Abstract:A biosensor was fabricated by immobilizing glucose oxidase (GOD) into nanostructured graphene (GRA)-conducting polyaniline (PANI) nanocomposite, which was based on electrochemical polymerization of aniline in GRA synthesized by using electrochemical expansion of graphite in propylene carbonate electrolyte. Scanning electron spectroscopy (SEM), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were used to characterize the morphology and performance of the as-prepared biosensor, respectively. Amperometric measurements were carried out to optimize test conditions (pH and applied potential) of the biosensor. Under the optimal conditions, the biosensor showed a linear range from 10.0μM to 1.48mM (R2=0.9988) with a sensitivity of 22.1μAmM−1cm−2, and a detection limit of 2.769μM (S/N=3). The apparent Michaelis–Menten constant (KMa) was estimated to be 3.26mM. The interference from glycine (Gly), D-galactose (D-Gal), urea (Urea), L-phenylalanine (L-Phe), ascorbic acid (AA), and L-tyrosine (L-Tyr) was also investigated. The results indicated that the biosensor exhibit high sensitivity and superior selectivity, providing a hopeful candidate for glucose biosensing.

Abstract:We report the synthesis of a novel electrochemical biosensor comprising of cupric oxide (CuO) nanoparticles (NPs) mediated poly(hydroxybutyrate) (PHB) composite film with polyvinyl alcohol (PVA) as a binder/template support using the solution casting method for the detection of a biomolecule i.e., ascorbic acid (AA). The specimens were characterized for surface, chemical, mechanical, optical, and electrochemical attributes. The results expressed regular mediation of CuO NPs in the PHB/PVA matrix towards nanobiocomposite formation with enhanced crystallinity, inter-molecular interactions, mechanical, and electrochemical attributes, and decreased hydrophilicity and bandgap, thus being useful in potential optoelectronic devices. The synthesized biocomposite film exhibited a tensile strength of 86.24 ± 4.10 N which might be due to reinforcement/uniform dispersion of the CuO nanofiller in the PHB-based matrix. The PHB/CuO composite, then, deposited on a glassy carbon electrode surface exhibited good electrocatalytic activity towards the AA in the aqueous media even at low analyte concentrations. Such modified electrode surfaces with metal/biopolymer complex could find possible applications in the detection of other bioactive molecules.
A graphene oxide-gold nanostar hybrid based-paper biosensor for label-free SERS detection of serum bilirubin for diagnosis of jaundice

Abstract:We report a paper-based surface-enhanced Raman spectroscopy (SERS) biosensor integrating the enrichment capability, namely enPSERS biosensor, for the sensitive, label-free detection of free bilirubin in blood serum for the accurate diagnosis of jaundice and its related diseases. This biosensor comprises multifunctional graphene oxide-plasmonic gold nanostar (GO-GNS) hybrids decorated on the filter paper, which integrates the high sensitivity of SERS detection, enrichment for serum bilirubin and fluorescence superquenching capability of GO-GNS hybrids for sensitive detection of serum bilirubin. The study of adsorption kinetics reveals that both electrostatic and π-π interactions between the GO-GNS hybrids and targets are responsible for the enrichment of bilirubin, and the adsorption process follows the pseudo-second-order kinetic model. The results of SERS detection of bilirubin in blood serum show two differential linear response ranges from 5.0 to 150 μM and 150-500 μM with the detection limit as low as 0.436 μM. The comparison of the results obtained from our present enPSERS biosensor with the commercial diazo reaction method for determination of free bilirubin in blood serum reveals the clinical effectiveness and suitability of the developed paper-based SERS biosensor. We believe that this sensitive and label-free SERS biosensor holds considerable promise for clinical translation in accurate diagnosis of jaundice.

Abstract:The functionalized graphene nanosheets (PDDA-G) with poly(diallyldimethylammonium chloride) (PDDA) were synthesized and used to combine with room temperature ionic liquid (RTIL). The resulting RTIL/PDDA-G composite displayed an enhanced capability for the immobilization of hemoglobin to realize its direct electrochemistry. Moreover, the RTIL/PDDA-G based biosensor exhibited excellent electrocatalytic activity for the detection of nitrate with a wide linear range from 0.2 to 32.6μM and a low detection limit of 0.04μM at 3σ. This work opens a new way to functionalized graphene nanosheets with good biocompatibility and solubility in biosensors.
Multi-dimensional plasmonic coupling system for efficient enrichment and ultrasensitive label-free SERS detection of bilirubin based on graphene oxide-Au nanostars and Au@Ag nanoparticles

Abstract:Rapid and sensitive detection of free bilirubin (BR) is essential for early diagnosis of jaundice and other hepatobiliary diseases. Inspired by sandwich immunoassay strategy, a multi-dimensional plasmonic coupling SERS platform composed of graphene oxide-Au nanostars nanocomposites (GANS NCs) and nanoparticles (NPs) was designed for label-free detection of BR. Specifically, GANS NCs were first prepared, and their excellent SERS activity was ascribed to synergistic enhancement effect of electromagnetic enhancement and chemical enhancement. Furthermore, SERS spectroscopy was used to monitor the adsorption process of BR. Subsequently, secondary reinforcing NPs were directly added, ultimately resulting in a multi-dimensional plasmonic coupling effect. The SERS enhancing mechanism of coupled system was discussed through electromagnetic field simulations. Interestingly, the high-density hotspots generated by strong plasmonic coupling in substrate could lead to more extraordinary SERS enhancing behavior compared to GANS NCs. Sensing efficiency of the SERS platform was examined by BR with a detection limit down to 10 −11 M. Besides, NCs performed high uniformity and reproducibility. This work not only opens up a new avenue for construction of multi-dimensional plasmonic coupling system, but also offers a new biosensing technology for label-free diagnosis of BR-related diseases, thereby expecting to be applied in clinical diagnosis.

Abstract:The authors report on the synthesis of carboxy functionalized graphene oxide (fGO) decorated with magnetite (Fe3O4) nanoparticles. The resulting nanomaterial was used to prepare a composite with polyaniline (PANI) which was characterized by UV-vis, Fourier transform-infrared and Raman spectroscopies. Its surface morphologies were characterized by atomic force and scanning electron microscopies. A screen-printed carbon electrode was then modified with the nanocomposite to obtain an enzyme-free glucose sensor. The large surface of fGO and Fe3O4 along with the enhanced charge transfer capability of PANI warrant a pronounced electrochemical response (typically measured at 0.18 V versus Ag/AgCl) which is suppressed in the presence of glucose. This reduction of current by glucose was used to design a sensitive method for quantification of glucose. The response of the modified SPCE is linear in the 0.05 μM - 5 mM glucose concentration range, and the lower detection limit is 0.01 μM. Abstract:Polyaniline and its composites with nanoparticles have been widely used in electrochemical sensor and biosensors due to their attractive properties and the option of tuning them by proper choice of materials. The review (with 191 references) describes the progress made in the recent years in polyaniline-based biosensors and their applications in clinical sensing, food quality control, and environmental monitoring. A first section summarizes the features of using polyaniline in biosensing systems. A subsequent section covers sensors for clinical applications (with subsections on the detection of cancer cells and bacteria, and sensing of glucose, uric acid, and cholesterol). Further sections discuss sensors for use in the food industry (such as for sulfite, phenolic compounds, acrylamide), and in environmental monitoring (mainly pesticides and heavy metal ions). A concluding section summarizes the current state, highlights some of the challenges currently compromising performance in biosensors and nanobiosensors, and discusses potential future directions.Schematic presentation of electrochemical sensor and biosensors applications based on polyaniline/nanoparticles in various fields of human life including medicine, food industry, and environmental monitoring. The simultaneous use of suitable properties polyaniline and nanoparticles can provide the fabrication of sensing systems with high sensitivity, short response time, high signal/noise ratio, low detection limit, and wide linear range by improving conductivity and the large surface area for biomolecules immobilization.

Abstract:Biosensors with high sensitivity, selectivity and a low limit of detection, reaching nano/picomolar concentrations of biomolecules, are important to the medical sciences and healthcare industry for evaluating physiological and metabolic parameters. Over the last decade, different nanomaterials have been exploited to design highly efficient biosensors for the detection of analyte biomolecules. The discovery of graphene has spectacularly accelerated research on fabricating low-cost electrode materials because of its unique physical properties, including high specific surface area, high carrier mobility, high electrical conductivity, flexibility, and optical transparency. Graphene and its oxygenated derivatives, including graphene oxide (GO) and reduced graphene oxide (rGO), are becoming an important class of nanomaterials in the field of biosensors. The presence of oxygenated functional groups makes GO nanosheets strongly hydrophilic, facilitating chemical functionalization. Graphene, GO and rGO nanosheets can be easily combined with various types of inorganic nanoparticles, including metals, metal oxides, semiconducting nanoparticles, quantum dots, organic polymers and biomolecules, to create a diverse range of graphene-based nanocomposites with enhanced sensitivity for biosensor applications. This review summarizes the advances in two-dimensional (2D) and three-dimensional (3D) graphene-based nanocomposites as emerging electrochemical and fluorescent biosensing platforms for the detection of a wide range of biomolecules with enhanced sensitivity, selectivity and a low limit of detection. The biofunctionalization and nanocomposite formation processes of graphene-based materials and their unique properties, surface functionalization, enzyme immobilization strategies, covalent immobilization, physical adsorption, biointeractions and direct electron transfer (DET) processes are discussed in connection with the design and fabrication of biosensors. The enzymatic and nonenzymatic reactions on graphene-based nanocomposite surfaces for glucose- and cholesterol-related electrochemical biosensors are analyzed. This review covers a very broad range of graphene-based electrochemical and fluorescent biosensors for the detection of glucose, cholesterol, hydrogen peroxide (H2O2), nucleic acids (DNA/RNA), genes, enzymes, cofactors nicotinamide adenine dinucleotide (NADH) and adenosine triphosphate (ATP), dopamine (DA), ascorbic acid (AA), uric acid (UA), cancer biomarkers, pathogenic microorganisms, food toxins, toxic heavy metal ions, mycotoxins, and pesticides. The sensitivity and selectivity of graphene-based electrochemical and fluorescent biosensors are also examined with respect to interfering analytes present in biological systems. Finally, the future outlook for the development of graphene based biosensing technology is outlined.
Folic acid capped bimetallic nanoparticle based fluorescence sensor for the nanomolar determination of bilirubin

Abstract:Folic acid capped Silver-Copper bimetallic nanoparticle (FA-AgCu BNP) based turn-off fluorescence sensor has been developed for the rapid and facile determination of a prominent disease biomarker, Bilirubin. FA-AgCu BNP has been characterised via Fluorescence spectroscopy, EDX, TEM, DLS, Zeta potential, UV–visible spectroscopy, FTIR spectroscopy and XPS techniques. Fluorescence quenching of FA-AgCu BNP has been observed in the presence of bilirubin which is due to the combined effect of static quenching and Inner filter effect. Different analytical parameters were optimised and under these conditions, the sensor showed good selectivity towards bilirubin and attained a wide linear range from 2.00 × 10-5M to 1.00 × 10-7M with a limit of detection of 5.53 × 10-9 M. Practical utility of the proposed sensor has been illustrated in synthetic physiological fluids by spike recovery analysis and the obtained results were validated using classical spectrophotometric method.

Abstract:Lead halide perovskites are an advanced class of materials with high photoluminescence and optoelectronic properties. However their instability towards moisture and lead toxicity hinders their applications especially in biological sensing. Hence the need for novel lead free perovskite nanocrystals are of utmost important. Herein we synthesized a lead free, highly efficient, stable, cost effective Cs 2 ZnCl 4 perovskite nanocrystals (PNCs) and extends its application in biosensing. The PNCs exhibits bright blue fluorescence upon UV irradiation and was able to selectively detect bilirubin (BR) - a clinically important jaundice biomarker. The intense fluorescence emission of the sensor was completely quenched upon the addition of bilirubin. The sensor could detect bilirubin even in the presence of other interfering biomolecules with a good limit of detection of 26.10 nM. The sensor could successively detect BR in human serum samples. Furthermore, for the visual detection of bilirubin, a paper strip sensor was also designed making the sensor more economical and adaptable.

Abstract:Bilirubin (BR) is a potent biomarker for jaundice and liver malfunction. However, its quantitative determination remains a bottleneck due to the interference of numerous biomolecules present in the blood serum. To overcome this, a conjugated polyfluorene derivative, poly1,1′-((2,7-dimethyl-9H-fluorene-9,9-diyl)bis(hexane-6,1-diyl))bis(1H-benzo[d]imidazole) (PFBZ), was synthesized by incorporating it with a specific receptor, benzimidazole, for BR detection using a low-cost and straightforward oxidative coupling polymerization. The PFBZ polymer spontaneously forms nanoparticles in an aqueous medium and unveils excellent sensitivity toward BR in an aqueous PBS medium with a limit of detection of 6.9 pM, which is far less than the clinically relevant range. The sensing mechanism is based on probe–analyte interaction chemistry and Förster resonance energy transfer, which were confirmed from both experimental and theoretical studies. This platform offers excellent sensitivity and selectivity, which motivated us to successfully explore the quantitative determination of BR in real serum samples. Abstract:A method is described for the preparation of a nanocomposite material consisting of cuprous oxide/polyaniline/reduced graphene oxide (Cu2O/PANI/rGO). Aniline was employed as both the precursor for PANI and the reducing agent for Cu2+ and graphene oxide. A glassy carbon electrode was modified with the nanocomposite material. Chronoamperometric studies with the modified electrode showed it to enable an efficient electroreduction of hydrogen peroxide at −0.2 V vs. saturated calomel electrode. All measurements were performed in the absence of oxygen. Figures of merit include a wide linear response range (0.8 μM to 12.78 mM) and a low limit of detection of 0.5 μM (S/N = 3).Graphical abstractCuprous oxide/polyaniline/reduced graphene oxide nanocomposites were synthesized through one-step process for fabricating an nonenzymatic electrochemical sensor for hydrogen peroxide.

Abstract:Electrochemical voltammetric sensors are some of the most promising types of sensors for monitoring various physiological analytes due to their implementation as non-invasive and portable devices. Advantages in reduced analysis time, cost-effectiveness, selective sensing, and simple techniques with low-powered circuits distinguish voltammetric sensors from other methods. In this work, we developed a Cu2O-based non-enzymatic portable glucose sensor on a graphene paste printed on cellulose cloth. The electron transfer of Cu2O in a NaOH alkaline medium and sweat equivalent solution at very low potential (+0.35 V) enable its implementation as a low-powered portable glucose sensor. The redox mechanism of the electrodes with the analyte solution was confirmed through cyclic voltammetry, differential pulse voltammetry, and electrochemical impedance spectroscopy studies. The developed biocompatible, disposable, and reproducible sensors showed sensing performance in the range of 0.1 to 1 mM glucose, with a sensitivity of 1082.5 ± 4.7% μA mM−1 cm−2 on Cu2O coated glassy carbon electrode and 182.9 ± 8.83% μA mM−1 cm−2 on Cu2O coated graphene printed electrodes, making them a strong candidate for future portable, non-invasive glucose monitoring devices on biodegradable substrates. For portable applications we demonstrated the sensor on artificial sweat in 0.1 M NaOH solution, indicating the Cu2O nanocluster is selective to glucose from 0.0 to +0.6 V even in the presence of common interference such as urea and NaCl.

Abstract:Functionalized nanocomposites based on various type of graphene nanomaterials including graphene, graphene oxides (GOs), and doped graphene (oxides) are widely used as materials for various sensors that can display high sensitivity, selectivity and stability. This review with 347 references summarizes advances in the preparation and functionalization of graphene nanocomposites for the application of electrochemical sensors and biosensors. Following a general introduction into the field, the article is divided into subsections on (a) the synthesis and functionalization of nanocomposites (made from graphene, various kinds of GOs, heteroatom-doped GOs), (b) on methods for functionalization of composites (with other carbon nanomaterials, metal nanoparticles, metal oxide and metal sulfide nanoparticles), (c) on functionalization with inorganic materials including polyoxometalates, hexacyanoferrates, minerals), (d) on functionalization with organic materials such as amino acids, surfactants, organic dyes, ionic liquids, macrocycles (including cyclodextrins, crown ethers and calixarenes), and (e) on functionalization with organometallics and with various other organic compounds, (f) on functionalizations with polymers such as conventional polymers, polyelectrolytes, conducting polymers, molecularly imprinted polymers, (g) on functionalization with biomolecules including proteins and nucleic acids. Abstract:Biofunctionalized graphene-gold nanoparticle (AuNP) hybrids were prepared using a facile approach of in situ growth, with homogeneous distribution of AuNPs on the graphene nanosheets. Hemoglobin (Hb) was immobilized on the graphene-AuNP composites to fabricate biosensors for determination of nitrite (NO2(-)). A pair of well-defined redox peaks was observed for Hb immobilized on the graphene-AuNP hybrids with a formal potential (E(0')) of -0.314 V in 0.1 M phosphate buffered saline (0.15 M NaCl, pH 7.0). The novel biosensors exhibited many advantages, such as wide linear response range (from 0.05 to 1000 µM, R(2)=0.997), low detection limit (0.01 µM, a signal to noise ratio of 3), high sensitivity (0.15 μA μM(-1) cm(-2)), and excellent selectivity. These constructed biosensors were further used for determination of nitrite in pickled radish. The results obtained were in good agreement with those using spectrophotometry based on the National Food Safety Standard (GB 5009.33-2010), which indicates that these novel and sensitive biosensors have promising application for determination of nitrite in food.
Bimetallic MOF‐Based Fibrous Device for Noninvasive Bilirubin Sensing

Abstract:Bimetallic FeCo metal–organic framework films are prepared on the surface of carbonized electrospun nanofibers via a stepwise growth strategy involving an induction effect of oxide nanomembrane produced by atomic layer deposition. The synergistic interaction between the bimetallic active sites on the MOF film and the carbonized nanofibers enhances the electronic conduction network, significantly enhancing the catalytic activity of the flexible electrode for bilirubin. Noninvasive and real‐time detection of bilirubin concentration enables rapid assessment of liver health status and the diagnosis of jaundice‐related diseases. Herein, the study proposes a strategy to uniformly grow bimetallic FeCo metal–organic framework (MOF) films onto the surface of carbonized electrospun nanofibers via a stepwise growth strategy involving an induction effect of oxide nanomembrane prepared by atomic layer deposition, to realize high‐performance flexible nanofiber bilirubin sensors. Compared with monometallic MOF, the FeCo bimetallic MOF can attain a larger specific surface area, thus augmenting the exposed active sites. Furthermore, the synergistic interaction between the bimetallic active sites on the MOF film and the carbonized nanofibers enhances the electronic conduction network, thereby significantly enhancing the catalytic activity of the flexible electrode for bilirubin. The current nanofibers sensor exhibits excellent performance in bilirubin detection, in terms of high sensitivity, large linear detection range, and low detection limit. The synthesis of bimetallic MOF films on flexible fibers holds promise for the development of highly sensitive electrodes for wearable bilirubin detection devices.

Abstract:It is known that concentration of unbound (free) bilirubin (BR) instead of the total bilirubin is the critical determinant of cellular uptake and toxicity of bilirubin. Thus, developing fast, effective and sensitive technique for detecting BR, a unique and important biomarker of liver dysfunction, has a long-standing demand. In this study, attempts were made to explore if biocompatible graphene quantum dots (GQDs) could be employed to probe BR at an ultra-low level in aqueous environment through optical techniques. Thus, understanding the possible interactions involved between GQDs and BR is important and necessary to develop protocol for probing BR. The detailed spectral analysis shows that BR molecule quenches the fluorescence of GQDs which may be attributed to a possible fluorescence resonance energy transfer (FRET) involved between the excited state of GQDs and BR. The micro-calorimetric and circular dichroic (CD) measurements have provided binding and structural information in the process of interactions. It is established that with the help of synthesized GQDs, BR can be detected as low as 0.15 nM selectively and this appears to be the most sensitive amongst other available methods. Interestingly, it is observed that the addition of bovine serum albumin (BSA) facilitates the recovery of the BR-induced loss in fluorescence intensity of GQDs. The adopted strategy demonstrates that GQD may act as a sensitive fluorescent "turn off–on" probe for detection of BR. The proposed optical method based protocol validated with serum samples isolated from blood for rapid detection of BR at ultra-low level may help in developing a suitable sensing device in future.

Abstract:This review will unveil the development of a new generation of electrochemical sensors utilizing a transition-metal-oxide-based nanocomposite with varying morphology. There has been considerable discussion on the role of transition metal oxide-based nanocomposite, including iron, nickel, copper, cobalt, zinc, platinum, manganese, conducting polymers, and their composites, in electrochemical and biosensing applications. Utilizing these materials to detect glucose and hydrogen peroxide selectively and sensitively with the correct chemical functionalization is possible. These transition metals and their oxide nanoparticles offer a potential method for electrode modification in sensors. Nanotechnology has made it feasible to develop nanostructured materials for glucose and H2O2 biosensor applications. Highly sensitive and selective biosensors with a low detection limit can detect biomolecules at nanomolar to picomolar (10-9 to 10-12 molar) concentrations to assess physiological and metabolic parameters. By mixing carbon-based materials (graphene oxide) with inorganic nanoparticles, nanocomposite biosensor devices with increased sensitivity can be made using semiconducting nanoparticles, quantum dots, organic polymers, and biomolecules.

Abstract:This study presents the development of a novel non-enzymatic electrochemical biosensor for the real-time detection of hydrogen peroxide (H2O2) based on a composite of cerium metal-organic frameworks (Ce-MOFs), hemin, and graphene oxide (GO). The Ce-MOFs served as an efficient matrix for hemin encapsulation, while GO enhanced the conductivity of the composite. Characterization techniques including scanning electron microscopy (SEM), X-ray diffraction (XRD), Fourier-transform infrared (FTIR) spectroscopy, UV-vis spectroscopy, and thermogravimetric analysis (TGA) confirmed the successful integration of hemin into the Ce-MOFs. The Ce-MOFs@hemin/GO-modified sensor demonstrated sensitive H2O2 detection due to the exceptional electrocatalytic activity of Ce-MOFs@hemin and the high conductivity of GO. This biosensor exhibited a linear response to H2O2 concentrations from 0.05 to 10 mM with a limit of detection (LOD) of 9.3 μM. The capability of the biosensor to detect H2O2 released from human prostate carcinoma cells was demonstrated, highlighting its potential for real-time monitoring of cellular oxidative stress in complex biological environments. To further assess its practical applicability, the sensor was tested in human serum samples, yielding promising results with recovery values ranging from 94.50 % to 103.29 %. In addition, the sensor showed excellent selectivity against common interfering compounds due to the outstanding peroxidase-like activity of the composite.
Surface-Enhanced Raman Scattering-Active Gold-Decorated Silicon Nanowire Substrates for Label-Free Detection of Bilirubin

Abstract:Bilirubin (BR) is a product of hemoglobin breakdown, and its increasing levels in the blood may indicate liver disorders and lead to jaundice. Kernicterus is most dangerous in newborns when the unconjugated BR concentration can quickly rise to toxic levels, causing neurological damage and even death. The development of an accurate, fast, and sensitive sensor for BR detection will help reduce diagnostic time and ensure successful treatment. In this study, we propose a new method for creating a surface-enhanced Raman scattering (SERS)-active substrate based on gold-decorated silicon nanowires (Au@SiNWs) for sensitive label-free BR detection. Gold-assisted chemical etching of crystalline silicon wafers was used to synthesize SiNWs, the tops of which were then additionally decorated with gold nanoparticles. The low detection limit of model analyte 4-mercaptopyridine down to the concentration of 10-8 M demonstrated the excellent sensitivity of the obtained substrates for SERS application. The theoretical full-wave electromagnetic simulations of Raman scattering in the Au@SiNW substrates showed that the major contribution to the total SERS signal comes from the analyte molecules located on the SiNW surface near the gold nanoparticles. Therefore, for efficient BR adsorption and SERS detection, the surface of the SiNWs was modified with amino groups. Label-free detection of BR using amino modified Au@SiNWs with high point-to-point, scan-to-scan, and batch-to-batch reproducibility with a detection limit of 10-6 M has been demonstrated. Artificial urine, mimicking human urine samples, was used as the matrix to get insights into the influence of different parameters such as matrix complexity on the overall BR SERS signal. The signal stability was demonstrated for 7 days after adsorption of BR with a concentration of 5 × 10-5 M, which is the required sensitivity for clinical applications.
