What problem does this paper attempt to address?
The paper aims to address the question of how the human V2 vasopressin receptor (V2R) is recognized and activated by its endogenous ligand arginine vasopressin (AVP). V2R belongs to the vasopressin (VP)/oxytocin (OT) receptor subfamily, which plays a crucial role in controlling water homeostasis, social behavior, and sexual reproduction. However, due to the lack of structural information, the specific mechanism by which AVP interacts with V2R and triggers its active state has remained unclear, limiting drug design targeting V2R. By resolving the high-resolution cryo-electron microscopy structure of V2R in complex with AVP, researchers have revealed the precise binding mode of AVP to V2R and its activation mechanism, providing an important structural basis for understanding V2R function and developing related drugs.
