sensitivity intact. These channels have important implications for hyperthermia research and may help to identify previously unexplored mechanisms in different tissues that are responsive to thermal stress.
