Abstract:Soil salinity is a common environmental stress factor that limits agricultural production worldwide. Plants have evolved different strategies to achieve salt tolerance. miR393 has been identified as closely related to biotic and abiotic stresses, and targets F-box genes that encode auxin receptors. The miR393-TIR1/AFB2/AFB3 regulatory module was discovered to have multiple functions that manipulate the auxin response. This study focused on miR393 and one of its targets, TIR1, and found that they played potential roles in response to salt stress. Our results showed that overexpression of a miR393-resistant TIR1 gene (mTIR1) in Arabidopsis clearly enhanced salt stress tolerance, which led to a higher germination rate, less water loss, reduced inhibition of root elongation, delayed senescence, decreased death rate and stabilized Chl content. These plants accumulated more proline and anthocyanin, and displayed enhanced osmotic stress tolerance. The expression of some salt stress-related genes was altered, and sodium content can be reduced in these plants under salt stress. We proposed that highly increased auxin signaling by overexpression of mTIR1 may trigger auxin-mediated downstream pathways to enhance plant salt stress resistance by osmoregulation and increased Na+ exclusion.

Abstract:Transcription factors play vital roles in stress signal transduction and gene expression modulation during plant growth and development. Sequence analysis showed that OsAP21 contained an AP2/ERF domain of 57 amino acids. By comparison of deduced amino acid sequences of AP2/ERF-related proteins, we deduced that OsAP21 is a transcription factor gene, which belonging to rice AP2/ERF family CBF/DREB subfamily. Further, we report that transgenic Arabidopsis thaliana plants expressing the OsAP21 gene exhibited stronger growth than wild type plants under salt/drought stress. Analysis of RT-PCR for RD29B gene implied that OsAP21 over-expressed plants had a higher expression level of RD29B gene than wild type plants, and drought and salt treatments could enlarge these differences. Collectively, our results indicate that OsAP21 may play an important role in the response of transgenic Arabidopsis plants to salt/drought stresses.

Abstract:Small auxin-upregulated RNAs（SAURs） are genes regulated by auxin and environmental factors. In this study, we identified a SAUR gene in wheat, Ta SAUR75. Under salt stress,Ta SAUR75 is downregulated in wheat roots. Subcellular localization revealed that Ta SAUR75 was localized in both the cytoplasm and nucleus. Overexpression of Ta SAUR75 increased drought and salt tolerance in Arabidopsis. Transgenic lines showed higher root length and survival rate and higher expression of some stress-responsive genes than control plants under salt and drought stress. Less H 2 O 2 accumulated in transgenic lines than in control plants under drought stress. Our findings reveal a positive regulatory role of the auxin-responsive gene Ta SAUR75 in plant responses to drought and salt stress and provide a candidate gene for improvement of abiotic stress tolerance in crop breeding.

Abstract:Abstract Background High soil salinity can cause significant losses in rice productivity worldwide, mainly because salt inhibits plant growth and reduces grain yield. To cope with environmental changes, plants have evolved several adaptive mechanisms that involve the regulation of many stress-responsive genes. Results In this study, we identified OsSTAP1 , which encodes an AP2/ERF-type transcription factor, was rapidly induced by ABA, ACC, salt, cold, and PEG treatments. OsSTAP1 is localized to the nucleus and acts as a transcriptional activator in plant cells. Compared with wild type, transgenic lines overexpressing OsSTAP1 exhibited increased tolerance to salt stress with higher SOD, POD, and CAT activities, and lower Na + /K + ratios in the shoots. In addition, many other stress-responsive genes, including other ERF- and peroxidase-encoding genes, were upregulated in the OsSTAP1 -overexpression lines. Conclusion This study suggests that OsSTAP1 functions as an AP2/ERF transcriptional activator, and plays a positive role in salt tolerance by decreasing the Na + /K + ratio and maintaining cellular redox homeostasis.

Abstract:Haloxylon ammodendron is a typical desert xerophyte with worthy genetic resources, therefore, it is necessary to explore the resistance genes of H. ammodendron, and to improve the stress tolerance of other plant species through genetic engineering. In this study, a heat shock transcription factor gene, HaHSFA1, was firstly cloned from H. ammodendron and overexpressed in Arabidopsis thaliana. The sensitivity of transgenic plants to exogenous ABA, osmotic stress and salt stress were analyzed; the effects of overexpression of HaHSFA1 in Arabidopsis on salt tolerance were deeply investigated. The results indicated that overexpression of HaHSFA1 increased chlorophyll content in Arabidopsis, and the growth of plants was promoted. The leaf soluble sugar and proline contents in OE lines were significantly increased compared with those in WT. OE lines maintained lower osmotic potential and higher shoot water content. Overexpression of HaHSFA1 in Arabidopsis also enhanced antioxidant activity, which may be important for protecting plasma membrane integrity in cells. Overexpression of HaHSFA1 in Arabidopsis improved root activity and increased selective absorption and transport capacity for K+ over Na+, which was of great significance to reduce the toxic effect of Na+. This study indicated that overexpression of HaHSFA1 from H. ammodendron significantly elevates the salt tolerance of Arabidopsis, and HaHSFA1 has potential application values in improving abiotic stress tolerance in crops by genetic engineering.

Abstract:CCCH-type zinc-finger proteins constitute a large family playing key roles during plant development and growth. In the present study, we investigated the involvement of the CCCH-type zinc finger protein of AtZFP1 (At2g25900) in flowering and salt stress response in Arabidopsis. Compared with the wild type (WT), bolting and flowering were delayed in AtZFP1-overexpressing plants. Real-time PCR analysis of floral regulating genes in overexpressing Arabidopsis revealed that enhanced expression of FLC decreased the expressions of FT and SOC1. The Fv/Fm of overexpressing Arabidopsis lines was unchanged under salt stress. In contrast, ΦPSII activity and PSI oxidoreduction decreased in WT, overexpressing and mutant strains under salt stress conditions, with the smallest reduction in these parameters observed in the overexpressing strains. These results suggest that the CCCH zinc-finger protein AtZFP1 primarily controls flowering time by changing the expression of flowering genes under long-day conditions. The overexpression of this protein delayed flowering and increased the content and double-bond index of unsaturated fatty acids. Elevation of unsaturated fatty acid content might play important role in protecting the photosynthetic apparatus and maintaining the membrane function at salt stress by alleviating PSII and PSI photoinhibition.

Abstract:Aquaporin (AQP) proteins have been shown to transport water and other small molecules through biological membranes, which is crucial for plants to combat salt stress. However, the precise role of AQP genes in salt stress response is not completely understood in plants. In this study, a PIP1 subgroup AQP gene, designated TaAQP8, was cloned and characterized from wheat. Transient expression of TaAQP8-green fluorescent protein (GFP) fusion protein revealed its localization in the plasma membrane. TaAQP8 exhibited water channel activity in Xenopus laevis oocytes. TaAQP8 transcript was induced by NaCl, ethylene and H2O2. Further investigation showed that up-regulation of TaAQP8 under salt stress involves ethylene and H2O2 signaling, with ethylene causing a positive effect and H2O2 acting as a negative factor. Overexpression of TaAQP8 in tobacco increased root elongation compared with controls under salt stress. The roots of transgenic plants also retained a high K+/Na+ ratio and Ca2+ content, but reduced H2O2 accumulation by an enhancement of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) activities under salt stress. Further investigation showed that whole seedlings from transgenic lines displayed higher SOD, CAT and POD activities, increased NtSOD and NtCAT transcript levels, and decreased H2O2 accumulation and membrane injury under salt stress. Taken together, our results demonstrate that TaAQP8 confers salt stress tolerance not only by retaining high a K+/Na+ ratio and Ca2+ content, but also by reducing H2O2 accumulation and membrane damage by enhancing the antioxidant system.
Analysis of resistance to alkaline salt stress in Arabidopsis thaliana with overexpression of OsiSAP1 gene

Abstract:The purpose of this study was to study the stress effects of rice ISAP transcription factor family containing zinc finger proteins of A20/AN1 domain and the mechanism of saline-alkali tolerance during rice growth and development. In this study, the ORF reading frame of OsiSAP1 (LOC_Os09g31200) gene 513bp was cloned from the leaves of rice Oryza sativa. L11, and the homology ratio NCBI 170 amino acids was as high as 71.51% with Panicum miliaceum L. The phylogenetic tree based on the nucleotide comparison of the open reading frame shows that it is on the same branch as Oryza officinalis. Through qRT-PCR detection in various tissues and organs of rice Oryza sativa. L11: root, stem, leaf and anther, it was found that the expression of mRNA in root was higher, and Gene gun-transformed onion epidermal cell method detected fluorescence signal of 35S-OsiSAP1-GFP fusion protein localized in the nucleus. The analysis of salt-alkali tolerance of rice overexpressing OsiSAP1 gene showed that the fresh weight and root length of transgenic lines were better than those of wild-type lines at germination stage and late germination stage under NaHCO3 treatment. It is speculated that OsiSAP1 transcription factors are involved in the regulation of saline-alkali tolerance in rice.

Abstract:Drought and salt are two major abiotic stresses that severely impact plant growth and development, as well as crop production. A previous study showed that OsOSCA1.4 , one of eleven rice OSCAs ( OsOSCAs ), complements hyperosmolality-induced [Ca 2+ ] cyt increases (OICI cyt ), salt stress-induced [Ca 2+ ] cyt increases (SICI cyt ) and the associated growth phenotype in Arabidopsis osca1 ( reduced hyperosmolality-induced [Ca 2 + ] cyt increase 1 ). In this study, Except for OsOSCA2.3 and OsOSCA4.1 , we generated independent transgenic lines overexpressing eight other OsOSCAs in the osca1 to explore their functions in osmotic Ca 2+ signalling, stomatal movement, leaf water loss, and root growth in response to hyperosmolality and salt stress. Similar to OsOSCA1.4 , overexpression of OsOSCA1.1 or OsOSCA2.2 in osca1 complemented OICI cyt and SICI cyt , as well as stomatal closure and root growth in response to hyperosmolality and salt stress treatments, and drought-related leaf water loss. In addition, overexpression of OsOSCA1.2 , OsOSCA1.3 or OsOSCA2.1 in osca1 restored OICI cyt and SICI cyt , whereas overexpression of OsOSCA2.5 or OsOSCA3.1 did not. Moreover, osca1 overexpressing these five OsOSCAs exhibited various abiotic stress-associated growth phenotypes. However, overexpression of OsOSCA2.4 did not have any of these effects. These results indicated that multiple members of the OsOSCA family have redundant functions in osmotic sensing and diverse roles in stress adaption.

Abstract:SUMMARY Soil salinization is becoming a great threat that reduces crop productivity worldwide. In this study, we found that rice allantoate amidohydrolase (OsAAH) expression was significantly upregulated by salt stress, and its overexpression conferred salt tolerance at the seedling stage. Compared to wild type (WT), the contents of ureides (allantoin and allantoate) were significantly increased in Osaah mutants and reduced in OsAAH overexpression lines both before and after salt treatments. Exogenous allantoin significantly promoted salt tolerance in OsAAH overexpression, but not in Osaah mutants. Subcellular localization showed that OsAAH was also localized to the peroxisomes in addition to the previously reported endoplasmic reticulum (ER). The differential expression of peroxisome‐related genes was identified between Osaah mutants and WT. Furthermore, the contents of H2O2 and malondialdehyde (MDA) were significantly accumulated in Osaah mutants and reduced in OsAAH overexpression lines. The activities of antioxidant enzymes were significantly reduced in Osaah mutants and enhanced in OsAAH overexpression under NaCl treatment. The transcription factor OsABI5 could directly bind to OsAAH promoter and activate OsAAH expression. Our findings reveal that OsAAH could be induced by salt stress through the activation of OsABI5 and then confer salt tolerance by enhancing the scavenging capacity of reactive oxygen species (ROS), which contributes to rice breeding in salt tolerance.

Abstract:MYB-type transcription factors (TFs) play essential roles in plant growth, development and respond to environmental stresses. Role of MYB-related TFs of rice in drought stress tolerance is not well documented. Here, we report the isolation and characterization of a novel MYB-related TF, OsMYB48-1, of rice. Expression of OsMYB48-1 was strongly induced by polyethylene glycol (PEG), abscisic acid (ABA), H2O2, and dehydration, while being slightly induced by high salinity and cold treatment. The OsMYB48-1 protein was localized in the nucleus with transactivation activity at the C terminus. Overexpression of OsMYB48-1 in rice significantly improved tolerance to simulated drought and salinity stresses caused by mannitol, PEG, and NaCl, respectively, and drought stress was caused by drying the soil. In contrast to wild type plants, the overexpression lines exhibited reduced rate of water loss, lower malondialdehyde (MDA) content and higher proline content under stress conditions. Moreover, overexpression plants were hypersensitive to ABA at both germination and post-germination stages and accumulated more endogenous ABA under drought stress conditions. Further studies demonstrated that overexpression of OsMYB48-1 could regulate the expression of some ABA biosynthesis genes (OsNCED4, OsNCED5), early signaling genes (OsPP2C68, OSRK1) and late responsive genes (RAB21, OsLEA3, RAB16C and RAB16D) under drought stress conditions. Collectively, these results suggested that OsMYB48-1 functions as a novel MYB-related TF which plays a positive role in drought and salinity tolerance by regulating stress-induced ABA synthesis.

Ectopic Expression of OsSta2 Enhances Salt Stress Tolerance in Rice

Abstract:Salt stress can severely reduce crop yields. To understand how rice (Oryza sativa) plants respond to this environmental challenge, we investigated the genes involved in conferring salt tolerance by screening T-DNA tagging lines and identified OsSta2-D (Oryza sativa Salt tolerance activation 2-Dominant). In that line, expression of OsSta2 was enhanced by approximately eightfold when compared with the non-transformed wild type (WT). This gene was highly expressed in the callus, roots, and panicles. To confirm its role in stress tolerance, we generated transgenic rice that over-expresses OsSta2 under a maize ubiquitin promoter. The OsSta2-Ox plants were salt-tolerant at the vegetative stage, based on our calculations of chlorophyll fluorescence (Fv/Fm), fresh and dry weights, chlorophyll concentrations, and survival rates. Under normal paddy field conditions, the Ox plants were somewhat shorter than the WT control but had improved agronomic traits such as higher total grain yield. They were also more tolerant to osmotic stress and hypersensitive to abscisic acid. Based on all of these results, we suggest that OsSta2 has important roles in determining yields as well as in conferring tolerance to salt stresses.
OsIAA18, an Aux/IAA Transcription Factor Gene, Is Involved in Salt and Drought Tolerance in Rice

Abstract:Auxin/indoleacetic acid (Aux/IAA) proteins play an important regulatory role in the developmental process of plants and their responses to stresses. A previous study has shown that constitutive expression of OsIAA18 , an Aux/IAA transcription factor gene of rice improved salt and osmotic tolerance in transgenic Arabidopsis plants. However, little work is known about the regulatory functions of the OsIAA18 gene in regulating the abiotic stress tolerance of rice. In this study, the OsIAA18 gene was introduced into the rice cultivar, Zhonghua 11 and the OsIAA18 overexpression in rice plants exhibited significantly enhanced salt and drought tolerance compared to the wild type (WT). Moreover, overexpression of OsIAA18 in rice increased endogenous levels of abscisic acid (ABA) and the overexpression of OsIAA18 in rice plants showed hypersensitivity to exogenous ABA treatment at both the germination and postgermination stages compared to WT. Overexpression of OsIAA18 upregulated the genes involved in ABA biosynthesis and signaling pathways, proline biosynthesis pathway, and reactive oxygen species (ROS)-scavenging system in the overexpression of OsIAA18 in rice plants under salt and drought stresses. Proline content, superoxide dismutase (SOD), and peroxidase (POD) activities were significantly increased, whereas malonaldehyde (MDA), hydrogen peroxide (H 2 O 2 ), and superoxide anion radical (O 2 – ) content were significantly decreased in the transgenic plants under salt and drought stresses. Taken together, we suggest that OsIAA18 plays a positive role in drought and salt tolerance by regulating stress-induced ABA signaling. The OsIAA18 gene has a potential application in genetically modified crops with enhanced tolerance to abiotic stresses.

Abstract:Gibberellin (GA) is an important phytohormone that regulates plant growth and development. GA-2-oxidases (GA2oxs) have been found to affect plant height, tillering and stress tolerance by inactivating endogenous bioactive GAs. The present study aimed to isolate GA2oxs genes from tall fescue (Festuca arundinacea), analyze their expression level under abiotic stresses (10% PEG6000 induced drought stress, 150 mM NaCl induced salt stress, heat stress at 38 °C, and cold stress at 4 °C) and search for valuable genes for improving stress tolerance. Among eight FaGA2ox genes, FaGA2ox4 was found to represent a highest expression level at 24 h of PEG-induced drought stress and was chosen for further analysis. Compared with wildtype plants, transgenic rice plants overexpressing FaGA2ox4 exhibited a dwarf and high-tillering phenotype and the dwarf phenotype was partially compensated by foliar application of 10 μM GA3. In addition, transgenic plants showed improved drought stress tolerance, as manifested by higher leaf relative water content and lower electrolyte leakage than wildtype. Further qRT-PCR analysis demonstrated that the enhanced drought stress tolerance may be associated with decreased expression levels of genes involved in chlorophyll degradation (OsSGRL) and GA synthesis (OsGA3ox1) as well as increased expression of ABA-synthesizing gene (OsNCED3) in transgenic plants overexpressing FaGA2ox4.

Abstract:Sumoylation is one of the post translational modifications, which affects cellular processes in plants through conjugation of small ubiquitin like modifier (SUMO) to target substrate proteins. Response to various abiotic environmental stresses is one of the major cellular functions regulated by SUMO conjugation. SIZ1 is a SUMO E3 ligase, facilitating a vital step in the sumoylation pathway. In this report, it is demonstrated that over-expression of the rice gene OsSIZ1 in Arabidopsis leads to increased tolerance to multiple abiotic stresses. For example, OsSIZ1-overexpressing plants exhibited enhanced tolerance to salt, drought, and heat stresses, and generated greater seed yields under a variety of stress conditions. Furthermore, OsSIZ1-overexpressing plants were able to exclude sodium ions more efficiently when grown in saline soils and accumulate higher potassium ions as compared to wild-type plants. Further analysis revealed that OsSIZ1-overexpressing plants expressed higher transcript levels of P5CS, a gene involved in the biosynthesis of proline, under both salt and drought stress conditions. Therefore, proline here is acting as an osmoprotectant to alleviate damages caused by drought and salt stresses. These results demonstrate that the rice gene OsSIZ1 has a great potential to be used for improving crop's tolerance to several abiotic stresses.

Abstract:Drought and salinity are major abiotic stresses affecting rice production. To improve plant tolerance to salinity and drought, we overexpressed rice Na+/H+ exchangers (OsNHX1) and H+-pyrophosphatase in tonoplasts (OsVP1) in a japonica elite rice cultivar, Zhonghua 11. Compared with our wild-type control, transgenic plants overexpressing both genes incurred less damage when exposed to long-term treatment with 100 mM NaCl or water deprivation. Under high-saline conditions, the transformants accumulated less Na+ and malondialdehyde in the leaves, thereby allowing the plants to maintain a low level of leaf water potential and reduce stress-induced damage. Those transgenics also had higher photosynthetic activity during the stress period. Under those conditions, they also showed an increase in root biomass, which enabled more water uptake. These results suggest that OsVP1 and OsNHX1 improve the tolerance of rice crops against drought and salt by employing multiple strategies in addition to osmotic regulation.

Abstract:Late embryogenesis abundant (LEA) proteins are involved in diverse abiotic stresses tolerance in many different organisms. Our previous studies have shown that the heterologous expression of OsLEA1a interfered with the resistance of Escherichia coli to abiotic stresses. However, in the present study, based on growth status and physiological indices of rice plant, the overexpression of OsLEA1a in rice conferred increased resistance to abiotic stresses compared with the wild-type (WT) plants. Before applying abiotic stresses, there were no significant differences in physiological indices of rice seedlings. After NaCl, sorbitol, CuSO4 and H2O2 stresses, the transgenic lines had lower relative electrical conductivity, malondialdehyde and lipid peroxidation, greater the contents of proline, soluble sugar and glutathione, and higher the activities of superoxide dismutase, catalase and peroxidase than the WT plants. The results indicate that the OsLEA1a gene is involved in the protective response of plants to various abiotic stresses by inhibiting cell membrane damage and enhancing reactive oxygen species scavenging capacity. It was speculated that post-translational modification causes OsLEA1a functional differences in E. coli and rice. The present study shows that OsLEA1a could be a useful candidate gene for engineering abiotic stress tolerance in cultivated plants.

Abstract:Trehalose plays an important role in metabolic regulation and abiotic stress tolerance in a variety of organisms. In plants, its biosynthesis is catalyzed by two key enzymes: trehalose-6-phosphate synthase (TPS) and trehalose-6-phosphate phosphatase (TPP). The genome of rice (Oryza sativa) contains 11 OsTPS genes, and only OsTPS1 shows TPS activity. To demonstrate the physiological function of OsTPS1, we introduced it into rice and found that OsTPS1 overexpression improved the tolerance of rice seedling to cold, high salinity and drought treatments without other significant phenotypic changes. In transgenic lines overexpressing OsTPS1, trehalose and proline concentrations were higher than in the wild type and some stress-related genes were up-regulated, including WSI18, RAB16C, HSP70, and ELIP. These results demonstrate that OsTPS1 may enhance the abiotic stress tolerance of plants by increasing the amount of trehalose and proline, and regulating the expression of stress-related genes. Furthermore, we found that overexpression of some Class II TPSs also enhanced plant tolerance of abiotic stress. This work will help to clarify the role of trehalose metabolism in abiotic stress response in higher plants.
Over-expression of an Arabidopsis Δ-Oat Gene Enhances Salt and Drought Tolerance in Transgenic Rice

Abstract:δ-OAT, ornithine-δ-aminotransferase, is the key enzyme involved in proline biosynthesis. In this study the Arabidopsis δ-OAT gene was transferred into rice ( Oryza sativa L. ssp japonica cv. Zhongzuo 321), whose successful integration was demonstrated by PCR and Southern blot analysis. The over-expression of the gene in transgenic rice was also confirmed. Biochemical analysis showed that, under salt or drought stress conditions, proline contents in the leaves and roots in transgenic rice plants were 5- to 15-fold of those in non-transgenic controls. Under stress conditions, germinating rate of transgenic lines is higher than that of controls. Although the growth of rice plants tested were more and more retarded with the increasing of NaCl concentration, the transgenic plants grow faster compared to the controls under the same stress condition. Meanwhile, the resistance to KC1 and MgSO 4 stresses was also found enhanced in transgenic rice. Furthermore, the over-expression of δ-OAT also improved the yield of transgenic plants under stress conditions. The average yield per plant of transgenic lines increases about 12%–41% more than that of control lines under 0.1 mol/L NaCl stress. These data indicated that the over-expression of δ-OAT , with the accumulation of proline, resulted in the enhancement of salt and drought tolerance and an increase of rice yield, which is of significance in agriculture.

Abstract:Tall fescue (Festuca arundinacea Schreb.) is a cool-season perennial grass, which has been conventionally grown in the temperate area. However, as a major type of cool-season turf grass, its growth has been extended to the sub-tropical climate or even to the transitional climate between the sub-tropical and the tropical, and, in some cases, to heavily salinized lands. The extended growth imposes a serious challenge to its tolerance to the abiotic stress, particularly to drought, salt and high temperature. Here, we report a successful introduction of Arabidopsis AtHDG11 into the tall fescue via Agrobacterium-mediated transformation. The ectopic overexpression of AtHDG11 under the control of CaMV 35S promoter with four enhancers resulted in significantly enhanced tolerance to drought and salt stress. No obvious adverse effects on growth and development were observed in the transgenic plants. The enhanced stress tolerance was associated with a more extensive root system, a lower level of malondialdehyde, a nearly normal Na+/K+ ratio, a higher level of proline and a kinetically accelerated induction of SOD and CAT activities observed in the transgenic plants during drought and/or salt stress, indicating that an enhanced ROS scavenging capability might play a significant role in the acquired tolerance to the abiotic stress.
