Abstract:In recent years, fundamental and applied researches on cellulase enzyme have not only generated significant scientific knowledge but also have revealed their enormous potential in biotechnology. Growing attention has been devoted to its bioconversion of biomass into fuel ethanol, considered the cleanest liquid fuel alternative to fossil fuels. Significant advances have been made towards the production and alteration technology of cellulase enzyme. This review simply introduces cellulose and cellulase enzyme, gives a broad overview of the current research status of cellulase enzyme, briefly refers to its applied fields, and lastly summarizes its promising prospects.
Impact of High-Shear Extrusion Combined With Enzymatic Hydrolysis on Rice Properties and Chinese Rice Wine Fermentation

Abstract:To better understand how high-shear extrusion combined with enzymatic hydrolysis affects the rice properties and subsequent fermentation, a twin screw extruder incorporated with rice treatment with thermostable α-amylase was used for simultaneous saccharification and fermentation (SSF) of Chinese rice wine. Seven pretreatment methods for rice were compared, and their impacts on rice properties and quality indices of final wine were studied. Enzymatic extruded rice had higher water solubility index (WSI), extent of gelatinization (GE), but lower water absorption index (WAI) and viscosity than those of other treated rice due to a strong synergy of shear action and enzymatic hydrolysis on the degradation and gelatinization of starch. The modification of enzymatic autoclaved rice was smaller than that of enzymatic extruded rice mainly for the lack of mechanical shearing force. Although extrusion with smaller particle size tended to have higher susceptibility to enzyme, the effect was not dramatic. This implied that manipulating particle size had limited effect on the modification of rice properties. Correlation analysis showed that the ethanol yield (EY), fermentation efficiency (FE), amino acid (AA), and soluble solid content (SSC) of finished rice wine increased as rice properties improved by enzymatic extrusion. Overall, simultaneous high-shear extrusion and enzymatic hydrolysis can be an attractive alternative to obtain modified rice with desirable properties for manufacturing Chinese rice wine or other fermentation products.

Abstract:Bamboo leaves extract (BLE) contains various effective ingredients, including phenolic compounds. In this study, the effect of BLE on ethyl carbamate (EC) formation was investigated in Chinese yellow rice wine brewing with three different fermentation starters (Saccharomyces cerevisiae, Saccharomyces cerevisiae and Lactobacillus brevis, and Chinese yeast). As a result, BLE showed significant inhibition effect on EC in multi-microbial fermented rice wine, by preventing the reactions between urea/citrulline and ethanol. We found that BLE had influence on arginine transport (GAP1, CAN1, ALP1, and VBA2 gene) in Saccharomyces cerevisiae (S. cerevisiae), which significantly up-regulated arginine uptake gene expression in vacuole (VBA2 gene) so that inhibited arginine metabolism. Besides, the presence of BLE could improve the overall quality of Chinese yellow rice wine. Consequently, it was worthwhile applying BLE to Chinese rice wine fermentation, especially the wine brewing with S. cerevisiae and Lactobacillus brevis starter.
Characterization of Volatile Flavor Compounds in Chinese Rice Wine Fermented from Enzymatic Extruded Rice

Abstract:Enzymatic extrusion, instead of traditional steam cooking, to treat rice is an efficient and alternative pretreatment for Chinese rice wine fermentation. In order to determine the formation of volatiles in enzymatic extrusion-processed rice wine (EE), and to confirm its characteristic flavor compounds, headspace solid-phase micro-extraction followed by GC-MS was used. A total of 66 volatile compounds were identified in EE. During fermentation, most volatiles generated from enzymatic extruded rice had the similar trends with those from steam-cooked rice, but the differences in the concentration of volatiles indicated a changed balance of flavors release caused by enzymatic extrusion. Besides, the concentrations and sorts of volatiles in EEs fermented from different rice particle sizes, were not dramatically different. By principal component analysis, EE could be distinctly separated from other traditional Chinese rice wines according to its characteristic volatiles, namely, 2-heptanol, 1-octen-3-ol, ethyl 4-hydroxybenzoate, methylpentyl 2-propenoate, -hexalactone, and 4-vinylguaiacol. Practical Application Enzymatic extrusion liquefaction has been a popular thermal treatment for cereals, and gradually being applied in fermentation and liquor-making industry all over the world. The characterization of volatile flavor compounds in Chinese rice wine processed by enzymatic extrusion liquefaction pretreatment, might be made use not only for a better understanding of this new-type rice wine, but for the further utilization of enzymatic extrusion in other wine or alcohol production as well.
Identification and expression of bifunctional acid urea-degrading enzyme/urethanase from Enterobacter sp. R-SYB082 and its application in degradation of ethyl carbamate in Chinese rice wine (Huangjiu)

Abstract:Background: Ethyl carbamate (EC) is a potential carcinogen existing in fermented foods such as Chinese rice wine (Huangjiu). Since urea is an important precursor of EC, the degradation of urea could be an effective way to reduce EC in foods. Results: In this study, an Enterobacter sp. R-SYB082 with acid urea degradation characteristics was obtained through microbial screening. Further research isolated a new acid urea-degrading enzyme from R-SYB082 strain - ureidoglycolate amidohydrolase (UAH) - which could degrade EC directly. The cloning and expression of UAH in Escherichia coli BL21 (DE3) suggested that the activity of urea-degrading enzyme reached 3560 U L-1 , while urethanase activity reached 2883 U L-1 in the optimal fermentation condition. The enzyme had the dual ability of degrading substrate urea and product EC. The removal rate of EC in Chinese rice wine could reach 90.7%. Conclusion: This study provided a new method for the integrated control of EC in Chinese rice wine and other fermented foods. © 2022 Society of Chemical Industry.

Abstract:SummaryTo promote the industrial production of Chinese rice wine made by enzymatic extrudates, the first is to ensure the consistency of the raw material. That is, an evaluation standard of enzymatic extrudate should be set up. It was studied for the first time. By studying the changes of seven indexes of samples at different sites of extruder barrel and the die along the extrusion direction and making the correlation analysis to select the representative indexes. The results showed that the change trends of dextrose equivalent (DE), molecular weight distribution (MWD) and oligosaccharide composition (OC) were the same, and these indexes were more affected by mechanical force than by temperature. The ability of degradation of different zones was: conveying area < kneading area << exit of the die. There was little difference in extent of gelatinisation (GE) at the temperature where the enzyme activity was relatively high. Different from DE value, water solubility index (WSI) rose gradually. Surprisingly, DE was negatively correlated with GE, and positively correlated with oligosaccharides except DP1 and DP6, and had no significant correlation with water absorption index (WAI) and WSI. In conclusion, DE, WSI, DPD (especially DP6) could be used as a comprehensive evaluation index of enzymatic extrudates.

Abstract:Glucoamylase, which catalyses the hydrolysis of the alpha-1,4 glycosidic bonds of starch, is an important industrial enzyme used in starch enzymatic saccharification. In this study, a glucoamylase gene from Aspergillus awamori, under the control of the promoter of seed storage protein Gt1, was introduced into rice by Agrobacterium-mediated transformation. Significant glucoamylase activity was detected specifically in the seeds but not other tissues of the transgenic rice lines. The highest enzymatic activity was found in the transgenic line Bg17-2, which was estimated to have about 500 units per gram of seeds (one unit is defined as the amount of enzyme that produces 1 micromol of reducing sugar in 1 min at 60 degrees C using soluble starch as substrate). The optimum pH for the activity of the rice produced enzyme is 5.0-5.5, and the optimum temperature is around 60 degrees C. One part of this transgenic glucoamylase rice seed flour fully converted 25 parts of corn starch pre-liquefied by an alpha-amylase also produced by a transgenic rice into glucose in 16 h incubation. This study suggests that this hydrolysis enzyme may substitute commercial fermentation enzymes for industrial starch conversion.

Abstract:In order to make full use of the protein resource in rice wine grains (RWG), the extraction technology of protein from RWG was optimized, through single factor experiment and Orthogonal experiment. The optimum conditions for enzymic extraction of protein were determined: enzyme active unit 6 000 u (enzyme dosage [E]/[S]=1%), the ratio of stuff to water 1∶8, pH 10, reaction time 3 h, temperature 50 ℃. Under these conditions, the protein extraction rate was 74.42%.
Optimized Solid-State Fermentation of Aspergillus Niger for Production of Feed-Purpose Β-Glucanase and Xylanase Using Rice Residues from Mass Production of Fungal Biocontrol Agents

Abstract:Rice residues from mass production of fungal biocontrol agents against insect pests were explored for use in the production of feed-purpose β-glucanase and xylanase,which is usually achieved by solid-state fermentation (SSF) of Aspergillus niger with wheat bran. A mixture of 70% wheat bran and 30% milled rice was selected as an acceptable substrate in the trials of 10 ratios of the two components and yielded 353.8 and 343.8 U·g-1 substrate for β-glucanase and xylanase after 3-day SSF,respectively. Factors found to significantly affect the enzyme yields in the mixture during SSF were the type of metal ion as trace nutrition,the content of (NH4)2SO4 as supplementary nitrogen,and the concentration of Tween-80 and pH in the water used to stir the mixture for SSF. Combinations of the selected factors were further optimized for enzyme production in triple four-factor,three-level orthogonal experiments. The optimized combination of the rice-bran mixture consisted of 0.1% KH2PO4,1.0% (NH4)2SO4 and the water containing 0.3% Tween-80 with pH 5. When this optimized mixture was used as substrate in three repeated trials of 54-h SSF at 25 ℃ for confirmation,the mean (±SD) yields of β-glucanase and xylanase reached (716.1±9.2) and (453.0±62.5) U·g-1,which were greatly enhanced compared to those before optimization. The results highlight an alternative approach to utilizing the rice residues for the enzyme production at the reduced cost of 30% wheat bran and would facilitate to establish a rice-based,environment-friendly technology for bulk production of fungal biocontrol agents.
Study on the Relationship Between the Degradation Degrees of Glutinous Rice Starch Extruded with Different Α‐amylases and the Qualities of Chinese Rice Wine

Abstract:SUMMARYIn view that enzymatic extrusion can change the degradation degree of raw materials, this paper studied the regulation rules of four indexes of degradation degree of enzymatically extruded glutinous rice flour and the correlation between these indexes and the quality indexes of brewed Chinese rice wine. The results showed that at appropriate water content, dextrose equivalent (DE) and water solubility index (WSI) were linearly related to the enzyme content in extrusion with thermostable α‐amylase (TS‐αA) and mesophilic α‐amylase (MS‐αA), respectively. But water absorption index (WAI) was not significantly affected by enzyme content. The percentages of each degree of polymerisation (DP) (1–7) and that of the sum of the rest DPs called other DP were the fundamental reasons for different DE values, WSI, WAI and quality indexes of Chinese rice wine. DP1 was significantly positively correlated with alcohol degree, and negatively correlated with total acid. DP2–DP7 (except DP4) was significantly positively correlated with amino acid nitrogen. Besides, other DP was significantly positively correlated with total sugar and total acid. The percentages of DP1, DP2–DP7 and other DP could be respectively adjusted by changing enzyme content in extrusion with MS‐αA, MS‐αA/TS‐αA and TS‐αA to control the related quality indexes of Chinese rice wine.

Abstract:The thermostable endo-1,4-β-glucanase (E1) from Acidothermus cellulolyticus, is a useful enzyme for commercial hydrolysis of cellulose into glucose. A codon-optimized synthetic gene encoding this enzyme was transformed into rice (Oryza sativa L. ssp. japonica) under the control of the rice seed storage protein Gt1 promoter. The transgenic line C19 was identified as the one with the highest endoglucanase activity among the total of 36 independent transgenic lines obtained. The cellulase activity in the C19 seeds was estimated at about 830U/g of dried seeds using CMC as substrate. The enzymes produced in the seeds had an optimum pH of 5.0 and optimum temperature of 80°C, which is similar to the enzymes produced by the native bacterium host. This study demonstrates that the transgenic rice seeds could be used as a bioreactor for production of enzymes for cellulosic biomass conversion.

Abstract:Xylanases have broad applications in the food industry to decompose the complex carbohydrate xylan. This is applicable to enhance juice clarity, improve dough softness, or reduce beer turbidity. It can also be used to produce prebiotics and increase the nutritional value in foodstuff. However, the low yield and poor stability of most natural xylanases hinders their further applications. Therefore, it is imperative to explore higher-quality xylanases to address the potential challenges that appear in the food industry and to comprehensively improve the production, modification, and utilization of xylanases. Xylanases, due to their various sources, exhibit diverse characteristics that affect production and activity. Most fungi are suitable for solid-state fermentation to produce xylanases, but in liquid fermentation, microbial metabolism is more vigorous, resulting in higher yield. Fungi produce higher xylanase activity, but bacterial xylanases perform better than fungal ones under certain extreme conditions (high temperature, extreme pH). Gene and protein engineering technology helps to improve the production efficiency of xylanases and enhances their thermal stability and catalytic properties.

Abstract:As functional materials,chitin and chitosan distributes widely and has a unique set of characteristics: non-toxicity,gel forming properties,biocompatibility,biodegradability to harmless products,remarkable affinity to proteins and so on.Owing to these characteristics,chitin-and chitosan-based materials,as yet underutilized,are predicted to be widely exploited in the near future,among others as enzyme immobilization supports.Based on recent research,we summarized recent advances in the physical and chemical characteristic of chitin and chitosan,prepration methods of support for enzyme immobilization,utilization on food and non-food industry,environmental protection,enzyme purification and medical treatment.

Abstract:In order to solve the problem of single aroma with rice wine brewed by uncooked material method, Zanthoxylum bungeanum, Glycyrrhiza uralensis Fisch, Osmanthus fragrans, glutinous rice and other materials were used to brew rice wine. Taking sensory evaluation and alcohol content as indexes, the suitable fermentation process parameters of compound rice wine with uncooked material method were determined by single factor test and response surface test. The results showed that when 1.7% Z. bungeanum seed, 1.0% G. uralensis Fisch, 3.0% O. fragrans and 1.5% koji were added to glutinous rice, the compound rice wine with 90.8 score of sensory evaluation and 9.1%vol alcohol content could be obtained by 18 days fermentation at 26.9 °C. The compound rice wine had bright yellow transparent, typical harmonious fruity aroma of the wine, mellow taste. The contents of limonene and linalool were 5.87 mg/L and 3.14 mg/L, respectively. Therefore, a theoretical basis for industrialized production was provided.

Abstract:An acid urease from Providencia rettgeri JN-B815 was purified via ultrasonication, ethanol precipitation, and DEAE ion-exchange column chromatography. It was found that the enzyme exhibits not only urease activity, but also urethanase activity, which made it possible to reduce EC already existed or would produce and its precursor urea at the same time. Then, crosslinked enzyme aggregates of P. rettgeri urease (PRU-CLEAs) were prepared using genipin as crosslinking agent. The purification process of acid urease, the effects of genipin concentration, and crosslinking time on PRU-CLEAs activity were investigated. The crosslinking was performed at pH 4.5 for 2.5 h, using 0.3% genipin as crosslinking agent, and 0.3 g · L(-1) bovine serum albumin as protein feeder. Using the obtained PRU-CLEAs, the removal rate of urea was up to 9.31 mg · L(-1) · h(-1). The removal rate of urea was still up to 7.56 mg · L(-1) · h(-1) after PRU-CLEAs was re-used for 6 times. When PRU-CLEAs were applied in a batch stirred and membrane reactor, the removal rate of urea in rice wine reached 5.16 mg · L(-1) · h(-1) and the removal rate of EC was 9.21 μg · L(-1) · h(-1). Furthermore, the treatment with PRU-CLEAs revealed no significant change of volatile flavor substances in Chinese rice wine. Thus PRU-CLEAs have great potential in the elimination of EC in Chinese rice wine.

Abstract:The effect of simultaneous extrusion and enzymatic hydrolysis on the phenolics and antioxidant activities of glutinous rice and further fermented Chinese rice wine was investigated. Great modification on rice properties (water absorption/solubility index) related to higher total phenolic content (TPC) retention (87.73%) was determined in enzymatically extruded rice compared to traditionally cooked or extruded rice (37.61–50.85% TPC retention). A total of ten phenolic acids (in free/bound forms) with great loss rates were identified in different treated rice, except the enzymatically extruded rice showing effective inhibition of phenolics loss (only 2.66, 2.04, 22.49, 1.89 and 6.37% loss for total gallic, chlorogenic, vanillic, syringic, and ferulic acids, respectively). After fermentation, the level of p-hydroxybenzoic and p-coumaric acids increased predominantly, while chlorogenic acid dramatically decreased to an extremely low level, indicating a changed chemical proportion of phenolics in Chinese rice wine. Compared with raw rice, enzymatically extruded rice achieved a higher antioxidant activity (DPPH, ABTS, reducing capacity and FRAP), probably due to the synergy of phenolic retention and Maillard-derived products formation. Moreover, this advantage of simultaneous extrusion and enzymatic hydrolysis could be further exhibited in Chinese rice wine products via fermentation processing.
